      • 네오마이신 인산기전위효소 유전자의 F-9 세포 內 형질전환

        쥐 배종양세포, F-9, 22에 aminoglycoside 계통의 항생제 G-418 저항성을 갖도록 형질전환 시키었다. 즉, aminoglyucoside phosphotransferase 유전자를 갖고 있는 포유동물세포 발현 백터 pHβ APr-1-neo를 F-9, 22 세포에 electrophoration 법을 이용하여 transfection시키었다. 이 형질전환된 세포를 여러차례 subcultrue한 다음 neo 유전자 저항 능력이 소멸되지 않고 아주 안정하게 유지됨을 보았다. neo유전자로 형질전환된 클론은 RNAse protection assay로 관찰하였으며, 이를 위하여 BSpMC1rp를 합성하였다. A Plasmid PH βAPr-1-neo containing phosphotransferase gene (neo) was introduced into murine embryonal carcinoma (EC) cells (F-9, 22) by electroporation (250 volts/cm, 10 millisecond).. Colonies resistant to the drug G-418 were selected as plausible clones transfected by the neo. Elevated levels of the neo in the putative clones were confirmed by RNAse protection experiments, one of which named "F-9/neo". A 241 bp fragment of neo cDNA (EcoRl-Narl segment of the plasmid pMClneoA) was ligated to generate BSpMC1rp for the neo riboprobe synthesis. Transcription of EcoRl cut BSpMC1rp in the presence of 32pGTP yielded a 384 by probe, of which 284 by were protected by endogenous neo mRNA in the F-9 /neo clone.

      • JunB cDNA의 쥐 배양세포에서의 발현

        JunB cDNA was inserted into a beta-actin promoter expression vector naming LK4JunB. The LK4JunB was introduced into F9 cells by electroporation. Some 5,000 clones resistant to G418 were identified. Three of 12 G418 resistant clones expressed elevated levels of JunB RNA. The two highest expressing isolates were subcloned. All of eight subclones of the first isolate were found to express elevated levels of JunB RNA ensuring the homogeniety in expression of the JunB RNA among subclones. The overexpression of JunB on the retinoic acid induced differentiation of F9 cells was assessed by RNA analysis of cells treated with the inducer for various lengths of time. Elevated JunB expression was evident in both experimental clones regardless of the time of treatment with retinoic acid. In the parental F9 cells, elevated JunB RNA was detected after three days of treatment but not before.

        This is a clinical study on 113 patients, who were treated for parotid tumors at Chonnam National University Hospital over a period of 5 years from January 1996 to December 2000. The results were as follow, 1. The mean ages were 42.6 years old (12~73 years) for benign parotid tumor patients and 43.9 years old (2~74 years) for malignant patients. 2. The ratio of male to female was 1:2 in malignant parotid tumors, but there was no difference in sex distribution of benign tumor. 3. There was no difference in mean mass size and duration of illness between benign and nalignant tumor patients. 4. The combined use of fine needle aspiration biopsy and computed tomograph was common as the diagnostic method of parotid tumor 5. The parotid tumors were commonly located in the superficial lobe(69%). 6. The ratio of benign and malignant tumors was 4:1. The most common benign tumor was pleomorphic adenoma (72% of benign tumor), followed by Warthin's tumor and monomorphic adenoma. The most common malignant tumor was rnucoeplderrnoid carcinoma(45% of malignant tumor). followed by adenoid cystic carnoma. carcinoma ex pleomorphic adenoma, and adenocarcinoma. 7. The most common operations were superficial parotidectomy (72%) for benign parotid tumor and total parotidectomy (45%) for malignant tumor 8. The most common postoperative complication was temporary facial nerve weakness (82%), followed by Frey's syndrome (14%) and permanent facial palsy (4%). 9. Postoperative radiation therapy was done in 12 patients and the mean radiation dose was 5550±620cGy. 10. Of 113 cases. tumor recurrence developed in 12 cases (10%). These results suggest that the successful management of parotid gland tumors needs adequate surgical treatment on the basis of knowledge of parotid gland anatomy and preoperative diagnosis.

