      • Induction of heme oxygenase-1 protects against podocyte apoptosis under diabetic conditions

        Heme oxygenase-1 (HO-1) is an anti-oxidant enzyme normally upregulated in response to oxidant injury. Here we determined the role of HO-1 in podocyte apoptosis in glomeruli of streptozotocin-treated rats and in immortalized mouse podocytes cultured in media containing normal or high glucose. HO-1 expression, its activity, the ratio of Bax/Bcl-2 protein, and active caspase-3 fragments were all significantly higher in isolated glomeruli of diabetic rats and in high glucose–treated podocytes. These increases were inhibited by zinc protoporphyrin treatment of the rats or by HO-1 siRNA treatment of the podocytes in culture. The number of apoptotic cells was also significantly increased in the glomeruli of diabetic rats and in high glucose–treated podocytes. Inhibition of HO-1 accentuated the increase in apoptotic cells both in vivo and in vitro. Our findings suggest that HO-1 expression protects against podocyte apoptosis under diabetic conditions.

        측두하악관절 골관절염 환자의 진단에서 Cone Beam 전산화 단층촬영의 유용성

        This study is designed to assess Relationship between clinical diagnosis of Temporomandibular Joint Disorder and diagnostic finding of Cone Beam Computed Tomography(CBCT) The author performed clinical examination for TMD Patients who visited Orofacial pain clinic, Jin-ju ooo Dental office. CBCT(Cone beam computed tomography) was taken for 190 joints in 95 subjects. A Oral medicine and Oral radiologist evaluated CBCT each other. then we compared with that result, Condyle bony changes were classified by no bone change, flattening, erosion and osteophyte. The obtained results were as follow: 1. The Kappa index of the diagnosis between oral medicine and oral radiogist were high, the index of diagnosis by degenerative joint disease were more higher. 2. The Kappa index of panoramic view and CBCT was low, more condylar bone chages were observed by CBCT diagnosis 3. Condylar bone changes of the 54.2% of non-DJD group clinicaly was observed by CBCT diagnosis and no bone changes of the 15.3% of DJD group.was observed by CBCT 4. TMJ pain was associated with erosion of condyle bone change of TMJ. Crepitation and longest duration of TMD were associated with osteophytic bone change.

        Kin Cnang-Saeng's theory of self-discipline is characteristic of "Shindok". It is based on his hard self-discipline ; according to Kim Chang-Saeng's attitude man cannot act without considerations of self-discipline. Man should practice moral principles to " Tien-Li." The bases of such act are performed under complete recognition of "Li" through "Ching" and "Tsun-yang." And it is equivalent to Kim Cnang-Saeng's attitude toward the study of philosophy. Unlike scholars before him, he particularly regards "Hsiao-Hsueh" and "Chia-Li" as primary sources. He thinks that man can achieve moral act through internal self-discipline directly coming from pracitcal act. This self-discipline of Kim Chang-Saeng's may result from complete "Cnontong" (keeping orthodoxy).

        요추부 유합수술 후 가동인접분절의 후기변화

        The spinal fusion in a lumbar region may influence biomechanically in the remained mobile segment to take over the lost motions Therefore the stress concentration on the adjacent segments may accelerate the degenerative changes, and then various late changes could occur. The aim of this study was to evaluate the late changes occuring in the adjacent segments to lumbar fusions. A retrospective review of radiographs and medical records was undertaken for 67 consecutive patients who had undergone various fusions in the lumbar region for various pathologic conditions. Included in this study were the patients who had performed active daily livings without any significant pain or disability after fusion and followed for a minimum of five years(up to 18 years with a mean of 8.3 years). The results of this study were as follows : The instability, disc narrowing, spinal stenosis, vertebral slipping, or isthmic defect were found in the adjacent segments to fusions in the patients who were followed longer, in L4-5 segment and above segment of fusion, and in the adjacent segments where a degenerative changes existed before surgery and where the angular motion increased considerably in a few years after fusion. The symptoms of these patients were responded well with conservative theraphy and no patient needed any surgical theraphy during these follow-up periods.

      • Thermoplasma acidophilum의 20S Proteasome에 의한 ATPase-활성적 단백질분해에 관한 연구

        The eukaryotic 26S proteasome is an ATP/ubiquitin-dependent proteolytic complex consisting of the 20S core particle and 19S ATPase complex. However, because of its complexity and unstable properties, this study was carried out to present more simple and stable model for the ATP-activated proteolytic complex in prokaryotes which can take the place of the eukaryotic 26S proteasome. For this purpose, recombinant Thermoplasma 20S proteasome (T20S) and Methanococcus MS4, a sequence homolog of one ATPase subunit in the 19S ATPase complex, were successfully isolated from Escherichia coli (E. coli). The α and β subunits of T20S expressed in E. coli could assemble for themselves, and showed the peptide-hydrolyzing activity. Whereas both T20S and R20S (the 20S complex from rabbit skeletal muscle) had the highest peptidase activity against Suc-LLVY-AMC, a good substrate for chymotrypsin-like peptidase activity, the specific activity of T20S was slightly lower than that of R20S. In addition, several reagents such as KCI, SDS, and ovalbumin were shown to have different effects on the peptidase activities between T20S and R20S. When the ATPase activity of the purified MS4 were assayed , the Km for ATP was about 0.5 mM, and casein could stimulate the activity more than 2 fold without the change in Km. This result implicates that protein-activated ATPase may induce the conformational change of casein, and therefore suggests that MS4 ATPase may activate the proteolytic activity of the 20S proteasome via accelerating the recognition and translocation of the protein substrates.

