        Differential Manifestation of Ocular Phenotypes in TALEN-mediated p19arf Knockout FVB/N and C57BL/6J mouse lines

        Background p19arf, primarily known as a tumor suppressor, has also been reported to play an essential role in normal development of mouse eyes. Consistently, lack of p19arf has been associated with ocular defects, but the mixed background of the knockout (KO) mouse strain used raised a concern on the accuracy of the phenotypes observed in association with the targeted gene due to genetic heterogeneity. Object We carried out a study to investigate into the efect of genetic background on the manifestation of p19arf KO associated phenotypes. Methods We characterized the phenotypes of novel p19arf KO mouse lines generated in FVB/N and C57BL/6J using a transcription activator-like efector nuclease (TALEN) system in comparison to the reported phenotypes of three other p19arf-defcient mouse lines generated using homologous recombination. Results Ninety-fve percent of FVB/N-p19arf KO mice showed ocular opacity from week 4 after birth which worsened rapidly until week 6, while such abnormality was absent in C57BL/6J-p19arf KO mice up to the age of 26 weeks. Histopathological analysis revealed retrolental masses and dysplasia in the retinal layer in FVB/N-p19arf KO mice from week 4. Besides these, both strains developed normally from birth to week 26 without increased tumorigenesis except for a subcutaneous tumor found in a C57BL/6J-p19arf KO mouse. Conclusion Our fndings demonstrated surprisingly variable manifestation of p19arf-linked phenotypes between FVB/N and C57BL/6J mice, and furthermore between our mouse lines and the established lines, indicating a critical impact of genetic background on functional study of genes using gene targeting strategies in mice.

      • BEZ235 (PIK3/mTOR inhibitor) Overcomes Pazopanib Resistance in Patient-Derived Refractory Soft Tissue Sarcoma Cells

        <P><I>BACKGROUND:</I> Although pazopanib treatment has become the standard chemotherapy in salvage setting for metastatic sarcoma patients, most patients progress after pazopanib treatment in 4 to 6 months. After failure to pazopanib, patients have limited options for treatment. Therefore, subsequent therapy in patients who failed to pazopanib is urgently needed and the use of patient derived cells or patient derived tumors for accompanying testing with various pharmacological inhibitors could offer additional treatment options for these patients. <I>METHODS:</I> Patient derived tumor cells were collected from ascites at the time of progression to pazopanib and a 13-drug panel was tested for drug sensitivity. We confirmed the results using <I>in vitro</I> cell viability assay and immunoblot assay. We also performed the genomic profiling of PDX model. <I>RESULTS:</I> The growth of patient derived tumor cells was significantly reduced by exposure to 1.0 μM AZD2014 compared with control (control versus AZD2014, mean growth = 100.0% vs 16.04%, difference = 83.96%, 95% CI = 70.01% to 97.92%, <I>P</I> = .0435). Similarly, 1.0 μM BEZ235 profoundly inhibited tumor cell growth <I>in vitro</I> when compared to control (control versus BEZ235, mean growth = 100.0% vs 7.308%, difference = 92.69%, 95% CI = 78.87% to 106.5%, <I>P</I> < .0001). Despite the presence of CDK4 amplification in the patient-derived tumor cells, LEE011 did not considerably inhibit cell proliferation when compared with control (control vs LEE011, mean growth = 100.0% vs 80.23%, difference = 19.77%, 95% CI = 1.828% to 37.72%, <I>P</I> = .0377). The immunoblot analysis showed that BEZ235 treatment decreased pAKT, pmTOR and pERK whereas AZD2014 decreased only pmTOR. <I>CONCLUSION:</I> Taken together, upregulation of mTOR/AKT pathway in sarcoma patient derived cells was considerably inhibited by the treatment of AZD2014 and BEZ235 with downregulation of AKT pathway (greater extent for BEZ235). These molecules may be considered as treatment option in STS patient who have failed to pazopanib in the context of clinical trials.</P>

        초등학교 5학년 실과 『우리 생활과 전기·전자』 단원의 ICT활용 교수-학습과정안 개발

        The purpose of this study was to raise the effectiveness of learning and teaching through developing lesson plans based on ICT for electrical · electronic units in elementary practical arts of 5th grade at elementary school in the 7th national curriculum. This study was conducted by dividing into four stage(the analysis stage, the design stage, the development stage, the evaluation stage) for this purpose. For the analysis stage, after the units required to develop were selected and teaching objectives were identified, objects that lesson plans based on ICT were applied to and teaching-learning setting were grasped and the learners' final behaviors were described in detail and the content analysis were systematized. For the design stage, teaching guidance based on ICT for accomplishing learning objectives was made and its application to elementary practical arts education was invested. For the development stage, Power-Point 2000 as the writing tool which is easy to make out, correct, and edit their presentation was used. For the evaluation stage, After self evaluation and expert evaluation were conducted to identify the problems and content's errors of the developed lesson plans based on ICT, lesson plans were corrected, made up for, and completed by their results. The results of this study tested by the experts were as follows : As these lesson plans were gotten the positive marks more than the average in the three domains of the teaching plan domain, the teaching process domain, and the evaluation & the rest, it showed that they were effective materials giving a high learning drive. According to this, lesson plans based on ICT as materials to raise teaching-learning effectiveness are required.

        This study investigated the relationship between self-esteem, perceived competence and participation of masters swimming. Participants(N=322) were randomly selected from masters swimming program of swimming pool in seoul and Kyung-Kee. Results indicated that the meaning significant found the representation of duration of contingencies and participation of swimming competition in masters swimming. In sum, firstly, in the participation of masters swimming, every time participation in swimming competition assigned greater than the other participant to perceived competence, and in duration of contingencies of participation, 4-7 years participation group assigned greater than the order participant group to perceived competence. Secondly, in the participation of masters swimming, every time participation in swimming competition assigned greater than the other participant to self-esteem, and in duration of contingencies of participation, 2-4 years participation group assigned greater than the participant group to self-esteem. Thirdly, in the participation of masters swimming, perceived competence is postitively influence on self-esteem.

