        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.         염증성 피부질환 개선 기능 소재로서 둥근전복(Haliotis discus) 유래 항균펩타이드 Ab4-7의 항염증 효과

        서해안에 서식하는 해양생물인 전복은 항산화 및 항염증 효과가 있으며, 항생제 개발 및 화장품 원료 등 다양한 산업에 활용될 수 있는 잠재력을 가진 자원이다. 본 연구에서 우리는 서해안의 다양한 해양 생물중에서 전복을 선택하였으며, 전복에서 분리한 항균펩타이드(AMP)를 이용하여 항균펩타이드 유도체인 Ab4-7를 규명하고 생리활성을 연구하였다. Ab4-7의 항염증 효능을 확인하기 위해 염증이 유도된 RAW 264.7에 Ab4-7을 처리한 결과, 염증성 사이토카인, TLR4, TNF-α, IL-1β, COX-2, iNOS 등을 억제하고 항산화와 관련된 유전인자인 HO-1의 mRNA 발현을 증가시켰다. Ab4-7의 강력한 항산화 및 항염증 효과를 바탕으로, 염증이 유도된 RAW 264.7 세포에서 Ab4-7이 extrocellular matrix metalloproteinase (ECM) 분해에 관여하여 다양한 염증성 피부질환을 유도하는 유전자인 matrix metalloproteinase (MMPs)를 조절하는 효과를 RT-PCR을 통해 확인하였다. Ab4-7은 강력한 항염증 및 항산화 효과를 가지고 있으며, 항염증 및 항산화 효과를 통한 MMPs 관련 유전인자 조절을 통해 다양한 염증성 피부질환 치료제의 기능성 재료로 활용될 수 있다고 사료된다. Abalone, a marine organism inhabiting the west coast of Korea, has antioxidant and anti-inflammatory effects, and is a resource with potential to be used in various industries such as antibiotic development and cosmetic raw materials. In this study, we chose abalone among various marine lives on the west coast. Antibacterial peptide (AMP) was separated from abalone and its derivative Ab4-7 was identified and its physiological activity was studied. The treatment of Ab4-7 in inflammatory RAW 264.7 to check the anti-inflammatory efficacy nhibited inflammatory cytokines, TLR4, TNF-α, IL-1β, COX-2, and iNOS and increased mRNA manifestation of HO-1, genes related to antioxidants. Based on the strong antioxidant and anti-inflammatory effects of Ab4-7, the effects of Ab4-7 in the inflammatory RAW 264.7 cells were identified through RT-PCR, which regulates the gene Matrix Metalloproteinase (MMPs) that induces a variety of inflammatory skin diseases by engaging in the decomposition of the extrocellular matrix metalloproteinase (ECM). Taken together, it is concluded that Ab4-7 has a powerful anti-inflammatory and antioxidant effect and can be used as a functional material for various inflammatory skin disease treatments by controlling the genes associated with matrix metalloproteinase (MMPs).

      • 원자로 스터드 볼트의 비파괴 평가를 위한 초음파 Phased Array 기법 적용

        The stud bolt is one of crucial parts for safety of reactor vessels in nuclear power plants. Cracks initiation and propagation were reported in stud bolts using closure of reactor vessel and head. Stud bolts are inspected by ultrasonic technique during overhaul periodically for the prevention of stud bolt failure and radioactive leakage from nuclear reactor. In conventional ultrasonic testing for inspection of stud bolts, crack was detected by using shadow effect. It take too much time to inspect stud bolt by using conventional ultrasonic technique. In addition, there were numerous spurious signal reflected from every thread. In this study, the advanced ultrasonic phased array technique was introduced for inspect stud bolts. The phased array technique provide fast inspection and high detectability of defects. There are sector scanning and linear scanning method in phased array technique, and these scanning methods were applied to inspect stud bolt and detectability was investigated.

