Identification of Genes Modulated by High Extracellular Calcium in Coculture of Mouse Osteoblasts and Bone Marrow Cells by Oligo Chip Assay

Calcium concentration in the bone resorption lacunae is high and is in the mM concentration range. Both osteoblast and osteoclast have calcium sensing receptor in the cell surface, suggesting the regulatory role of high extracellular calcium in bone merabolism. In vitro, high extracellular calcium stimulated osteoclastogenesis in coculture of mouse osteoblasts and bone marrow cells. Therefore we examined the genes that were commonly regulated by both high extracellular calcium and 1,25(OH)_(2)vitaminD_(3)(VD3) by using mouse oligo 11 K gene chip. In the presence of 10 mM[Ca^(2+)]e or 10 nM VD3, mouse calvarial osteoblasts and bone marrow cells were co-cultured for 4 days when tartrate resistant acid phosphatase-positive multinucleated cells start to appear. Of 11,000 genes examined, the genes commonly regulated both by high extracellular calcium and by VD3 were as follows; 1) the expressions of genes which were osteoclast differentiation markers or were associated with osteoclastogenesis were up-regulated both by high extracellular calcium and by VD3; trap, mmp9, car2, ctsk, ckb, atp6b2, tm7sf4, rab7, 2) several chemokine and chemokine receptor genes such as sdf1, scya2, scyb5, scya6, scya8, scya9, and ccr1 were up-regulated both by high ectracellular calcium and by VD3, 3) the genes such as mmp1b, mmp3 and c3 which possibly stimulate bone resorption by osteoclast, were commonly up-regulated, 4) the gene such as c1q and msr2 which were related with macrophage function, were commonly down-regulated, 5) the genes which possibly stimulate osteoblast differentiation and/or mineralization of extracellular matrix, were commonly down-regulated;slc8a1, admr, plod2, lox, fosb, 6) the genes which possibly suppress osteoblast differentiation and/or mineralization of extracellular matrix, were commonly up-regulated;s100a4, npr3, mme, 7) the genes such as calponin 1 and tgfbi which possibly suppress osteoblast differentiation and/or mineralization of extracellular matrix, were up-regulated by high extracelluar calcium but were down-regulated by VD3. These results suggest that in coculture condition, both high extracellular calcium and VD3 commonly induce osteoclastogenesis but suppress osteoblast differentiation/mineralization by regulating the expression of related genes.

The diagnostic requirements were suggested and explained regarding the systems of differentiation of symptoms and signs in the third year study of standardization and unification of the terms and conditions used for diagnosis in oriental medicine. The systems were as follows : - analyzing and differentiating of epidemic febrile disease - analyzing and differentiating in accordance with the Sasang constitution medicine based on four-type recognition - differentiation of disease according to pathological changes of Chong and Ren channels - standards for diagnosis of women's disease - standards for diagnosis of children's disease - standards for diagnosis of motor and sensor disturbance (-muscle. born, joint, etc.) - standards for diagnosis of neuropsychiatric disease - standards for diagnosis of five sense organ disease - standards for diagnosis of external disease The indivisual diagnosis pattern was arranged by the diagnostic requirements in the following order : another name, notion of diagnosis pattern, index of differentiation of symptoms and signs, the main point of diagnosis, analysis of diagnosis pattern, discrimination of diagnosis pattern, prognosis, a way of curing a disease, prescription, herbs in common use, disease appearing the diagnosis pattern, documents. The standards for diagnosis of each disease was arranged by the diagnostic requirements in the following order : another name, notion of disease, the main point of diagnosis, analyzing and differentiating of disease, analysis of disease, discrimination of disease, prognosis, a way of curing and prescription of disease, disease in western medicine appearing the disease in oriental medicine, documents.

음향방출 신호처리에 의한 플라즈마용사코팅 피막부식재의 파손해석

This paper was to investigate of a adhesiveness of the plasma sprayed coating materials did a salt spray by acoustic emission method in tensile loading. The powders used for the coating were nickel aluminum composite powder Ni-4.5wt.%Al and titanium dioxide powder TiO2. These powders were coated on a carbon steel S45C by plasma spray method. The salt solution was a 5% NaCl and the salt spray times were 2, 5 and 10 hours respectively. The salt solution penetrated into the surface of the substrate through pore of the coating layer built in the process of plasma spay. Corrosion productions formed on the surface of substrate. The adhesiveness between the substrate and the coating layer is weaken by corrosion and the exfoliation initiated chiefly at the corrosion surface of the substrate. The AE events and energy of the corroded coating specimens decreased as the salt spray times increased. The AE amplitudes increased at strain range of 6%-11%.

