Analysis of <i>Staphylococcus aureus</i> cell viability, sublethal injury and death induced by synergistic combination of ultrasound and mild heat

        Li, Jiao,Suo, Yuanjie,Liao, Xinyu,Ahn, Juhee,Liu, Donghong,Chen, Shiguo,Ye, Xingqian,Ding, Tian Elsevier 2017 Ultrasonics sonochemistry Vol.39 No.-

<P><B>Abstract</B></P> <P>This study was designed to investigate the combined effects of ultrasound and mild heat on the viability of <I>S. aureus</I> in association with the cell membrane integrity and intracellular enzyme activity. Cells were treated by ultrasound under 55°C for 3, 5, 7, 10, and 15min. The dynamic changes of <I>S. aureus</I> cell viability, sublethal injury and death were evaluated using flow cytometric assay. Microscopies were applied to identify the morphological appearance, ultrastructure and topography of <I>S. aureus</I>. The results showed the membrane damage was synchronous with esterase inhibition during the exposure to sonication, leading to the immediate lethal effect. On the other hand, bacteria under the mild heat at 55°C were inactivated via a sublethal injury process. The different lethal modes were observed between sonication and mild heat treatments, which could synergistically inactivate <I>S. aureus</I>. The antibacterial value of thermo-sonication was greater than the sum of the individual treatments. The thermo-sonication combination synergistically reduced the number of sublethal cells and also resulted in severe cell damage.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The lethal modes induced by ultrasound and mild heat were different. </LI> <LI> Two sterilization techniques were synergistic in cell inactivation. </LI> <LI> Cell viability, sublethal injury and death were distinguished by flow cytometry. </LI> <LI> Thermo-sonication treatment reduced the number of sublethal cells. </LI> <LI> Microscopies revealed <I>S. aureus</I> morphological changes and bactericidal effect. </LI> </UL> </P>

Association of CAST Gene Polymorphisms with Carcass and Meat Quality Traits in Chinese Commercial Cattle Herds

        Li, Jiao,Zhang, Lu-Pei,Gan, Qian-Fu,Li, Jun-Ya,Gao, Hui-Jiang,Yuan, Zheng-Rong,Gao, Xue,Chen, Jin-Bao,Xu, Shang-Zhong Asian Australasian Association of Animal Productio 2010 Animal Bioscience Vol.23 No.11

Calpastatin (CAST), an endogenous inhibitor of the calpains, plays an important role in post-mortem tenderization of meat. The objectives of this study were to investigate single nucleotide polymorphisms (SNPs) in the bovine CAST gene and association with carcass and meat quality traits. A total of 212 cattle from commercial herds were tested in this study including 2 pure introduced breeds, 4 cross populations, and 3 pure Chinese native breeds. Five SNPs were identified at position 2959 (A/G), 2870 (G/A), 3088 (C/T), 3029 (G/A) and 2857 (C/T) in the CAST gene (GenBank Accession No. AF159246). Allele frequencies of SNP2959 and SNP2870 were 0.701 (A) and 0.462 (A), respectively. A general linear model was used to evaluate the associations between the two markers and 7 traits. The results showed that both SNP2959 and SNP2870 were significantly (p<0.01) associated with the Warner-Bratzler shear force (WBSF), while they had no significant association with the other 6 traits in the whole population. However, in Chinese native pure breeds, only SNP2870 had significant association with WBSF (p<0.05). The simultaneous analysis of two-marker genotype effects indicated animals containing the A/G haplotype (A for SNP2959 and G for SNP2870) tended to have lower shear force than those containing the G/A haplotype, and, especially, animals homozygous for the A/G haplotype had approximately 2 kg lower shear force than those homozygous for the G/A haplotype (p<0.01). These results suggested that both markers may be effective for the marker-assisted selection of meat quality traits in Chinese commercial herds, especially SNP2870 which can be used for Chinese native cattle.

      • Diagnostic Value of Endorectal Ultrasound in Preoperative Assessment of Lymph Node Involvement in Colorectal Cancer: a Meta-analysis

        Li, Li,Chen, Shi,Wang, Ke,Huang, Jiao,Liu, Li,Wei, Sheng,Gao, Hong-Yu Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.8

