        Cloning and Characterization of a Na+/H+ Antiporter Gene of the Moderately Halophilic Bacterium Halobacillus aidingensis AD-6T

        A gene encoding a Na+/H+ antiporter was obtained from the genome of Halobacillus aidingensis AD-6T, which was sequenced and designated as nhaH. The deduced amino acid sequence of the gene was 91% identical to the NhaH of H. dabanensis, and shared 54% identity with the NhaG of Bacillus subtilis. The cloned gene enable the Escherichia coli KNabc cell, which lack all of the major Na+/H+ antiporters, to grow in medium containing 0.2 M NaCl or 10 mM LiCl. The nhaH gene was predicted to encode a 43.5 kDa protein (403 amino acid residues) with 11 putative transmembrane regions. Everted membrane vesicles prepared from E. coli KNabc cells carrying NhaH exhibited Na+/H+ as well as Li+/H+ antiporter activity, which was pH-dependent with the highest activity at pH 8.0, and no K+ /H+ antiporter activity was detected. The deletion of hydrophilic C-terminal amino acid residues showed that the short C-terminal tail was vital for Na+/H+ antiporter activity.

        Antimony Selenide Nanorods Decorated on Reduced Graphene Oxide with Excellent Electrochemical Properties for Li-Ion Batteries

        <P>A promising anode material for lithium-ion batteries (LIBs) consisting of Sb2Se3 nanorods and reduced graphene oxide (rGO) sheets has been prepared by an effective solvothermal approach. The synergetic effect between Sb2Se3 nanorods and rGO matrix provides not only high conductivity paths and strong electron contact interface, but also alleviates the volume change of Sb2Se3 nanorods, resulting in excellent lithium-storage performance. When tested as an anode material for LIBs, a high capacity of 868.30 mAh g(-1) can be retained after 100 cycles at 200 mA g(-1). Even at 2000 mA g(-1), a satisfactory capacity of 430.40 mAh g(-1) after long 550 cycles can be delivered. Ex situ X-ray diffraction study suggests that the Sb2Se3/rGO composite follows the combined Li+ intercalation, conversion reaction and alloying reaction mechanism.       • Well-designed Te/SnS<sub>2</sub>/Ag artificial nanoleaves for enabling and enhancing visible-light driven overall splitting of pure water

        <P><B>Abstract</B></P> <P>To produce hydrogen and oxygen from photocatalytic overall splitting of pure water provides a promising green route to directly convert solar energy to clean fuel. However, the design and fabrication of high-efficiency photocatalyst is challenging. Here we present that by connecting different nanostructures together in a rational fashion, components that cannot individually split water into H<SUB>2</SUB> and O<SUB>2</SUB> can work together as efficient photocatalyst with high solar-to-hydrogen (STH) energy conversion efficiency and avoid the use of any sacrificial reagent. Specifically, Te/SnS<SUB>2</SUB>/Ag artificial nanoleaves (ANLs) consist of ultrathin SnS<SUB>2</SUB> nanoplates grown on Te nanowires and decorated with numerous Ag nanoparticles. The appropriate band structure of Te/SnS<SUB>2</SUB> p-n junctions and the surface plasmon resonance of Ag nanoparticles synergistically enhance the quantum yield and separation efficiency of electron-hole pairs. As a result, Te/SnS<SUB>2</SUB>/Ag ANLs enable visible-light driven overall water-splitting without any sacrificial reagent and exhibit high H<SUB>2</SUB> and O<SUB>2</SUB> production rates of 332.4 and 166.2μmolh<SUP>−1</SUP>, respectively. Well-preserved structure after long-term measurement indicates its high stability.       • Neutrophil Count and the Inflammation-based Glasgow Prognostic Score Predict Survival in Patients with Advanced Gastric Cancer Receiving First-line Chemotherapy

        Purpose: To explore the value of systemic inflammatory markers as independent prognostic factors and the extent these markers improve prognostic classification for patients with inoperable advanced or metastatic gastric cancer (GC) receiving palliative chemotherapy. Methods: We studied the prognostic value of systemic inflammatory factors such as circulating white blood cell count and its components as well as that combined to form inflammation-based prognostic scores (Glasgow Prognostic Score (GPS), Neutrophil-Lymphocyte Ratio (NLR), Platelet Lymphocyte Ratio (PLR), Prognostic Index (PI) and Prognostic Nutritional Index (PNI)) in 384 patients with inoperable advanced or metastatic gastric cancer (GC) receiving first-line chemotherapy. Univariate and multivariate analyses were performed to examine the impact of inflammatory markers on overall survival (OS). Results: Univariate analysis revealed that an elevated white blood cell, neutrophil and/or platelet count, a decreased lymphocyte count, a low serum albumin concentration, and high CRP concentration, as well as elevated NLR/PLR, GPS, PI, PNI were significant predictors of shorter OS. Multivariate analysis demonstrated that only elevated neutrophil count (HR 3.696, p=0.003) and higher GPS (HR 1.621, p=0.01) were independent predictors of poor OS. Conclusion: This study demonstrated elevated pretreatment neutrophil count and high GPS to be independent predictors of shorter OS in inoperable advanced or metastatic GC patients treated with first-line chemotherapy. Upon validation of these data in independent studies, stratification of patients using these markers in future clinical trials is recommended.

