      • 黃柏에서 抽出한 多糖類의 抗癌作用에 대한 硏究

        In order to investigate antitumor activities of the extracted polysaccharides from Phellodendron chinense SCHNEID, the following studies were carried out. Phellodendron chinense SCHNEID was extracted with hot water and alkali, and the extracted polysaccharides showed antitumor activities against mice Sarcoma-180 solid tumors in parameters of tumor weight, inhibition ratio, thymidilate synthetase(TS) and thymidine kinase(TK) activities. The results were as follows : 1. Delipidated extracts from Phellodendron chinense SCHNEID, were systematically fractionated, the heteropolysaccharide contains D-N-acetylglucosamine, D-galactose, D-mannose, D-glucose. The molecular weight distribution of the sample was found to be nearly homogeneous (about from 200 to 1000 kDa) in gel permeation analysis. 2. Among the polysaccharide fractions, the water extracted fractions, particularly Fr.-2, showed a potent growth-inhibitory activity against implanted in mice Sarcoma 180 solid tumor (inhibition, 95.3%, at 2 ㎎/100g), but the cold alkali extract (Fr.-4) showed a very low activity (inhibition; 4.7%). Fr.-7 and Fr.-8 showed the highest tumor inhibitory activity (inhibition; 96.4 and 98.2%, respectively). 3. TS activity in the Fr.-2 given oral administration of propolis was markedly suppresed to 73.7% of that in the control (p<0.01 by Student's t-test). However, there was little difference in TK activity with a slight decrease in Fr.-2 only.

        마른명태 식해의 향기성분과 기능성

        The volatile compounds of Myungtae (Alaska pollack) sikhae obtained by simultaneous steam distillation and extraction(SDE) apparatus were separated by gas chromatography(GC) and gas chromatography mass spectrometry(GC/MS). The totals of 155 volatile flavor components was identified in traditional Kyungsangdo Myungtae (Alaska pollack) sikhae, respectively. α-Zingihirene(11.03%) (E)-di-2-propenyl disulfide(7.95%) β-cironellol(6.02%), methyl allyl disulfide(3.58%), cryptone(3.39%), camphene(3.23%), pentano(3.21%), penadecanal(2.66%) and β-phellandrene(2.06%) were contained as the main compounds of Myungatae shikae. The fraction obtained from sikhae were tested for electron donating ability, angiotensin converting enzyme and xanthine oxidase inhibitory activity. There was no electron donation abilities(SC_50) of hexane and water fraction. On the other hand, the abilities of ethylacetate fraction and butanol fraction showed 310.64 ㎍.mL, 1096.49 ㎍/mL, respectively. Angiotensin converting enzyme inhibitory activities(IC_50) of ethylacetate fraction and butanol fraction were 1.623 mg/mL, 1.303 mg/mL, respectively. Xanthine oxidase inhibitory activities(IC_50) of ethylacetate fraction and butanol fraction were 3.591 mg/mL, 2.803 mg/mL, respectively.

        This study was to understand the neuromuscular coordination, analyzing the relationship among 25m dash, simple reaction time (SRT) and choice reaction time (CRT) of whole body and finger, and movement time (MT) of middle and high school sprinters (N=24). There were significant relationships among simple reaction time (SRT), choice reaction time (CRT), movement time (MT), and interval(Int) lap times of middle and high school sprinters: the faster SRT, the faster MT, Int 1,2,3,4,5, and 25m running time for all sprinters; The faster CRT, the faster Int 1,2,3,4,5, and 25m running time of all sprinters. Also, same to SRT and CRT by hand(s). While sprinters' MT in high school was significantly related to Int 1,2,3,4,5, and 25m running time, sprinters' MT in middle school was not related to any Int 1,2,3,4,5, and 25m running time. In addition, while 25m running time of high school sprinters was predicted by Int 1(RT+MT), in turn, Int 3,2, gender, SRT and CRT, 25m running time of middle school sprinters was predicted by Int 3, in turn, Int 1(RT+MT), and 2. Even though male sprinters in high school showed faster SRT and CRT female sprinters in lab, male sprinters were not significantly better than female sprinters on reaction time (RT) during starting phase. There was gender difference on each running time of Int 2,3, and 4 (5-15m), but not on RTs

        도시공공시설의 지역간 입지불균형에 따른 공간 분석 : 서울시 초등학교와 어린이공원을 중심으로 A Case Study of Elementary School and Children's Playground in Seoul

