        운동강도의 차이가 대장에서의 Heme oxygenase-1 발현에 미치는 영향: Oligonucleotide chip microarray analysis

        이 연구의 목적은 운동강도 차이에 의한 대장조직이 Heme oxygenase-1(HO-1)의 mRNA 발현에 어떠한 영향을 미치는지 규명하기 위해, Sprage-Dawley계 흰쥐를 대상으로 통제그룹(CON)과 저강도 운동그룹(LIE), 고강도 운동그룹(HIE)으로 구분하여 4주 동안 트레드밀운동 후 48시간 이후에 High through-put microarray analysis방법으로 다음과 같은 실험을 실시하였다. 본 실험은 Rat ABI oligo chip 26,857 개의 유전자 중 filtering을 통하여 12.079개 유전지를 선택하였고, 이 중 유의성 있는(p<.05) 유전자 12,072개를 선별하였으며, clustering분석 방법과 면역조직화학염색법에서도 HO-l이 운동강도에 따라 유의하게 발현하였다(p<.05). Microarray분석 후 RT-PCR로 확인한 결과 HO-I의 유전자 발현양상이 일치하는 결과를 얻었다. 이 실험의 결과로서 운동이 대장에서 HO-1 발현에 영향을 주는 것을 알 수 있었다. 운동이 대장에서의 mRNA 발현이 저강도 운동그룹(LIE)보다 고강도 운동그룹(HIE)에서 더 많이 발현되어지는 것으로 보아 고강도 운동에 의해 발생되는 산소라디칼을 HO-l의 유도를 통하여 적절히 제거함으로써 스스로 항상성을 유지하고 있는 것으로 생각되어진다. The purpose of this study is to identify how large intestine tissue effects to mRNA expression in Heme oxygenase-l (HO-1) due to differences in exercise intensity. To perform this experiment we divided Sprage-Dawley rats into 3 groups (control group (CON). low-intensity exercise group (LIE) and high-intensity exercise group (HIE)), and in 48 hours after 4 weeks treadmill exercise. the following experiment was performed with "High through-put micro array analysis methods" . 12,079 genes were chosen by filtering of the Rat ASI oligo chip 26.857 genes in this experiment Among 12.079 genes, we selected significant genes (p<.05) that were also expressed in the ways of clustering analysis and immunohistochemistry. With results of RT-PCR confirming and microarray analyzing, we derived that gene expression profiles of HO-l had been consistent. As a result of this experiment, we certainly identified that exercise effects to HQ-l expression in large intestine. mRNA expression was more expressed in high-intensity exercise group CHIE) than low-intensity exercise group (LIE), it seems that homeostasis maintains itself by properly removing oxygen radical. caused by high intensity exercise. through reduction of HO-l.

        국산 Lidocaine 알칼리화를 위한 Bicarbonate 혼합시 침전유발 최소량에 대한 고찰

        Background : When local anesthetics for regional anesthesia is used, usually small amounts of bicar-bonate are added for rapid onset. This addition gives the mixed solution a more alkaline pH. The following result is an increased unionized form of the local anesthetic and rapid penetration of the drug into tiissue. Unfortunately, no data about adequate mixing volumes of domestic lidocaine and bicarbonate is available. Methods : We examined six mixing pairs of two kinds of 2% lidocaines and three kinds of 8.4% bicarbonates for minimum volumes of bicarbonate to cause a precipitation of 2% 20 ml lidocaine. Results : The mean volumes of bicarbonate to cause precipitation were 1.54 ml for Kwang-Myung lidocaine with Kwang-Myung bicarbonate, 2.90 ml for Kwang-Myung lidocaine with Dae-Won bicarbonate, 2.73 ml for Kwang-Myung lidocaine with Je-Il bicarbonate, 0.97 ml for Je-Il lidocaine with Kwang-Myung bicarbonate, 1.26ml for Je-Il lidocaine with Dae-Won bicarbonate and 1.39 ml for Je-Il lidocaine with Je-Il bicarbonate. Conclusions : We conclude that the Kwang-Myung lidocaine and the Je-Il lidocaine could cause precipitation when mixing with a smaller bicarbonate volume than foreign textbook recommended.       • Hypoxia-Responsive MicroRNA-101 Promotes Angiogenesis <i>via</i> Heme Oxygenase-1/Vascular Endothelial Growth Factor Axis by Targeting Cullin 3

