        한우, 칡소 및 제주 흑우 Calpain-Calpastatin 유전자 다양성

        The aim of study was to investigate genetic diversity for the calpain/calpastatin gene in three Hanwoo breeds [(Brown (n=62), Brindle (n=81) and Jeju Black (n=30)]. Random samples from three breeds of Hanwoo were selected and genotyped for the 7 SNPs of calpain/calpastatin using TaqMan method. Allele frequencies were investigated for CAPN1/CAST gene. Allele frequency of CAST2 SNP was 0.75, 0.59 and 0.22 for Brown, Brindle and Jeju black, respectively. The CAST3 revealed allele frequency of 0.59 and 0.57 in Brown and Jeju Black, while it showed very low allele frequency (0.07) in Brindle. In particular, favorable allele (G allele) for the CAPN1-2 SNP which was shown a strong association with tenderness in Taurine and Indicine cattle revealed 16% and 17% higher allele frequency in Brown Hanwoo (0.82) comparing Brindle (0.66) and Jeju Black Hanwoo (0.65). AMOVA demonstrated that among population variance occupied only 10% of total variance and among individual variance was 0%, while within individual variance was 90% of total variance. This result showed that population effect contributed very small portion of genetic to these three Hanwoo breeds, while within individual variance contributed large portion of genetic diversity within these Hanwoo breeds. In conclusion, three Hanwoo breeds (Brown, Brindle and Jeju black) showed a genetically homogeneous based on the 7 SNPs of CAPN1/CAST gene and it came from same ancestor to form modern Hanwoo breed.

        프레탈^(R)정(실로스타졸 50mg)에 대한 실로졸^(R)정의 생물학적 동등성

        Bioequivalence of two cilostazol tablets, the Pletaal^(R)(Korea Otsuka Pharmaceutical Co., Ltd.) and the Cilozol^(R)(Hanmi Pharmaceutical Co., Ltd.), was evaluated according to the guideline of KFDA, Sixteen normal male volunteers(age 20~29 years old) were divided into two groups and a randomized 22 cross-over study was employed. After two tablets containing 50㎎ of cilostazol were orally administered. blood was taken at predetermined time intervals and the concentration of cilostazol in plasma was determined with an HPLC method using UV detector. The pharmacokinetic parameters(C_(max), T_(max) and AUC_(t)) were calculated and ANOVA was utilized for the statistical analysis of parameters. The results showed that the differences in C_(max), T_(max) and AUC_(t) between two tablets were 4.99%, 1.74% and 7.68%, respectively. The powers(1-β) for C_(max), T_(max) and AUC_(t) were83.92%, 80.12% and 85.03%, respectively. Detectable differences(Δ) and confidence intervals were all less than 20%, and confidence interval of all the parameters were also less than 20% at the significance level(α) of 0.05. All of these parameters met the criteria of KFDA for bioequivalence, indicating that Cilozol^(R) tablet is bioequivalent to Pletaal^(R) tablet.

        Objectives : Sipjundaebo-tang is prepared by ten medical herbs that tone the blood and vital energy, and strengthen health. This prescription has long been used traditionally against anemia, anorexia, extreme exhausion and fatigue. The purpose of this thesis was to review the effects of Sipjundaebo-tang that are about immunopotentiation effects, antitumor activities and potentiation, detoxification of antitumor drugs Methods : We studied the research methods and results of experiments which were selected from Korean, Japaness and American theses were on the topic of immunopotentiation effects, antitumor activities and potetiation, detoxification of antitumor drugs from Sipjundaebo-tang Results and conclusions : Sipjundaebo-tang not only potentiates the effects of the combined use of chemotherapy, but also reduces and elimimate the immunotoxicity of antitumor drugs and radiotherapy. And it strengthen immunity and improve QOL, S/OH of cancer patients.

      • GIS를 이용한 부여 서부지역의 산사태 연구

        지구정보시스템(GIS)을 이용하여 충청남도 부여군 서부지역의 산사태 발생 가능성을 분석하였다. 산사태 취약성 분석을 위하여 산사태 발생에 영향을 미치는 사면경사, 토양침식율, 지질, 토지이용도 등과 같은 환경지질학적 요인들을 선정한 후 산사태 발생지역에 대한 통계분석을 행하였다. 분석 결과를 이용하여 database를 구축하고 각각의 자료들을 중첩시켜 산사태 취약성 지수(LSI)를 산출한 다음 이 지수와 분포 면적에 대한 누적곡선을 작성하여 산사태 취약성도를 작성하였다. 산사태 취약성은 연구지역 남쪽과 북쪽 경계부, 그리고 은산면 중앙부 지역에 매우 높게 나타났으며, 은산면 북서쪽과 내산면 일대는 비교적 높은 산사태 취약성을 보였다. 그러나 규암면 일대와 구룡면, 옥산면을 잇는 수계하천을 따라서는 산사태 취약성이 매우 낮게 나타났다. This study used GIS to analyze the possibility of landslide occurrence in the western part of Puyeo-kun, Chung Nam Province. For the regional analysis of landslide, environmental and geological factors such as topographic slope, soil erodibility, geology, landuse which have influence on the landslide occurrence were extracted, and then analyzed statistically for the area of landslide occurrence. With the result obtained by statistical analysis, databases were constructed. LSI were calculated by overlapping each factor. Landslide susceptibility map was obtained by use of LSI and cumulative curve of area. The results of this study show that each border of the south and north regions and the center of Eunsan-myeon have the very high susceptibility and northwest of Eunsan-myeon and side area of drainage between Gurong-myeon and Oksan-myeon have the very low landslide susceptibility.

