        Use of Antimicrobial Food Additives as Potential Dipping Solutions to Control Pseudomonas spp. Contamination in the Frankfurters and Ham

        This study evaluated the effect of sodium diacetate and sodium lactate solutions for reducing the cell count of Pseudomo-nas spp. in frankfurters and hams. A mixture of Pseudomonas aeruginosa (NCCP10338, NCCP10250, and NCCP11229),and Pseudomonas fluorescens (KACC10323 and KACC10326) was inoculated on cooked frankfurters and ham. The inoc-ulated samples were immersed into control (sterile distilled water), sodium diacetate (5 and 10%), sodium lactate (5 and10%), 5% sodium diacetate + 5% sodium lactate, and 10% sodium diacetate + 10% sodium lactate for 0-10 min. Inoculatedfrankfurters and ham were also immersed into acidified (pH 3.0) solutions such as acidified sodium diacetate (5 and 10%),and acidified sodium lactate (5 and 10%) in addition to control (acidified distilled water) for 0-10 min. Total aerobic platecounts for Pseudomonas spp. were enumerated on Cetrimide agar. Significant reductions (ca. 2 Log CFU/g) in Pseudomo-nas spp. cells on frankfurters and ham were observed only for a combination treatment of 10% sodium lactate + 10% sodiumdiacetate. When the solutions were acidified to pH 3.0, the total reductions of Pseudomonas spp. were 1.5-4.0 Log CFU/g. The order of reduction amounts of Pseudomonas spp. cell counts was 10% sodium lactate > 5% sodium lactate ≥ 10%sodium diacetate > 5% sodium diacetate > control for frankfurters, and 10% sodium lactate > 5% sodium lactate > 10%sodium diacetate > 5% sodium diacetate > control for ham. The results suggest that using acidified food additive antimicro-bials, as dipping solutions, should be useful in reducing Pseudomonas spp. on frankfurters and ham.

        Expression and Characterization of a Novel 2-Deoxyribose-5-phosphate Aldolase from Haemophilus influenzae Rd KW20

        A codon-optimized 2-deoxyribose-5-phosphate aldolase (DERA) gene from Haemophilus influenzae Rd KW20 was synthesized and expressed in Escherichia coli, and the biochemical properties of its product were investigated. DERA was purified using affinity chromatography and characterized using 2-deoxyribose-5-phosphate as the substrate. Specific activity of the recombinant DERA was $34.1Umg^{-1}$. The pH and temperature optima were 7.5 and $40^{\circ}C$, respectively. Additionally, the recombinant enzyme retained stability up to temperatures below $50^{\circ}C$. Maximal enzyme activity was observed in presence of 300 mM of acetaldehyde. The apparent $K_m$ and $V_{max}$ of purified enzyme towards 2-deoxyribose-5-phosphate were 0.14 mM and $70.42{\mu}mol\;min^{-1}mg^{-1}$ and towards 2-deoxy-D-ribose were 24.77 mM and $1.94{\mu}mol\;min^{-1}mg^{-1}$, respectively. For synthesis of statin intermediates, the bioconversion process for production of (3R, 5S)-6-chloro-2,4,6-trideoxy-erythro-hexose from chloroacetaldehyde and acetaldehyde using the recombinant DERA was studied and this process took 3 h for maximal conversion. This recombinant DERA could be potentially applied in the production of (3R, 5S)-6-chloro-2,4,6-trideoxy-erythro-hexose.

      • High-performance Liquid Chromatographic Analysis for Quantitation of Marker Compounds of Artemisia capillaris Thunb

