Backgrounds Arctigenin derived from the seeds of Arctium lappa Linnaeus is known as an anticancer drug candidate by targeting various pathways involved in anticancer therapy. Methods Using 2D monolayer and 3D spheroid culture systems in nasal septum carcinoma RPMI-2650 cells, the effects of arctigenin and dexamethasone on cell viability, ROS levels, ATP level, mitochondrial function, apoptosis and necroptosis were examined. Results The combination treatment of both compounds induced strong cytotoxicity, accompanied by increases of sub-G0/ G1 peak, annexin V-PE-positive cells, and ROS levels, loss of mitochondrial membrane potential, and decrease of cellular ATP content. These changes were observed as simultaneous induction of DNA damage, apoptosis, and necroptosis. A series of changes by arctigenin and dexamethasone were efficiently restored by decreasing ROS levels or supplementing ATP. Treatment of 3D spheroids with arctigenin and dexamethasone decreased cell viability in the spheroids, but it was slightly resistant than cells under 2D conditions. In addition, this phenomenon was accompanied by an increase in mediators for both apoptosis and necroptosis. Conclusion Results of this study suggest that the apoptosis and necroptosis-inducing effects of arctigenin are associated with ATP depletion due to oxidative mitochondrial dysfunction.

<P>An all‐solution‐processed flexible piezoelectric nanogenerator, composed of polycrystalline ZnO thin film and functional polymer layers such as P3HT/PCBM and PEDOT:PSS, generates energy through a mechanical rolling and muscle stretching system. On page 6022, Youn Sang Kim, Sang‐Woo Kim, and co‐workers show that this all‐solution‐processed nanogenerator is feasible as a piezoelectric patchable device and is promising for use in future energy harvesters such as wearable human patches and mobile electronics. </P>

<B>Graphic Abstract</B> <P>The cover shows an image of fully rollable transparent nanogenerators synthesized using chemical vapor deposition grown large-scale graphene sheets as transparent electrodes and piezoelectric ZnO nanorod arrays. Sang-Woo Kim, Jae-Young Choi, and co-workers report on p. 2187 the electrical and structural stability of the nanogenerators, with excellent charge scavenging performance under external mechanical loads such as bending and rolling. It is well-known that Titanium (Ti) and Ti alloys demonstrate the most biocompatibility. The current research examined the initial ossification and formations of bone contacts upon implanting 4 types of implants with different surface treatments. Total of 20 samples were used during the experiment and categorized into sand-blasted large grit acidetched (SLA) surface treatment group, SLA+ surface treatment group, Active surface treatment group and Zn-Anodizing 340 V surface treatment group. Two implants were placed on the left side and right side each of tibia and sacrificed after 4 weeks period. Tibias of rabbits were produced into specimen slides with a thickness of 15 µm instead of traditional thickness of over 50 µm and we could get the good resolution. In histomorphometrically measured bone implant contact rates, significant differences of SLA+ and Zn-Ano are recognized, and the Zn-Anodizing surface treatment group displayed higher results than all other groups. In the results of histomorphometrically measuring and comparing the surface area rates, differences between of SLA and SLA+ are significantly recognized than comparisons of other groups, and the SLA surface treatment group scored the highest. It may be concluded that the period for implant masticatory functions may be shortened when using both of Zn-Anodizing and SLA implants and suggested that the semi-thin section produced the good result.

<P>Cigarette smoking is known to aggravate Graves' orbitopathy (GO) severity by enhancing adipogenesis. We investigated the effect of quercetin, an antioxidant, on adipocyte differentiation induced by cigarette smoke extract (CSE) in primary cultured orbital fibroblasts (OFs) from GO patients. Freshly prepared CSE was added to the cells and H(2)O(2) was used as a positive control. Intracellular reactive oxygen species (ROS) generation and adipogenesis were measured. The expressions of proteins peroxisome proliferator-activated receptor (PPAR) γ, CCAAT-enhancer-binding proteins (C/EBP) α and β, and heme oxygenase-1 (HO-1), an antioxidant enzyme, were examined during adipogenic differentiation. In result, CSE and H(2)O(2) dose-dependently stimulated intracellular ROS production in normal and Graves' OFs. The effect of 2% CSE was similar to that of 10?μM H(2)O(2); both concentrations were noncytotoxic and were used throughout the experiment. Quercetin pretreatment reduced the ROS generation stimulated by either CSE or H(2)O(2) in preadipocyte OFs. CSE and H(2)O(2) stimulated adipocyte differentiation in cultured OFs. The addition of quercetin (50 or 100?μM) suppressed adipogenesis. Quercetin also suppressed ROS generation in differentiating OFs during adipogenesis stimulated by CSE and H(2)O(2). Additionally, the expressions of PPARγ, C/EBPα, and C/EBPβ proteins were reduced in the quercetin-treated OFs. Quercetin also reduced the CSE- and H(2)O(2)-induced upregulation of ROS and HO-1 protein in differentiated OFs and preadipocyte OFs. As shown in this study, quercetin inhibited adipogenesis by reducing ROS in vitro, supporting the use of quercetin in the treatment of GO.</P>