        The substrate removal rate in a biofilm was investigated by the measurement of substrate concentration in an inverse fluidized bed operated batchwisely. When the biofilm thickness was below 60㎛, the overall reaction rate was found as first order but it turned out to zero order at the thickness of above 60㎛. The reaction rate constant and the effective diffusivity could be calculated by using a mass balance and a diffusion model in the biofilm and a typical effective diffusivity of zero and first order reaction were calculated as 5.49×??㎠/s and 2.3×??㎠/s, respectively. In the region of zero order reaction, the effect of diffusion on the effectiveness factor increased with increasing biofilm thickness. Therefore in the zero order region, the diffusion step controlled the system while the reaction step controlled in the first order region.

        이산화탄소 재호흡법을 이용한 비침습적 심박출량 측정법

        N/A Objetive: The measurement of cardiac output by CO2 rebreathing method is noninvasive procedure using indirect Fick equation. In order to compare the result of cardiac output measured by CO2 rebreathing method with that by thermodilution technique, this study was performed. Methods: Simultaneous measurement of cardiac output by CO2 rebreathing method and thermodilution technique was performed in 13 mitral stenosis patients. The subjects were 4 men and 9 women, with mean age of 41.15±11.01 year. The cardiac output (CO) can be calculated from indirect Fick equation using the CO2 rebreathing method, CO=CO2 production/CvCO2-CaCO2. The CO2 production was obtained by collecting expired gas and multiply its volume by CO2 concentration and the arterial PCO2 was estimated from the end tidal PCO2. The mixed venous PCO2 was obtained from rebreathing plateau during O2 and mixture gas breathing through rebreathing bag. Results: 1) The average cardiac output was 3.41±0.45(L/min) by CO2 rebreathing method and 3.45±0.37(L/min) by thermodilution technique. 2) The result of cardiac output measured by CO2 rebreathing method was highly correlated to that by thermodilution technique (r=0.82). 3) The equation relating two measurements was Y =-0.01±0.99X. (Y: CO2 rebreathing method, X: thermodilution technique) Conclusion: This study showed that the result of cardiac output by CO2 rebreathing method was correlated well with that by standard thermodilution technique, so it is thought that CO2 rebreathing method could provide valid estimate of cardiac output for evaluation of cardiac function. It is simple to perform, easily repeatable and essentially risk free. Therefore this method could be very useful for clinical use.

        한국홍삼(韓國紅蔘)과 북미삼(北美蔘)의 환경호르몬 독성방어(毒性防禦) 활성(活性) 및 화학성분(化學成分) 비교(比較)

        Objective: This study was carried out to reveal differences between Korean red ginseng(Panax ginseng C.A. Meyer) and American ginseng(Panax quinquefolius L.) on the protective effects against environmental hormones-induced toxicity. Methods: For the assessment of experiment, male Sprague Dawley rats were divided to normal control, TCDD only and TCDD plus saponins (40mg/kg)-administered groups. A single dose of TCDD (50㎍/kg) was intraperitoneally injected to rats on the seventh day. Results: Administrations of Korean red ginseng saponin fraction reduced TCDD-induced weight loss and histopathological lesions of liver and testis. In contrast, administration of American ginseng did not. A percentile of ginsenoside Rh1 was peculiarly high in Korean red ginseng and administration of ginsenoside Rh1 (20mg/kg) attenuated the TCDD-induced weight loss and histopathological lesions of liver and testis were also reduced. Conclusions: The saponin of Korean red ginseng might prevent the endocrine disruptor-induced toxicity whereas American ginseng did not. It is also not able that the difference of pharmacologocal activity between Korean red ginseng and American ginseng may be originated from their different chemical compositions.

        <P>We demonstrate a highly sensitive THz molecular imaging (TMI) technique involving differential modulation of surface plasmons induced on nanoparticles and obtain target specific in vivo images of cancers. This technique can detect quantities of gold nanoparticles as small as 15 µM in vivo. A comparison of TMI images with near infrared absorption images shows the superior sensitivity of TMI. Furthermore, the quantification property of TMI is excellent, being linearly proportional to the concentration of nanoparticles. The target specificity issue is also addressed at the ex vivo and cell levels. The high thermal sensitivity of TMI can help extend photonic-based photothermal molecular imaging researches from the in vitro level to the in vivo level. The TMI technique can be used for monitoring drug delivery processes and for early cancer diagnosis.</P>

        흰쥐의 발목염좌에 대한 전침, 온침 및 고주파온침 자극별 진통 효과의 비교

        Objectives : The objectives of this study is to compare the effects of electroacupuncture(EA), warm needling(WN) and Radio Frequency warm needling(RFWN) stimulations on the acupoints at the artificially damaged ankles of Sprague-Dawley rats, which could be classified as the Grade 3. Methods : The foot weight bearing force ratio(FWBFR) of ankle sprain was measured first at 24 hours after without any other stimulations. Pain Recovery Index(PRI) represents the analgesic level, and modified Pain Recovery Index(mPRI) shows the accumulated recovery level. PRI was measured at 2 hours after each stimulus on GB34, GB39 and GB42, and mPRI was during 7 days. Results : EA stimulation of GB34 and GB39 acupoint in grade 3 ankle sprain showed a marked analgesic and recovery effect. RFWN of GB42 exhibited significant analgesic and pain recovery effect. RFWN of GB34 resulted in pain recovery effect but not analgesic effects, and RFWN of GB39 resulted in analgesic effect 2 hours after but not pain recovery. However, WN did not affect the pain recovery among three acupoints at all. Conclusions : In the Grade 3 ankle sprain model, the difference of analgesic effects were explained by the acupoints and the stimulation methods according to the accumulated recovery effects during 7 days. It is insufficient to determine that a certain acupoint has a specific analgesic effect depending on the stimulation method by the results of this study. Therefore, the effects of each stimulation on the acupoints in any other meridians at the sprained ankle should be compared and analyzed.
