        Aspergillus candidus F1484 균주가 생산하는 항진균 화합물의 분리 및 특성

        Candida albicans의 효모형태에 대해 활성을 나타내는 항진균 물질의 탐색 중에 Aspergillus candidus F1484 균주의 발효액으로부터 항진균 화합물 F1484를 단리하고 ethyl acetate 추출, silica gel column chromatography, ODS column chromatography 및 preparative HPLC를 행하여 분리정제하였다. F1484 화합물은 EI-MS, ^13C-, ^1H-NMR, DEPT, HMQC 및 HMBC에 의한 구조분석을 행한 결과 이 화합물은 항진균 물질인 chloroflavonin과 동일한 구조를 나타내었으며 효모형태의 Candida albicans에 대한 항진균 활성뿐만 아니라 여러 종류의 인체 종양 세포주에 대해서도 세포독성을 나타내었다. In the course of screening for the antifungal compounds against Candida albicans, an antifungal compound (F1480) was isolated from the culture broth of Aspergillus candidus F1484. Isolation and purification of compound F1484 were performed using ethyl acetate extraction, silica gel column chromatography, ODS column chromatography, and preparative HPLC. The structure of compound F1484 was determined by the spectroscopic analyses of EI-MS, ^13C-, ^1H-NMR, DEPT, HMQC, and HMBC. This compound appeared to have a structure of antifungal agent, chloroflavonin. In addition to antifungal activities against the yeast phase of Candida species, compound F1484 showed cytotoxic effect against various human tumor cell lines.

        클럽 샤프트(Club Shaft) 특성에 따른 골프 스윙(Golf Swing)동작 분석

        본 연구는 클럽 샤프트의 대표적인 재질인 그라파이트(graphite)의 유연한(flexible) 클럽 샤프트(club shaft)의 특성에 따라 피험자가 운동학적(kinematics) 요인이 되는 관절의 각변위, 각속도, 각가속도, 클럽헤드의 속도와 가속도와 같은 변인들이 어떻게 적응하는지 비교 분석하여 보다 효율적인 드라이버 선택에 도움을 주며 샤프트 특성에 따른 신체관절의 움직임에 대한 자료를 제시하고자 하였다. 고속 카메라 2대의 속도는 500fps로 하였고 각 regylar,stiff, x-stiff, 자신의 클럽을 포함 각 4개의 클럽을 사용하여 각 클럽당 3번씩 촬영하였으며 목표방향에서 20m이상 벗어나는 경우의 촬영은 다시 촬영하였다. 본 연구에서는 디지타이징(digitizing)을 신체 9개 마커는 강체로 가정된 클럽과 신체분절 모델로 정의하였으며 2 대의 카메라(500fps)로부터 얻은 avi화일을 컴퓨터에 저장하고 자료로부터 Butterworth 6th order recursive digital filter를 사용하여 1차 자료를 smoothing 하고 DLT를 이용하여 3차원 좌표를 구성하도록 한다. 좌표값을 얻기 위하여 kwon3d v3.0을 이용하였다. 본 실험은 피험자 스스로 클럽의 특성에 따라 스윙의 속도를 달리 하기 때문에 스윙의 시간이 달라지며 어느 정도 클럽이 강성에 따라 스윙시간이 빨라지는 결과로 나타났다. 이것은 피험자가 샤프트가 강성(stiffness)에 따라 스윙 속도를 빨리 하게 되는 원인이 되는 것으로 생각된다. 어깨의 각변위는 클럽이 regular의 경우 임팩트에서 각속도를 계속 유지하고 있으며 stiff, x-stiff의 경우에는 어깨의 움직임이 임팩트에서 급격하게 감소되는 것을 알 수 있다. 이것은 팔의 동작과 클럽의 힘을 크게 하기 위한 동작으로 생각된다. 어깨 각속도는 클럽이 stiff할수록 각속도가 큰 감속하는 것으로 나타났다. 손목속도는 regular 클럽의 경우 손목의 감속이 늦게 되고 임팩트에서 손목의 감속이 적게 하는 것으로 나타났으며 stiff와 x-stiff의 클럽에서 임팩트 시에 순간적인 감가속으로 인해 클럽의 속도를 증가시키고 있다. 임팩트 시에 손목의 감가속은 클럽헤드의 임팩트 시 속도를 증가시키는 결과를 보였다. 클럽헤드는 regular 클럽이 임팩트전에는 속도 증가가 커지는 결과와 일치된 결과를 보이고 있다. The purpose of this study was to find the rational method to analyze golf swing with specific property of club shaft. Three subjects were filmed by two high speed digital cameras with 500fps. The phase analyzed was downswing of each subject. The three-dimensional coordinates of the anatomical landmarks were obtained with motion analysis system Kwon3d 3.0 version and smoothed by lowpass digital filter with cutoff frequency 6Hz. From these data, kinematic and kinetic variables were calculated using Matlab(ver 5.0) The variables for this study were angular velocity and accelerations, which were calculated and following conclusions have been made : 1) Golf swing time of stiff club is faster than that of regular club. 2) In shoulder joint motion of swing with the stiff club, x-stiff showed mort rapid negative acceleration than that of regular club. 3) In regular club, the velocity of club head would be more effective velocity, which was increasing, than those of other clubs before impact. 4) In wrist joint motion of swing with stiff club, x-stiff club showed faster than regular club in the downswing and impact more rapid negative acceleration.

