An HPLC method was employed for the determination of ethambutol in human plasma. After addition of internal standard (IS, octylamine, 2 μg/mL) and alkalinization of the plasma with 5 M sodium hydroxide, the drug and IS were extracted into the mixture of chloroform and diethyl ether (40:60, v/v). Following a 15-min vortex-mixing and a 10-min centrifugation, the organic phase was spiked with 100 pL of phenylethylisocyanate (2000 μg/mL) for chemical derivatization, mixed for 5 min and evaporated to dryness under a stream of nitrogen. The residue was reconstituted with 100 μL of mobile phase and 20 pL was injected into Cl8 column with a mobile phase consisting of methanol:water (70:30, v/v). The samples were detected utilizing an ultraviolet detector at 200 nm. The method was specific and validated with a limit of 0.15 μg/mL. Infra- and inter-day precision and accuracy were acceptable for all quality control samples including the lower limit of quantification. The applicability of this method was demonstrated by analysis of human plasma after oral administration of a single 1200-mg dose to 20 healthy subjects. From the plasma ethambutol concentration vs. time curves, the mean AUC was 9.61 ± 1.64 μg hr/mL. and Cmax of 2.68 μg/mL reached 2.73 hr after administration. The mean biological half-life of ethambutol was 3.46 ± 1.21 hr. Based on the results, this simple and validated assay could readily be used in any pharmacokinetic studies using humans.

The purpose of this study was to gather and categorize the data of electrical energy sources for public office buildings in Taegu, South Korea. The fossil energy sources are not enough for all of us, as we know, to use. In a few days, it will be hard to utilize or find them neither under the earth nor the sea or around the world. New energy sources should be recommended and solar energy has been referred to substitute these limited sources. Prior to studying solar energy sources applied to public office buildings, necessary data should be systematically surveyed and lined up. This research ,therefore, will be a very important foundation for studying next thesis, solar energy project.

To study the distribution, contents and structure of inclusion in 17Cr stainless steel, the samples were obtained with elapes time and Fe-Si addition after melted down in the high frequence induction furnace. And the distribution of inclusions, inclusion contents and the structure of inclusions were studied by means of optical microscopy, Br-Methanol method and X-ray technique. From the above experiments, the following conclusions could be summerized. 1) The inclusions of holding samples after melted down were spheroidal shape. And with the increase of holding time, the inclusion contents increased from 0.073% to 0.125%. And the distribution of fine inclusions decreased and the coarse inclusions increased from 2.0-3.ou:76% to 2.0-3.0:40%. 2) The inclusions of Fe-Si treated sample were dispersed and granular type. And the inclusion contents were increased compared with the holding samples after melted down. And the inclusions were distributed to fine one. 3) The inclusions were composed of Fe3O4, Cr2O3 and amorphous compound of Ni,Mo, Mn etc.

Transglutaminase(TGase) catalyzes the Ca^2+ dependent acyl transfer reaction to from γ-glutamyl lysine cross-links between substrate proteins. we have examined the expression of TGase 1 and TGase 2 enzyme activity and protein expression in differentiation induced salivary gland cells(HSG), and human salivary adenocarcinoma cell lines(SGT)in reation to anchorage independent growth. The TGase1 and TGase 2 activity in SGT was higher than in HSG, and there is no significant difference between TGase 2 and TGase1 cells. HSG cells showed no increase in TGase 1 and TGase 2 upon reaching post confluence and in 1.8mM Ca^2+. Expression of the TGase 1 and TGase 2 was induced by high cell density or by high calcium in SGT. No induction of TGase 1 & 2 protein was observed in postconfiuent HSG cells, but there was notable increases in TGase 1 & 2 protein in post confiuent SGT cells. This pattern of TGase 1 & 2 protein expression correlates with the pattern of particulate activity. SGT cells showed more colony size and colony forming activity in soft agar rather than HSG cells. The ability of SGT cells, containing high levels of TGase 2 and TGase1,to form colonies in soft agar suggests a role for this enzyme in anchorage independence.

Multidrug resistance (MDR) of cancer cells is, at least in part, associated with the overexpression of P-gly-coprotein (P-gp). Many studies hve demonstrated that natural compounds obtained from fruits, vegetables, teas and medicinal plants may modulate P-gp activity. The objective of the present investigation was to examine the effect of seven natural compounds on the P-gp activity in human uterine sarcoma cell line, MES-SA/DX5. Daunomycin uptake was significantly increased by biochanin A and silymarin (p<0.0001) whereas it was reduced by morin (p<0.01). The efflux of daunomycin from the cells was significantly inhibited by biochanin A, morin, cephalotaxine, berberine (p<0.05) and silymarin (p<0.0001). Biochanin A, berberine and silymarin significantly decreased IC_(50) value of daunomycin (p<0.05) while morin increased it (p<0.05). These results suggest that some natural compounds such as biochanin A and silymarin may inhibit P-gp funciton and can by developed as MDR reversing agents to improve the efficacy of chemotherapeutic drugs when administered concomitantly.
2003년 국내 중증급성호흡기증후군 진료 현황 및 문제점 분석

