        ABSTRACTMechanical alloying of Al-8wt%Ti and Al-12wt%Ti alloy powders has been studies to understand the mechanical alloying process.Various analytical techniques including microvickers hardness test, SEM, optical microscopy, and X-ray diffraction were used to find optimum conditious for mechanical alloying.Homogeneity of Al-8wt%Ti composite metal powders was obtained at the following process condition: addition of 1.5wt% stearic acid as PCA, the milling velocity of 300rpm and the milling time more that 10hours.At the above conditions it was also found that the mechanical alloyed powders reached steady state.At the steady state the particles have a uniform size distribution, a saturation hardness, an equiaxed shape, a composite structure and a chemical homogeneity.       • 기계적 합금화에 의한 Nb-35at%AI혼합분말의 상변화

        To investigate the phase transformation behaviors of mechanically alloyed Nb-35at%AI powders, the mixed Nb-35at%AI powders were mechanically alloyed in SPEX 8000 Mixer/Mill. Mechanical alloying(MA) time was varied between 1 hour and 20 hours. The phase formation behaviors of these mechanically alloyed powders were examined using X-ray diffraction(XRD), scanning electron microscopy(SEM) and differential thermal analysis(DTA), Appreciable amorphization started from 10 hours of MA. The powders mechanically alloyed for 10 hours were in almost amorphous phase. DTA results showed that the powders mechanically alloyed for 10 hours had a strong exothermic peak about 600℃.

        Dummy-Bar Head is used for continuous casting equipment. It is heated up at high temperature and cooled down repeatedly in using. Then the thermal extension by thermal stress and high temperature oxidation occur to it. This easily results in heating damage because Head, contact with melting metal, appears in micro-crack and tension force. In other words, it is material that gives the greatest influence in endurance life. The method of study is to compare and evaluate present material and improved material by the new alloy design. In conclusion, the results are as follows : 1) I think that the following range is suitable to chemical component. ◁표 삽입▷(원문을 참조하세요) 2) Micro-crack occurs in repeated heating and cooling owing to decarburization and thermal extension. Adding carbide stability elements-Cr, Mn, and V-shows martensite and less micro-crack. 3) Dummy-Bar Head is a large cast steel, An effective heat treatment condition is to heat step by step and to cool by force at 880℃. After that, It is effective to cool at 600∼680℃. 4) Improved product maintains the even hardness value. And impact value is over three times higher than present product for repeated heating and cooling. 5) The transition range over the fracture toughness value has smaller in improved product than present product for repeated heating and cooling. Through the fracture toughness evaluation, the characteristics of change is evaluated in materials. And we can make the predictable criteria of life with Refrica method. According to the analysis and investigation of the real condition is use, the improved product is accomplished in optimal alloying design and product processing, and satifies the required condition. I think that it is widely used as proper materials to a higher temperature concerning the economy.

        Aequorin Based Functional Assessment of the Melanin Concentrating Hormone Receptor by Intracellular Calcium Mobilization

        Melanin concentrating hormone is a neuropeptide highly expressed in the brain that regulates several physiological functions mediated by receptors in the G-protein coupled receptor family, especially plays an important role in the complex regulation of energy balance and body weight mediated by the melanin concentrating hormone receptor subtype 1 (MCH1). Compelling pharmacological evidence implicating MCH1 signaling in the regulation of food intake and energy expenditure has generated a great deal of interest by pharmaceutical companies as MCH1 antagonists may have potential therapeutic benefit in the treatment of obesity and metabolic syndrome. Although fluorescence-based calcium mobilization assay platform has been one of the most widely accepted tools for receptor research and drug discovery, fluorescence interference and shallow assay window limit their application in high throughput screening and have led to a growing interest in alternative, luminescence-based technologies. Herein, a luminescence-based functional assay system for the MCH1 receptor was developed and validated with the mitochondrial targeted aequorin. Aequorin based functional assay system for MCH1 presented excellent Z' factor (0.8983) and high signal-to-noise ratio (141.9). The nonpeptide MCH1 receptor antagonist, SNAP 7941 and GSK 803430, exhibited $IC_{50}$ values of 0.62 ${\pm}$ 0.11 and 12.29 ${\pm}$ 2.31 nM with excellent correlation coefficient. These results suggest that the aequorin based assay system for MCH1 is a strong alternative to the traditional GPCR related tools such as radioligand binding experiments and fluorescence functional determinations for the compound screening and receptor research.