        Two stable high-performance liquid chromatography (HPLC) methods were developed that could quantitatively analyze 10 major marker compounds of Artemisia capillaris Thunb and could also distinguish among `Injinho` and `Myeon-injin` and `Haninjin`--A. capillaris collected in autumn, A. capillaris collected in spring and A. iwayomogi, which can be misused as `Injinho` in Korean herbal drug markets. The first HPLC method was a reversed-phase chromatography using a C18 column with an isocratic solvent system of phosphoric acid (0.05%) and acetonitrile at the flow rate of 1.0 mL/min, ultraviolet (UV) detection wavelength at 254 nm and column temperature at 40°C. Calibration andquantitation were made by using acetaminophen as an internal standard (I.S-A) and chlorogenic acid (1) was determined within 20 min. The second HPLC method was a reversed-phase chromatography using a C18 column with a gradient solvent system of phosphate buffer (0.015 M, pH 6) and acetonitrile at the flow rate of 1.0 mL/min, UV detection wavelength at 254 nm and column temperature at 40°C. Calibration and quantitation were made by using ethylparaben as an internal standard (I.S-B) and 3,5-di-O-caffeoylquinic acid (2), 3,4-di-O-caffeoylquinic acid (3), 4,5-di-O-caffeoylquinic acid (4), hyperoside (5), isoquercitrin (6), isorhamnetin 3-O-robinobioside (7), isorhamnetin-3-O-galactoside (8), isorhamnetin-3-O-glucoside (9) and scoparone (10) were determined within 60 min. Pattern recognitionanalysis of data from the 60 samples classified them clearly into three groups. These assay methods could be applied for QA/QC of A. capillaris and Artemisia iwayomogi.

        위절제술 환자의 표준진료지침 개발 및 적용 효과

        Background : Gastric cancer is the most common malignant tumor in Korea. surgical operation is one of the major treatment modalities for gastric cancer patients. Therefore, gastrectomy is one of the most common procedures in General Surgery. There were variation in length of hospital stay and medical treatment for gastrectomy between three surgeons at Asan Medical Center. Clinical pathways have received considerable attention as a tool for recucing the medical practice variation, increasing the efficiency of care process, and improving the quality of care. The aim of this study was to evaluate the effect of a clinical pathway for gastrectomy in gastric cancer patients. Methods : The clinical pathway for gastrectomy was developed and implemented by a multidisciplinary group in Asan Medical Center. A computerized clinical pathway program was developed and revised after a pilot test. A total of 145 patients underwent gastrectomy by three surgeons at Asan Medical Center. We compared the length of hospital stay, patient satisfaction, and unplanned readmission rate between the pre-pathway group(n=67) and the post-pathway group(n=78). We also investigated the degree of satisfaction among the physicians and nurses who were main end-users of the clinical pathway. Results : The clinical pathway was applied to all target patients. The average length of hospital stay was shortened from 12.7days to 10.6days(p<0.01). The degree of patient satisfaction with the care process changed from 90.3% to 89.2% after the implementation of the clinical pathway, but the difference was of satistically significant(p=0.761). Unplanned readmission rate was 2.9% in the pre-pathway group. More than 90% of physicians and nurses answered that the clinical pathway had been a useful tool in their medical practice. Conclusions : The findings of the study demonstrated that implementation of the clinical pathway for gastrectomy produced substantial reduction in the length of hospital stay while improving the quality of patient outcomes. The computerized clinical pathway program can be used as one of the powerful patient management tools for reducing the practice variations and increasing the efficiency of care process in Korea hospital settings.

      • Improving the subcritical water extraction of flavonoids narirutin and hesperidin from Citrus unshiu peel by pulsed electric fields

        Citrus fruit is important source of flavonoids, mainly flavanones which are narirutin and hesperidin. Those citrus flavonoids have been found to have health-related properties including antioxidant, anticancer, and anti-inflammatory. The main purpose of this study was to verify that the extraction of narirutin and hesperidin from Citrus peel can be more effective by combining pulsed electric field (PEF) pre-treatment and subcritical water extraction (SWE). Citrus unshiu peels were treated with PEF under conditions of electric field strength (3 kV/cm) and times (1 and 2 min). Subsequent SWE was conducted by using a Dionex Accelerated Solvent Extractor (ASE, Model 350) at extraction temperature 170°C for 10 min. The total flavonoids content was measured by using the aluminum chloride colorimetric method and the antioxidant capacity was analyzed by the Ferric reducing antioxidant power (FRAP) assay using spectrophotometer. The concentrations of narirutin and hesperidin were increased as PEF pre-treatment time increased. The highest concentrations of narirutin and hesperidin were 13.41 mg narirutin/g dry citrus peel and 141.16 mg hesperidin/g dry citrus peel at PEF pre-treatment condition of 3 kV/cm and 2 min. The total flavonoids contents of the extracts increased 105.2% and 123.1% for citrus peel PEF treated at 1 and 2 min, respectively. In addition, compared to the untreated sample, PEF pre-treatments of 1 and 2 min increased the antioxidant capacity of the extracts 109.2% and 160.8%, respectively. Therefore, the results demonstrate the potential of PEF pre-treatment to improve the SWE of flavonoids from citrus unshiu peel.

