      • MLH1 Polymorphisms and Cancer risk: a Meta-analysis Based on 33 Case-control Studies

        Objective: Cumulative evidence suggests that MLH1, the key component in the mismatch pathway, plays an important role in human cancers. Two potential functional polymorphisms (-93G>A and I219V) of MLH1 have been implicated in cancer risk. The aim of this meta-analysis was to summarize the evidence for associations. Methods: Eligible studies were identified by searching the electronic literature PubMed, ScienceDirect and Embase databases for relevant reports and bibliographies. Studies were included if of case-control design investigating MLH1 polymorphisms (-93G>A and I219V) and cancer risk with sufficient raw data for analysis. Odds ratios (OR) and 95% confidence intervals (95% CI) were used to evaluate the strength of associations. Results: Our meta-analysis from 33 published case-control studies showed the variant A allele of -93G>A polymorphism to be associated with increased risk in all genetic models (AA vs. GG: OR = 1.22, 95% CI: 1.03-1.44), especially among non-Asians (AA vs. GG: OR = 1.28, 95% CI: 1.04-1.58). For the I219V polymorphism, however, there was no main effect associated with overall cancer risk in any genetic model. Conclusions: The meta-analysis suggested that the MLH1 -93G>A polymorphism may be a biomarker of cancer susceptibility. Large sample association studies and assessment of gene-to-gene as well as gene-to-environment interactions are required to confirm these findings.

        Emotional Rationality in Confucian Ethics

        Confucian ethic is characterized by emotional rationality. It conceptualizes emotions, which are subjective and individual, by intelligence. Given this, confucian ethics constructs ethical principles. At the beginning stage, confucian ethic takes “benevolence” as its core, “filial piety” and “fraternal duty” as its beginning, “love the multitude at large” as its goal, and “rest mind” as its ground. Following this, Mencius identifies sympathy as human nature. Therefore, he overcomes the uncertainty of ethical principles bound up with blood relationships, emphasizing that benevolence and love are universal. In Song and Ming dynasties, two schools, which are distinguished in terms of their metaphysics, emerged: one is characterized by “principle”, the other by “mind”. Confucian ethic states that benevolence and love are the characteristics of humans and what makes humans humans, and believes that every human being shares the common emotions and sympathy. In addition, it maintained that benevolence and love are not only possessed by human beings but also the foundation of a confucian metaphysics about the universe. It is therefore concluded that confucian ethic, which highlights emotional rationality, provides people with a variety of moral choices: individuals have diverse comprehensions of benevolence and love, so, there are higher and lower levels of spiritual states. The point is not only to differentiate between moral and nonmoral behaviours, but also to identify which sort of behaviours embodies higher moral value.

        Tourism service quality is the lifeline for the survival of tourism enterprises , which is the core embodiment of the competitiveness of tourism enterprises. Based on the perspective of tourists perceived value, this paper aimed to analyze the quality of rural tourism of Shandong province, it indentified the effect factors of tourist service quality from two research perspective: the impact to the expectations before the tourists travel process and the experience during tourists travel the process. And then, through the analysis of problems existing in the rural tourism service of Shandong province, it put forward the relative countermeasures.

        Fatigue strength predictions of FOD dents using ΔK threshold methods considering residual stresses

        Fatigue strength predictions of TC4 FOD-dent specimens using ΔK threshold methods considering residual stresses were the main emphasis in this paper. Both the experimental simulation and the fatigue test were conducted to evaluate the HCF performance of the real FOD dent. The geometrical models of FOD dents were established for the elastic stress concentration analysis. Residual stresses around the dent were numerical simulated or reservedly solved from the laboratory test data and subsequently established as a function of the dent depth. With numerical simulated residual stresses, the prediction results fall into an error interval of about ±30 %. With the reserved residual stress, the method has a smaller error interval of about ±20 %. Both the two procedures can fortunately meet the exit criteria which is probably because the potential micro-damages like micro crack and adiabatic shear band etc. could be counted as sharp cracks.

      • Genetic Variants of NBS1 Predict Clinical Outcome of Platinum-based Chemotherapy in Advanced Non-small Cell Lung Cancer in Chinese