XML/EDI를 이용한 인터넷쇼핑몰 구축관련연구

인터넷 이용의 급증에 따라 전자상거래(Electronic Commerce)가 지식, 정보기반시대의 새로운 패러다임으로 부각됨에 따라, 시장규모도 이례 없이 증가되고 있다. 인터넷의 판도를 바꾸어준 HTML(Hyper Text Markup Language)의 한계성을 극복하기 위한 사용자의 다양한 요구는 XML(eXtensible Markup Language)를 전자상거래의 표준으로 자리잡게 하였으며, XML의 등장으로 기존의 EDI 시스템을 통합하고 그 효율성을 최대화시키고자 하는 노력으로 XML/EDI 기술도 눈부신 발전을 하고 있다. 이 기술은 별개가 아닌 각각의 기술적 특성과 장점을 이용한 통합 시스템으로 정의할 수 있다. 본 연구에서는 XML/EDI를 기반으로 하는 인터넷 전자상거래 시스템의 구축현황 및 방안에 대해 논의하면서, 이러한 서비스를 바탕으로 제공하는 기존의 인터넷쇼핑몰 시스템을 새로운 방식으로 설계하여 실제적으로 사용할 수 있게 구현 하였다. In relevance to the increasing usage of the Internet, Electronic Commerce, a new paradigm in this knoeledge, information society, is increasing in market share accordingly. The various demands of users have brought a change from HTML(Hyper Text Markup Language) to the standardized XML(eXtensible Markup Language). With the development of XML, EDI system was integated and in order to maximize efficiency, XML/EDI thchology was developed at an astonis hing rate. This thchology can be characterized not as separate entities, but as an assimilation of each thchological advencement and strengths as a integrated system. During this research, I discussed the establishment procedures and plans of XML/EDI based internet electronic commerce system, establishing and designing a practical and usable computer shopping-mall system based on these systems.

자기공명영상으로 진단된 바이러스성 능형뇌염

Viral neuritis due to herpes simplex type II has been well documented. The virus lies dormant within the gasserian ganglion of the trigeminal verve. Retrograde spread into the brainstem is rare. This has been referred to as rhombencephalitis. The reported cases of rhombencephalitis detected by MR are very rare. So we report a case of viral rhombencephalitis which was diagnosed by MR. A 35 year-old female patient was admitted due to dizziness, nausea, tinnitus(Lt.), hearing impairment(Lt.), and hypoesthesia on the left face, which were similar to the symptoms of acoustic neuroma. MR showed the enhanced high signal intensity in left middle cerebellar peduncle. One month after treatment, the symptoms resolved. Follow-up MR showed the decreased high signal intensity. We report this case with review of the literature.

Propranolol이 흰쥐 해마박편에서 Gamma-aminobutyric acid 및 Glutamic acid유리에 미치는 영향에 관한 연구

Present study was performed to clarify the effect of propranolol on the release of amino acid neurotransmitters employing hippocampal slices. Hippocampal slices (300-400㎛ thick) were prepared by the method of Spencer et al.(1976) and pre-equilibrated in Krebs-bicarbonate medium (KBM, pH 7.4) for 1 hr at 37℃. Pre-equilibrated slices were incubated in fresh KBM and then veratrine (25μM)-containing KBM for 10 min period. Basal and veratrine-induced release of GABA and glutamic acid were determined from recovered medium by HPLC. After 30 min resting period, slices were reincubated in propranolol-containing KBM and propranolol plus veratrine-containing medium consecutively for 10 min period each to investigate the effect of propranolol on basal or veratrine-induced amino acid release from hippocampal slices. The observed results were as follows : 1. The release of GABA induced by the first and second 10min-exposure of 25μM veratrine was 246.9±16.82 nmol and 209.9±20.84 nmol, respectively. When compared with released amounts of GABA during the corresponding spontaneous periods, these were 7.3 and 7.3-fold increase respectively. 2. the release of glutamic acid induced by the first and second 10min-exposure of 25μM veratrine was 426.3±63.16 nmol and 377±61.83 nmol, respectively. When compared with released amounts of glutamic acid during the corresponding spontaneous periods, these were 9.9 and 10.2-fold increase respectively. 3. Propranolol (2-8 mM) inhibited veratrine-stimulated GABA release in dose-dependent manner. 4. Propranolol (2-6 mM) enhanced veratrine-stimulated and spontaneous release of glutamic acid in dose-dependent fashion.

유압피로시험기의 제작

A hydraulic fatigue test machine was manufactured using a personal computer and a proportional valve, a hydraulic system. This machine was controlled loading waves and stress ratios, frequency by personal computer. A numerical simulation for the dynamic quality of this machine was done by 4 order Runge-Kutta method. Corrosion fatigue crack growth behaviors of SPH41 in 3% NaCl aqueous solution were investiged with this machine. Corrosion fatigue crack growths of this steel were reduced than the uncoated steel by anti-corrosion effect of zinc.

Corrosion fatigue crack growth behaviors of Zinc coating steel, SGH41, in 3% NaCl aqueous solution was investiged with a hydraulic fatigue testing machine. A hydraulic fatigue test machine was manufactured using a personal computer, a proportional valve one, a hydraulic system. Corrosion fatigue crack growth rates of SGH41 is reduced than that of the uncoated steel by anti-corrosion effect of zinc. Pans rule, da/dN=C(ΔK)^(m) is applied about the relation between the stress intensity factor range, ΔK, and crack growth rate, da/dN, of both the coated specimen and the uncoated one. Where the slope of the correlation, m, is 2.24 for the coated specimen and 3.66 for the uncoated one. The corrosion sensitivity, (da/dN)_(corr)/(da/dN)_(air), of the coated specimen was smaller than that of the uncoated specimen.
