Investigation on Polychlorinated Dibenzo-p-dioxins, Polychlorinated Dibenzofurans and Dioxin-like Polychlorinated Biphenyls in Korean Fruits and Dietary Intake Estimation

        Choi, Geun-Hyoung,Choi, Dal-Soon,Hong, Su-Myeong,Kwon, Oh-Kyoung,Eun, Hee-Soo,Kim, Jeong-Han,Kim, Jin-Hyo The Korean Society for Applied Biological Chemisty 2012 Journal of Applied Biological Chemistry (J. Appl. Vol.58 No.3

Contamination levels of 17 polychlorinated dibenzo-p-dioxins (PCDDs)/polychlorinated dibenzofurans (PCDFs) and 12 dioxin-like polychlorinated biphenyls (DL-PCBs) were investigated on six major fruits consumed in Korea. Dioxin level was monitored on whole fruits. Average concentrations of PCDD/Fs were 0.042 pg/g fresh weight (f.w.) (apple), 0.0094 pg/g f.w. (grape), 0.1843 pg/g f.w. (mandarin orange), 0.2282 pg/g f.w. (peach), 0.1491 pg/g f.w. (pear), and 0.1330 pg/g f.w. (persimmon). For DL-PCBs, the average concentrations were 0.0932 pg/g f.w. (apple), 0.2649 pg/g f.w. (grape), 1.5437 pg/g f.w. (mandarin orange), 0.1942 pg/g f.w. (peach), 0.4591 pg/g f.w. (pear), and 0.4396 pg/g f.w. (persimmon). The major toxic equivalency (TEQ) contributors were identified as 1,2,3,7,8-pentachlorodibenzo-p-dioxin in PCDD and 2,3,4,7,8-pentachlorodibenzofuran in PCDFs. Most residual DL-PCBs showed no contribution to total TEQ values. The lowest TEQ level was recorded in peach (0.0161 pg-$TEQ_{WHO/g}$ f.w.), and the highest TEQ level was found in mandarin orange (0.0448 pg-$TEQ_{WHO/g}$ f.w.). Total estimated daily intake (EDI) of dioxins from the six fruits was calculated at 3.8159 pg-$TEQ_{WHO}/day$, which is 1.59% of Korean tolerable daily intake based on 60 kg body weight.

Resveratrol relieves hydrogen peroxide-induced premature senescence associated with SIRT1 in human mesenchymal stem cells

Choi, Mi Ran,Han, Dal Mu Ri,Kim, Sun Hwa,Ohn, Takbum,Jung, Kyoung Hwa,Chai, Young Gyu THE KOREAN SOCIETY OF TOXICOGENOMICS AND TOXICOPRP 2014 MOLECULAR AND CELLULAR TOXICOLOGY Vol.10 No.1

Cellular senescence of mesenchymal stem cells (MSCs) is often induced during in vitro expansion, by multiple experimental stimuli including oxidative stress. In this study, we investigated expression of senescence-associated proteins including SIRT1after inducing premature senescence of MSCs with hydrogen peroxide ($H_2O_2$). We also analyzed the effect of resveratrol (RSV) on premature senescence. We found that $H_2O_2$ triggered the recruitment of RCK (p54) to P-bodies in MSCs. Premature senescence of MSCs in response to $H_2O_2$ induced a decrease in SIRT1expression and activity (indirectly identified by measuring H3-K9ac). Cellular expression of p21 and phosphorylation of ERK1/2 and p38 kinases were increased in response to $H_2O_2$, whereas phosphorylation of pRb was decreased. In contrast, RSV pretreatment resulted in a decrease in the premature senescence of MSCs. In addition, RSV pretreatment before exposing cells to $H_2O_2$ not only alleviated changes in the levels of proteins that were sensitive to the $H_2O_2$ treatment (SIRT1, p21,ERK1/2 and p38) but also inhibited the decrease of SIRT1 induced by nicotinamide (NAM). Our results suggest that MSCs may exhibit an increased tolerance for $H_2O_2$-induced oxidative stress via the senescence-associated proteins that are regulated by RSV.

