Development and validation of FRAT code for coated particle fuel failure analysis

TRISO-coated particle fuel is widely used in high temperature gas cooled reactors and other advanced reactors. The performance of coated fuel particle is one of the fundamental bases of reactor safety. The failure probability of coated fuel particle should be evaluated and determined through suitable fuel performance models and methods during normal and accident condition. In order to better facilitate the design of coated particle fuel, a new TRISO fuel performance code named FRAT (Fission product Release Analysis Tool) was developed. FRAT is designed to calculate internal gas pressure, mechanical stress and failure probability of a coated fuel particle. In this paper, FRAT was introduced and benchmarked against IAEA CRP-6 benchmark cases for coated particle failure analysis. FRAT's results agree well with benchmark values, showing the correctness and satisfactory applicability. This work helps to provide a foundation for the credible application of FRAT.
Enhanced β-carotene production by Rhodotorula glutinis using high hydrostatic pressure

High hydrostatic pressure (HHP) technology was used for improving the ability of β-carotene biosynthesis of Rhodotorula glutinis R68. After the treatments of five repeated cycles at 300MPa for 15 min, the barotolerant mutant PR68 was obtained. After 72 h of culture, the biomass of mutant PR68 was 21.6 g/L, decreased by 8.5% compared to the parent strain R68, but its β-carotene production reached 19.4 mg/L, increased by 52.8% compared to the parent strain R68. The result of restriction fragment length polymorphism analysis suggested that mutant strain PR68 was likely to change in nucleic acid level, and thus enhanced β-carotene production in this strain was a result of gene mutation induced by HHP treatment. HHP technology seems a promising approach for improving industrial microbes

High levels of extracellular xylanase activity (211.79 IU/mg) produced by Paenibacillus sp. NF1 were detected when it was submerged-cultured. After three consecutive purification steps using Octyl-Sepharose, Sephadex G75, and Q-Sepharose columns, a thermostable xylanase (XynNF) was purified to homogeneity and showed a molecular mass of 37 kDa according to SDS-PAGE. The specific activity of the purified XynNF was up to 3,081.05 IU/mg with a 14.55-fold purification. The activity of XynNF was stimulated by Ca2+, Ba2+, DTT, and β-mercaptoethanol, but was inhibited by Fe3+, Zn2+, Fe2+, Cu2+, SDS, and EDTA. The purified XynNF displayed a greater affinity for oat spelt xylan with the maximal enzymatic activity at 60°C and pH 6.0. XynNF, which was shown to be cellulose-free, with high stability at high temperature (70°C-80°C) and low pH range (pH 4.0-7.0), is potentially valuable for various industrial applications. The enzyme hydrolyzed oat spelt xylan to yield mainly xylooligosaccharides (95.8%) of 2-4 degree of polymerization (DP2-4). Moreover, the majority of the xylooligosacharides (DP2- 4) products was xylobiose (61.5%). The thermostable xylanase (XynNF) thus seems potentially usefull in the production of xylooligosaccharides.