        The reaction of 3-methoxycarbonylmethylene-2-oxo-1,2,3,4-tetrahydroquinoxaline(14) with hydrazine hydrate gave 3-hydrazinocarbonyl-methylene-2-oxo-1,2,3,4-tetrahydroquinoxaline(15), whose reaction with phenyl isothiocyanate provided 3-(4-phenyl-3,4-dihydro-3-thioxo-2H-1,2,4-triazol-5-yl)methylene-2-oxo-1,2,3,4-tetrahydroquinoxaline(16). The tautomeric behavior of compound 15 was investigated on the basis of the tautomer ratio determined by the ¹H-nmr spectral data. The structures of the synthesized compounds were confirmed on the basis of ir, ¹H-nmr and mass spectral data.

        Identification of the Gene Products Responsible for F Plasmid Partitioning

        DNA subfragments, sopA, sopB and sopC which help to maintain the stability of an on C plasmid, were derived from a mini-F plasmid DNA (EcoRI restriction fragment f5) after digestion with restriction endonuclease, and cloned in the vector plasmid pBR322. The recombinant plasmids obtained were introduced into E. coli KY7231 and E. coli CSR603 strains, and proteins specified by the mini-F fragments were analysed by SDS-PAGE. Two proteins encoded by the F fragments were detected, and their molecular weights were 41,000 and 37,000 daltons. Fluorography after one and two dimensional gel electrophoresis of the lysates showed that these two proteins had been overproduced in the cells which were allowed to incorporate radioactive amino acid after plasmid amplification by chloramphenicol treatment. The isoelectric points of sopA and sopB proteins were 6.6 and 7.0, respectively.

      • 2형 당뇨 환자에 있어서 C형 간염의 유병률에 관한 연구

        Background: We have doubt about the truth that Type 2 DM is associated with chronic hepatitis C infection. The aim of our study was to prove a close relationship between chronic HCV infection and type 2 DM in korean patients. Methods: We reviewed the out-patient's medical records of 466 patients with type 2 DM who visit the Sung-ae hospital from March 2000 to August 2001. Diabetic patients were analyzed for the following variables: age, sex, waist circumference, weight, waist/hip ratio, using of self monitoring of blood glucose, method of therapy, duration of disease, experience of previous transfusion, major surgical procedure, accompanied disease, liver function tests (LFT), HbA1 c. We ruled out patients with liver cirrhosis. Results: Hepatitis C virus infection was observed more frequently in patient with Type 2 DM. Of 466 Type 2 DM patients, 13 patients were infected with Hepatitis C virus (2.79%). Compared with mean prevalence of Korean population (1.3~1.6%), it's value is very high. the other hand, 21 patients were infected with Hepatitis B virus (4.5%). Such results are similar to mean prevalence of the general public (4~5%). Conclusions: Prevalence of hepatitis C was closely associated in korean patients with type 2 DM. And an increased prevalence is independent of cirrhosis.