Background: Nodal invasion by colorectal cancer is a critical determinant in estimating patient survival and in choosing appropriate preoperative treatment. The present meta-analysis was designed to evaluate the diagnostic value of endorectal ultrasound (EUS) in preoperative assessment of lymph node involvement in colorectal cancer. Materials and Methods: We systematically searched PubMed, Web of Science, Embase, and China National Knowledge Infrastructure (CNKI) databases for relevant studies published on or before December 10th, 2014. The sensitivity, specificity, likelihood ratios, diagnostic odds ratio (DOR) and area under the summary receiver operating characteristics curve (AUC) were assessed to estimate the diagnostic value of EUS. Subgroup analysis and meta-regression were performed to explore heterogeneity across studies. Results: Thirty-three studies covering 3,016 subjects were included. The pooled sensitivity and specificity were 0.69 (95%CI: 0.63-0.75) and 0.77 (95%CI: 0.73-0.82), respectively. The positive and negative likelihood ratios were 3.09 (95%CI: 2.52-3.78) and 0.39 (95%CI: 0.32-0.48), respectively. The DOR was 7.84 (95%CI: 5.56-11.08), and AUC was 0.80 (95%CI: 0.77-0.84). Conclusions: This meta-analysis indicated that EUS has moderate diagnostic value in preoperative assessment of lymph node involvement in colorectal cancer. Further refinements in technology and diagnostic criteria are necessary to improve the diagnostic accuracy of EUS.

      • Associations of IL-10 Gene Polymorphisms with Acute Myeloid Leukemia in Hunan, China

        Yao, Chen-Jiao,Du, Wei,Chen, Hai-Bing,Xiao, Sheng,Wang, Cheng-Hong,Fan, Zi-Li Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.4

We investigated the possible association of interleukin-10 (IL-10) single nucleotide polymorphisms (SNPs) and susceptibility to acute myeloid leukemia (AML) in 115 patients and 137 healthy controls. Genetic analysis of IL-10 SNPs at -819 and -592 was carried out with the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) approach. The IL-10 mRNA expression of AML patients and controls with different genotype was detected by real-time quantitative polymerase chain reaction (RT-PCR). Genetic analysis of IL-10 revealed that the -819AA genotype frequencies and the -819A allele frequencies in the AML group were higher than in the controls (59.1% vs 40.9%; 75.6% vs 63.9%, respectively); there were remarkable differences in -819T/C and -592A/C gene distribution (P<0.05) and the TA haploid frequencies were higher in the AML group (75.6% vs 63.9%, P<0.05). IL-10 mRNA expression in incipient AML patients was obvious higher than the non-tumor group and the remission group ($7.78{\times}10^{-3}$ vs $2.43{\times}10^{-3}$, $3.64{\times}10^{-3}$, P<0.05).The study suggested that the haploid TA and genotype TA/TA may be associated with AML in Han people in Hunan province. The IL-10 SNPs at -819 and -592 sites were associated with AML and may affect IL-10 mRNA expression in AML patients.

Bioconversion of conjugated linoleic acid by Lactobacillus plantarum CGMCC8198 supplemented with Acer truncatum bunge seeds oil

        Dong-Ju Chen,Li-Hua Yan,Qian Li,Cai-jiao Zhang,Chuan-Ling Si,Zhong-Yuan Li,Ya-Jian Song,Hao Zhou,Tong-Cun Zhang,Xuegang Luo 한국식품과학회 2017 Food Science and Biotechnology Vol.26 No.6

Conjugated linoleic acid (CLA) isomers, c9, t11- CLA and t10, c12-CLA, have been proved to exhibit excellent biomedical properties for potential use in anticancer applications and in reducing obesity. Acer truncatum Bunge (ATB), which is rich in unsaturated fatty acids, including oleic acid, linoleic acid, and nervonic acid, is a new resource for edible oil. In the present study, we developed a new method for producing two CLA isomers from ATB-seed oil by fermentation using Lactobacillus plantarum CGMCC8198 (LP8198), a novel probiotics strain. Polymerase chain reaction results showed that there was a conserved linoleate isomerase (LIase) gene in LP8198, and its transcription could be induced by ATBseed oil. Analyses by gas chromatography–mass spectrometry showed that the concentration of c9, t11-CLA and t10, c12-CLA in ATB-seed oil could be increased by about 9- and 2.25-fold, respectively, after being fermented by LP8198.

Complete Genome Sequence of Salmonella enterica Serovar Pullorum Multidrug Resistance Strain S06004 from China(s)

( Qiu Chun Li ),( Ya Chen Hu ),( Yin Fei Wu ),( Xiao Chun Wang ),( Xiao Lei Xie ),( Ming Xin Tao ),( Jun Lei Yin ),( Zhi Jie Lin ),( Yang Jiao ),( Li Juan Xu ),( Xinan Jiao ) 한국미생물 · 생명공학회 2015 Journal of microbiology and biotechnology Vol.25 No.5

As Salmonella enterica serovar Pullorum remains a major economic problem for the poultry industries of countries with no efficient control measures, we presented a multidrug resistance strain S06004 (isolated from a clinically sick chicken in China in 2006) for genome sequencing. The genome comparison showed that the strain contained two prophages, the ST104 and prophage-4 (Fels2) of E. coli LF82, which were not detected in the only published genomes of S. Pullorum RKS5078 and CDC1983-67. In addition, the GyrA Ser83 point mutation, drugresistant genes, and many antibiotic pump systems that are present in S06004 may be contributing to the multidrug resistance of this strain.