      • Kinematic Analysis of Locomotory Recovery Following Unilateral Hemisection of Spinal Cord in the Adult Rat

Choi, Chang Rak,Kim, Dal Soo,Cho, Kyoung-Suok CATHOLIC MEDICAL CENTER 1995 Bulletin of the Clinical Research Institute Vol.23 No.1

Spinal cords posses a remarkable capacity for locomotory recovery in the rat. For example adult rats with dorsally hemisected thoracic (T6) spinal cord recover hindimb locomotor function within a few weeks. The objectves of this study was, therefore, to investigate recovery of locomotion after unilateral hemisection spinal cord in the rat. Especially, we were interested in comparing the locomotion of the limbs between the hemisected side and the intact side. Adult Sprague-Dawley rats (290-310 grams) were used. Unilateral hemisection on right spinal cord at T7 was performed on a total of 10 rats under strile condition. Foot trajectories of treadmill walking animals were video-taped and analyzed using a computerized motion analysis system. The foot trajectories, stride length, vertical displacement, peak velocities, duration of swing phase, and limb coordination were calculated and plotted. Following the right hemisection, all these kinematic variables were initially disrupted but returned to prelesion values with in three to six weeks. Eventhough the lesion was unilateral, locomotory behavior was disturbed bilaterally. Amplitudes (1.44±0.26cm) and velocities (19.24±3.6cm/sec) of vertical movement of the hindfoot in the hemisected side was signficantly (P<0.01) lower than those of the prelesion values (ampitude 1.96±0.21cm; velocity 30.60±2.75cm/sec) and these values did not recover to prelesion levels. The difference in locomotory behavior between the limbs of the injured and uninjured side could not be easily detectable without using detailed kinematic analysis of the limb trajectories. In conclusion, asymmetry of the limb kinematics was found in the vertical movement of the hindlimbs of the unilateral hemisection model.

급성 뇌허혈증에 있어 만니톨의 효과에 대한 실험적 연구

조경석,홍용길,백민우,김달수,강준기,최창락 대한신경외과학회 1994 Journal of Korean neurosurgical society Vol.23 No.8

The development of postischemic irreversible brain damage depends upon the length of ischemia and its severity during arterial occlusion, although release of the occlusion always leads to restoration of normal or above n o d cerebral blood flow. This experiment was planned to determine the effects of mannitol on cerebral ischemia on subsequential izgional cerebral blood flow(rCBF) and somatosensory evoked potential(SEP) following reperfusion after ischemia and also define the proper time of vascular occlusion without irreversible brain damage. Cerebral ischemia was induced in cat by transorbital occlusion of the left MCA with a Sugita clip for period of 30 minutes and the ischemic brain was reperfused for 180 minutes by removing the clip. Foty adult cats, weighing 25 to 4.0㎏ were divided into 5 groups : control(Gmup Ⅰ, n=8), permanent MCA occlusion(Group Ⅱ, n= 8), permanent MCA occlusion with mannitol infusion (Group Ⅲ, n=8), permanent MCA occlusion with mannitol infUsion(Group Ⅲ, n=8), 30 minutes MCA clipping followed by reperfusion(Gmup Ⅳ, n= 8) and 30 minutes MCA clipping with mannitol infusion followed by reperfusion groups((group Ⅴ, n=8) respedvely. The rCBF and SEP measurents were carried out in each animal immediately, after MCA occlusion, at 30 minutes, 60 minutes, 90 minutes, 120 minutes, 150 minutes and 180 minues followed by reperfusion. The rCBF was measured by the hydrogen clearance technique. Mando1 was given in bolus of l a g body weight as a 25% solution delivered via the femoral vein The results were as follows: 1) Gradual elevation of intracranial pressure(1CP) and systolic blood pressure were observed after MCA occlusion. Treatment with mannitol in MCA occlusion animals(Group Ⅲ & Ⅴ) decreased ICP immediately a k r idusion of mannitol. 2) Normal control rCBF(ml/100g/min) were 51.94±5.05 in the left temporal(LT) and 50.80± 4.87 in the left perietal lobes(LP). 3) The MCA occlusion resulted in a reduction of the blood flows to 72% of the normal control ones(LT : 14.29± 4.81ml/100g/min) at the left temporal area immediately after occlusion and also a reduction of flows to 80.4% of the normal control ones(LT : 1024± 3.69ml/100g/min) at 180 minutes after occlusion in Group Ⅱ. 4) In the mannitol-treated group(group Ⅳ), reperfwd animals, with removal of the clip on MCA had an improved postischemic recovely of blood flow and ipsilateral cerebral blood flows were restored to 42.5% of the normal control ones(LP : 212±4.13ml/100g/min) at 180 minutes after occlusion in Lt parietal CBF. 5 ) In the reperfusion group(group Ⅳ), reperfused animals, with removal of the clip on MCA had an increase in rCBF to the level of 833% of the control value(LT : 45.78± 6.80ml/100g/min) at 180 minutes after reperfusion and also reperfused animals, with treated mannitol had further increase the blood flow up to the level of 92s % of the control value(LT : 49.04± 43.6 ml/100g/min) at 180 minutes after repexfwion. 6) After the MCA occlusion, the SEP was present but markedly altered in shape and particularly the early components of the SEP were suppressed in the ipsilateral occlusion hemisphere. 7) In the mannitol-treated reperfusion group(Group Ⅴ) the amplitude of the SEP was restored to 80% of control value. The SEP was significantly suppressed if the rCBF fell below 10- 14ml/100g/min. These results suggest that provided CBF can be restored to above the 40% threshold well within 30 minutes, prevention of ischemic brain damage can be expected and also the mannitol may of benefit in prolongation of the time threshold for the formation of the cerebral ischemia after vessel occlusion.

