        젊은 습관성 음주자에서 도플러 심초음파를 이용한 좌우심실 이완 기능에 관한 연구

        N/A Objectives: It is well known that many chronic alcoholics manifest diastolic dysfunction of left ventricle in its early stage. But the effects of chronic alcohol drinking on right ventricular function are not well understood. Thus left ventricular filling impairment and its effects to right ventricular diastolic function were evaluated in young chronic alcoholics. Methods: For the evaluation of left and right ventricular diastolic function in chronic alcoholics, 30 young chronic alcoholics and 28 control subjects were studied by pulsed Doppler echocardiography at the left and right ventricular inflow. Peak E velocity, peak A velocity, E/A velocity ratio, acceleration time and deceleration time were measured as diastolic filling parameters. Results: 1) In the chronic alcoholics, the interventricular septum and posterior wall were thicker and left ventricular muscle mass was significantly increased than that in controls. 2) Among 30 cases of chronic alcoholics, 8 cases(26.7%) showed that the E/A ratio of the left ventricle was less than 1.0 and the deceleration time of the left ventricle was more than 240msec, where- as 12 cases(40.0%) showed that the E/A ratio of the left ventricle was less than 1.0. 3) Among 30 cases of chronic alcoholics, 8 cases (26.7%) showed that the E/A ratio of the right ventricle was less than 1.0 and the deceleration time of the right ventricle was more than 232msec, whereas 14 cases(46.7%) showed that the E/A ratio of the right ventricle was less than 1.0. 4) The HV E/A ratio was significantly correlated with the LV E/A ratio(r=0.697, p<0.001). 5) Blood pressure, ejection fraction, left ventricular mass, E/A ratio and deceleration time of both ventricles were not significantly different in comparison with the daily average amount, duration of ingestion, and total lifetime dose of alcohol. Conclusion: In all chronic alcoholics less than 50 years of age, the left ventricular systolic function was normal. But Doppler echocardiography showed that diastolic dysfunction of the left and right ventricles was present in 8 cases of 30 chronic alcoholics. Right ventricular diastolic dysfunction was closely related with left ventricular diastolic dysfunction. In conclusion, diastolic dysfunction of both ventricle in chronic alcoholics may be the earliest functional sign of preclinical alcoholic cardiomyopathy.

        90-Day Inhalation Toxicity of Dimethylamine in F344 Rats

        Dimethylamine (DMA) is a widely used commodity chemical with few toxicity data. Groups of 10 male and female F-344 rats were exposed by inhalation to 0, 5, 10, 20, 40 and 80 ppm of DMA for 6 hrs/day, 5 days/week for 90 days. The changes of body weight, organ weight, hematology, clinical chemistry, and histopathological changes were evaluated after the exposure. As the results, the body weight was significantly decreased at 80 ppm in male and female rats (p<0.05). The absolute lung weight showed no statistically significant changes in any group. In contrast, the relative lung weight significantly increased at 80 ppm in male and female rats (p<0.05). Erythrocytes, mean cell hemoglobin, leukocytes, neutrophil, and platelet numbers were significantly increased in male and female at 40 or 80 ppm of DMA (p<0.05, p<0.01). In addition, the serum values of total protein, urea nitrogen were increased in male and creatine kinase, total protein were increased in female rats at 40 or 80 ppm (p<0.05, p<0.01). Histopathological examinations of the male and female lung samples showed slight hyperplasia and congestion at 80 ppm. Taken together, our study revealed that maximum tolerated dose of DMA would be over 40 ppm

        심율동전환을 시행하는 환자에서 정맥마취를 위해 병용한 Midazolam, Fentanyl, Propofol 의 효과

Midazolam, fentanyl and propofol on hemodynamic changes and side effects were studied. Methods: Thirty patients scheduled for an elective cardioversion in the coronary care unit received propofol 500 ㎍/kg I.V., followed by a continuous infusion with 25-75 ㎍/kg/min until they lost consciousness as as determined by cessation of response to verbal command. All patients received mi-dazolam 0.02 mg/kg and fentanyl 2 ㎍/kg I.V. 5 minutes before induction In addition, the cardiologist administrated the electric shock. Blood pressures and heart rates were measured prior to induction, after the administration of midazolam and fentanyl, after cardioversion, 5 minutes after cardioversion and recovery. The recovery time from termination drug administration to awakening, and the total dose of propofol were recorded. Patients were observed for side effects. Results: Systolic, mean, and diastolic blood pressures were significantly decreased 5 minutes after cardioversion and recovery. Heart rates were significantly decreased after cardioversion, 5 minutes after cardioversion and recovery. Recovery times were 22.1 ?? 14.9 minutes and the total dose of propofol was 0.79 ?? 0.39 mg/kg. Incidence of apnea (> 30 s) was 30%. Conclusions: We concluded that midazolam, fentanyl and propofol in patients undergoing an electric cardioversion decreased blood pressure and heart rate, but the cardiovascular status was maintained within clinically acceptable levels.       • Variable-number tandem repeat loci-discriminating <i>Pleurotus ostreatus</i> cultivars

