      • Relationship Between PTEN and Livin Expression and Malignancy of Renal Cell Carcinomas

        Objectives: This study focused on PTEN and Livin expression and associations with malignancy in human renal clear cell carcinomas (RCCC). Methods: PTEN and Livin expression was assessed in 100 RCCC tissue samples, 50 paracarcinoma cases, and 20 normal renal tissue samples using the immunohistochemical Streptavidin proxidase (SP) method. The relationships between binding and corresponding biological characteristics, such as histological grade, lymph node metastases, and clinical stages were analyzed. Results: Positive PTEN expression in RCCC was significantly lower than in renal tissue adjacent to carcinoma tissue and normal renal tissue (P<0.01). Livin expression in the renal tissue adjacent to the carcinoma and normal renal tissues exhibited only low levels, whereas overall Livin expression in RCCC was statistically significant (P<0.01). In RCCC, PTEN expression rate gradually decreased with an increase in clinical stage, whereas that of Livin increased to statistically significant levels (P<0.01), PTEN and Livin levels being negatively correlated (r=-0.395, P<0.01). Conclusions: PTEN and Livin are important in RCCC development. The two factors combined are expected to provide indices for estimating RCCC malignancy and progression levels, as well as references for RCCC diagnosis and treatment.

        Preparation and characterization of hydrophilic polydopamine-coated Fe3O4/oxide graphene imprinted nanocomposites for removal of bisphenol A in waters

        Bisphenol A (BPA), a known endocrine disruptor, is of global concern because it poses serious threats to the ecological environment and human health. In this work, hydrophilic polydopamine-coated Fe3O4/oxide graphene (IPDA@MGO) magnetic imprinted nanocomposites were prepared by the self-polymerization of dopamine on the surface of Fe3O4/GO in Tris-HCl buffer using BPA as a template for selective adsorption of BPA in water. IPDA@MGO showed specific recognition toward BPA with a high imprinting factor of 3.2 compared with nonimprinted polymer. The capacity of IPDA@MGO toward BPA was 41.2mg/g and the adsorption reached equilibrium within 30 min. The adsorption agreed well with the Freundlich and pseudo-second order kinetic models. The good adsorption performance was attributed to the abundant binding sites and good dispersibility of IPDA@MGO nanocomposites derived from its excellent hydrophilicity. The nanocomposites could be removed rapidly by an external magnet and regenerated for repeated adsorption of BPA in water. The proposed method has potential applications for efficient removal of BPA in environmental waters.

        SIMULATION-BASED DESIGN METHOD FOR ROOM AIR CONDITIONER WITH SMALLER DIAMETER COPPER TUBES

        Promoting the use of smaller diameter tube in room air conditioner is bene¯cial to reduce copper consumption and refrigerant charge, but may cause reduction of air conditioner performance, so a design method is needed. This paper presents a simulation-based design method for air conditioner with smaller diameter tube. The new method combines heat exchanger simulator and knowledge-based evolution method optimizer for designing air conditioner heat exchanger with smaller diameter tube. The simulation-based design method is illustrated in detail by an air conditioner of replacing 7mm tube indoor unit heat exchanger and 9.52mm tube outdoor unit heat exchanger with 5mm tube. Case study shows that the cost of the designed air conditioner with 5mm copper tube is 17.3% lower than that of the original one while the performance deviation between these two air conditioners is less than 0.7%.

        High-Quality Coarse-to-Fine Fruit Detector for Harvesting Robot in Open Environment

        Fruit detection in orchards is one of the most crucial tasks for designing the visual system of an automated harvesting robot. It is the first and foremost tool employed for tasks such as sorting, grading, harvesting, disease control, and yield estimation, etc. Efficient visual systems are crucial for designing an automated robot. However, conventional fruit detection methods always a trade-off with accuracy, real-time response, and extensibility. Therefore, an improved method is proposed based on coarse-to-fine multitask cascaded convolutional networks (MTCNN) with three aspects to enable the practical application. First, the architecture of Fruit-MTCNN was improved to increase its power to discriminate between objects and their backgrounds. Then, with a few manual labels and operations, synthetic images and labels were generated to increase the diversity and the number of image samples. Further, through the online hard example mining (OHEM) strategy during training, the detector retrained hard examples. Finally, the improved detector was tested for its performance that proved superior in predicted accuracy and retaining good performances on portability with the low time cost. Based on performance, it was concluded that the detector could be applied practically in the actual orchard environment.

        Mest Attenuates CCl4-Induced Liver Fibrosis in Rats by Inhibiting the Wnt/β-Catenin Signaling Pathway

        Background/Aims: The Wnt/β-catenin signaling pathway has been reported to play an important role in liver fibrosis. This study was designed to investigate whether mesoderm-specific transcript homologue (Mest), a strong negative regulator of Wnt/β-catenin signaling, could inhibit liver fibrosis. Methods: pcDNA-Mest was transfected into hepatic stellate cells (HSCs) and rats. Rats were randomly divided into four groups: normal group (normal saline), treatment group (pcDNA- Mest+CCl4), control group (pcDNA-neo+CCl4), and model group (normal saline+CCl4). Changes in liver pathology were evaluated by hematoxylin and eosin and Masson’s trichrome staining. The levels of alanine transaminase, aspartate transaminase, lactic dehygrogenase, hyaluronic acid, and laminin in the serum and hydroxyproline in the liver were detected by biochemical examination and radioimmunoassay, respectively. The expression and distribution of β-catenin, α-smooth muscle actin (α-SMA), Smad3, and tissue inhibitor of metalloproteinase type I were determined, and the viability of the HSCs was tested. Results: Our data demonstrate that Mest alleviated CCl4-induced collagen deposition in liver tissue and improved the condition of the liver in rats. Mest also significantly reduced the expression and distribution of β-catenin, α-SMA and Smad3 both in vivo and in vitro, in addition to the viability of HSCs in vitro. Conclusions: We found that Mest attenuates liver fibrosis by repressing β-catenin expression, which provides a new therapeutic approach for treating liver fibrosis.
