        A 9 months intact male American bull dog with chief complaints of dry cough, dyspnea and fever was referred to the Veterinary Teaching Hospital, Chungnam National University. Based on the findings of physical examination, radiological examination, and blood and blood chemical examinations, the present patient was diagnosed into pulmonary edema secondary to primary bacterial and aspiration pneumonia. The patient was hospitalized and treated with buscopan, cephazolin and furosemide for 1 week. His activity and appetite was returned to normal and thoracic radiographs revealed that Jung was recovered to the normal condition after treatment for 7 days. The present patient was a case with pulmonary edema showed favorable therapeutic responses to proper medication.

      • HMG-CoA 환원효소 억제제에 의한 ICAM-1 유전자의 발현조절

        Background : ICAM-1 act as one of major adhesion molecules in the atherosclerotic lesion. ICAM-1 expression is mainly regulated at the level of transcription and depend on IFN-γ signal transduction pathway in which the STAT1 transcrption factor is a critical intermediate. IFN-γreceptor not only initiates tyrosine 701 phosphorylation of STAT1 by Jak1 and Jak2, but also phosphorylates serine 727 through the activation of Raf-1/MAP kinases. HMG-CoA reductase inhibitors have anti-atherosclertic effects, beyond normalization of hypercholesterolemia, by directly acting on endothelial cells, macrophages and vascular smooth muscle cells. HMG-CoA reductase inhibitors suppress the synthesis of isoprenoid intermediates such as geranylgeranyl-pyrophosphate or farnesylpyrophosphate. These effects results inhibitors suppress the synthesis of isoprenoid intermediates such as geranylgeranyl-pyrophosphate or farnesylpyrophosphate. These effects results inhibition posttranslational farnesylation and geranyl-geranylation processing of small GTP-binding preoteins and inhibition of normal signaling activities. Method : We made several 5'-deletion constructs of rat ICAM-1 promoter and analyzed the promoter activities by measuring the luciferase activity after transfection into ECV304 cells and smooth muscle cells. We checked the level of total and phosphorylated STAT1 protein by immunoblot analysis using specific antibodies. Results : Lovastatin inhibits IFN-γ-induced ICAM-1 gene expression in the ECV304cell. The cells pretreated with PD98059, MEKK inhibitor showed significantly low ICAM-1 RNA induction with IFN-γ stimulatio. IFN-γ induced phosphorylation of tyrosine 701 was not significantly changed by the pretreatment of lovastatin. But lovastatin suppresses IFN-γ-induced phosphorylation of ERK1/ERK2 which are responsible for the seine 727 phosphorylation in STAT1. Conclusion : We showed that HMG-CoA reductase inhibitors, lovastatin, suppresses IFN-γ mediated ICAM-1 gene expression through the inhibition of transcription. HMG-CoA reductase inhibitor suppresses IFN-γ-induced phosphorylation of serine 727 in STAT1 through the modulation of MAP kinases.

        야뇨증 환아들의 심리사회적 특성에 대한 다기관 연구 : 행동 및 정서 문제를 중심으로

        Objectives : The aim of this study was to examine the behavioral and emotional problems associated with nocturnal enuresis in Korean children. Methods : Three hundred eighteen children with nocturnal enuresis, together with their parents, completed the Child Behavior Checklist (CBCL), Disruptive Behavior Disorder Scale according to DSM-IV (DBDS), Children's Depression Inventory (CDI), State-Trait Anxiety Inventory for Children (STAIC), and Piers-Harris Children's Self-Concept Scale (PHCSC). Ninety-three normal students were selected as the control group. Results : Compared to the normal control group, the mean scores with regard to the withdrawn, social problems, attention problems, delinquent behavior, aggressive behavior, externalizing problems and total problems profiles were significantly higher in the nocturnal enuresis group according to the CBCL results. The nocturnal enuresis group also scored significantly higher in the ADHD and ODD profiles of the DBDS. The nocturnal enuresis group was more depressed and anxious than the control group according to the results of the CDI and STAI. The mean score of the PHCSC was significantly lower in the nocturnal enuresis group when compared to the normal control group. Conclusion : The results of this study suggest that children with nocturnal enuresis in Korea have clinically relevant behavioral and emotional problems. The findings support the link between nocturnal enuresis and psychopathology in Korean children.

