        In this paper, the image binarization method using standard specifications of the vehicle license plate to recognize characters on the plates is proposed. The global binarization method that had a threshold over entire image pixels cannot acuratelly binarize in case that local average brightness are varied to each plates characters regions. The proposed method uses the characters regions in the standard specifications of the korean vehicle license plates as binarization blocks. For installed cameras on national roads and parking areas, the computer simulation results showed average characters recognition rate about 5% higher than the otzu global binarization method.

      • 표정인식을 위한 PCA와 템플릿 정합을 사용한 얼굴 요소 추출

        This paper proposed an extracting algorithm of human facial components for the recognition of facial expressions. First face image is acquired using the Haar-like feature mask from an input image. The face image is divided into two images. One is the upper image including eye and eyebrow. The other is the lower image including mouth and jaw. The extraction of facial components, such as eye and mouth, begins getting eye image and mouth image. Then eigenfaces are produced by the PCA training process with learning images. An eigeneye and an eigenmouth are produced from eigenfaces. Result eye image is obtained by template matching the upper image with the eigeneye, and the mouth image is obtained by template matching the lower image with the eigenmouth. The simulation results show that the proposed method has superior extraction ratio than previous method.

      • Bioconverted Orostachys japonicas Extracts Suppress Angiogenic Activity of Ms-1 Endothelial Cells

        <P>Orostachys japonicus A. Berger (), known as Wa-song in Korea, has been reported to exert various biological effects, such as anti-tumor, anti-oxidant, and anti-febrile effects. However, the anti-angiogenic effects of O. japonicus extracts remain to be investigated. In the present study, we demonstrated the anti-angiogenic effects of bioconverted O. japonicus extract (BOE) in Ms-1 mouse endothelial cells and compared them with the bioactivities of O. japonicus extract (OE). BOE, but not OE, were found to exert anti-angiogenic effects, including inhibition of cell migration, cell adhesion, tube formation of Ms-1 cells, and blood vessel formation of matrigel plug assay in vivo. Furthermore, protein levels of phosphorylated Src kinase were lower in BOE-treated cells than in OE-treated cells. Treatment with OE or BOE did not influence cell viability during the experimental period. Bioconverted extract of O. japonicus have anti-angiogenic effects in vitro and vivo, but non-bioconverted extract do not. We suggest that these observed anti-angiogenic effects are caused by the changes in the composition of bioactive compounds in the extracts as a result of biological conversion.</P>

        Anti-Metastasis Effects of Ginsenoside Rg3 in B16F10 Cells

        Ginsenoside Rg3 is a bioactive ginseng constituent that has been reported to have diverse pathological and physiological effects, including anti-inflammatory and anti-metastatic activities. Metastasis is one of the most important factors involved in patients with melanoma. However, the molecular mechanism underlying the anti-metastatic activities of Rg3 in malignant melanoma cancer has not been fully elucidated. In this study, we have evaluated that Rg3 effectively inhibits metastasis of B16F10 melanoma cancer cells. We found that Rg3 significantly suppresses the migration, invasion, wound healing, and colony-forming abilities of B16F10 cells in a dose-dependent manner. Mechanistically, we demonstrate that Rg3 suppresses B16F10 cell metastasis by inhibiting MMP-13 expression. These results indicate that Rg3 suppresses the metastasis of B16F10 mouse melanoma cancer cells via MMP-13 regulation. Importantly, MMP-13 downregulation may influence the migration and invasion capabilities of melanoma cells and has been correlated with melanoma progression. Therefore, Rg3 is a potential therapeutic candidate that could be used to treat patients with metastatic melanoma.

        Assessment of the effect of cooking on speciation and bioaccessibility/cellular uptake of arsenic in rice, using <i>in vitro</i> digestion and Caco-2 and PSI cells as model

        <P><B>Abstract</B></P> <P> <I>In vitro</I> digestion/Caco-2 or pig small intestinal epithelium cell line (PSI) uptake models were used to study the bioaccessibility and cellular uptake of arsenic (As) in cooked white rice and brown rice. The arsenite(AsIII), was the predominant species in cooked rice and in its bioaccessible fractions. The percentage of total As bioaccessibility in white rice (75%) was slightly higher (p=0.061) than that in brown rice(66%). However, there was no difference in the inorganic As (iAs) bioaccessibility between white rice (95%) and brown rice (96%). In Caco-2 cell monolayer, total As retention was 7-31%, transport was 4-25%, and uptake (sum of retention and transport) was 16-38%. In PSI cell model, the retention, transport, and uptake of tAs were 10-28%, 14-31%, and 29-50%, respectively. In both cells, the cellular uptake of tAs in brown rice was 1.4-1.5 folds lower (p<0.05) than that of white rice. These results indicate that the cellular uptake of As can be affected by nutritional compositions.         Numerical study on the effect of the tip clearance of a 100 HP axial fan on the aerodynamic performance and unsteady stall characteristics

