      • Expression of Endogenous Hypoxia Markers in Vulvar Squamous Cell Carcinoma

        Objective: To investigate the expression of endogenous hypoxia-related markers identified as being involved in vulvar squamous cell carcinoma (VSCC). Methods: We performed immunohistochemical staining of hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$), glucose transporter-1 (GLUT-1), carbonic anhydrase 9 (CA-9) and vascular endothelial growth factor (VEGF), on tissue sections of 25 VSCC patients, 10 vulvar intraepithelial neoplasia (VIN) patients and 12 healthy controls. Results: HIF-$1{\alpha}$ expression was found in all sections, with no significant difference between controls, VIN and VSCC sections (all P<0.05). Glut-1 expression was found in 25% of control, 90% of VIN and 100% of VSCC sections. A significant difference between control and VIN or VSCC was observed (all P<0.05), while no difference was found between VIN and VSCC sections (P>0.05). CA-9 expression was negative in control sections, but it was found in 30% of VIN sections and 52% of VSCC sections with strong staining. Similarly, CA-9 expression also showed obvious differences between controls and VIN or VSCC sections (all P<0.05). However, there was no significant difference between VIN and VSCC (P>0.05). There were only 25% of control sections with weak VEGF expression, while strong staining was found in about 60% of VIN sections and 25% of VSCC sections (all P<0.05). In addition, a difference was also found between VIN and VSCC sections (P<0.05). Conclusion: Expression of endogenous hypoxia markers (HIF-$1{\alpha}$, GLUT-1, CA-9 and VEGF) might be involved in the malignant progression of VSCC.

        Controllable synthesis of zone-distributed Pd over CeO2–ZrO2/Al2O3 as advanced three-way catalyst

        Two optimally designed catalysts (Ce0.7Zr0.3O2/Pd/Al2O3, Pd/Ce0.7Zr0.3O2/Pd/Al2O3) were synthesized and compared with conventional Pd/Ce0.7Zr0.3O2/Al2O3. The characterization results reveal that for Pd/Ce0.7Zr0.3O2/Al2O3, Pd species undergo severe agglomeration and encapsulation during thermal aging process, leading to dramatic catalyst deactivation. While for the two optimized catalysts, some Pd species initially distributed on Al2O3 would migrate to the surface of CexZr1−xO2(CZ) grains upon aging treatment, resulting in enhanced Pd–CZ interaction, thus the two aged samples possess higher dispersion, larger amount of oxidized Pd2+ species and more oxygen vacancies compared with conventional Pd/Ce0.7Zr0.3O2/Al2O3-a. Consequently, modified reducibility and improved three-way catalytic performance are obtained, especially for Pd/Ce0.7Zr0.3O2/Pd/Al2O3-a.

        Theoretical Study of the Reaction Mechanism for SiF2 Radical with HNCO

        The reaction mechanism of SiF2 radical with HNCO has been investigated by the B3LYP method of density functional theory(DFT), while the geometries and harmonic vibration frequencies of reactants, intermediates, transition states and products have been calculated at the B3LYP/6-311++G** level. To obtain more precise energy result, stationary point energies were calculated at the CCSD(T)/6-311++G**//B3LYP/6-311++G** level. SiF2+HNCO→IM3→TS5→IM4→TS6→OSiF2CNH(P3) was the main channel with low potential energy, OSiF2CNH was the main product. The analyses for the combining interaction between SiF2 radical and HNCO with the atom-in-molecules theory (AIM) have been performed.

        Novel Phage Display-Derived H5N1-Specific scFvs with Potential Use in Rapid Avian Flu Diagnosis

        The highly pathogenic avian influenza A (HPAI) viruses of the H5N1 subtype infect poultry and have also been spreading to humans. Although new antiviral drugs and vaccinations can be effective, rapid detection would be more efficient to control the outbreak of infections. In this study, a phage-display library was applied to select antibody fragments for HPAI strain A/Hubei/1/2010. As a result, three clones were selected and sequenced. A hemagglutinin inhibition assay of the three scFvs revealed that none exhibited hemagglutination inhibition activity towards the H5N1 virus, yet they showed a higher binding affinity for several HPAI H5N1 strains compared with other influenza viruses. An ELISA confirmed that the HA protein was the target of the scFvs, and the results of a protein structure simulation showed that all the selected scFvs bound to the HA2 subunit of the HA protein. In conclusion, the three selected scFVs could be useful for developing a specific detection tool for the surveillance of HPAI epidemic strains.