        <P>Aims: Hypoxia induces expression of various genes and microRNAs (miRs) that regulate angiogenesis and vascular function. In this study, we investigated a new functional role of new hypoxia-responsive miR-101 in angiogenesis and its underlying mechanism for regulating heme oxygenase-1 (HO-1) and vascular endothelial growth factor (VEGF) expression. Results: We found that hypoxia induced miR-101, which binds to the 3 ' untranslated region of cullin 3 (Cul3) and stabilizes nuclear factor erythroid-derived 2-related factor 2 (Nrf2) via inhibition of the proteasomal degradation pathway. miR-101 overexpression promoted Nrf2 nuclear accumulation, which was accompanied with increases in HO-1 induction, VEGF expression, and endothelial nitric oxide synthase (eNOS)-derived nitric oxide (NO) production. The elevated NO-induced S-nitrosylation of Kelch-like ECH-associated protein 1 and subsequent induction of Nrf2-dependent HO-1 lead to further elevation of VEGF production via a positive feedback loop between the Nrf2/HO-1 and VEGF/eNOS axes. Moreover, miR-101 promoted angiogenic signals and angiogenesis both in vitro and in vivo, and these events were attenuated by inhibiting the biological activity of HO-1, VEGF, or eNOS. Moreover, these effects were also observed in aortic rings from HO-1(+/-) and eNOS(-/-) mice. Local overexpression of miR-101 improved therapeutic angiogenesis and perfusion recovery in the ischemic mouse hindlimb, whereas antagomiR-101 diminished regional blood flow. Innovation: Hypoxia-responsive miR-101 stimulates angiogenesis by activating the HO-1/VEGF/eNOS axis via Cul3 targeting. Thus, miR-101 is a novel angiomir. Conclusion: Our results provide new mechanistic insights into a functional role of miR-101 as a potential therapeutic target in angiogenesis and vascular remodeling. Antioxid.         사례보고 : 하수오 복용 후 발생한 재발성 독성 간염 1예

        Toxic hepatitis has been reported as a major cause of acute hepatitis, but its potential induction by herbal remedies and/or health foods is usually neglected. We experienced a case of toxic hepatitis associated with Polygoni multiflori, a Chinese herb commonly known as Ho-Shou-Wu. A 54-year-old woman consumed Ho-Shou-Wu for 1 month, after which she experienced fatigue and overall weakness. A diagnosis of toxic hepatitis was made based on her clinical history, the findings for viral markers and other laboratory data, and ultrasonography. Her condition improved considerably after she stopped taking Ho-Shou-Wu. However, she resumed taking Ho-Shou-Wu immediately after discharge from hospital, which aggravated her symptoms and liver function. She was immediately readmitted and stopped taking Ho-Shou-Wu. Her relapse into hepatitis immediate after resuming consumption of the herb is strongly indicative of the validity of Koch`s postulate in this case.

