        鹿脛骨이 卵巢摘出로 誘發된 흰쥐의 骨多孔症에 미치는 影響

        The purpose of this study is to examine what are the effects of the Deer Tibia on the postmenopausal osteoporosis. In order to observe the effects of the Deer Tibia on the osteoporosis induced from ovariectomy, administered the Deer Tibia in p.o to the ovariectomized Rats, and measured the variance of weight related to osteoporosis, the variance of osteocalcin, calcium, ALP, phosphorus, GOT and GPT in serum, the variance of creatinine, calcium to creatinine rate in urine and urinary volume, the variance of the organs in weight and the variance of the bones. The results of the experiment are as follows: 1. The level of serum ALP, S-GOT and S-GPT in Deer Tibia group was showed a significant decrease in comparison with control group. The level of serum osteocalcine, calcium, phosphorus in Deer Tibia group was showed no efficiency in comparison with control group. 2. The level of urinary volume in Deer Tibia group was showed a significant increase in comparison with control group, and calcium to creatnine ratio in Deer Tibia group showed a significant increase. 3. The weight of tibia ash and the length of femur in Deer Tibia group showed no significant change. The bone density in Deer Tibia group showed a significant increased in comparison with control group. 4. Body weight in Deer Tibia group was increased in comparison with control group, but showed no efficiency. The weight of heart, liver, spleen, kidney and uterus in Deer Tibia group showed no significant change in comparison with control group. According to the above results, effects of the Deer Tibia on the aged ovariectomized rat model showed a efficiency in elevation of bone density by increasing bone mass and in prevention of renal function failure by ovariectomy. So Deer Tibia can be used for prevention and curing of the postmenopausal osteoporosis, as for it's effects, continuous clinical study is needed.

        구척이 흰쥐의 장골 길이성장에 미치는 영향

        In this study, we searched some herbal medicines on promoting the heights of longitudinal bone and found that the root of Cibotium barometz promotes longitudinal bone growth. The longitudinal bone growth was measured in the proximal tibia by fluorescence microscope with tetracycline. Tetracycline plays a chelate with calcium ion which is accumulated in bone forming-zones. The accumulated tetracycline made fluorescent lines corresponding to two injections. The growth rate of bones are evaluated by measuring the length between two lines. The longitudinal bone growth rate of control group for two days was 489.0±70.1 ㎛ but, C. barometz administration significantly promoted the longitudinal bone growth(100 ㎎/㎏, 764.2±41.1 ㎛, p<0.01). And the heights of proliferative and hypertrophic zones were also increased in C. barometz administered group measured by Nissl staining. To evaluate its growth promoting mechanisms, the induction of local insulin-like growth factor-1(IGF-1) and bone morphogenetic protein-2(BMP-2) were measured by using immunohistochemical method. C. barometz increased local induction of BMP-2 in hypertrophic and ossification zones, whereas IGF-1 did not increase. These results showed that administration of the extracts of C. barometz promoted longitudinal bone growth and its mechanism could be explained in part by regulation of BMP-2 induction in growth plates.

        다엽(茶葉)의 4 - VO 로 유발한 흰쥐뇌허혈의 신경방어효과

        Green tea(茶葉), the leaves of Camellia sinensis (L.) O. Kuntze (Thea sinensis L.), whose property, flavour and channel tropism is bitter and sweet in flavour(苦甘), cool in property(?), acting on the heart, lung, stomach and kidney channels(歸心???經), has the effects of clearing the head and eyes(淸?目), removing the thirst(???), digesting the food(?食), resolving the dampness(化?), facilitating the urination(利?), neutralizing a poison(解?). And this is also one of the most popular beverages in the world. So, this study was planned to check the neuroprotective effect of green tea on the global ischemia induced by 4-vessel occlusion in Wistar rats. Tea extract was lyophilized after extraction with water. We induced 4-vessel occlusion for 10 minutes and reperfused again. The number of CA1 pyramidal neurons were counted after 7 days of reperfusion under the cresyl violet staining. The results obtained that in 4-VO ischemia, Green tea showed significant neuroprotective effects(20 and 100 ㎎/㎏ of green tea extracts, p<0.05 and p<0.001 respectively) compared with control group. Each neuroprotective ratio was about 53.8%, 97.6% respectively. We estimated the amount of the main component (-)epigallocatechin gallate(EGCG) by HPLC, found out its net content in extract was about 9%. However, EGGG given at 10 ㎎/㎏ had HPLC, found out its net content in extract was about 9%. However, EGGG given at 10 ㎎/㎏ had no effect on global ischemia, indicating that EGGG may not be the only main active component of green tea. Immunohistochemical data partially explained the protective effects of green tea via promotion of bcl-2 induction in hippocampus. Consequently, green tea has neuroprotective effects on the global ischemia induced by 4-vessel occlusion in Wistar rats. This work was supported by Korea Research Foundation Grant.         麻黃의 사람 비점막 섬유아세포 monocyte chemotactic protein 중 MCP - 1 , MCP - 2 , 및 MCP - 3 분비에 대한 효과