      • 갑상선 세포에서 전사보조활성인자인 CBP와 CIITA의 인터페론-감마 활성 부위에 대한 조절 : CBP and CIITA on GAS(Interferon- γ Activated Site)-Mediated Transcription in Thyroid Cells

        Background : In the previous stuides, we identified that the interferon-r activated sequence (GAS) in the 5-flanking region of rat ICAM-1 gene is major element for interferon-r-inducible expression of the gene in rat thyroid cells, FRTL-5. We here, investigated the role of transcriptional coactivators, CBP(CREB binding protein) and CⅡTA (class Ⅱ transactivator) in the modulation of the activity of GAS which could interacts with signal transducers and activators of transcription-1 and 3 (STAT1 and STAT3). Methods : The expression of CBP RNA and protein were quantitated in FRTL-5 after stimulation with interferon-r (IFN-r), thyroid stimulating hormone (TSH), forskolin and methimazole. Direct association of CBP with STAT were analyzed by immunoprecipitation. The transcriptional roles of CBP and CⅡTA in the regulation of GAS were assessed by the cotransfection with their expression vectors with reporters; 5-deletion constructs of rat ICAM-1 promoter or 8xGAS-luc constructs, into FRTL-5 thyroid cells. Results : The level of CBP RNA and protein were not changed by the treatment with TSH, IFN-r, forskolin and methimazole in FRTL-5, FRT and BRL liver cells. The CBP could be directly associated with STAT1. Furthermore, the overexpression of CBP significantly increases the both promoter activities; rat ICAM-1 gene promoter which has GAS element and 8xGAS-luc cassette constructs. However, the cotransfection of CⅡTA decreased the constitutive and CBP-mediated transactivation of rat ICAM-1 promoter and 8xGAS-luc cassette constructs. Conclusion : We identified that the two tanscriptional coactivators; CBP and CⅡTA has differential roles in the regulation of transcriptional activity of GAS drived promoter. CBP increases the GAS activity through the direct binding with STAT1, but CⅡTA inhibited the CBP-mediated transactivation of GAS activity.

      • 고기능 치재용 시멘트 및 세척용 화학용액 개발

        치과용 도구세척을 위한 화학용액을 개발하기 위하여, 상용 치과재료용 화학용액의 주성분을 바탕으로 분해성능에 영향을 미치는 성분, 조성, 조건에 대한 연구를 수행하였다. 분해성능은 온도가 높을수록, 초음파의 세기가 클수록 향상되었다. 화학용액에 첨가된 계면활성제는 분해를 촉진시켰으며 계면활성제의 종류에 따라 부내성능에 큰 차이를 보였다. 알콜류는 분해성능을 저하시키는 나타났으며 시멘트류는 NaOHtndyddor에, 석고류는 sodium bicarbonate 수용액에서 뛰어난 분해성능을 보였다. Chemical solutions were prepared and the effects of chemical composition of decomposition ability of cleaning solution for dental instruments were studied. With high temperature and ultrasonic intensity, decomposition ability of cleaning solution were improved. Certain surfactants(Miwon Commercial Co. Ltd) such as amine oxides and alkanol amides accelerated decomposition rate of cleaning solution. Alcohols added to solution made worse the decomposition capability of cleaning solution. Dental cements and gypsums were well decomposed in sodium hydroxide aqueous solution and sodium bicarbonate aqueous solution respectively.

      • 인체 대장암에서 72kDa 제 4형콜라겐 분해효소의 발현

        수술 절제한 대장암 45예에서 Northern blot analysis와 gelatin zymography를 시행하여 72kDa 제 4 형콜라겐 분해효소의 mRNA치와 활성도를 조사하였다. 대장암에서 주위 정상조직에 비해 이 효소의 mRNA치가 증가하였으며 단백질 합성단계에서 효소의 활성도와도 연관되었다. 특히 활성형인 62kDa의 활성도가 크게 증가하였다. 암진행에 따른 이 효소의 활성도 변화는 T2와 T3에서 암조직과 정상조직사이에서 mRNA치와 효소활성도에 차이가 있었으나 전반적으로 암의 침윤도, 림프절 전이, 듀크스 병기에 따른 유의한 차이는 볼 수 없었다. 암침윤과 전이에는 제 4 형콜라겐 분해효소 외에 딴 metalloproteinase들, plasminogen activator등 여러 가지 다른 단백질 분해효소도 관여하며, 암종류에 따라 이들 효소의 작용정도가 차이가 날 수 있다. 또 조직 억제제인 tissue inhiitors of metalloproteinase(TIMPs)와의 균형관계도 고려해야 한다. 그러므로 대장암에서 72kDa 제 4 형콜라겐 분해효소의 증가된 활성양상을 이들 다른 인자들과 함께 관찰하면 암의 진행정도를 나타낼 수 있는 보다 뚜렷한 지표를 만들 수 있을 것으로 생각된다. Type Ⅳ collkagenases are thought to play a role in the proteolysis of the basement membrance and the extracellular matrix by the malignant cells that occur early in the development of invasive tumors. In several human tumors, including breast, colonic, ovarian, and gepatocellular carcinomas, Type Ⅳ collagenase expression has been positively correlated with tumor invasiveness. In this study, we examined the expression and the activation of 72kDa Type Ⅳ collagenase in cancerous and normal tissues of the colon and, we attempted to clarify its clinical significance in 45 cases of human colorectal cancer by using northern blot analysis and gelatin zymography. This study showed that the level of 72kDa Type Ⅳ collagenase mRNA was increased in colorectal cancer and that this increase was associated with increased enzymatic activity. While there was a difference in the expression and the activation of this enzyme between T2 and T3, this enzyme activity did not show a definite correlation with cancer progress as measured by the depth of tumor invasion and lymph node metastasis.