        Effects of dietary supplementation of Pediococcus pentosaceus strains from kimchi in weaned piglet challenged with Escherichia coli and Salmonella enterica

        Escherichia coli (E. coli) and Salmonella enterica (SE) infections in pigs are major source associated with enteric disease such as post weaning diarrhea. The aim of this study was to investigate the effects of Pediococcus pentosaceus in weaned piglets challenged with pathogen bacteria. In Experiment.1 90 weaned piglets with initial body weights of 8.53 ± 0.34 kg were assigned to 15 treatments for 2 weeks. The experiments were conducted two trials in a 2 × 5 factorial arrangement of treatments consisting of two levels of challenge (challenge and non-challenge) with E. coli and SE, respectively and five levels of probiotics (Control, Lactobacillus plantarum [LA], Pediococcus pentosaceus SMFM2016-WK1 [38W], Pediococcus acidilactici K [PK], Lactobacillus reuteri PF30 [PF30]). In Experiment.2 a total of 30 weaned pigs (initial body weight of 9.84 ± 0.85 kg) were used in 4 weeks experiment. Pigs were allocated to 5 groups in a randomized complete way with 2 pens per group and 3 pigs per pen. Supplementation of LA and 38W improved (p < 0.05) growth performance, intestinal pathogen bacteria count, fecal noxious odor and diarrhea incidence. In conclusion, supplementation of 38W strains isolated from white kimchi can act as probiotics by inhibiting E. coli and SE.

        SV40 T 항원의 온도조건부 변이형 유전자가 포함된 Amphotropic Retrovirus 에 의한 사람 태아 간세포의 불멸화

        N/A Human cells are almost never spontaneously immortalized in vitro. We tried to immortalize human fetal hepatocytes (h-FH) and evaluate the differentiational status and its change. Methods : Hepatocytes were isolated from a liver fragment of 20 week old fetus and infected with amphotropic recombinant retrovirus containing a temperature- sensitive mutant of SV40 large T antigen and neomycin phosphotransferase gene. G418 resistant colonies were cloned and expanded. The cells which were able to divide more than 30 times were used to analyze various functions. Results : The immortalization rate was 3.3 x 10-8 and two cell lines (C11, D21) were established. C11-60, C11-80, D21-30 and D21-60 (suffix number means the cell division counts) were evaluated. D21-30 was thougt to be imcompletely immortalized because a considerable portion of cells died during culture. The morphology was similar to that of epithelial cells except for D21-30 which looked like fibroblast. The cells grew rapidly at 33oC but stopped growing at 39oC. T antigen and p53 was expressed at 33oC but disappeared at 39oC, which suggest that T antigen binds to p53. Chromosomal changes were so marked that it was impossible to discriminate exact number. Albumin was secreted as about 1/10 as that of h-FH, but alpha-fetoprotein secretion stopped after immortalization. Telomerase was activated in both cell lines except for the incompletely immortalized cells D21-30. Telomere was elongated in competely immortalized cell lines, but it was rather shortened in D21-30 compared to that of h-FH. Macroscopic colonies did not develop in soft agar assay. Conclusions : We successfully immortalized human fetal hepatocytes. Although the cells are not likely to have oncogenicity, the functions are not so good, possibly due to marked chromosomal changes which are thought to occur before telomerase is activated during immortalization step.

        신부식전 유치 법랑질표면의 기계적 처리가 레진접합력에 미치는 영향에 관한 연구

        The purpose of this study was to determine the effect of the grinding method on the enamel of primary tooth on the bond strength of composite resin. Methods of tooth preparation investigated were 1.unground enamel, exposure to 37% phosphoric acid for 60 sec. 2.unground enamel, exposure to 37% phosphoric acid for 120 sec. 3.ground enamel with a diamond bur, exposure to phosphoric acid for 60 sec. 4.ground enamel with a carbide bur, 37% phosphoric acid for 60 sec. 5.ground enamel with a green stone, 37%phosphoric acid for 60 sec. 6.ground enamel with a disk, 37% phosphoric acid for 60 sec. The shear bond strength of composite resin was tested a Instron and surface tomography was examined with a SEM. The results were as follows: 1.With grinding of outer enamel surface,etch-time to produce a favorable bonding surface in primary teeth was shortened. 2.In SEM study,unground surface often had a prismless layer obscuring the rod pattern.Ground surface showed more effective removal of prismless layer more uniformly etched prism patterns. 3.Grinding enhanced the bond strength the enamel surface and composite resin. 4.Grinding enamel surface with a coarse diamond bur resulted in the highest bond strength.There was no significanct difference in the bond strength between other grinding methods.