        단신 : 실리콘의 염소화반응에 의한 사염화규소 제조

        직경 25 mm의 파이렉스 튜브 내에서 실리콘의 유동층 염소화 반응이 수행되었다. 반응기에 공급되는 질소 유량0.8~1.0 L/min, 염소 유량 0.2 L/min, 반응온도 450℃, SiCl4 응축기의 냉매온도는 -5℃로 설정하였다. 반응기에 도입되는 가스 내 염소의 몰분율이 증가하면 SiCl4의 수율이 증가하였다. 반응가스 중 염소의 몰분율 0.2의 조건에서 SiCl4의 수율은 28% 이었다. 염소의 몰분율 증가는 반응열 상승에 의해 반응온도 상승을 가져옴으로써 안전을 고려하여 염소의 몰분율을 0.2 이상으로 올리지 못했다. 실리콘의 유동층 염소화 반응에 의한 사염화실리콘의 제조 가능성이 입증되 었으며, 향후 보다 가혹한 조건에서의 실용화 연구를 위한 기초로 활용될 수 있을 것으로 기대된다. The chlorination of a metallurgical-grade silicon was carried out in a fluidized bed reactor, 25 mm in diameter. The flow rate of the chlorine admitted into the reactor was 0.2 L/min and that of the carrier nitrogen was 0.8~1.0 L/ min. The reactor temperature was maintained at 450℃ and the temperature of the coolant at the SiCl4 condenser was at -5℃. The SiCl4 yield increased with increasing the mole fraction of chlorine in the feed gas, exhibiting 28% at the mole fraction of 0.2. Further increase of the chlorine mole fraction was not attempted in a worry that the reactor might be failed due to the high exothermicity of the reaction. The production of SiCl4 from silicon by fluidized bed chlorination was demonstrated on a laboratory scale, which is a stepping stone for future studies under more severe conditions toward industrial application.