      • 신생아 황달질환에서의 RDW의 진단적 의의

        목적 : 신생아 적혈구는 그 모양이 불규칙하여 여러 측정치가 성인 및 소아와 다르다, 저자들은 국내에서 보고된 바 없는 신생아의 Red cell distribution width(RDW)를 조사하여 신생아 정상 측정치를 제시하는 동시에 신생아 시기의 적혈구에 손상을 미치는 질환의 RDW를 분석하여 진단상 의의를 조사하였다. 대상 및 방법 : 신생아 집중치료실에 입원한 만삭아중 감염소견이 없었던 78명과 ABO부적합증 및 패혈증 환아에서 hemoglobin, hematocrit, MV, RDW 및 망상적혈구를 측정하였다. 결과 : 생후 1일부터 7일까지의 만삭신생아의 RDW 17.4±1.5로 소아 정상치 13.4±1.2%에 비해 증가되어 있었으나 출생후 7일까지 hemoglobin, hematocrit는 의의있게 감소한 반면 RDW는 변동이 없었다. 같은 연령의 ABO 부적합증 및 패혈증 환아군은 대조군에 비해 의의있게 hemoglobin과 hematocrit가 낮았으나 RDW는 대조군과 질병군 및 각 질병군 사이에 차이를 보이지 않았다. RDW와 망상적혈구는 ABP 부적합증에서만 상관계수 0.91 (p<0.001)의 상관관계를 보였다. 결론 : 출생후 7일까지 계측한 정상 신생아의 RDW는 소아에 비해 증가되어 있으나 출생후 변화는 없었다. ABO 부적합증 및 패혈증 환아군의 RDW는 대조군과 질병군 및 각 질병군 사이에 차이를 보이지 않아 이들 질환의 진단적 의의가 없었다. ABO 부적합증에서 RDW가 망상적혈구 수와 높은 상관관계를 나타내므로서 자동 분석기로 측정된 RDW로서 망상적혈구 수를 추정할 수 있었다. The RBC distribution width(RDW) has been reported to be of value in the early and differential diagnosis of several RBC disorders, with no sufficient studies on the newborn population. Seventy-eight normal fullterm newborns were studied to establish normal values for RDW of 1st week of life. The RDW of 43 patients with ABO incompatibility or sepsis was then evaluated for the usefulness of the RDW in diagnosis of Jaundice. The RDW was 17.7 ± 1.2 and 17.5 ± 0.8 at 1st and 4-7th days of life, revealing no differences de pending on the postnatal age. The RDW of newborns with ABO incompatibility, sepsis with and without jaundice were 18.4 ± 2.2, 18.2 ± 1.1 and 17.3 ± 0.9 respectively. The RDW was found to be consistently elevated in all these newborn infants-the normal newborns and the newborns with ABO incompatibility or sepsis-when compared with normal older children at our hospital. That reveals a physiologic state of anisocytosis in the newborn, showing no significant differences between these infants. Our results suggest that RDW alone cannot be used as an indicator to distingish between jaundice induced by hemolysis and by other causes. In this study all parameters examined except the reticulocyte counts of ABO incompatibility, which showed, by regression analysis, no correlation with the RDW. High RDW in ABO incompatibility is consistent with high reticulocyte count. One clinical value of the RDW therefore may lie in its capacity for reflecting active erythropioesis in ABO incompatibility. Thus the study has confirmed that red blood cell anisocytosis, as determined by RDW, has no value to differentiate the etiology of jaundice in the newborn period but it seems that RDW plays a role in determining the reticulocyte count in newborns with ABO incompatibility.