        Selection of Reference Genes for Real-time Quantitative PCR Normalization in the Process of Gaeumannomyces graminis var. tritici Infecting Wheat

        Gaeumannomyces graminis var. tritici is a soil borne pathogenic fungus associated with wheat roots. The accurate quantification of gene expression during the process of infection might be helpful to understand the pathogenic molecular mechanism. However, this method requires suitable reference genes for transcript normalization. In this study, nine candidate reference genes were chosen, and the specificity of the primers were investigated by melting curves of PCR products. The expression stability of these nine candidates was determined with three programs-geNorm, Norm Finder, and Best Keeper. TUBβ was identified as the most stable reference gene. Furthermore, the exopolygalacturonase gene (ExoPG) was selected to verify the reliability of TUBβ expression. The expression profile of ExoPG assessed using TUBβ agreed with the results of digital gene expression analysis by RNA-Seq. This study is the first systematic exploration of the optimal reference genes in the infection process of Gaeumannomyces graminis var. tritici.

        Single‑wire power transfer method and verification

        As a novel power supply method, single-wire power transfer has gained a great deal of attention. However, most of the related studies were based on using loops to realize a two-way power transfer. To further develop single-wire power transfer technology, a single-wire power transfer method is proposed in this paper, which does not rely on loops for power transfer. Through theoretical derivation, its transfer principle is analyzed, and a single-wire power transfer structure is designed. In addition, the theoretical derivation is verified by simulation analysis. Finally, the factors and laws that affect transfer efficiency are obtained through simulations and experiments. The research conducted in this paper is conducive to the development of single-wire power transfer methods, and lays a foundation for further research on transfer characteristics.

        Associations of Genetic Variations in Mismatch Repair Genes MSH3 and PMS1 with Acute Adverse Events and Survival in Patients with Rectal Cancer Receiving Postoperative Chemoradiotherapy

        Purpose Mismatch repair (MMR) deficiency plays a critical role in rectal cancer. This study aimed to explore the associations between genetic variations in seven MMR genes and adverse events (AEs) and survival of patients with rectal cancer treated with postoperative chemoradiotherapy (CRT). Materials and Methods Fifty single nucleotide polymorphisms in seven MMR (MLH1, MLH3, MSH2, MSH3, MSH6, PMS1 and PMS2) genes were genotyped by Sequenom MassARRAY method in 365 patients with locally advanced rectal cancer receiving postoperative CRT. The associations between genotypes and AEs were measured by odds ratios and 95% confidence intervals (CIs) by unconditional logistic regression model. The associations between genetic variations and survival were computed by the hazard ratios and 95% CIs by Cox proportional regression model. Results The most common grade ! 2 AEs in those 365 patients, in decreasing order, were diarrhea (44.1%), leukopenia (29.6%), and dermatitis (18.9%). Except 38 cases missing, 61 patients (18.7%) died during the follow-up period. We found MSH3 rs12513549, rs33013, and rs6151627 significantly associated with the risk of grade ! 2 diarrhea. PMS1 rs1233255 had an impact on the occurrence of grade ! 2 dermatitis. Meanwhile, PMS1 rs4920657, rs5743030, and rs5743100 were associated with overall survival time of rectal cancer. Conclusion These results suggest that MSH3 and PMS1 polymorphisms may play important roles in AEs prediction and prognosis of rectal cancer patients receiving postoperative CRT, which can be potential genetic biomarkers for rectal cancer personalized treatment.