        Elementary schools are one of the important public facilities as the basis of student out-door activities as well as residential community activities in a region. Thus, the location of elementary schools must be closely related to student out-door facilities: playgrounds within schools, playgrounds in apartment complexes near schools, and community playgrounds in the region. In view of urban planning, the student out-door facilities must be planned and built as a key residential component by regulations of community site planning. However, the location of elementary schools is not associated with that of student out-door facilities in many cities because elementary schools and student out-door facilities are not planned by same institutions with same target years and objectives. Seoul, the capital of Korea, is a very dynamic city in terms of population, educational facilities, and public facilities. The population of Seoul was at the highest point of 10,969,862 in year 1992, and has slowly decreased to 10,280,523 in year 2002. The number of elementary schools in Seoul has increased from 481 to 542 during the same period. Seoul is a very dense city with the area of 605.52㎢ and the population density of 16,978 person to a square kilometer. Thus, the playgrounds for elementary school students are always lack in terms of number and size, though the number of elementary schools is increasing in Seoul. The objectives of this research are (1) to investigate elementary school students in order to evaluate a change of educational environment, (2) to identify the association between the location of elementary schools and playgrounds for out-door activities of elementary school students in Seoul, and (3) to evaluate the impacts of three types of outdoor facilities elementary school playgrounds, playgrounds , and community playgrounds. To do this, this research uses various statistical analysis methods. The elementary school data sets are obtained from Seoul statistical books and educational public service books, GIS.

      • Identification of Redox Sensitive Proteins in Prdx II Deficient Red Blood Cells

        Red blood cells (RBCs) have been studied as models for infectious diseases, various symptoms of anemia, hemolysis, and erythrocyte aging. Although do not directly affect RBCs, other diseases may cause RBC physiological alterations that could be advanced for diagnostic aim or to convince better understanding of a certain pathological pattern. In this study, comparative RBC proteomics between healthy and abnormal conditions involve to promote aging related‐biomarker discovery. Peroxiredoxin II (Prdx II; a typical 2‐Cys Prdx) has been originally isolated from erythrocytes, and its structure and peroxidase activity have been adequately studied. Mice absent to Prdx II proteins had heinz bodies in their peripheral blood, and morphologically aged cells were detected in the dense RBC fractions, which contained markedly higher levels of reactive oxygen species (ROS). In addition, a labeling experiment with the thiol‐modifying reagent biotinylated iodoacetamide (BIAM) in Prdx II‒/‒ mice revealed that a variety of RBC proteins were highly oxidized. To identify oxidation‐sensitive proteins in Prdx II‒/‒ mice, we performed RBC comparative proteome analysis by nano‐UPLC‐MSE shotgun proteomics with relative protein quantitative analysis. We found oxidation‐sensitive 18 membrane and 41 cytosol proteins from 32 and 85 peptides containing cysteine oxidation, and analyzed comparative expression pattern in healthy RBCs of Prdx II+/+ mice (W1), healthy RBCs of Prdx II‒/‒ mice (K1), and abnormal RBCs of Prdx II‒/‒ mice (K2). These proteins belonged to cellular functions related with RBC lifespan maintain, such as cytoskeleton, stress‐induced proteins, amino acid/nucleic acid metabolic enzymes, signal transduction, and molecular transporters. Furthermore, protein networks among identified oxidation sensitive proteins were analyzed to associate with aging consequence. Consequently, we expected that RBC proteome may provide clues to understand redox‐imbalanced diseases.

        Histiocytic medullary reticulosis (HMR) is a malignant noeplastic process, with tumor cells belonging to the histiomonocytic line, and originally described by Scott and Robb-Smith in 1939 characterized by its acute onset, progression to death within a few months and its clinical features are fever, lymphadenopathy, hepatosplenomegaly, jaundice, and purpura common laboratory findings are characterized by severe anemia, leukopenia, thrombocytopenia and histologically by systemic progressive invasion of morphologically atypical histiocytes or of their precursor. 61 and 37-year-old males were admitted with camplaints of high fever and chilling sensation and were treated by cyclophosphamide, vincristine, prednison and antiobiotics, but patients all died 17 and 14 days after admission due to probably sepsis and respiratory failure.