        Allyl isothiocyanate may reverse the expression of MRP1 in COPD rats via the Notch1 signaling pathway

        In the present study, the roles of AITC in upregulatingthe MRP1 expression and its relationship with theactivation of the Notch1 signaling pathway were investigatedby combining the in vivo and in vitro experiments. AITCwas administered to the COPD model rats and normal ratsto explore the association between Notch1 and MRP1. Thehuman bronchial epithelial cells were treated with DAPT,the Notch1 signaling pathway inhibitor, to verify the effectof Notch1 on the expression of AITC-induced MRP1. Comparedwith the control group, the expressions of Notch1,Hes1 (the target gene of Notch1) and MRP1 in the lung tissueof the COPD model group were significantly inhibited. In contrast to the COPD model group, the expressions ofMRP1, Hes1 and Notch1 dramatically up-regulated followingthe treatment with Low/High doses of AITC. The expressionof MRP1 in the 16 HBE cells was down-regulated bythe inhibition of Notch in a DAPT concentration-dependentmanner. Additionally, the AITC-induced up-regulation ofthe MRP1 expression was markedly impaired following the inhibition of Notch1. The above results indicated that thepulmonary function and the expression of MRP1 in COPDrats could be improved by AITC, which was partly dependenton the Notch1 signaling pathway. Therefore, targetingthe Notch signaling pathway may present as an effectivetherapeutic strategy for COPD treatment.

        Isolation, Culture and Identification of Porcine Skeletal Muscle Satellite Cells

        The objective of this study was to establish the optimum protocol for the isolation and culture of porcine muscle satellite cells. Mononuclear muscle satellite cells are a kind of adult stem cell, which is located between the basal lamina and sarcolemma of muscle fibers and is the primary source of myogenic precursor cells in postnatal muscle. Muscle satellite cells are a useful model to investigate the mechanisms of muscle growth and development. Although the isolation and culture protocols of muscle satellite cells in some species (e.g. mouse) have been established successfully, the culture system for porcine muscle satellite cells is very limited. In this study, we optimized the isolation procedure of porcine muscle satellite cells and elaborated the isolation and culture process in detail. Furthermore, we characterized the porcine muscle satellite cells using the immunofluorecence. Our study provides a reference for the isolation of porcine muscle satellite cells and will be useful for studying the molecular mechanisms in these cells.

      • Impact response of ultra-high performance fiber-reinforced concrete filled square double-skin steel tubular columns

        This paper studies the lateral impact behavior of ultra-high performance fiber-reinforced concrete (UHPFRC) filled double-skin steel tubular (UHPFRCFDST) columns. The impact force, midspan deflection, and strain histories were recorded. Based on the test results, the influences of drop height, axial load, concrete type, and steel tube wall thickness on the impact resistance of UHPFRCFDST members were analyzed. LS-DYNA software was used to establish a finite element (FE) model of UHPFRC filled steel tubular members. The failure modes and histories of impact force and midspan deflection of specimens were obtained. The simulation results were compared to the test results, which demonstrated the accuracy of the finite element analysis (FEA) model. Finally, the effects of the steel tube thickness, impact energy, type of concrete and impact indenter shape, and void ratio on the lateral impact performances of the UHPFRCFDST columns were analyzed.