        Objective: NBS1 plays a key role in the repair of DNA double-strand break (DSB). We conducted this study to investigate the effect of two critical polymorphisms (rs1805794 and rs13312840) in NBS1 on treatment response and prognosis of advanced non-small cell lung cancer (NSCLC) patients with platinum-based chemotherapy. Methods: Using TaqMan methods, we genotyped the two polymorphisms in 147 NSCLC patients. Odds ratios (ORs) and their 95% confidential intervals (CIs) were calculated as a measure of difference in the response rate of platinum-based chemotherapy using logistic regression analysis. The Kaplan-Meier and log-rank tests were used to assess the differences in progression-free survival (PFS) and overall survival (OS). Cox proportional hazards model was applied to assess the hazard ratios (HRs) for PFS and OS. Results: Neither of the two polymorphisms was significantly associated with treatment response of platinum-based chemotherapy. However, patients carrying the rs1805794 CC variant genotype had a significantly improved PFS compared to those with GG genotype (16.0 vs. 8.0 months, P = 0.040). Multivariable cox regression analysis further showed that rs1805974 was a significantly favorable prognostic factor for PFS [CC/CG vs. GG: Adjusted HR = 0.62, 95% CI: 0.39-0.99; CC vs. CG/GG: Adjusted HR = 0.56, 95% CI: 0.32-0.97). Similarly, rs13312840 with a small sample size also showed a significant association with PFS (CC vs. CT/TT: Adjusted HR = 25.62, 95% CI: 1.53-428.39). Conclusions: Our findings suggest that NBS1 polymorphisms may be genetic biomarkers for NSCLC prognosis especially PFS with platinum-based chemotherapy in the Chinese population.

        Synthesis and Characterization of pH and Temperature Sensitive Hydrogel Based on Poly(N-isopropylacrylamide), Poly(ε-caprolactone), Methylacrylic Acid, and Methoxyl Poly(ethylene glycol)

        In this paper, a novel biodegradable and pH/thermo-sensitive hydrogel based on poly(ε-caprolactone),methoxyl poly(ethylene glycol), methylacrylic acid and N-isopropylacrylamide was prepared by UV-initiated free radical polymerization. The hydrogels were characterized by Fourier transforms infrared ray. The thermal responsibility was investigated with the help of differential scanning calorimetry. Swelling behavior in aqueous medium with different pH value was studied in detail. When the pH value of the aqueous medium was increased from 1.2 to 7.2,the swelling ratio of the hydrogels increased accordingly. The morphology was observed by scanning electron microscopy, and the hydrolytic degradation behavior in different aqueous media (pH 1.2 and pH 7.2) was also investigated in detail. The prepared biodegradable pH/thermo-sensitive hydrogel based on poly(ε-caprolactone), methoxyl poly(ethylene glycol), methylacrylic acid and N-isopropylacrylamide hold great promise in the development of a smart drug delivery system.

      • Expression and Significance of Microsomal Prostaglandin Synthase-1 (mPGES-1) and Beclin-1 in the Development of Prostate Cancer

        The aim of this study was to investigate the expression and significance of microsomal prostaglandin synthase-1 (mPGES-1) and Beclin-1 in the development of prostate cancer (PCa). Immunohistochemistry was performed on paraffin-embedded sections with rabbit polyclonal against mPGES-1 and Beclin-1 in 40 PCa, 40 benign prostatic hyperplasia (BPH) and 10 normal prostate specimens for this purpose. Quantitative real-time polymerase chain reaction (qRT-PCR) was applied for mRNA expression of mPGES-1 and Beclin-1, while MTT assays were used to ascertain the best working concentration of the mPGES-1 inhibitor (CAY10526). The effect of CAY10526 treatment on expression of Beclin-1 in DU-145 cells was studied using Western blot analysis. Localization of Beclin-1 and mPGES-1 was in endochylema. Significant differences in expression was noted among PCa, BPH and normal issues (P<0.05). Beclin-1 expression inversely correlated with mPGES-1 expression in PCa tissue (P<0.05). CAY10526 could significantly block mPGES-1 expression and the proliferation of DU-145 cells (P<0.05), while increasing Beclin-1 levels (P<0.05). Overexpression of mPGES-1 could decrease the autophagic PCa cell death. Inhibiting the expression of mPGES-1 may lead to DU-145 cell death and up-regulation of Beclin-1. The results suggest that inhibition of mPGES-1 may have therapeutic potential for PCa in the future.

      • Thermal post-buckling of graphene platelet reinforced metal foams doubly curved shells with geometric imperfection

        In the present work, thermal buckling and post-buckling behaviors of imperfect graphene platelet reinforced metal foams (GPRMFs) doubly curved shells are examined. Material properties of GPRMFs doubly curved shells are presumed to be the function of the thickness. Reddy’ shell theory incorporating geometric nonlinearity is utilized to derive the governing equations. Various types of the graphene platelets (GPLs) distribution patterns and doubly curved shell types are taken into account. The nonlinear equations are discretized for the case of simply supported boundary conditions. The thermal postbuckling response are presented to analyze the effects of GPLs distribution patterns, initial geometric imperfection, GPLs weight fraction, porosity coefficient, porosity distribution forms, doubly curved shell types. The results show that these factors have significant effects on the thermal post-buckling problems.