        <P><B>Abstract</B></P> <P> <I>Pleurotus ostreatus</I> is one of the most important edible mushrooms. Many cultivars have been bred to meet consumer needs. The identification of cultivars based on the morphological characteristics is restricted because fruiting bodies are frequently capricious due to environmental conditions; accordingly, sequence-based methods are required. A total of 546 simple sequence repeat primers derived from the <I>P. ostreatus</I> genome were screened, and one primer, JHH_SSR-184, was found to show polymorphisms on the major cultivars in Korea. The sequences of the polymorphic loci showed variable-number tandem repeat loci-like features enabling cultivar specificity. Thus, these loci might be applicable to discriminate <I>P. ostreatus</I> cultivars.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The SSR marker discriminated the Korean major <I>Pleurotus ostreatus</I> cultivars. </LI> <LI> The cultivar specific sequences showed variable number tandem repeat loci-like feature. </LI> <LI> Nine loci with variable copy number of the repeat conferred the cultivar specificity. </LI> </UL> </P>

        EDXRF에 의한 용접흄 중의 중금속의 비파괴 정량

        The EDXRF(energy Dispersive X-ray Fluorescence Spec=trometer) technique was applied to the determination of heavy metals in welding fume. The EDXRF method designed in this study was a non-destructive analysis method. Samples were analyzed directly by EDXRF without any pre-treatment such as digestion and dilution. The samples used to evaluate such as digestion and dilution. the samples used to evaluate this method were laboratory samples exposed in a chamber connected with a welding fume generator. The samples were first analyzed using a non-destructive EDXRF method. The samples subsequently were analyzed using AAS method to verify accuracy of the EDXRF method. The purpose of this study was to evaluate the possibility of the non-destructive analysis of heavy metals in welding fume by EDXRF. The results of this study were as follow: 1. When the samples were collected under the open-face sampling condition, a surface distribution of welding fume particles on sample filters was uniform, which made non-destructive analysis possible. 2. The method was statistically evaluated according to the NIOSH(National Institute for Occupational Safety and Health) and HSE(Health and Safety Executive) Method. 3. The overall precision of the EDXRF method was calculated at 3.45% for Cr, 2.57% for Fe and 3.78% for Mn as relative standard deviation(RSD), respectively. The limits of detection were calculated at 0.46 ㎍/sample for Cr, 0.20 ㎍/sample for Fe and 1.14 ㎍/sample for Mn, respectively. 4. A comparison between the results of Cr, Fe, Mn analyzed by EDXRF and AAS was made in order to assess the accuracy of EDXRF method. The correlation coefficient between the results of EDXRF and AAS was 0.9985 for cr, 0,995 for Fe and 0.9982 for Mn, respectively. The overall uncertainty was determined to be ±12.31% , 8.64% and 11.91% for Cr, Fe and Mn, respectively. In conclusion, this study showed that Cr, Fe, Mn in welding fume were successfully analyzed by the EDXRF without any sample pre-treatment such as digestion and dilution and a good correlation between the results of EDXRF and AAS was obtained. It was thus possible to use the EDXRF technique as an analysis method of working environment samples. The EDXRF method was an efficient method in a non-destructive analysis of heavy metals in welding fume.

        線蟲 寄生 天敵 眞菌의 接種源 濃度와 溫度條件에 따른 線蟲感染 및 集團의 減少效果

        Five nematophagous fungi were evaluated for their nematicidal effect in vitro on Rhabditis sp. and Meloidogyne hapla in soil. Inocula of Arthrobotrys arthrobotryoides, A. conoides, A. oligospora, Dactylella lobata, and Fusarium oxyaporum were grown in moistened corn-sandy soil and chopped potato-sandy soil media, and incubated at 26 for one week. The prepared inocula were incorporated in autoclaved sandy soil, mixing thoroughly at rates equ-invalent to 1:50, 1:100, 1:200, and 1:400, repectively, before 80g of the mixture carrying 100 Rhabditis sp. was put into petri plates. Nematophagous fungi effectively teduced the popuation of Rhabditis sp. in soil in a week or two following treatment of the incula at concentration of 1:50 and 1:100. The optimum was at for nematicidial effect as high as 80-100%. The at the rate of 1:100 prepared incula were incorporated in auto-claved soil, where 100 Juveniles M. hapla were introduced per 80% soil. All fungi infected the M. hapla effectively in soil, caysing more than 90% mortality within one week. This result indicated the potential value of these fungi as promising biocontrol agents

        Generation and Biological Characterization of a Neutralization-Resistant Mutant of Newcastle Disease Virus

        A neutralization-resistant mutant of Newcastle disease virus (NDV) Kr005 strain belonging to class II genotype VII was generated using a neutralizing monoclonal antibody and its biological effects were assessed. The mutant showed single amino acid substitution (E to K) at position 347 of the hemagglutinin-neuraminidase (HN) protein (E347K mutant). The E347K mutant exhibited marked rounding of the cells and few syncytia in infected chicken embryofibroblast (CEF) cells. The hemadsorption and neuraminidase activities of the E347K mutant of the wild-type virus were 118% and 166%, respectively. The mutant produced a rapid elution pattern whereas the wild type had a slow elution pattern. Growth kinetics studies showed that the E347K mutant produced an 80-times higher yield of extracellular virus in CEF cells compared with the wild-type virus. The time-course virus titer showed a marked increase in mutant-infected cells from 6 h to 12 h post infection (pi), which was consistent with the titer pattern time-course for NA activity. The E347K mutant virus showed a slight decrease in virulence compared to the wild-type virus, but there was no change in pathotype when measured by in vivo pathogenicity testing. These results suggest that an E347K mutation in HN protein might be associated with increased NA activity and subsequent enhancement of virus release from infected cells without change in viral pathotype.