      • Incidence and Mortality of Female Breast Cancer in Jiangsu, China

        Objectives: The aim of this study was to describe and analyze the incidence and mortality of female breast cancer in Jiangsu Province of China. Methods: Incidence and mortality data for female breast cancer and corresponding population statistics from eligible cancer registries in Jiangsu from 2006 to 2010 were collected and analyzed. Crude rates, age-specific rates and age-standardized rates of incidence and mortality were calculated, and annual present changes (APCs) were estimated to describe the time trends. Results: From 2006 to 2010, 11,013 new cases and 3,068 deaths of female breast cancer were identified in selected cancer registry areas of Jiangsu. The annual average crude incidence and age-standardized incidence by world population (ASW) were 25.2/ and 17.9/100,000 respectively. The annual average crude and ASW for mortality rates were 7.03/ and 4.81/100,000. The incidence was higher in urban areas than that in rural areas, and this was consistent in all age groups. No significant difference was observed in mortality between urban and rural areas. Two peaks were observed when looking at age-specific rates, one at 50-59 years and another at over 85 years. During the 5 years, incidence and mortality increased with APCs of 4.47% and 6.89%, respectively. Compared to the national level, Jiangsu is an area with relatively low risk of female breast cancer. Conclusion: Breast cancer has become a main public health problem among Chinese females. More prevention and control activities should be conducted to reduce the burden of this disease, even in relatively low risk areas like Jiangsu.

      • Efficacy of Prophylactic Entecavir for Hepatitis B Virus-Related Hepatocellular Carcinoma Receiving Transcatheter Arterial Chemoembolization

        Background and Aims: Hepatitis B virus (HBV) reactivation was reported to be induced by transcatheter arterial chemoembolization (TACE) in HBV-related hepatocellular carcinonma (HCC) patients with a high incidence. The effective strategy to reduce hepatitis flares due to HBV reactivation in this specific group of patients was limited to lamivudine. This retrospective study was aimed to investigate the efficacy of prophylactic entecavir in HCC patients receiving TACE. Methods: A consecutive series of 191 HBV-related HCC patients receiving TACE were analyzed including 44 patients received prophylactic entecavir. Virologic events, defined as an increase in serum HBV DNA level to more than 1 log10 copies/ml higher than nadir the level, and hepatitis flares due to HBV reactivation were the main endpoints. Results: Patients with or without prophylactic were similar in host factors and the majorities of characteristics regarding to tumor factors, HBV status, liver function and LMR. Notably, cycles of TACE were parallel between the groups. Ten (22.7%) patients receiving prophylactic entecavir reached virologic response. The patients receiving prophylactic entecavir presented significantly reduced virologic events (6.8% vs 54.4%, p=0.000) and hepatitis flares due to HBV reactivation (0.0% vs 11.6%, p=0.039) compared with patients without prophylaxis. Kaplan-Meier analysis illustrated that the patients in the entecavir group presented significantly improved virologic events free survival (p=0.000) and hepatitis flare free survival (p=0.017). Female and Eastern Cooperative Oncology Group (ECOG) performance status 2 was the only significant predictors for virological events in patients without prophylactic antiviral. Rescue antiviral therapy did not reduce the incidence of hepatitis flares due to HBV reactivation. Conclusion: Prophylactic entecavir presented promising efficacy in HBV-related cancer patients receiving TACE. Lower performance status and female gender might be the predictors for HBV reactivation in these patients.

      • Curcumin Inhibits MHCC97H Liver Cancer Cells by Activating ROS/TLR-4/Caspase Signaling Pathway

        Curcumin can inhibit proliferation of liver cancer cells by inducing apoptosis, but the specific signaling pathways involved are not completely clear. Here, we report that curcumin inhibited proliferation of MHCC97H liver cancer cells by induction of apoptosis in a concentration dependent manner via stimulating intracellular reactive oxygen species (ROS) generation. Also, we showed that increased intracellular ROS formation activated the TLR-4/MyD-88 signaling pathway, resulting in activation of caspase-8 and caspase-3, which eventually led to apoptosis in MHCC97H cells. These results showed that as an prooxidant, curcumin exerts anti-cancer effects by inducing apoptosis via the TLR-4/MyD-88 signaling pathway.
