사티오시아노코발트[Ⅱ]산 이온을 이용한 植物性鹽基成分의 確認 및 定量

A Coloring and spectrophotometric method using tetrathiocyanatocobaltate(Ⅱ) ion as a complexation reagent was developed for the identification of Coptidis Rhizoma and Phellodendri Cortex in herb medicine. Identification tests could be carried out coloring in test tube and ultraviolet-visible spectrophotometry. Coptidis Rhizoma and Phellodendri Cortex: with a solution of 1,2-dichloroethane in thst tube - yellowish green. The ultraviolet - visible spectrum of base components in 1,2-dichloroethane exgibits maxima at 270, 340, 442 and 624nm and was developed for the determination of berberine or palmatine in Coptidis Rhizoma and Phellodendri Cortex. Optimum conditions for good determination was determined: Maximum wavelength 442nm; Determination of berberine was possible to the range of 0.01mg/ml-0.10mg/ml in 1,2-dichloroethane. Calibration curve showed that the coefficient of variation was 0.999% for berberine. And determination of palmatine was possible to the range of 0.10mg/ml - 0.60mg/ml in 1,dichloroethane. The method was rapid, accurate. It was considered that spectrophotometry could be greatly applied to the analysis of base components in Coptidis Rhizoma and Phellodendri Cortex containing relatively small amount of gerb medicine extract.
국내에서 채집한 진드기에서 중합효소연쇄반응을 이용한 라임병균 및 Ehrlichiosis 원인체의 검출

국내에서 채집한 진드기의 라임병 및 ehrlichiosis원인체 보균 상태를 조사하기 위하여 총 516마리 (Ixodes spp. 22마리, Haemaphysalis spp. 494마리)의 ixodid 진드기를 봄과 가을에 걸쳐 강원도 고산지대 일원에서 채집하였다. 수집한 진드기에서 DNA를 추출 ·정제한 후 추출한 DNA를 template로 이용하여 , Borrelia burgdorferi sensu lato 및 Ehrlichia spp.에 특이하게 반응하도록 제작한 primer를 이용한 중합효소연쇄반응 (polymerase chain reaction, PCR)을 실시하였으며, 이 결과를 oligonucleotide probe를 사용한 southern blotting을 통하여 다시 확인하였다. 총 516마리의 진드기중 B.garinii sensu lato DNA 양성 인 진드기는 68(13.2%)마리 (B. burgdorferi sensu stricto,2; B. afzelii,1; B. garinii,33; B.tanukii,8; B.turdae, 4)로 나타났으며 이 중 37 (7.2%)마리의 진드기 는 southern blot analysis에서도 양성으로 확인되었다. 또한 101 (19.2%)마리의 진드기가 Ehrlichia spp.에 대한 PCR에서 양성 이었으나, 이들 중 25 (4.8%)마리 만이 southern blot analysis에서 양성으로 확인되었다. 그러나 사람의 병인체로 추정 되는 human granulocytic ehrlichiosis (HGE) agent DNA를 보균한 진드기는 확인되지 않았다. 한편 라임병균과 ehrlichiosis원인체를 동시에 보균한 것으로 밝혀진 진드기가 3마리에서 (0.6%)확인되어,국내에서도 진드기 교상시 이들 두 가지 열성질환이 동시에 감염될 가능성이 있는 것으로 사료됨으로 진드기 매개성 열성질환에 대한 적절한 진단법 등을 보다 체계적으로 연구하여야 할 것으로 판단된다. To investigate the distribution of Borrelia Burgdorferi and human granulocytic ehrlichiosis(HGE) agent in ticks, adult ixodid ticks of Ixodes spp. and Haemaphysalis spp. were collected from the high mountain areas of Kangwon Province. Using DNAs extracted and purified in the collected ticks, polymerase chain reaction (PCR) was performed to amplify the specific nucleotide sequences of both agents. Of the 516 ticks, a total of 68 (13.2%) ticks was positive for Borrelia burgdorferi sensu lato with PCR analysis (2 for B. burgdorferi sensu stricto; 1 for B. afzelii; 33 for B. garinii; 8 for B.tanukii; 4 for B. turdae). However a little more than half of PCR-positive ticks (37/68) was found to be positive in the southern blot analysis with Bl6S oligonucleotide probe. One hundred and one (19.2%) ticks were positive for Ehrlichia spp. in PCR, and a quarter of them (25/101) was positive in southern blot with E16S oligonucleotide probe. But none of them was found to be the DNA of HGE agent. And 0.6% (3/516) ticks were positive for both of B. burgdorferi sensu lato and Ehrlichia spp. These findings might implicate the possibility of the outbreak of lyme borreliosis and ehrlichiosis in Korea, and more extensive studies may be need for the diagnosis of multiple tick-borne diseases.