        This study departs from the recognition of the absence of public sphere in the present time, when the human beings have become alienated by the rapid growth of technology and the progression of the society. The study intends to reexamine the public through the discourse of 'body' with its extension through media. The argument is focused on investigating the territories extended by the changes in the recognition of body, reviewing the public sphere through the body and its extension and the relationship with the others, and exploring any applicabilities of the new concept of the public sphere into today's city and architecture.

        Remodeling projects are on a increasing trend because of the old buildings built in 1960-70's, the preference for energy saving and the changes of social demands and government policies, etc. And also discount-store projects, which are rapidly increasing, are characterized by the fact that they are repetitive and have analogous scale and structure. Therefore, it is necessary to develop the concept discount-store model and apply the appropriate project delivery system for a successful execution of the projects. The purpose of this study is to analyze the characteristics of the discount-store remodeling projects and propose CM at Risk for the optimum delivery system of those projects.

        Hypereosinophilic syndrome is characterized by persistent blood eosinophilia of 1,500/ul or more in the absence of known causes and multiorgan dysfunction by eosinophil-related tissue damage. In Korea, some cases of hypereosinophilic syndrome with hepatic involvement have been described with prolonged benign clinical courses. Most of them were diffuse or multifocal lesions in imaging studies, and a few case presenting as a single mass has been described. Here we report a case of hypereosinophilic syndrome with hepatic involvement in a 65-year-old man who presented with multifocal mass lesions. By abdominal US and CT scan, multiple, ill-defined nodules were detected in throughout the liver. Liver biopsy revealed severe eosinophilic hepatitis, suggestive of hypereosinophilic syndrome. Hepatic mass lesions improved after steroid administration for several weeks.

      • 말기신부전 환자의 혈색소 순환 : 투석 방식에 따른 양상 비교 및 관련 인자들에 대한 고찰

        혈색소 순환은 말기신부전 환자에서 인체 재조합 적혈구 조혈 인자를 투여하여 빈혈을 치료하는 동안 빈번히 관찰되며 이는 환자의 입원과 사망률과 관계가 있다고 알려져 있다. 이에 저자는 투석중인 말기 신부전 환자를 대상으로 혈색소 순환 양상과 이와 연관된 인자들을 알아보고자 하였다. 혈색소 순환의 관련 인자들을 알아보기 위하여 투석 치료를 받고 있는 환자 40명 (혈액투석 환자군: 20형, 복막투석 환자군: 20명)을 대상으로 투석 시작부터 1년 동안의 환자의 나이와 성별, 투석시작 1년간의 평균 Hb, 수혈 횟수, 총입원 횟수, 감염으로 인한 입원 횟수, 혈청 ferritin 농도, 혈청 transferrin saturation (TSAT), 철분제제 복용 유무, 일주일 동안 투여된 rHuEPO 양, rHuEPO의 변화량, 알부민, C-reactive protein (CRP) 및 입원 원인 등을 후향적으로 조사하였다. 전체 환자 40명 중 33명 (82.5%)에서 혈색소 순환이 관찰되었다. 혈액투석 환자군에 비해 복막투석 환자군에서 혈색소 순환 첫수가 유의하게 높았다 (p=0.037).혈색소 순환 발생군과 혈색소 순환 미발생군의 관련인자들을 비교하였을 경우 혈색소 순환 발생군에서 총 입원 첫수 (p=0.01), 감염으로 인한 입원 횟수 (p=0.036)가 유의하게 많았으며, 혈중 알부민 수치는 유의하게 낮았다 (p=0.003).그러나 두 군간의 나이, 성별, 수혈 횟수, 혈청 ferritin농도, 혈청 transferrin saturation (TSAT) 및 혈청 철분 농도는 유의한 차이가 없었다. 혈색소 순환의 빈도는 일주일 동안 투여된 r-HuEPO변화량과 양의 상관관계를 보였고 (p=0.003),혈중 알부민 수치와 음의 상관관계를 보였다 (p=0.013). 복막투석 환자군에서 혈액투석 환자군에 비해 혈색소 순환이 유의하게 높게 발생하였다. 일주일 동안 투여된 r-HuEPO 변화량, 입원 및 혈청 알부민 수치는 투석 시작부터 1년 동안의 혈색소 순환과 관련이 있었다. Hemoglobin cycling is frequently observed during treatment of anemia with recombinant human erythropoietin (r-HuEPO) in patients with end stage renal disease (ESRD) it is also reported to be associated with hospitalization and morbidity. We analyzed hemoglobin cycling and potential associated factors in patients on dialysis. Forty new dialysis patients (20 hemodialysis, HD and 20 peritoneal dialysis, PD) were recruited during their first year. Age, gender, mean hemoglobin level, frequency of transfusion, frequency of hospital admission, frequency of admission due to infection, serum ferritin, transferrine saturation, iron levels, rHuEPO dose per week, serum albumin, and c-reactive protein were retrospectively evaluated in order to examine the possible correlations with hemoglobin cycling. Thirty-three patients (82.5%) were identified in the hemoglobin cycling group. The PD group had a higher frequency of hemoglobin cycling than did the HD group (p=0.037). The hemoglobin cycling group showed a significantly higher frequency of hospital admission (P=0.001), admission due to infection (p=0.036) and a lower serum albumin level (p=0.003) than the other group, but there was no significant difference between the groups for age, gender, frequency of transfusion, serum ferritin, transferrine saturation, and iron levels. The frequency of hemoglobin cycling showed a positive correlation with the i-HuEPO dose variation per week (p=0.003) and a negative correlation with serum albumin level (p=0.013). The PD group had a higher frequency of hemoglobin cycling than the HD group, and the change of rHuEPO dose per week, hospitalization and serum albumin were associated with the hemoglobin cycling during the first year of dialysis.