        경상도 전통 마른 명태 식해의 품질특성

        The quality characteristics of traditional Kyungsangdo Myungtae (Alaska pollack) sikhae including optimal composition of the product were investigated. In the sensory evaluation, the Myungtae sikhae made from Gampo accepted to be best for traditional Kyungsangdo Myungtae sikhae and it is called standard sikhae. The standard sikhae was prepared as follows: dried-Myungtae 200g, cooked rice 230g, red pepper 92g, garlic 112g, ginger 100g, radish 1.6kg, salt 20g, and malt meal 65g. The Myungtae sikhae made from Gampo showed higher values in total sugar, free reducing sugar, and nitrogen compounds than the Myungtae sikhae made from another locations. The sugar of standard sikhae consisted of four kinds including glucose. And, the contents of free amino acid was increased in the order of glutamic acid, alanine, and methionine. The composition of free amino acid in Myungatae sikhae contained 17 kinds, and the contents was increased in order of glutamic acid, aspartic acid, and proline. The organic acid of standard Myungtae sikhae consisted of 7 kinds including lactic during fermentation. The contents of citric acid, succinic acid, and lactic acid gradually increased during fermentation, and especially, increasing of lactic acid remarkably increased than the different organic acid.

        인천 인현동 호프집 화재 피해자 분석

        Purpose : This study was conducted to develop field triage, transportation, distribution, and prehospital care at a fire disaster by analyzing the victims of the fore that broke out at a bar in Incheon. Method : We analyzed the cases of the victims of a fire in Incheon in Oct. 1999. We determined the primary care hospital, the arrival time, the burn size, the outcome, and the injury type from the medical records, the concerned organ records, and interviews with concerned persons. Result : The total number of victims was 137: 56 prehospital deaths, 1 hospital death, and 80 survivals. The Pearson correlation coefficient between the burn size and the severity was -0.175. There were 121(89.6%) cases of inhalation injury, 59 (43.7%) cases of flame burns, 66 (48.9%) cases of hypoxic brain damage, and 16 (11.9%) cases involving other types of injury. Conclusion : The causes of death of the fire victims were inhalation injury and hypoxic brain damage due to CO poisoning and other toxic inhalants. We propose the use of a simple triage and rapid treatment(START) system and a reassessment the delayed category in fire disasters.

      • CRP* 의존성 maltose 대사 촉진 유전자 sfs4의 클로닝 및 염기배열 결정

        CRP (cAMP receptor protein)은 cAMP와 결합하여 cAMP-CRP 복합체를 형성하여 전사조절의 조절인자로서 작용한다. crp 유전자에 변이를 도입하여 cAMP의 비존재 상태에서 cAMP-CRP와 비슷한 기능을 가진 crp* 유전자가 도입된 대장균 MK2001 (crp*¹, cya::km)을 숙주로 사용하여 cAMP 혹은 cGMP의 비존재하에서도 mal 유전자의 발현을 촉진시키는 유전자 sfs (sugar fermentation stimulation) 수 종을 클로닝하였다. 본 실험에서는 이미 밝혀진 nlp (Ner like protein) 유전자와 같이, sfs의 새로운 유전자를 탐색하여, 그 중 sfs4의 2126bp 전 염기배열을 결정하고, 잠정적인 sfs4의 promoter 영역에는 CRP 단백질과의 결합 DNA 공통 염기배열(5' AAT TGTGA ACACCA TCACC CGT 3')이 존재함을 확인했다. lacZ 융합 유전자를 작성하여 TP2010R1과 MK2001의 균주에서 cAMP를 첨가할 경우 각각 2.3배, 1.8배의 β-galactosidase 활성이 증가 하는 것으로 보아 sfs4는 cAMP-CRP에 의해 발현 조절을 받는 것으로 나타났다 In Escherichia coli, CRP forms a complex with cAMP and acts as a transcriptional regulator of many genes, including sugar metabolism operons. The E. coli MK2001, which is introduced the altered crp*¹, is functional in the expression of lac, ara and man, in the absence of cAMP. However, the expression of mal gene is fully activated by the addition of cAMP or cGMP. The object of the study is cloning of the sfs (sugar fermentation stimulation) genes, which was involved in regulation of mal gene expression with the altered crp*¹gene, and structural analysis and characterization of the genes at the molecular level. We have cloned 5 different E. coli genes which stimulate the maltose metabolism in a crp*¹, cya::km (MK2001) background. Newly identified genes were designated as sfs. One of the sfs genes (pPC1), located at the 53.2 min map position in the E. coli chromosome, was further analyzed. Expression of the genes, which is involved in maltose metabolism, malQ (amylomaltase), was increased to 5.8-fold in the presence of a plasmid, pAP5, containing the subcloned sfs4 gene. The nucleotide sequence of a common 2,126bp segment of the pPCM1 was determined and two open reading frames (ORF1 and ORF2) were detected. The ORF1 encodes the sfs4 gene and ORF2 encodes a truncated protein. Potential CRP binding site is located in the upstream of the putative promoter in the regulatory region. Expression of the cloned sfs4 gene was positively regulated by the cAMP-CRP complex.