In this paper, we propose a nonlinear system identification method using genetic algorithm. We represent the nonlinear system as a parameter vector and a measurement vector. In order to identify the nonlinear system, we find the parameter vector using genetic algorithm. The parameter vector is regarded as a chromosome of gene. The error between the desired output and estimated output every sampling period is used to calculate the fitness of one gene. The simulation results showed the effectiveness of using the genetic algorithm in the nonlinear system identification.

This paper considers identical parallel machine scheduling problem. Each job has a processing time, due date, weight and family type. If a different type of job is follow by prior job, a family setup is incurred. A two phased heuristic is presented for minimizing the sum of weighted tardiness. In the first phase, using rolling horizon technique, group each job into same family and schedule each family. In the second phase, assign each job to machines using schedule obtained in the first phase. Extensive computational experiments and comparisons among other algorithms are carried out to show the efficiency of the proposed algorithm.

The prevalence of PPNG among pretreated gonorrhea cases isolated at the STD clinic of Choong-Ku Public Health Center in Seoul has been studied and reported annually since 1981. In 1991, 123 strains of N.gonorrhoeae were isolated, among which 58(47.1%) were PPNG. In 1992, 98 starains of N.gonorrhoeae were isolated, among which 51(52.0%) were PPNG. In all, 109(49.3%) strains were found to be PPNG among 221 strains isolated between 1991-1992. The prevalence of PPNG in Seoul showed increased tendency till 1989, thereafter, it has been stationary or slightly decreasing.

The objective of this study was to select an effective microbial strain to improve the functional qualities of Eisenia bicyclis water extract by fermentation. For this purpose, several microorganisms isolated from traditional Korean fermented foods were inoculated and cultivated in E. bicyclis water extract. Ultimately, yeast strain YM-1 was selected for further study based on its total phenolic compound (TP) content and antioxidant activity, which were enhanced by microbial fermentation. The extract fermented by YM-1 exhibited a superior TP content and 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity compared to extracts fermented by other microbes. The highest TP content and DPPH radical scavenging activity were observed after one day of YM-1 fermentation. Yeast strain YM-1 was identified as Candida utilis based on an analysis of its physiological characteristics. During fermentation of the extract by C. utilis YM-1, no significant difference was observed in the proximate composition, including moisture, crude lipid, crude protein, and crude ash. Fermentation by C. utilis YM-1 resulted in enhanced biological activity, including increases in the TP content and antioxidant activity. Thus, fermentation by C. utilis YM-1 is an attractive strategy for developing value-added food ingredients.

In an effort to discover alternative phytotherapeutic antimicrobial agents to combat Streptococcus parauberis, a fish pathogenic bacterium, we evaluated the antibacterial activity of seaweed extracts in vitro. A methanolic extract of Ecklonia cava exhibited strong antibacterial activity against S. parauberis isolated from olive flounder Paralichthys olivaceus. Furthermore, the n-hexane soluble (Hexane) fraction of the E. cava methanolic extract exhibited the greatest antibacterial effect on S. parauberis strains with a minimum inhibitory concentration (MIC) ranging from 256 to $1,024{\mu}g/mL$. In addition, the MIC values of oxytetracycline against antibiotic-resistant S. parauberis were markedly reduced up to 64-fold in combination with the Hexane fraction, suggesting that the antibacterial activity of the antibiotic was restored when combined with the Hexane fraction. The interaction between both antibiotics and the Hexane fraction was assessed by the fractional inhibitory concentration (FIC) index. The Hexane fraction and oxytetracycline combination against antibiotic-resistant S. parauberis strains resulted in a median ${\sum}FIC$ range of 0.502 to 0.516. Thus, the synergistic ranges of median ${\sum}FIC$ < 1 were observed for all combinations of the Hexane fraction and oxytetracycline against S. parauberis. To the best of our knowledge, this is the first report indicating the efficacy of an E. cava extract against fish pathogenic bacterium S. parauberis.