        Hot Wall Epitaxy 법에 의한 CdIn_2S_4 단결정 박막의 성장과 광전류 특성

        수평 전기로에서 Cdln_2S_4 다결정을 합성하여 HWE(Hot Wall Epitaxy)방법으로 Cdln_2S_4 단결정 박막을 반절연성 GaAs (100)기판에 성장시켰다. Cdln_2S_4 단결정 박막의 성장 조건은 증발원의 온도 630 ℃, 기판의 온도 420 ℃ 였고 성장 속도는 0.5 ㎛/hr였다. Cdln_2S_4 단결정 박막의 결정성의 조사에서 10 K에서 광발광(photoluminescence) 스펙트럼이 463.9 nm (2.6726eV)에서 exciton emission 스펙트럼이 가장 강하게 나타났으며, 또한 이중 결정 X-선 요동 곡선(DCRC)의 반폭치(FWHM)도 127 arcsec로 가장 작아 최적 성장 조건임을 알 수 있었다. Hall효과는 van der Pauw 방법에 의해 측정되었으며, 온도에 의존하는 운반자 농도와 이동도는 293K에서 각각 9.01 × 10 exp (16) /cm^3, 219 cm^2/V·s였다. Cdln_2S_4 /Sl(Semi-Insulated) GaAs(100) 단결정 박막의 광흡수와 광전류 spectra를 293K에서 10K까지 측정하였다. 광흡수 스펙트럼으로부터 band gap E_g(T)는 Varshni 공식에 따라 계산한 결과 2.7116eV - (7.74x10 exp (-4)eV/K)T^2/(T+434K)이었으며 광전류 스펙트럼으로부터 Hamilton matrix(Hopfield quasicubic mode)법으로 계산한 결과 crystal field splitting Δcr값이 0.1291 eV이며 spin-orbit Δso값은 0.0248 eV임을 확인하였다. 10K일 때 광전류 봉우리들은 n = 1일때 A_1-B_1-와 C_l-exciton 봉우리임을 알았다. A stoichiometric mixture of evaporating materials for Cdln_2S_4 single crystal thin films was prepared from horizontal electric furnace. To obtain the single crystal thin films, Cdln_2S_4 mixed crystal was deposited on thoroughly etched semi-insulating GaAs(100) substrate by the Hot Wall Epitaxy (HWE)system. The source and substrate temperatures were 630℃ and 420℃, respectively. The crystalline structure of the single crystal thin films was investigated by the photoluminescence and double crystal X-ray diffraction (DCXD). The carrier density and mobility of Cdln_2S_4 single crystal thin films measured with Hall effect by van der Pauw method are 9.01×10 exp (l6) cm^-3 and 219 cm^2/V·s at 293 K, respectively. The temperature dependence of the energy band gap of the Cdln_2S_4 obtained from the absorption spectra was well described by the Varshni's relation, E_g(T) = 2.7116 eV - (7.74× 10 exp (-4)eV)T^2/(T+ 434). The crystal field and the spin-orbit splitting energies for the valence band of the Cdln_2S_4 have been estimated to be 0.1291 eV and 0.0248 eV, respectively, by means of the photocurrent spectra and the Hopfield quasicubic model. These results indicate that the splitting of the Δso definitely exists in the Γ_5 states of the valence band of the AgInS_2/GaAs epilayer. The three photocurrent peaks observed at 10K areascribed to the A_l-, B_l-, and Cl-exciton peaks for n = 1.

      • 유전체 공진기를 이용한 개인휴대 통신용 대역통과 필터의 설계 및 구현

        In this paper, the band pass filter for personal communication services was constructed and designed using λ/4 TEM mode dielectric resonators with a (Zr0.65,Sn0.35)ATi1.04O4.04 ceramic of dielectric constant εr=37.8, Qxfo=48.600 temperature coefficient of resonant frequency rf=7ppm/˚C, and Unloaded-Q (Qo) of λ/4 TEM mode dielectric resonator with inner hole size 0.9㎜ and external size 3㎜ using silver electrode is 354.5 at 1950MHz. For the band pass filter design and construction, the design theory and simulation results of band pass filter using J-inverter theory have been studied. The parameters which are evaluated by design theory are practically applied to the filter construction and the simulation results are in agreement with the measured results after fine tunnings.

      • n-CdS_(0.46)Se_(0.54)/p-Cu_92-x)S_(0.46)Se_(0.54) 이종접합 태양전지의 제작과 그 특성에 관한 연구

        승화방법에 의해 CdS_0.46Se_0.54 단결정을 성장하여 결정구조를 조사하고, Van der Pauw 방법으로 Hall effect를 측정하여 carrier density의 온도 의존성과 mobility의 온도 의존성을 조사하였다. 성장된 CdS_0.46Se_0.54 단결정을 치환반응하여 n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 이종접합 태양전지를 제작하였다. Spectral response, 전류-전압특성 및 전력변환 효율을 조사하여 그 결과로부터 개방전압은 0.48V, 단락 전류 밀도는 21mA/㎠, fill factor와 전력변환효율은 각각 0.75와 9.5%를 얻었다. CdS_0.46Se_0.54 single crystal was grown by a sublimation method. The crystal structure and the temperature dependence of carrier density and mobility of CdS_0.46Se_0.54 single crystal were studied. Heterojunction solar cells on n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 were fabricated by the substitution reaction. The spectral response, the J-U characteristics and the conversion efficiency of the n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 heterojunction solar cells were studied. The open-cricuit voltage, short-circuit density, fill factor and conversion efficiency of n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 heterojunction solar cells under 80mW/㎠ illumination were found to be 0.48V, 21mA/㎠, 0.75 and 9.5%, respectively.