        Background : This study was development of moisture tolerance and high-yielding Rehmannia glutinosa cultivar. Methods and Results : Segang is developed by the medicinal crop breeding team of National Institute of Horticulture and Herbal Science(NIHHS), Rural Development Administration(RDA), during the period from 2005 to 2015. The reproduction of Rehmannia glutinosa has been accomplished mainly by vegetative propagation with its seedlings have many variants. The cultivar was selected from seedling of Jihwang 1(check variety). The plant type of Segang is some rising from ground. Regional yield trials conducted at three site from 2014 to 2015. The root yield of Segang was 21.1ton per hectare, which was increased 12% compared with Jihwang 1. Also, Segang has higher catalpol content and dried root ratio compared with Jihwang 1. Conclusion : Segang is a moisture tolerance and high-yielding Rehmannia glutinosa cultivar.

      • PCR-based S.pombe transformation for systematic deletion mutants and functional analysis of human gene

        S. pombe의 genomic sequence 염기서열의 분석결과에 따르면 유전자의 기능적 반복성이 적으며 S. pombe에는 진핵세포에서 중요한 세포의 기능과 질병에 관련된 유전자들이 많은 양 차지하고 있다고 보고되었다. 따라서 S. pombe는 약물개발을 위한 cell-based 시스템으로서 인간 유전자의 기능연구에 효율적인 방법으로 쓰일 수 있다. 본 실험에서는 S. pombe의 genome-wide systematic deletion mutant 제조를 완성하기 위한 기반을 마련하기 위하여 이미 deletion mutant가 제조 된 바 있는 두 개의 유전자인 sum1, gef3를 직접 deletion 하여 gene deleted S. pombe를 제조 해 보았다. sum1은 S. pombe에서 essential gene으로써 chromosome 1에 존재하고, gef3은 non-essential gene으로써 chromosome 2에 존재한다. 이렇듯 gene의 essentiality를 증명하기 위하여 deletion mutant를 만드는 과정 중 yeast transformation을 위한 DNA 단편의 제조를 완성 하였으며, agarose gel electrophoresis를 통하여 확인 할 수 있었다. 이와 같이 임의로 제조된 DNA 단편은 S. pombe에 transformation 시킴으로써 mutant를 만들기 위한 핵심 요소가 될 것이다.

      • Detection of calcium signaling in MCH1 receptor

        Melanin concentrating hormone(MCH)은 동물에서 외측 시상하부와 zona incerta에서 발현되는데 에너지 균형과 체중을 조절하는 중요한 매개체로 작용한다. 사람에서 MCH는 MCH receptor subtype 1 (MCH1)과 subtype 2 두 가지의 수용체를 통해 전달되는데 그 중 MCH1의 신호전달 과정을 통해 음식섭취와 에너지 소비를 조절하기 때문에 MCH1 antagonist로 비만, 대사 증후군의 치료제로 연구되고 있다. MCH1는 두 가지의 G protein-coupled receptors로써 신호 전달되는데 Gi- or Gq-dependent 경로를 다중 활성화한다. 본 실험에서는 Gq 경로를 통해 변화하는 세포 내 칼슘 이온을 형광염료를 이용해 확인하기 위하여 MCH1이 발현되어있고, Gq pathway를 활성화하는 Gα16 단백질이 동시에 발현되어 있는 세포를 사용하였다. MCH 가 결합했을 때 MCH1의 Gq 경로를 통해 소포체에서 세포질로 방출되는 칼슘 이온이 형광 염료와 결합하여 형광을 띄는 adherent 세포를 현미경을 통해 관찰하여 MCH를 첨가하였을 때 세포에서 형광의 변화를 동영상으로 얻을 수 있었다. 그리고 다기능 플레이트 신호측정장비인 EnVison을 사용하여 세포 내 칼슘 변화를 형광 신호 값으로 측정 함으로써 서로 다른 농도의 MCH 첨가에 따른 농도 의존성 반응 곡선을 구할 수 있었다.

        Changes in Adrenal Angiotensin II Receptors in Renin-dependent Hypertensive Rats

        The changes in blood pressure may relate to the alterations of the responsiveness to vasoconstrictors and vasodilators, and these alterations can arise the modifications in the properties of angiotensin II (AII) receptor. In order to examine the changes of AII receptor in the hypertensive mechanism of renin-dependent hypertensive rats (RHRs; two-kidney, one-ligated type), we compared the equilibrium binding characteristics of $[^3H]$All in adrenal cortex and medulla from RHRs and normotensive rats. The dissociation constants of AII binding in both tissues of RHRs were very similar to those in the respective tissue of normotensive rats. However, the maximum binding was increased from 805 to 1050 fmole/mg protein in the adrenal cortex of RHRs, and decreased from 172 to 126 fmole/mg protein in the adrenal medulla of RHRs. These results imply that the up- and down-regulation of the All receptor population on the cell surface of adrenal glands from RHRs are consorted with the elevation of blood pressure and the activation of renin-angiotensin system.