Experimental study of the effect of internal waves on the rotational hydrodynamics of underwater vehicle

        Chen Sheng-tao,Li Dong-ju,Tian Hao,Hou Jiao-yi,Ning Da-yong,Gong Yong-jun 대한조선학회 2022 International Journal of Naval Architecture and Oc Vol.14 No.1

Maneuverability, which is closely related to navigation safety, is an important part of the overall performance of an underwater vehicle. The purpose of this study is to analyze the dynamic response of an underwater vehicle to internal waves at different angles and speeds. A combination of experiments and numerical simulations is used to compare the forces on the underwater vehicle. In terms of fluidstructure coupling, the internal wave interface is tracked by the VOF method. In this paper, waves of the same amplitude ratio as the numerical results are simulated using a wave-making sphere. The validity of the numerical method is verified by comparing with the rotation moment test of the underwater vehicle. The results show that different angles of attack have significant effects on the forces and moments of the underwater vehicle and that choosing the right angle of attack and rotation speed can limit the dangerous depth. The conclusion is of certain significance for the safety of underwater navigation.

Enhanced D-ribose production by genetic modification and medium optimization in Bacillus subtilis 168

        Chen Zhao,Xiang-Ying Zhao,Jian-Jun Liu,Jun-Jiao Zhang,Jia-Xiang Zhang,Li-He Zhang 한국화학공학회 2018 Korean Journal of Chemical Engineering Vol.35 No.5

D-ribose, a five-carbon sugar with diverse applications, is mainly produced by transketolase (tkt)-deficient Bacillus subtilis (B. Subtilis). We constructed B.subtilis SFR-3A by replacing the corresponding sites of B. subtilis 168 with the mutation site of tkt in the “wild” D-ribose high-producing strain B. subtilis SFR-4, resulting in 5.29 g/L of Dribose. In the meantime, B.subtilis SFR-159 was constructed by completely removing the tkt gene of B. subtilis 168 with a higher production of 6.21 g/L. Through medium optimization, batch fermentation of SFR-3A and SFR-159 gave the best result of 27.56 g/L and 29.89 g/L, which corresponds to productivity of 0.51 g/L/h and 0.41 g/L/h, respectively. In this work, SFR-3A showed more latent capacity over SFR-159 as to productivity and had greater potential to serve as a platform for D-ribose production.

Silicate Ions Derived from Calcium Silicate Extract Decelerate Ang II-Induced Cardiac Remodeling

        Li Xin,Zhang Yanxin,Jin Qishu,Song Qiaoyu,Fan Chen,Jiao Yiren,Yang Chen,Chang Jiang,Dong Zhihong,Que Yumei 한국조직공학과 재생의학회 2023 조직공학과 재생의학 Vol.20 No.5

BACKGROUND: Pathological cardiac hypertrophy is one of the main activators of heart failure. Currently, no drug can completely reverse or inhibit the development of pathological cardiac hypertrophy. To this end, we proposed a silicate ion therapy based on extract derived from calcium silicate (CS) bioceramics for the treatment of angiotensin II (Ang II) induced cardiac hypertrophy. METHODS: In this study, the Ang II induced cardiac hypertrophy mouse model was established, and the silicate ion extract was injected to mice intravenously. The cardiac function was evaluated by using a high-resolution Vevo 3100 small animal ultrasound imaging system. Wheat germ Agglutinin, Fluo4-AM staining and immunofluorescent staining was conducted to assess the cardiac hypertrophy, intracellular calcium and angiogenesis of heart tissue, respectively. RESULTS: The in vitro results showed that silicate ions could inhibit the cell size of cardiomyocytes, reduce cardiac hypertrophic gene expression, including atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP) and b-myosin heavy chain (b-MHC), decrease the content of intracellular calcium induced by Ang II. In vivo experiments in mice confirmed that intravenous injection of silicate ions could remarkably inhibit the cardiac hypertrophy and promote the formation of capillaries, further alleviating Ang II-induced cardiac function disorder. CONCLUSION: This study demonstrated that the released silicate ions from CS possessed potential value as a novel therapeutic strategy of pathological cardiac hypertrophy, which provided a new insight for clinical trials.