        영일동맹 이후 주한영국공사의 활동

        영일동맹 이후 주한영국공사의 활동 현 광 호 영국은 1902년 1월 일본과 영일동맹을 체결했다. 일본은 동맹체결 이후 대한제국에 대한 침략을 강화했다. 조단은 영일동맹 이후 대한제국의 내정에 한층 깊숙이 개입했다. 조단은 대한제국의 자주적 개혁에 제동을 걸었고, 일본의 대한제국 진출에 중요한 역할을 담당했다. 조단은 러시아인의 용암포활동을 주시했다. 조단은 러시아인의 활동을 벌목작업으로 인정하지 않았다. 조단은 러시아인의 활동이 대한제국의 주권을 침해하는 것으로 인식했다. 조단은 대한제국에 대해 압록강의 개방을 요구했다. 조단은 경제발전론을 제기하여 고종을 설득했다. 다음으로 조단은 고종에게 국가주권문제를 거론했다. 끝으로 조단은 고종에게 압록강지역을 개항하여 러시아를 견제할 것을 제의했다. 조단은 대한제국이 용암포조차협정을 체결할 경우 영국은 자유행동을 취할 것을 경고했다. 그 결과 대한제국은 러시아와의 용암포조차협정 체결에 부정적인 입장이 되었다. 조단은 일본, 미국 외교관들과 공조하여 개항을 추진했다. Jordan demanded to the Great Han Empire opening of Yalu River(鴨綠江). Jordan pursuaded King Kojong with Theory of economic development. Secondly Jordan made Theory of sovereignty (國權論) to win King Kojong(高宗) over. Finally Jordan pursuaded King Kojong checking aggression of Russia by opening of Yalu River. Jordan gave a warning of having a free hand in Great Han Empire if Great Han Empire would conclude Yongampo lease treaty with Russia. As a result Great Han Empire a took a negative attitude for lease treaty with Russia. Jordan propelled opening of Yongampo by coalition of Japanese diplomat and American diplomat.

        Stain improvement in the white button mushroom ‘Seolgang’ and its varietal characteristics in Agaricus bisporus

        The button mushroom (Agaricus bisporus) is one of the most widely cultivated important edible mushroom species. In the breeding of new button mushroom, ‘Seolgang’ was developed by crossing two monokaryons ‘CM020913-27’ and ‘SSU423-31’. Because of the secondarily homothallism, only a small percentage of the basidia produce 3 or 4 spores, which are mostly haploid (n) and do not fruit. Single spore cultures derived from these types of spores produce a vegetative mycelium that also contain a variable number of genetically identical nuclei per cell called monokaryon. The lack of clamp connections between monokaryon and dikaryon required a series of mycelial culture and fruiting test. After crossing, hybrids were cultivated on a small scale and on a commercial scale at a farm. For this, the spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of ‘Seolgang’ on CDA was better at 20℃ and 25℃ when it was compared with that of ‘505 Ho’. The mature cap shape of new strain ‘Seolgang’ is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 41.2 mm on average. In comparison with white strain ‘505 Ho’, the strain had a yield that was 9% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 19% firmer with a good shelf life. Days of fruiting body were 3-4 days later than those of ‘505 Ho’. The physical characteristics such as elasticity, chewiness, adhesiveness were better than that of ‘505 Ho’. Genetic analysis of the new strain ‘Seolgang’ showed different profiles compared to ‘505 Ho’, CM02913-27, SSU413-31, when RAPD primers A02 and O04 were used.

        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

        In this paper, the relation between the local stress and the fatigue behavior at gusset plate connected with main girder and floor beam in arch bridges was obtained. The conclusion was as follows : (1) In-plane bending moment and out-of-plane bending moment are higher than other internal stress resultants. (2) When a vehicle crosses the bridges, the change of out-of-plane bending moment corresponds with the change of relative difference in displacement in the longitudinal direction between the tie-girder and stringer. (3) The change of in-plane bending moment corresponds with the change of the displacement in the transverse direction of arch-rib. (4) Fatigue crack can be initiated by in-plane bending stress that Is produced by out-of-plane bending moment at the toe of gusset plate. (5) Fatigue crack can be initiated by tension stress that is produced by in-plane bending moment at the toe of gusset plate.