        Recent reports have proposed that Ephedrae Herba may modulate the immune response on allergy or asthma. Human nasal mucosal fibroblasts are a rich source cytokines, inflammatory mediators, and chemokines. Chemokines are important for the recruitment of leukocytes to sites of infection, which is essential in host defense. Objectives : The objective of this study was to investigate the effect of Ephedrae Herba on the release of the chemokines such as monocyte chemotactic protein(MCP)-1, MCP-2, and MCP-3 in human nasal mucosal fibroblasts after stimulation with cytokines like tumor necrosis factor -α(TNF-α), interferon-γ(IFN-γ), and interleukin -1β(IL-1β).

        사람 비만세포주에서 사이토카인발현에 대한 茶葉 主成分 Epigallocatechin - 3 - Gallate 의 억제효과

        The human mast cell is an effector cells in allergic inflammation and secrete a number of cytokines. Many allergic inflammations could be reduced with green tea treatments. The aim of this study was to investigate the potential effects of epigallocatechin-3-gallate(EGCG), the main polyphenol component in green tea, on pro-inflammatory cytokine productions(IL-1 β, IL-6, TNF-α) and immune-regulating cytokine productions that promote the growth and differentiation of lymphocytes(IL-2, IFN-γ) in I uM calcium-inonphore-A23178-activated human mast cell line HMC-1. Cytokines are immune regulating proteins secreted by many types of cells in the body. The effects of cytokines are pleiotypic actions on the immune system and inflammation. In this study, the effect of EGCG on the expression of cytokines was examined by RT-PCR using the human mast cell line(HMC-1), which is known to secrete and expressed, and expression of IL-2 and IFN-γ were also increased. In order to investigate the protective effect of EGCG, HMC-1 cells were incubated with pretreatment of EGCG for 24 h. RT-PCR analyses of cytokine genes of cells pretreated with EGCG showed that expressions of IL-1 β, TNF-α, IL-6 and IL-2 genes in these cells were lower and IFN-γ was similar pattern compared to the calcium ionophore-treated group. This study would provide fundamental data on the possibility of the clinical treatment of green tea in immune diseases such as allergic disorder, asthma and atopic dermatitis, etc.

        조구등(釣鉤藤)의 4-VO로 유발한 흰쥐뇌허혈에 대한 신경방어효과

        Uncariae Ramulus et Uncus(UR, ???) has sweet in flavour and slightly cold in property, acting on the liver and pericardium channels. This drug was described in a medical classic as having the ability to remove "heat", check hyperfunction of the liver and relieve dizziness, tremors, and convulsion, and subdue "endogenous wind". So this study was planned to check the neuroprotective effect of UR on the global ischemia induced by 4-vessel occlusion in Wistar rats. and UR extract was lyophilized after extraction with 70% methanol. We induced 4-vessel occlusion for 10 minutes and reperfused again. The number of CA1 pyramidal neurons were counted after 7 days of reperfusion under the cresyl violet staining. In 4-VO ischemia model, UR showed significantly neuroprotective effects(1,000 and 500 ㎎/㎏ of UR extracts, p<0.05) compared with control group. Each neuroprotective ratio was about 23.0%, 19.0% respectively. Consequently, Uncariae Ramulus et Uncus has neuroprotective effects on the global ischermia induced by 4-vessel occlusion in Wistar rats. So we expect that Uncariae Ramulus et Uncus can be used as a drug for neurodegenerative disease.

        황금(黃芩)의 4-VO로 유발한 흰쥐뇌허혈에 대한 신경방어효과

        Scutellariae Radix(SR), the root of Scutellaria baicalensis G_EORGI, whose property, flavour and channel tropism is bitter in flavour, cold in property(寒), acting on the lung, gallbladder, stomach and large intestine channels(歸?,?,?,大??), has the effects of clearing away heat and dampness(淸熱??), purging fire, detoxicating(?火解?), stopping bleeding(止血) and preventing miscarriage(安?). This drug is one of the popular drugs in traditional Korean medicine, which has been used to antipyretic, antibacterial, antitoxic and antihypersensitive effects. So this study was planned to check the neuroprotective effect of SR on the global ischemia induced by 4-vessel occlusion in Wister rats. and SR extract was lyophilized after extraction with 70% methanol. We induced 4-vessel occlusion for 10 minutes and reperfused again. The number of CA1 pyramidal neurons were counted after 7 days of reperfusion under the cresyl violet staining. The result obtained that in 4-VO ischemia, SR showed significantly neuroprotective effects(1,000 and 500 ㎎/㎏ of SR extracts, p<0.05) compared with control group. Each neuroprotective ratio was about 27.4 %, 23.2 % respectively. Consequently, Scutellariae Radix has neuroprotective effects on the global ischemia induced by 4-vessel occlusion in Wistar rats.