      • 내시경적 점막절제술로 치험한 식도 과립상 세포종 1례

        과립상 세포종은 Schwann 세포 기원으로 생각되며 인체에 비교적 드물게 발생한다. 과립상 세포종은 전신 어느 곳에서나 발견될 수 있으나 주로 혀, 구강, 피부 혹은 유방 등에서 호발하며 드물게 위장관에서 발견된다. 위장관에서는 식도에서 가장 호발하며 다음으로 위, 대장 순이다. 과립상 세포종은 대부분, 특히 위장관에서는 양성이며 소수의 악성 병변이 보고되었다. 이러한 이유와 함께 수술 전의 진단이 어렵기 때문에 과립상 세포종에 대한 근본적인 치료는 현재까지 외과적 절제술이다. 최근에 시도되는 치료방법들로는 내시경적 레이저 치료, 용종절제술, 내시경적 점막 절제술 등이 있다. 저자들은 상부 소화관 내시경검사를 시행하여 식도 과립상 세포종을 진단하고 내시경적 점막 절제술을 시행하여 합병증 없이 퇴원하여 현재 재발없이 경과 관찰중인 1례를 경험하였기에 보고하는 바이다. Granular cell tumors, which occur infrequently, are probably of Schwann cell origin. They can occur almost anywhere in the body but usually affect the tongue, oral cavity, skin, or breasts and are rarely found in the gastrointestinal tracts. The esophagus is the most frequent gastrointestinal site, followed by the stomach and the colon. Granular cell tumors are generally benign, especially in the gastrointestinal tract, some malignant lesions have been reported. For this reason, and also because preoperative diagnosis is difficult, the standard treatment for granular cell tumor has until now been surgical excision. In recent years, other therapeutic methods is endoscopic laser therapy (ELT), polypectiomy, endoscopic mucosal resection (EMR). We report a case of esophageal granular cell tumor which was diagnosed by an endoscopy and managed using an endoscopic mucosal resection without complication.

        Original Article : Geographic difference of epidemiological features of HCV infection in Korea

        Background/Aims: The prevalence of hepatitis C virus (HCV) infection in Korea exhibits significant geographic variation, with it being higher in Busan and Jeonam than in other areas. The reason for this intranational geographic difference was investigated in this study by conducting a comparative analysis of the risk factors related to HCV infection among three geographic areas: the capital (Seoul), Busan, and the province of Jeolla. Methods: In total, 990 patients with chronic HCV infection were prospectively enrolled at 5 university hospitals located in Seoul (n=374), Busan (n=264), and Jeolla (n=352). A standardized questionnaire survey on the risk factors for HCV infection was administered to these three groups of patients, and a comparative analysis of the findings was performed. Results: The analysis revealed significant regional differences in exposure to the risk factors of HCV infection. By comparison with patients in Seoul as a control group in the multivariate analysis, patients in Busan had significantly more experience of invasive medical procedures, acupuncture, cosmetic procedures, and multiple sex partners. In contrast, patients in Jeolla were significantly older, and they had a higher prevalence of hepatocellular carcinoma, a lower prevalence of multiple sex partners, and had experienced fewer invasive procedures. Conclusions: There was a significant geographic difference in the exposure to potential risk factors of HCV infection between patients from the three studied regions. This may explain the regional variation of the prevalence of HCV infection in Korea, and should be taken into account when planning strategies for the prevention and management of HCV infection.         하악골에 발생된 Low grade 골육종:증례보고

        Osteosarcoma is a primary malignant tumor arising most often in the long bone, but rare in jaw bone. Especially osteogenic sarcoma of the mandible is a rare tumor, comprising less than 0.5% of all head and neck tumors. Osteosarcoma occurs cheifly in young persons, and presenting very survival rates. Histopathologically this tumor can be classified three types, osteoblastic, fibroblastic, chondroblastic and classified from low grade to high grade by anaplasia or mitosis of the tumor cells. Sometimes, the exact diagnosis of osteosarcoma is difficult especially in low grade osteosarcoma, even if clinical or radiographical findings suggest to osteosarcoma. So suspcious to malignant bone tumor in clinical or radiological findings, biopsy should be taked from deep portion and multiple area, and sharing the patient history and radiographs with pathologist will assist in the development of the diagnosis. We report a case of low grade osteosarcoma on the mandible initially difficulty in accurate diagnosis. The patient visited our hospital for routine dental treatment but radiographic findings displayed ill-defined radiolucency with osteoid formation on the mandible. Final diagnosis was difficulty in initial biopsy but subsequent biopsy taked from deep portion presented infiltrative growth and mitosis of the tumor cell in some area with small osteoid bone formation, so we could reach final diagnosis as low grade sarcoma. We treated this patient with bloc resection of the mandible and immediate reconstruction with iliac corticocancellous block.
