      • 소아에서 성장호르몬 결핍증 진단을 위한 혈중 IGF-1과 IGFBP-3의 절단값

        Background : It is essential to have reference criteria for the diagnosis of growth hormone deficiency in children. The objective of study was to decide the cut-off value for Insulin-like Growth Factor-Ⅰ(IGF-Ⅰ) and Insulin-like Growth Factor Binding Protein-3(IGFBP-3) for diagnosis of growth hormone deficiency. Methods : We studied the serum level of IGF-Ⅰ and IGFBP-3 in 231 subjects(131 boys and 100 girls) with idiopathic short stature(ISS) and 37 subjects(23 boys and 14 girls) with complete or partial growth hormone deficiency(GHD). IGF-Ⅰ were measured by radioimmunoassay(RIA) method and IGFBP-3 were measured by immnoradiometric assay(IRMA) method. We conducted chi-square test, Fisher's exact test, student t-test, receiver operating characteristic(ROC) curve and discriminant anaylsis using MedCalc and SPSS Program. Results : The ROC curves showed that the best IGF-Ⅰ and IGFBP-3 cut-off value. Cut-off value of serum IGF-Ⅰ concentration was 346ng/mL(sensitivity 86.49%, specificity 85.71%) and of serum IGFBP-3 concentration was 3196ng/mL(sensitivity 81.82%, specificity 81.66%). If discriminant value (Z=0.442+0.661×IGF-Ⅰ+0.469×IGFBP-3) was below -1.47, the subject is discriminated GHD respectively. Conclusion : Our study results support that the measurement of serum IGF-Ⅰ and IGFBP-3 concentration might be an essential supplementary tools in the diagnostic evaluation of children with GHD.

        원예치료를 통한 여성재소자들의 사회성 및 대인관계 변화 연구

        The purpose of this study was identify change for socialite and interpersonal relationship of female prisoner through horticultural therapy on 14 prisoners in ○○female prison located in Chung-Book. Measurement methods were used Scale of sociality, Scale of change for interpersonal relationship, Form for evaluation of horticultural therapy. The results were as follows; 1.After HTP, Sociality was increased on female prisoners. and there was statistically significant difference(p〈0.01). 2.According to low-ranking field of I-R, communication, sensitivity, openness were increased, satisfaction, confidence, a feeling of intimacy were decreased, understanding was the same after HTP. But the average was increased, and there was statistically significant difference(p〈0.01). 3.Form for evaluation of horticultural therapy showed a increase of participation, interest and assistance, verbal communication, interpersonal relationship. Through above result, horticultural therapy has positive effect far socialite and I-R on female prisoners.

      • 포도나무가지 추출물의 프리라디칼 소거 작용 및 염증 발현 매개인자 생성 억제 효과

        Free radical scavengers or quenching agents for reactive oxygen species(ROS) present in consumable fruits, vegetables, and beverages have received considerable attention as potential antioxidants, and thus uses for treatment of several human diseases. In this study, grapevine shoot extract(GSE) containing high concentration of resveratrol and viniferine was evaluated for antioxidant potential and inhibition of pro-inflam-matory medicator production. Utilizing 1, 1-diphenyl-2-picrylhydrazyl(DPPH) scavenging activity and xanthine oxidase(XOD) inhibition assay the GSE showed inhibitory effects of DPPH radical scavenging and XOD activity with the IC_50 values of 34.5 ad 155㎍/ml, respectively. In addition, GSE also exhibited the inhibition of prostaglandin E_2(PGE_2) and nitric oxide(NO) production in lipopolysaccharide(LPS)-induced mouse macrophage RAW264.7 cells with the IC_50 value of 6.4 and 14.5 ㎍/ml, respectively. This suggests that grapevine shoot extract has the potential activity as a natural antioxidant or antiinflammatory agent.

        디지털 및 일반 측방두부규격방사선사진에서 측정 방법에 따른 계측치의 비교

        Purpose : To compare cephalometric measurement between measuring methods in digital and conventional lateral cephalometric radiograph. Materials and Methods : Twenty digital and conventional lateral cephalometric radiographs were selected. In digital group, cephalometric measurements were performed manually using hardcopies and automatically using VCeph^(TM) program on the monitor. In conventional group, the same measurements were performed manually on conventional films, and for automatic measurement conventional films were digitized by scanner. All measurements were performed twice by 4 observers, and 24 cephalometric variables were calculated and the time spent for each measurement was recorded. The differences in measurements data and the time spent for each measurement were compared within each group. Intra-observer and inter-observer comparisons were performed. Results : In both groups, no statistically significant difference between manual and automatic measurements was observed and most of the variables didn’t show statistically significant differences between methods. The observer with less experience tended to show statistically significant differences of measurements between methods, and differences from other observers. The differences of measurements between methods in digital group were lesser than those of conventional group with statistical significance in 8 variables out of 24. With automatic method and in digital group, the spent time was shorter. Conclusion : With direct digital radiograph, automatic method using manually idenitified landmarks can be preferable in cephalometric analysis.