목적 : 2002년 말 중국에서 SARS가 발생한 이후 국내에서도 2003년 10월까지 총 3명의 추정환자, 17명의 의심환자가 보고되었다. 향후 추가적인 SARS의 유행이 우려되는 상황에서, 그간의 SARS 환자 진료에 있어서의 실질적인 준비사항, 진료 현황 등에 대한 조사를 통해 문제점을 파악하여, 향후 더 나은 대비가 될 수 있도록 개선점을 제시하고자 하였다. 재료 및 방법 : SARS로 의심되는 환자를 진료 경험이 있는 병원의료진을 대상으로 2003년 10월에 설문조사를 실시하였다. 설문에는 SARS 환자 진료 시의 실질적인 조치, 진료 현황, 병실, 응급실 및 외래에서의 격리 시설과 준비사항, 보건당국의 관리와 지원에 관한 사항을 포함하였다. 결과 : 대상이 되는 22개 병원 중 17개(17/22, 77.2%) 병원이 설문에 응하였다. SARS 환자를 위한 격리실은 응급실, 외래, 일반병실 및 중환자실에서 각각 9개(9/17, 52.9%), 5개(5/17, 29.4%), 15개(15/16, 93.7%), 4개(4/16, 25%) 병원에서 음압처리가 되어있지 않은 일인실 혹은 다인실이 사용되었고, 1개(1/16, 6.3%) 병원에서만 일반병실에서 음압격리실이 운영되었다. 입원환자의 진찰 시 개인보호구의 착용은 거의 모든 의료기관에서 이루어졌다. 보건당국에서 SARS지정병원의 시설 등을 사전에 확인한 곳은 1곳(1/12, 8.3%)이였고, 14개 병원(14/15, 93.3%)에서는 보건당국에 의뢰한 검사결과를 통하 받지 못하였다. 결론 : 의료기관에서 SARS 환자용 격리실뿐만 아니라 기존의 격리실 설비 등이 미흡하였으며, 특히 중환자실 및 외래의 준비가 더욱 부족하였다. 보건당국의 의료기관에 대한 종합적인 지원이 부족하였고, 병원과의 원활한 연계가 잘 이루어지지 않았다. SARS 만이 아닌 격리를 필요로 하는 질환의 적절한 진료를 위해 향후 병원 시설의 정비와 정부차원에서의 보다 구체적이고 실질적인 대책마련이 필요하다. Background : There was an worldwide outbreak of the Severe Acute Respiratory Syndrome (SARS) originated from China in late 2002. During that period three cases of suspected SARS and 17 cases of probable SARS were reported in Korea. With the concerns about the reemergence of SARS-coV transmission, it is important to be prepared for any possibility. So, this study is aimed to analysis the past measures in managing SARS and propose the amendatory plans to improve the preparedness. Materials & Methods : Questionnaires were collected among clinicians with any experience in managing the probable or suspected SARS cases in Oct. 2003. 17 out of 22 hospitals responded to the questionnaire. The contents in the questionnaire were practical activities, personal equipments, response plans, isolation facilities in emergency centers, outpatient clinics, general wards and intensive care units, and relationship with the public health department. Results : The dedicated isolation rooms in emergency centers, outpatient clinics, general wards, and intensive care units were prepared in 9 (9/17, 52.9%), 5 (5/17, 29.4%), 15 (15/16, 93.7%), and 4 (4/16, 25.0%) hospitals, respectively. Except for one hospital that newly made negative pressure room for SARS, single or multi-bed rooms without airborne infection control were used in all the other hospitals. The personal precaution principles were kept quite well in general wards. Before the designation of SARS hospital by the public health department prior evalution to see if the hospital was suitable for managing SARS was conducted in only 1 (1/12, 8.3%) hospital. The results of laboratory diagnosis were reported back in 1 (1/15, 6.6%) hospital. Conclusions : The isolation facilities which can control airborne infection were almost deficient not only for SARS but also for other respiratory transmissible diseases. For the infection control of transmissible diseases including SARS, more investment is needed on medical facilities and comprehensive support from the public health department required.

<▼1><P>Supplemental Digital Content is available in the text.</P></▼1><▼2><P>[<SUP>11</SUP>C]Acetate, a radiotracer for PET imaging, is a promising radiopharmaceutical for overcoming the limitation of 2-deoxy-2-[<SUP>18</SUP>F]fluoro-<SMALL>D</SMALL>-glucose in a number of cancers. Here, the optimized automatic synthesis of [<SUP>11</SUP>C]acetate using an in-house-developed module under different conditions has been reported for routine production. [<SUP>11</SUP>C]CO<SUB>2</SUB> was produced in a 16.4 MeV PETtrace cyclotron, and methyl magnesium chloride was used for synthesis. For product purification, ion-exchange solid-phase extraction cartridges were used, connected in series. High-performance liquid chromatography and gas chromatography were used to measure radiochemical and chemical purity. The Limulus amebocyte lysate test and the fluid thioglycollate medium test were performed for quality control of [<SUP>11</SUP>C]acetate. The total reaction time of [<SUP>11</SUP>C]acetate was within 15 min, and the overall decay-corrected radiochemical yield was 84.33±8.85%. Radiochemical purity was greater than 98% when evaluated on an analytical high-performance liquid chromatography system. No endotoxins or anaerobic bacteria were seen on quality control checks. Optimized production of [<SUP>11</SUP>C]acetate was achieved by the in-house module. Radiochemical and biological properties of the [<SUP>11</SUP>C]acetate produced were appropriate for clinical PET study.</P></▼2>
Electric-field-induced spin disorder-to-order transition near a multiferroic triple phase point

<P>The emergence of a triple phase point in a two-dimensional parameter space (such as pressure and temperature) can offer unforeseen opportunities for the coupling of two seemingly independent order parameters. On the basis of this, we demonstrate the electric control of magnetic order by manipulating chemical pressure: lanthanum substitution in the antiferromagnetic ferroelectric BiFeO3. Our demonstration relies on the finding that a multiferroic triple phase point of a single spin-disordered phase and two spin-ordered phases emerges near room temperature in Bi0.9La0.1FeO3 ferroelectric thin films. By using spatially resolved X-ray absorption spectroscopy, we provide direct evidence that the electric poling of a particular region of the compound near the triple phase point results in an antiferromagnetic phase while adjacent unpoled regions remain magnetically disordered, opening a promising avenue for magnetoelectric applications at room temperature.</P>