        Influence of Hydrodynamic Effect on Dynamic Response of Bridge with Complex Piers Submerged in Reservoir

        In order to clarify the influence of hydrodynamic on dynamic response of bridge with complex piers submerged in reservoir, based on the continuous beam of a 4-column pile column frame pier in Xiaolangdi Reservoir of Yellow River, taking 4-column pile column frame pier and bridge as the object separately, the numerical analysis model of bridge (pier) - water interaction is established by ANSYS. The flow around deep-water piers is analyzed. For the single pier (SP) and the pier of the bridge (BP), the dynamic responses of different flow velocities, water depths and wave heights and the combined action are studied. The research shows that detouring flow analysis shows that velocity field and pressure field of each column are asymmetric. Specifically, for this bridge, due to the columns interfere with each other, the velocity difference of 1#, 4# column is more obvious than that of 2#, 3# column. Maximum and stability values of the dynamic response of SP and BP increase with the increase of flow velocity and depth. Specifically, as the water flow velocity increases, the maximum response of the dynamic response increases linearly, while the stable response of the dynamic response increases in the form of a quadratic curve. When the water depth does not exceed 50 m, the maximum and stable values of the dynamic response gradually increase in the form of a quadratic curve. When the wave acts alone, the dynamic response of SP and BP is the largest at the wave crest and the smallest at the wave trough. The maximum dynamic response at the wave crest and trough increases with the increase of wave height. With the increase of water depth, the dynamic effects of waves on SP and BP are more significant. Under the combined action of wave and current, the dynamic response of SP and BP increases with the increase of wave height and water depth. For different flow velocities, the maximum value of dynamic response at wave crest increases with the increase of flow velocity, and the maximum absolute value of dynamic response at wave trough decreases with the increase of flow velocity. Dynamic response under the combined action of wave and current is not the superposition of the dynamic response under the separate action of wave and current. The maximum value of dynamic response appears in wave crest and wave trough, indicating that the current does not change the action of wave field, and the wave still plays a leading role. Specifically, the influence of water on the dynamic response of BP is: pier bottom bending moment > pier bottom shear force > pier top displacement and the influence of water on the dynamic response of SP is: pier bottom bending moment > pier top displacement > pier bottom shear force.

        Positive Association of TEAD1 With Schizophrenia in a Northeast Chinese Han Population

        Objective Schizophrenia is a complex and devastating psychiatric disorder with a strong genetic background. However, much uncertainty still exists about the role of genetic susceptibility in the pathophysiology of schizophrenia. TEA domain transcription factor 1 (TEAD1) is a transcription factor associated with neurodevelopment and has modulating effects on various nervous system diseases. In the current study, we performed a case–control association study in a Northeast Chinese Han population to explore the characteristics of pathogenic <i>TEAD1</i> polymorphisms and potential association with schizophrenia.Methods We recruited a total of 721 schizophrenia patients and 1,195 healthy controls in this study. The 9 single nucleotide polymorphisms (SNPs) in the gene region of <i>TEAD1</i> were selected and genotyped.Results The genetic association analyses showed that five SNPs (rs12289262, rs6485989, rs4415740, rs7113256, and rs1866709) were significantly different between schizophrenia patients and healthy controls in allele or/and genotype frequencies. After Bonferroni correction, the association of three SNPs (rs4415740, rs7113256, and rs1866709) with schizophrenia were still evident. Haplotype analysis revealed that two strong linkage disequilibrium blocks (rs6485989-rs4415740-rs7113256 and rs16911710-rs12364619-rs1866709) were globally associated with schizophrenia. Four haplotypes (C-C-C and T-T-T, rs6485989-rs4415740-rs7113256; G-T-A and G-T-G, rs16911710-rs12364619-rs1866709) were significantly different between schizophrenia patients and healthy controls.Conclusion The current findings indicated that the human <i>TEAD1</i> gene has a genetic association with schizophrenia in the Chinese Han population and may act as a susceptibility gene for schizophrenia.

        Red Deer Antler Extract Accelerates Hair Growth by Stimulating Expression of Insulin-like Growth Factor I in Full-thickness Wound Healing Rat Model

        In order to investigate and evaluate the effects of red deer antlers on hair growth in the full-thickness wound healing model, Sprague-Dawley rats were given incision wounds through the full thickness of their dorsal skin and deer antler was applied for 40 days. At specified intervals thereafter (4, 8, 16, 32 and 40 days), the animals were sacrificed and the wound site skins were excised, processed, and sectioned. At post-injury days 16, 32 and 40, longer and more active new hair appeared around the healing wound of antler-treated skin. Histological studies showed that the antler extract markedly increases the depth, size, and number of hair follicles. Expression of IGF-I (insulin-like growth factor) mRNA was detected by RT-PCR and real time RT-PCR. The result showed that the expression of IGF-I (days 16, 32, and 40) was obviously up-regulated in antler-treated skins compared to control skins. Similar results were seen in the ELISA analysis to quantify the IGF-I expression. These results support the notion that wound healing can cause hair growth by enhancing the expression of IGF-I. Deer antler extract appears to have the potential to promote hair growth and could be used in hair growth products.
