<P>Macrophages are crucial in controlling infectious agents and tissue homeostasis. Macrophages require a wide range of functional capabilities in order to fulfill distinct roles in our body, one being rapid and robust immune responses. To gain insight into macrophage plasticity and the key regulatory protein networks governing their specific functions, we performed quantitative analyses of the proteome and phosphoproteome of murine primary GM-CSF and M-CSF grown bone marrow derived macrophages (GM-BMMs and M-BMMs, respectively) using the latest isobaric tag based tandem mass tag (TMT) labeling and liquid chromatography-tandem mass spectrometry (LC-MS/MS). Strikingly, metabolic processes emerged as a major difference between these macrophages. Specifically, GM-BMMs show significant enrichment of proteins involving glycolysis, the mevalonate pathway, and nitrogen compound biosynthesis. This evidence of enhanced glycolytic capability in GM-BMMs is particularly significant regarding their pro-inflammatory responses, because increased production of cytokines upon LPS stimulation in GM-BMMs depends on their acute glycolytic capacity. In contrast, M-BMMs up-regulate proteins involved in endocytosis, which correlates with a tendency toward homeostatic functions such as scavenging cellular debris. Together, our data describes a proteomic network that underlies the pro-inflammatory actions of GM-BMMs as well as the homeostatic functions of M-BMMs.</P>

Oroxylin A is a flavone isolated from a medicinal herb reported to be effective in reducing the inflammatory and oxidative stresses. It also modulates the production of brain derived neurotrophic factor (BDNF) in cortical neurons by the transactivation of cAMP response element-binding protein (CREB). As a neurotrophin, BDNF plays roles in neuronal development, differentiation, synaptogenesis, and neural protection from the harmful stimuli. Adenosine A2A receptor colocalized with BDNF in brain and the functional interaction between A2A receptor stimulation and BDNF action has been suggested. In this study, we investigated the possibility that oroxylin A modulates BDNF production in cortical neuron through the regulation of A2A receptor system. As expected, CGS21680 (A2A receptor agonist) induced BDNF expression and release, however, an antagonist, ZM241385, prevented oroxylin A-induced increase in BDNF production. Oroxylin A activated the PI3K-Akt-GSK-3β signaling pathway, which is inhibited by ZM241385 and the blockade of the signaling pathway abolished the increase in BDNF production. The physiological roles of oroxylin A-induced BDNF production were demonstrated by the increased neurite extension as well as synapse formation from neurons. Overall, oroxylin A might regulate BDNF production in cortical neuron through A2A receptor stimulation, which promotes cellular survival, synapse formation and neurite extension.

Ulcerative colitis (UC) is an inflammatory bowel disease, which is a chornic gastrointestinal disorder. Zizyphus jujube (Z. jujube) has been used as a traditional medicine for treatment of inflammation. However, the regulatory effect of Z. jujube in intestinal inflammation have not been understood. The aim of this study is to investigate the protective effects of Z. jujube on dextran sulfate sodium (DSS)-induced colits. The colitis mice were induced by drinking water containing 5% DSS for 7 days. Our findings showed that Z. jujube attenuated the DSS-induced loss of body weight and shortening of colorectum and improved DAT score. Additionally, Z. jujube inhibited the interleukin (IL)-6 levels in colon tissue, which were elevated in DSS-treated group. Taken together, we provide experimental evidence that Z. jujubemight be a useful therapeutic drug for patients with UC.

<P>Si film electrodes were fabricated on surface-modified Cu current collectors (foils) by using an oxidation-reduction process and their electrochemical properties were investigated with various preoxidation times of 1, 3, and 6 h. The reduction of the preoxidized Cu foil formed Cu2O for 1 hand 3 h-preoxidized Cu foils and Cu for 6 h-preoxidized Cu foil as the reductive products on the surface, leading to the changes in the shape of nanostructures and to the decrease in the surface roughness. The Si film electrode with 1 h- and 3 h-preoxidized Cu foils exhibited the rapid capacity fading due to the presence of Cu2O. The Si film electrode with the 6 h-preoxidized (fully reduced) Cu foil demonstrated the good cycle performance, since the coral-like Cu nanostructures enhanced the adhesion with the Si film and the structural stability of the Si film electrode during the electrochemical reaction.</P>

A decrease in pass ratio of Korean exporting pears causes several negative effects including an increase in pesticide dependency. In this study, we attempted to establish the pest forecasting management system, composed of weekly field forecasting by pear farmers, meteorological data obtained by automatic weather station (AWS), newly designed internet web page (http://pearpest.jnu.ac.kr/) as information collecting and providing ground, and information providing service. The weekly field forecasting information on major pear diseases and pests was collected from the forecasting team composed of five team leaders from each pear exporting complex. Further, an abridged weather information for the prediction of an infestation of major disease (pear scab) and pest (pear psylla and scale species) was obtained from an AWS installed at Bonghwang in Naju City. Such information was then promptly uploaded on the web page and also publicized to the pear famers specializing in export. We hope this pest forecasting management system increases the pass ratio of Korean exporting pears throughout establishment of famer-oriented forecasting, inspiring famers` effort for the prevention and forecasting of diseases and pests occurring at pear orchards.

During the prostate cancer (PCa) development and its progression into hormone independency, androgen receptor (AR) signals play a central role by triggering the regulation of target genes, including prostate-specific antigen. However, the regulation of these AR-mediated target genes is not fully understood. We have previously demonstrated a unique role of HOXB13 homeodomain protein as an AR repressor. Expression of HOXB13 was highly restricted to the prostate and its suppression dramatically increased hormone-activated AR transactivation, suggesting that prostate-specific HOXB13 was a highly potent transcriptional regulator. In this report, we demonstrated the action mechanism of HOXB13 as an AR repressor. HOXB13 suppressed androgen-stimulated AR activity by interacting with AR. HOXB13 did neither bind to AR responsive elements nor disturb nuclear translocation of AR in response to androgen. In PCa specimen, we also observed mutual expression pattern of HOXB13 and AR. These results suggest that HOXB13 not only serve as a DNA-bound transcription factor but play an important role as an AR-interacting repressor to modulate hormone-activated androgen receptor signals. Further extensive studies will uncover a novel mechanism for regulating AR-signaling pathway to lead to expose new role of HOXB13 as a non-DNA-binding transcriptional repressor.