        Provisional implant의 임상적 적용례

        It has been reported that for implatns to become osseointegrated, they must heal in the absence of functional loads for 4 to 6 months. The provisional implants were designed to support provisional restorations and to allow for load-free osseointegration of definitive implants, while the was provided with immediate esthetic and function. In addition, the provisional implant protects graft site. Also the provisional implant provides anchorage for orthodontic treatment and reconstructs vertical dimension in edentulous condyle fracture to practice functional therapy. The provisional implant system is a sound and economical method to achieve patient's immediate masticatory and esthetic function. Three cases are presented to document and illustrate the clinical application.

        《傷寒論》ㆍ《金匱要略》에서 煩躁의 病理에 關한 文獻硏究

        煩躁가 실려있는《傷寒論》과《금궤요약》의 조문들을 대상으로 病理的인 연구를 하는 것은 煩躁 證候의 病理뿐만 아니라 外感·內傷 질병의 正邪와 發病의 연구를 위한 이론적 토대로서의 중요한 의의를 가지고 있다.《傷寒論》六經病 분류와《금궤요약》雜炳 분류, 病機別 분류에 따라 각 條文에서의 煩躁의 病理를 분석함과 아울러 正邪와 發病 機轉에 대해 考察하였다.《傷寒論》에서의 煩躁는 三陽病 경우 正邪相爭으로 인한 化熱의 병리기전을, 三陰病 경우 陰寒極盛에 의한 虛陽外越하는 병리기전을 반영하였고,《금궤요약》에서는《傷寒論》에서는 나오지 않는 濕邪에 의한 氣機阻滯로 인한 번조가 있었다. 三陽病경우는 단순한 陽熱過多로 인한 것과 病機가 熱化되면서 체내 수액대사 이상을 초래하거나 수액대사 이상에 의한 병리적 산물인 水飮, 痰飮과 熱이 互結되어 氣機를 阻滯하는 경우 및 腸內陰液을 燥竭시켜 大便不通되므로 생기는 熱獨上擾에 의한 煩躁ㆍ誤治로 인한 煩躁로 구분된다. 三陰病의 경우는 극심한 경우 陽機脫亡하는 危證경우와 陽氣回復으로 陰寒이 消退하여 病이 愈하는 徵兆로서의 煩躁로 구분된다. 또한《傷寒論》의 六經病에서의 煩躁는 사상체질의학적 측면에서 보면 陰寒極盛으로 인한 陽虛煩躁는 少陰人 病證에 해당되고, 襄熱에 의한 熱獨上擾한 煩躁는 少陽人 病證에 해당되며, 燥濕不調에 의한 燥熱한 煩躁는 太陰人 病證에 해당된다고 보여진다. In 《Shang han lun(傷寒論)》 and 《Chin K'uei Yao Lueh(금궤요약)》, in order to find out pathology of ‘figetiness(煩躁)’. First, clsssify figetiness into the six channels(六經) disease, and various disease, base on expression of stipulated text. Second, investigate and consider in the main process of pathology, the conclusion is runs as fallows; 1. In 《Shang han lun》, figetiness of disease of the three yang channels(三陽) reflects the process of pathogenicy fire resulted from fighting between the vital energy and the pathogenic facter. The inside, simple caused by excessive yang fever(陽熱), or process of pathology becoming feverish bring about abnormal water metabolism in the body, or pathologic products resulted from abnormal water metabolism; excessive fluid(水飮), phlegm retention(痰飮) and case of fever conjoin stop up functional activities of qi (氣機), and it drys up yin fluid in intestine(腸內陰液) causes stoppage of bowel movements. Therefor produse furbid stifling hot rise to confuse mentality case(熱獨上擾), besides, mistritment case are devided. 2. In 《Shang han lun》, figetiness of disease of the three yang channels(三陰) reflects upward floating of yang in deficiency(虛陽外越), caused by accumulation of severe pathogenic cold(陰寒極盛). In severe cases, it becomes dangerous case of exhaustion of vital energy, otherwise recover vital energy(陽氣), so go away pathogenic cold(陰寒) and get well's good omen of figetiness are devided. 3. As respects of Four Constitutional medicine, deficient yang of figetiness(陽虛煩燥) which is based on highly flourishing pathogenic cold(陰寒極盛) in the six channels(六經) disease of 《Shang han lun》 is applicable to the simptoms of Soum-in(少陰人), furbid stifling hot rise to confuse mentality case(熱獨上擾) based on heat of interior(襄熱) is applicable to the symptoms of Soyang-in(少陽人), and figetiness of dry heat(燥熱) based on unbalance between dryness and wet(燥濕不調) is applicable to the symptoms of Taeum-in 4. In 《Chin K'uei Yao Lueh》, there are cases of figetiness the duplicated 《Shang han lun》 texts; some are arthralgia (關節煩疼) causing by conjoint invasion of pathogenic wind-dampness(風濕相搏), some are figetiness of colic caused by ascaris(蛔厥), and the others not the figetiness acompanying exterior syndrome(表證) like in 《Shang han lun》 but the figetiness caused by damr pathogen(濕邪) in exterior syndrome, and the figetiness of the five viscera system appeared in pathologic process of various desease, women's postpartum of figetiness. 5. Figetiness in 《Shang han lun》·《Chin K'uel Yao lueh》 can be sorted out exterior and interior(表襄)·cold and heat(寒熱)·deficiency and excess(虛實), especially some cases are the signs of recovery of vital-qi(正氣恢復), some are the cases connected with abnormal water metabolism, besides the cases related to damr pathogen are devided apart. By this, we can know that grasp the pathologic process of figetiness has the diagnosis.