        Lactobacillus pentosus 발효에 의한 담쟁이덩굴 줄기 추출물의 항산화 및 세포보호 효과

        본 연구에서는 담쟁이덩굴 줄기 70% 에탄올 추출물과 발효균주 Lactobacillus pentosus를 이용하여 발효시킨 담쟁이덩굴 줄기 발효추출물에 대하여 항산화 및 세포보호 효과를 측정하였다. 1,1-Diphenyl-2-picrylhydrazyl (DPPH)를 이용한 자유라디칼 소거 활성(FSC<sub>50</sub>)은 담쟁이덩굴 줄기 추출물 및 발효추출물이 각각 42.3 및 34.5 μ g/mL로 발효 후의 라디칼 소거활성이 약 18.4% 더 높게 나타났다. Lumiol-의존성 화학발광법을 이용한 Fe<sup>3+</sup>-EDTA/H<sub>2</sub>O<sub>2</sub>계에서의 총 항산화능(OSC<sub>50</sub>) 평가에서도 담쟁이덩굴 줄기 추출물과 발효추출물은 각각 2.6 및 2.5 μ g/mL로 발효 후가 약 4.2% 정도 더 높은 총 항산화능을 나타냈다. <sup>1</sup>O<sub>2</sub>로 유도된 적혈구 세포손상에 있어서 추출물 및 발효추출물의 세포 보호 효과(τ<sub>50</sub>)는 50 μ g/mL에서 각각 126.4 및 173.0 min을 나타내어 발효 후 세포 보호 효과가 약 34.0% 더 높게 나타났다. 발효추출물은 지용성 항산화제로 알려진 (+)-α-tocopherol (43.4 min)보다도 3.9배 높은 세포 보호 활성을 보여주었다. 사람 섬유아세포인 Hs68을 대상으로 elastase 저해 활성을 조사하였다. Elastase 저해 활성(IC<sub>50</sub>)은 담쟁이덩굴 줄기 추출물과 발효추출물에서 각각 873.6 및 687.8 μ g/mL로 발효 후에 elastase 저해 활성이 약 21.3% 더 높은 것으로 나타났다. 이상의 결과들은 담쟁이덩굴 줄기 발효추출물이 항산화 작용과 더불어 주름개선 효과를 가지는 천연 화장품 소재로써 응용 가능성이 있음을 시사한다. In this study, the antioxidant activities, cellular protective effects, and inhibitory effects on elastase of non-fermented and fermented extracts of Parthenocissus tricuspidata (P. tricuspidata) stem using Lactobacillus pentosus were investigated. The free radical scavenging activities (FSC<sub>50</sub>) of non-fermented and fermented extracts were 42.3 and 34.5 μg/mL, respectively, in which the activity after fermentation was approximately 18.4% higher. Reactive oxygen species (ROS) scavenging activities (OSC<sub>50</sub>) in Fe<sup>3+</sup>-EDTA/H<sub>2</sub>O<sub>2</sub> system of non-fermented and fermented extracts were 2.6 and 2.5 μg/mL, respectively. The activity after fermentation was approximately 4.2% higher. In the <sup>1</sup>O<sub>2</sub>-induced cellular damage of erythrocytes, the cellular protective effects (τ<sub>50</sub>) of non-fermented and fermented extracts were 126.4 and 173.0 min at 50 μg/mL, respectively. The activity after fermentation was approximately 34.0% higher. The effect of fermented extract was 3.9 times higher than (+)-α-tocopherol (τ<sub>50</sub> = 43.4 min), known as a lipophilic antioxidant at 50 μg/mL. The inhibitory effect of elastase was investigated to predict the anti-wrinkle efficacy using Hs68 human fibroblasts cells. The elastase inhibitory activities (IC<sub>50</sub>) of non-fermented and fermented extracts were 873.6 and 687.8 μg/mL, respectively, and the activity after fermentation was approximately 21.3% higher. These results indicated that fermented extract of P. tricuspidata stem has potentials as natural cosmetic ingredients with antioxidant and anti-wrinkle effect.

      • 마늘 잼의 저장과정 중 성상변화

        In order to make a functional jam containing garlic, sugar, acid and pectin were mixed with garlic and concentrated by heating the mixture under atmosphere or vacuum conditions. And changes of some physico-chemical properties of the garlic jam(i.e., color, textural properties, vitamin C) were investigated during storage at 30℃. It was found that lightness(L) was higher in sample of vacuum concentration than that in atmosphere concentration and redness(a) and yellowness(b) were higher in sample of atmosphere concentration. During the storage lightness was found to decrease for all samples with more drastic decrease in sample of atmosphere concentration where browning was more severely observed after 8 weeks of storage. In terms of textural properties, hardness, chewiness and gumminess were higher in sample of atmosphere concentration than that in vacuum concentration. And springiness and cohesiveness were higher in sample of vacuum concentration. Although sensory score for garlic jam found to be lower than that for the existing strawberry jam, the present garlic jam might have high marketability if considering its physiological value. 마늘을 이용한 기능성 잼을 제조하기 위하여 마늘에 당, 산, 펙틴을 첨가하고 평압과 감압에서 농축하여 마늘잼을 제조한 후 색도, 조직특성을 조사하고 30℃에서 저장하면서 색도, 조직의 변화, 비타민 C의 변화를 측정한 결과는 다음과 같다. 원료마늘의 일반성분은 수분 57.2%, 탄수화물 23.6%, 조단백 1.9%, 조지방 0.75%, 회분0.93%이었고 비타민 C는 8.4 mg/l00g이었다. 명도는 진공농축한 것이 평압농축한 것보다 높았고 적색도와 황색도는 평압의 것이 높았다. 저장기간에 따라 명도는 모두 감소하였으나 평압에서 농축한 것이 감압보다 크게 감소하였으며 갈변화가 저장 8주에는 심하게 나타났다. 마늘잼의 조직특성은 hardness, chewiness, gumminess는 평압농축이 감압농축에 비하여 높았고 springiness, cohesiveness는 평압농축이 낮았다. 마늘잼의 기호성은 딸기잼에 비하여 떨어지지만 기능성을 감안할 때 시장성이 매우 높을 것으로 판단한다.