      • Proteomic Analysis of the Copper Ion-Induced Stress Response in a Human Embryonic Carcinoma Cell Line

Han, Dal Mu Ri,Choi, Mi Ran,Jung, Kyoung Hwa,Lee, Hyung Tae,Park, Ji Hyun,Ohn, Takbum,Chai, Young Gyu SAGE Publications 2012 International journal of toxicology Vol.31 No.4

Excessive exposure to copper, a redox-active metal, generates free radicals, which can cause cellular damage. In this study, we aim to identify the proteins that are up- or downregulated by copper exposure in human embryonic carcinoma (NCCIT) cells and to understand the mechanisms that play a role in the copper-induced stress response. After exposure to copper ions, the cells showed upregulated levels of 78 kDa glucose-regulated protein, fibrillin 1, CWC22 spliceosome-associated protein (KIAA1604), heat shock protein (HSP) 60, and HSP70, while the tumor necrosis factor receptor-associated factor 6, vimentin, 14-3-3 protein zeta, and RAC-beta (AKT2) serine/threonine protein kinase were downregulated. The GeneGo Process Networks of the proteins upregulated by copper ions were analyzed, and the 3 highest-scoring networks from the proteins upregulated by copper ions are presented here. In particular, the increased level of HSP70 in response to copper ions occurred in a dose-dependent manner, indicating that HSP70 could be a potential biomarker for copper toxicity in mammalian cells.

A food exchange system for diabetes is a useful tool for meal planning and nutritional education. The first edition of the Korean food exchange lists was developed in 1988 and the second edition was revised in 1995. With recent changes in the food marketplace and eating patterns of Koreans, the third edition of food exchange lists was revised in 2010 by the Korean Diabetes Association, the Korean Nutrition Society, the Korean Society of Community Nutrition, the Korean Dietetic Association and the Korean Association of Diabetes Dietetic Educators through a joint research effort. The third edition is based on nutritional recommendations for people with diabetes and focuses on adding foods to implement personalized nutrition therapy considering individual preferences in diverse dietary environment. Foods were selected based on scientific evidence including the 2007 Korea National Health and Nutrition Examination Survey data analysis and survey responses from 53 diabetes dietetic educators. While a few foods were deleted, a number of foods were added, with 313 food items in food group lists and 339 food items in the appendix. Consistent with previous editions, the third edition of the food exchange lists included six food categories (grains, meat, vegetables, fats and oils, milk, and fruits). The milk group was subdivided into whole milk group and low fat milk. The standard nutrient content in one exchange from each food group was almost the same as the previous edition. Korea Food & Drug Administration's FANTASY (Food And Nutrient daTA SYstem) database was used to obtain nutrient values for each individual food and to determine the serving size most appropriate for matching reference nutrients values by each food group. The revised food exchange lists were subjected to a public hearing by experts. The third edition of the food exchange lists will be a helpful tool for educating people with diabetes to select the kinds and amounts of foods for glycemic control, which will eventually lead to preventing complications while maintaining the pleasure of eating.