      • 졸겔법에 의한 알루미나 박막의 제조 및 특성(Ⅰ) ; 코팅용 알루미나 sol의 합성

        The properties of alumina sol for single layer Anti-Reflective film by dip-coating were investigated. Transparent alumina sol could be obtained by heating the sol within 2 hours after hydrolysis at room temperature. The particle size of sols decreased with increasing the amount of nitric acid, while the specific surface area and the total pore volume of heat treated gels increased, up to the ratio of 0.1 mol nitric acid per 1 mol alkoxide. Excess addition of nitric acid above 0.1 mol resulted in decrement of the specific surface area and the total pore volume, and broadness of the pore size distribution. The average pore diameter and the total pore volume of heat-treated gel at 400℃ with composition of 1 alkoxide·100 water·0.1 nitric acid in ratio were ?? and 0.315 cc/g, respectively.

      • 석탄 슬러리 기포탑에서 초음파에 의한 슬러리 입자의 축방향 분산 촉진

        In a semi-batch coal slurry bubble column(0.10m-IDx1.53 m-height),the effects of gas velocity(0.02-0.10 m/s),apparent slurry viscosity(1.2x10?-3.82x10?Pa·s),concentration of slurry particles(10.04-51.25kg-solid/m³-slurry)and ultrasound on the axial concentration distribution of slurry particles have been determined. Sedimentation-dispersion model and modified sedimentation-dispersion model have been applied to analyze axal concentration distribution of slurry particles. The axial distribution of slurry particles became uniform with increases in gas velocity, apparent slurry viscosity and slurry concentration,and by introduction of ultrasound. The axial dispersion coefficient of slurry particles increased with increasing gas velocity,but it decresed with increasing apparent viscosity of slurry. The magnitude of the axial dispersion coefficient increased to about 2-5 times by using ultrasound in comparison with that without ultrasound. The axial distriution of slurry particles from the experiments agreed well with that obtained from the modified sedimentation-dispersion model, in which the particle size distribution of slurry particles was considerd.