        In this study, a numerical analysis was conducted to investigate the effect of the tip clearance on the aerodynamic performance, internal flow characteristics, and stall region characteristics of an axial fan. Three-dimensional steady and unsteady Reynolds-averaged Navier-Stokes (RANS) calculations were conducted with a shear stress transport (SST) turbulence model. Tip clearance ratios of 0, 0.01, and 0.02 were applied to the impeller. As the tip clearance ratio increased, the aerodynamic performance of the axial fan decreased at both the design and the off-design conditions. The correlation between the tip leakage vortex (TLV) and the flow angle of the velocity triangle was presented for the difference in the tip clearance and flow rate. As the flow rate increased, the differences in the aerodynamic performance induced by the tip clearance ratio decreased. As the tip clearance ratio increased, the size of the TLV increased and gradually moved in the circumferential direction to interfere with the main flow at the low flow rate. Meanwhile, the size of the TLV was similar and gradually moved in the axial direction even if the tip clearance ratio increased at the high flow rate. The pressure fluctuations were observed by the fast Fourier transformation (FFT) analysis to compare and analyze internal flow characteristics at the stall region and design point. The static pressure was converted to the appropriate magnitude. The locations of the highest magnitude were shown to be different at the stall region and the design point, respectively.

        Autophagy mediates enhancement of proangiogenic activity by hypoxia in mesenchymal stromal/stem cells

        <P><B>Abstract</B></P> <P>Mesenchymal stromal/stem cells (MSCs) have been promising source for regenerative cell therapy in ischemic diseases. To improve efficacy of MSC therapy, various priming methods have been developed, and hypoxic priming has been reported to enhance therapeutic efficacy of MSCs by increasing secretion level of growth factors and cytokines. Recently, it has been reported that bone marrow MSCs primed with hypoxic condition show an increase of autophagy. Here, we addressed whether proangiogenic activity increased by hypoxic condition is associated with autophagy. Wharton's jelly derived MSCs primed with hypoxia showed increase of autophagy with increased hypoxia inducible factor-1α level, and conditioned medium (CM) derived from these cells showed increased levels of migration and tube formation of human umbilical vein endothelial cells (HUVECs) compared to non-primed MSCs-derived CM. Pretreatment with autophagy inhibitor 3-methyladenine or chloroquine prior to exposure of hypoxia resulted in reduction of migration and tube formation of HUVECs. CM obtained under hypoxic condition from MSCs in which autophagy activity was inhibited by ATG5 and ATG7 siRNA treatment also showed decrease of migration and tube formation of HUVECs. Accordingly, secretion levels of angiogenin and VEGF that were markedly increased upon hypoxia exposure was decreased by ATG5/7 knockdown.       • Analysis of QTL Associated with WBPH and Identification of WBPH Mediated Compounds in Rice (Oryza sativa L.)

        The White backed planthopper (WBPH), Sogatella furcifera (Horvath) is one of the serious insect pests in rice growing region in Asia. When rice is attacked by the insect it releases secondary metabolites for self-defense. In this study, we identified WBPH-mediated compounds from a cross ‘Cheongcheongbyeo/Nagdongbyeo’ doubled haploid (CNDH). The compounds were located in chromosome. Leaves and stem of CNDH lines were infected by 2∼3 insta of 3 weeks WBPH and samples were extracted by 90% methanol. Extracted compounds were analyzed through HPLC. TLC was used in separating the target compounds. QTL analysis of compound was done using winQTLcart 2.5 program. Chrysoeriol was highly contained in Cheongcheongbyeo. QTL location is found on chromosome by winQTLcart 2.5. QTL analcited with compound7 was detected on chromosome 4, 7 and 12. qFla4 was detected on chromosome 4 in RM280-RM6909 at LOD 3.5 with 30% of variation. qFla7 was detected on chromosome 7 in RM248-RM1134 with LOD 3.0 with 30% of variation. qFla12 was detected on chromosome 12 in RM1226-RM12 with LOD 2.7 with 40% of variation. Cochlioquinone was detected on chromosome8, qFla8 in RM23230-RM3689 with LOD 2.5 with 30% of variation. Chrysoeriol and Cochlioquinone separated to condition of (Chloroform: Methanol:1-Butanol:Water=4:5:6:4). Separated compounds were analyzed by LC/MS and NMR. These results, investigation is being done to ditermine how the secondary metabolites come lead to pathways of genes and its effect on WBPH relation.