        數 種 韓藥의 아세틸콜린에스터레이즈 酵素活性에 대한 효과

        아세틸콜린에스터레이즈(AChE)는 신경조직에 분포하는 효소로 신경전달물질 중 아세틸콜린(ACh)을 분해하는 효소로 시냅스 신호전달체계에 있어 중요한 역할을 하는 효소이다. 현재 알츠하이머병과 치매치료에 사용하는 서양약물은 AChE의 활성을 저해하는 작용기전을 가진다. 목적 : 전통적으로 기억력 감퇴, 치매 등 치료에 사용되어 온 한약의 AChE에 대한 작용을 밝히기 위해 본 실험이 구상되었다. 방법 : 생쥐 대뇌신경세포의 균질 혼합액으로부터 효소의 활성을 Ellman 방법을 개선한 엘리자(ELISA)법을 이용하여 측정하였다. 결과 : 실험결과 天花? 의 6종이 2.5㎎/㎖의 농도에서 우수한 효소활성저해력을 보였고, 50% 효소활성 저해농도(ID_50)를 구한 결과, 天花?의 ID_50은 1.5㎎/㎖이었고, ??? 1.2 ㎎/㎖, ?? 2.0 ㎎/㎖, ??草 1.2 ㎎/㎖, ??? 1.0 ㎎/㎖, ??生 1.7㎎/㎖, 및 大?皮 2.0 ㎎/㎖를 나타내어 忍冬?의 저해력이 가장 우수하였다. 결론 : 치매 및 기억력감퇴 증상을 치료하는 한약의 작용기전 중 AChE 효소활성억제효과는 忍冬? 의 6종에서 나타났으며, 이는 전통적인 한방치료에 과학적인 기초자료를 제공할 것으로 생각한다.

      • 사람 비만세포주에서 Clozapine과 Fluoxetine에 의한 케모카인 및 케모카인 수용체의 상이한 발현

        Objectives : It has been suggested that the immune system has a role in the pathophsiology of neuropsychiatric diseases. There are some reports that clozapine and fluoxetine affect on cytokine networks. Recently, it has been known that chemokines have chemotactic effect, and modulate a number of biological responses including the process of inflammation and the maintenance of CNS homeostasis. In the present study, we investigated the effect of clozapine or fluoxetine on the expression of chemokines and their receptors. Methods : Human mast cells(HMC-1) was incubated with phorbol myristate acetate(PMA, 100ng/㎖) and calcium ionophore A23187(350ng/㎖) for 3 hours. Colzapine(10^-7M) or fluoxetine(10^-7M) were pretreated for 1 hour. RT-PCR was performed for the detection of expression of various chemokines and chemokine receptors. Results : after treatment of clozapine, the expression of MCP-1, MIP-1β, CCR3,CXCR2, CXCR3 and CXCR4 were lower than the PMA/Ca ionophore-treated group, while the expression of MIP-1α, RANTES and IL-8 were not changed. However, fluoxetine was not affected the changes of expression of various chemokines and chemokine receptors. Conclusion : This result indicates that clozapine may affect on the expression of chemokines and chemokine receptors. Therefore, this study could, in part, provide the important basic data on the explanation of side effects of clozapine, such as fever and pancreatitis.

        Effects of Meretrix Extracts on the Collagenase Activity and Procollagen Synthesis in HS68 Human Fibroblasts and Tyrosinase Activity

        This study was designed to investigate the collagen metabolism and tyrosinase activity of Meretrix extracts (ME). The effect of ME on type I procollagen production and collagenase activity in human normal fibroblasts HS68 after UVB (312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment of ME was measured as well. Type I procollagen production was recovered by ME in UVB damaged HS68 cells. The increased collagenase activity after UVB damage was significantly recovered by ME. The tyrosinase activity and L-DOPA oxidation were significantly reduced as well. However, the effects on tyrosinase activity and L-DOPA oxidation were not powerful enough to be used as whitening agents. ME showed the anti-wrinkle effects and some whitening effects in vitro. These results suggest that ME may be a useful drug as an anti-wrinkle treatments.