        최승자 시의 애도 주체와 젠더 정치학

        본 논문은 최승자 초기 시에서의 ‘애도의 주체’를 분석하면서 그 미학적 특이성과 젠더 정치학의 문제를 제기하고자 한다. 최승자의 시는 애도의 주체가 ‘낳은 주체’ 즉 존재를 ‘생성’하고 ‘이행’시키는 존재가 되는 시적인 과정을 드러낸다. ‘너’와 함께 한 시간의 상실 앞에서 시적 주체는 기억을 보존하는 방식으로 ‘부재’속에 머문다. 기억은 몸에 새겨져 지워질 수 없는 것으로 재구성되고, 기억의 주체는 그 ‘주체화’의 자리에서 내려와 기억에 대해 수동적인 위치에 처하게 된다. 기억의 윤리학은 몸에 새겨진 시간을 ‘기억할 수밖에 없는’ 존재로서의 ‘이중 애도’의 주체가 된다. 기억의 주체가 ‘낳은 주체’로 전환될 때, 죽어 가는 ‘여성-몸’은 애도의 예외적인 형식을 드러낸다. 죽은 여성의 몸과 ‘사산’된 존재에 대한 애도는, ‘여성-몸’이 만드는 다른 생산의 ‘(불)가능성’으로 전환된다. 애도는 죽음을 넘어서는 ‘여성-몸’의 혁명적인 상상력과 조우한다. ‘낳는 주체’로서의 ‘여성-몸’의 잠재성은, 하나의 존재를 다른 존재로 ‘이행’시키는 것이며, 애도 주체로서의 여성 주체는 통과제의적인 장소가 된다. 최승자의 초기 시의 여성 주체는 시의 언어들 속에서 구성된 애도의 ‘수행자(遂行者)’로서의 시적 주체이다. 이 애도의 주체는 사랑과 기억의 대상을 포기하기를 거부하며, ‘죽은 몸’이 대상을 ‘다시 낳는’ 행위를 통해 대상을 ‘내’ 안에서 재생성한다. ‘낳은 주체’는 모성의 이데올로기로서의 여성을 규정하는 것이 아니라, ‘낳은 행위’ 자체를 통해 애도의 주체를 재구성한다. 정상적인 애도의 경제학이 애도를 탈정치화한다면, 최승자의 시적 주체는 제도화되고 권력화된 젠더 시스템을 파열시키고 애도를 재정치화 한다. This research aimed at re-contextualizing the problem of the esthetic specialty and gender politics while analyzing "the subject of condolence" in Choi, Seung-Ja"s early poem. Choi, Seung-Ja"s poem reveals the poetic process that becomes a being that "creates" and "transmits" the "born subject", i.e. the existence that the subject of condolence gave birth. At the loss of time with "you", poetic subject remains in "absence" in the method of preserving the memory. Memory is recomposed as something that cannot be erased by being carved in body, and the subject of memory stays in the passive position in memory by descending from the position of that "subjectivation". Ethics of memory becomes the subject of "double condolence" as the being that "cannot but remember" the time carved in body. When the subject of memory is changed to "the subject that gave birth", dying "woman-body" reveals the exceptional form of condolence. Condolences on dead woman"s body and "stillborn" being is changed to "(im)possibility" of another production and revolution that "woman-body" makes. Condolence encounters revolutionary imagination of "woman-body" beyond death. Potential of "woman-body" as "the subject that gives birth" "transmits" an existence to another one, and the woman subject as the subject of condolence becomes the place of passage offer. Woman subject in Choi, Seung-Ja"s early poem is the poetic subject as the "performer (遂行者)’ of condolence composed in poetic languages. Subject of this condolence refuses to give up the object of love and memory, and regenerate the object in "me" through the act of ‘giving birth to the object again’ of ‘dead body’. ‘Birth giving subject’ does not define woman category as the ideology of mother, but recomposes the subject of condolence through ‘birth giving act’ itself. If the economics of normal condolence ex-politicizes condolence, Choi, Seung-Ja"s poetic subject ruptures institutionalized and authorized gender system and re-politicizes condolence.