      • 실험쥐 모델에서 림프구 주입을 이용한 동종 장기이식에 대한 관용 유도

        연구배경: Mixed chimerism 상태에서는 공여 장기에 대해 특이적인 관용효과가 유도되어 면역억제제를 투여하지 않아도 거부반응이 나타나지 않는다. 본 연구에서는 조직적합항원(major histocompatibility complex)이 일치하지 않는 실험쥐를 대상으로 전신방사선조사나 이식 전 동종림프구를 주입하는 방법으로 donor chimerism 상태를 만들어 이식장기에 대한 관용효과를 유도하고자 하였다. 방법: C57BL/6 (H-2^(b))를 전처치 방법에 따라 7 Gy의 전신방사선조사를 시행한 군(1군)과 BALB/c (H-2^(d)) 비장 세포를 간문맥 내로 주입하여 이식세포에 대한 관용을 유도한 군(2군)으로 나눈 뒤 BALB/c 골수세포를 주사하여 유도된 chimerism을 분석하고 MLR을 시행하였으며, BALB/c 피부를 이식하여 생착 여부를 확인하였다. 결과: 1군에서는 말초혈액 내에서 donor chimerism이 확인되었고 2군에서는 PCR 방법으로 말초혈액뿐만 아니라 각 장기에서 microchimerism이 확인되었다 MLR 방법으로 1, 2군 모두에서 BALB/c 세포에 대해 동종림프구의 증식반응이 저하되었으며, 동종피부이식편이 평균 생착기간도 대조군의 6.6±0.8일에 비해 1군에서는 평균 90일 이상, 2군에서는 16.3±1.2일로 현저하게 증가된 것을 확인할 수 있었다. 결론: 방사선조사나 공여림프구 투여로 동종세포에 대한 면역학적 관용효과를 유도할 수 있었으며, 동종피부이식편의 생착을 증가시킬 수 있었다. Background: Because mixed allogeneic chimerism results in donor-specific tolerance, rejection doesn't occur without immunosupression. In this study, we investigate the induction of immune tolerance on mouse skin allograft in MHC-mismatched hosts that developed donor chimerism after conditioning with total body irradiation or a portal vein injection of donor splenocytes (SPLc), followed by an infusion of donor bone marrow cells (BMCs). Methods: In BALB/c (H-2^(d)) to C57BL/6 (H-2^(b)) combination, C57BL/6 recipients were irradiated at 7 Gy total body irradiation (TBI) prior to BMC infusion (Group 1) and given a portal vein injection with SPLc of BALB/c prior to BMCs infusion (Group 2). BMCs were infused from BALB/c donors to C57BL/6 recipients. Flow cytometry and PCR were used to assess donor-chimerism and standard skin grafting and mixed lymphocyte reaction (MLR) were used to assess donor-specific tolerance. Results: Under this protocol, it was confirmed that mixed chimerism was induced in group 1 and microchimerism was induced in group 2. The proliferation of lymphocyte was reduced in both group 1 and group 2 using MLR analysis. The graft survivals in group 1 (>90 days) and group 2 (16.33±1.2 days) were longer than the control (6.6±0.8 days), which suggested that tolerance was induced across the MHC barrier without further need for immunosuppression. Conclusion: In this donor chimerism model, functional tolerance was induced across the MHC barrier with combined protocol of total body irradiation/ donor BMC infusion and donor SPLc/ dornor BMC infusion. Mechanisms responsible for this tolerance induction require future elucidation.

        Pyruvate dehydrogenase(El) composed of two nonidentical subunits a & b has been isolated from Pyruvate dehydrogenase complex(PDC) (porcine kidney). Essential steps in this purification process are involved in fractionation of PDC component by sodium dodecyl sulfate -polyacrylamide gel electrophoresis (SDS -PAGE), electroelution of E1 (a, b) using dialysis membrane, and ethanol precipitation of eluate. Anti-El antiserum has been raised in a rabbit against El(a, b) purified from porcine kidney. The anti-El antiserum is immunoreacted with 20ng of Ela or Elb by a dot-blot assay.