      • 忍冬덩굴의 揷木繁殖에 관한 硏究

        The honeysuckles constitute a sturdy group of semievergreen vines, some which can be grown in everywhere of the native mountains. As a group, they are not susceptible to serve disease troubles, which are easily grown and will usually produce a wealth of foliage, flower and fruit with very little care. They are perenial and deciduous climbing plants with strong vines, and flowered from June to September that are considerd as a good ornamental plants for Landscaping. Plants of honeysuckle are ordinarily able to be rooted from softwood or hardwood cuttings, the cutting itself being about 4-5cm long. This study was carried out to determine the effectiveness. of various rate of IBA and NAA treatment on rooting of honeysuckles (Lonicera japonica, Lonicera japonica var. aureoreticulata, Lonicera sempervirens) in the mist room and closed moist chamber. And the results obtained were as follows. Treatment of IBA and NAA increase! in the rooting percentage, rootlet number and length of root on the rooting of honeysuckle cuttings. The rooting percentage of honeysuckle cuttings with IBA 2000pprn 10 sec dip are higher than other rooting hormone treatment. IBA showed more promoting .effect than NAA treatment on the rooting of leafy softwood cuttings. Rooting percentage and number of root tended to have the hightest root system in the closed moist chamber than the mist room.

        효모시스템에서 Human Transglutaminase C(TGase 2)의 발현에 관한 연구 : C-말단부위의 결손효과 An Effect of a Domain from Carboxyl Terminal Deletion of the Enzyme

        Transglutaminase(TGase Ⅱ)는 펩타이드-결합 글루타민 잔기와 펩타이드-결합 라이신잔기 및 1차 아민류를 포함하는 다양한 화합물과의 교차결합에 의하여 단백질의 변형을 촉매하는 효소이다. 동물세포에는 구별가능한 5종류의 TGase가 존재한다[TGase Ⅰ(또는 K), Ⅱ(C), Ⅲ(E), Ⅳ(P)와 혈장단백 ⅩⅢa]. 그들은 아미노산 서열에서 N-말단에 약 70%의 상동성을 갖고 있으며, C-말단의 경우에는 거의 상동성이 부위(N-말단 2/3)와 비상동성 부위(C-말단 1/3 부위)의 역할을 알아보기 위해 전체효소 또는 C-말단이 결손된 형태의 효소를 Saccharmoyces cerevisiae에서 발현 하고자 하였다. Southern blot, 역전사 중합 연쇄 반응, Western blot등을 통해 전체효소 및 C-말단 결손형 효소가 형질전환된 효모에서 발현되는 것을 확인하였다. Succinylated casein에 [1,4-^14C]-putrescine이 통합되는 반응을 이용한 활성 측정에서 C-말단이 결손된 TGase Ⅱ는 같은 세포 농도에서 전체 TGase Ⅱ 효소와 비교하여 유사한 수준의 활성을 나타내었다. 이러한 결과들은 사람 TGase Ⅱ의 경우 N-말단의 높은 상동성부위가 효소의 역가결정에 중요하다는 것을 나타내 준다. 효모시스템에서 발현한 결과 효소의 역가가 나타났으나 높지 않았으며 발현된 양이 극히 낮아서 단백질의 정제나 특성연구를 위하여 발현벡터의 개량을 포함한 방법의 개선이 필요하다고 사료된다. 흥미롭게도 TGase Ⅱ의 발현이 효모의 성장을 저해하는 것으로 나타나 이러한 현상이 표피세포의 각질화에 수반된 세포의 죽음과 유관한지의 가능성에 대한 보다 자세한 연구가 필요하다. In an effort to understand the role of the conserved domain and of the heterologous one-third part of the carboxy terminal domain of transglutaminase C (TGase Ⅱ), attempts were made to express TGase Ⅱ cDNA of human origin in yeast Saccharmoyces cerevisiae as in a full-length form as well as in a form of C-terminal truncation. The 2μ-based expression plasmids which contained the TGase Ⅱ cDNA under the gal inducible promoter were introduced into yeast and the maintenance of the full-length and truncated form of the TGase Ⅱ gene plasmids were confirmed by Southern blot. The expression of e results indicate that the N-terminal homologous domain of human TGase Ⅱ retains a catalvtically active domain. The level of TGase Ⅱ expressed in yeast, however, was far lower than satisfactory and other expression system should be sought further characterization of the enzyme. The negative effect of TGase Ⅱ on the growth of yeast is interesting with respect to the physiological effect of TGase Ⅱ in the cornification f epidermal keratinocytes.