        노인외상환자의 외상중증도계수와 동통감지력의 변화에 대한 임상적 고찰

        Because elderly individuals are participitating in social work as adult inspite of their physical handicapes such as hearing difficulty, visual difficulty and limit of motion, they are continually exposed to the risk of injury. Trauma is now fifth cause of death in elderly of over 65 years old, and the elderly suffer disproportionately high injury-related motality comparing with younger adults. In addition, sometimes elderly patients don`t complaint of pain on injured site because of high threshold to pain. We studied about occurence of neglected injury due to high threshold to pain in geriatric trauma patients. When we diagnosised injury that patients did not complaint of pain on injured site through physical examination and radiologic examination, we defined that as neglected injury. There were 8 (8%) geriatric patients who didn`t complaint of pain on injured site on admission to emergency center, but only 0.7% of adult patients had neglected injury. The severity was estimated by use of AIS(Abbreviated Injury Scale), and the severity of all neglected injuries were above AIS 2 point. The neglected injury were head injuries(4 in number), abdominal injury(1), pelvic injuries(2) and extremity injury(1). So if geriatric trauma patients were admitted to emergency center, we recommand full physical and radiological examinations although patients don`t complaint of pain.

        Locoregional recurrence is the most common type of recurrence in surgical operation of gastric adenocarcinoma, and peritoneal dissemination is one of the most difficult problems in advanced gastric adenocarcinoma treatment. Because the peritoneal cavity is the most common site of the first recurrence after gastric cancer resection, intraperitoneal chemotherapy seems a logical choice for cancer chemotherapy. In this study, Mitomycin C (MMC)-activated charcoal (CH)-alginate (ALG) beads were prepared by the mixtures of CH particles adsorbed with MMC as an anti-cancer drug and aqueous alginate solution. The alginate is recognized as biodegradable, nontoxic, and biocompatible. The release of MMC from the beads in 0.1 M Tris buffer was stable and continuous until about 1 week. The MMC-CH-ALG beads can be applied in the peritoneal cavity for intraperitoneal chemotherapy since they provide a good adhesiveness on the tissue and controlled release pattern of the drugs.

        "Biomaterials" are nonviable materials used in medical devices, intended to interact with biological systems. They should have "biocompativility", which is defined as the ability of materials to perform with an appropriate host response in a specific application. It includes many materials that are often classified as biomaterials. Metals, ceramics, polymers, glasses, carbons, and composite materials are some examples. Among them, biodegradable polymers are discussed in this revies article. Since a biodegradable polymeric implant does not have to be removed surgically once it is no longer needed, biodegradable polymers are of value in short-term applications that require only the temporary presence of a polymeric implant. Main short-term medical applications of biodegradable polymers include the temporary tissue scaffold, the temporary adhesion barrier, the drug delivery device, and the multifunctional implant. In this article, the properties, processing methods, and medical applications of the biodegradable polymers are reviewed.

      • 양배추 배양세포로부터 동조배양을 통한 체세포배 발생

        Some attempts were made in this investigation to establish an experimental system in which synchronous embryogenesis occurs in a suspension culture derived from calli of B. oleracea L. leaf tissue at a high frequency. Embryogenic calli were developed around 4 weeks after being subcultured in the MS medium containing 2, 4-D(0.5∼1.0 mg/l). The cells showed active proliferation 7 days after suspension culture from the calli. Synchronization of cells or cell clusters was achieved by sieving the suspended cells for 7 days in the medium containing 2, 4-D and by controlling KH_2PO_4. From the synchronized cells or cell clusters, uniform embryo formation occured at high frequency(75∼93%), but irregular embryos were formed from non-synchronized cells. In the exclusive treatment of cytokinins(BAP and Kinetin), somatic embryo formation occured more effectively than in the combination of auxin(2, 4-D) and cytokinin.

        The flora of Mt. Baekhaksan(615m) was surveyed in this study from the middle of May till the middle of October, 1999. The vascular plants collected from this mountain are as following: Totally 178 taxa those are 4 forms, 28 varieties, 146 species, 106 genera, 60 families, 28 orders, and 4 phyla.. There are 1 taxa in this mountain known distributed only in northern parts, and no taxa known distributed only in southern parts. And there are 7 taxa of special protected plants which was designated from Ministry Environment of Korea, those are 2 taxa of 4th grade, 1 taxon of 3rd grade, 4 taxa of 1st grade. There are 2 taxa needed more ecological study with detail that are Larix gmelini var. prinsipisruprechtii Pilger and Sorbus amurensis Koehne.

        This paper describes a design of trasducer for non-invasively detecting pressure radial pulse wave in aterial system and a recording system that for the striding the aterial pulse diagnosis of korean traditional medicine. The mechanism of transducer is composed of sensing mechanism, pressure sensor, conditioning amplifier. The variation of radial pulse pressure in the sensing mechanism is converted to the electric signal by piezo-resistive pressure sensor and it converted to the digital signal after preprocessing via A/D converter. The converted signals inputed to the computer as data files and then it display to the monitor for waveform watching and this datas can be used as the aterial pulse diagnosis data. This system effectively detect non-differential radial pulse wave and we conside that if analizing the recorded radial pulse wave, compared each other, it can be helpful in quantify radial pulse wave diagnosis of the Korean traditionl medicine.
