The optimal medium composition for the production of carotenoid pigment from Haloarcular sp. EH-1 as a dietary for fishes were 1.0% sucrose, 1.0% yeast extract, 25% sodium chloride, 0.3% sodium citrate, 0.2% potassium chloride, 2.0% magnesium sulphate, 0.002% ferric sulphate(pH7.0). The incubation temperate, aeration rate and agitation speed were 40℃, 100ml medium/500ml vol. shaking flask, and 180 rpm/min. The carotenoid pigment production was highest after 5 days of incubation with the light.

The purpose of this research is to examine the conditions, the utilizations and awareness of eco-educational spots that are the fittest to children. The findings of survey carried out to 648 of children(the 4th grade 262, the 6th grade 386) are as follows: The 4th graders(96%) and 6th graders(97%) replied that the environment has been polluted or seriously polluted. and that water pollution has been the highest, TV has been selected as the most common means to get various environmental informations. The 4th graders were taught the environmental education through ethics, society, science in order. while the 6th graders ethics, science, society in order The 4th graders, expect to get environmental education through ethics, optional time (or special activity time) and society in order. For the 6th graders, on the other hand, it has been ethics, optional time (or special activity time), science in order. The survey showed that the environmental education at field had been more effective than that in classroom The students have selected a pond, a flower bed, a garden, a zoo and a vegetable garden for the environment education Main inconveniences that respondents undergo in utilizing eco- educational spots have been a long-time round trip and strict control by persons in charge. Besides, the 4th graders have complained of lack in various lives, the 6th graders of lack in sufficient learning opportunities due to a lot of participants. The respondents(81.4%) have revealed their will to put environmental conservation in practice, and the 6th graders have been more positive in searching for concrete ways of environmental conservation than the 4th graders. And they have preferred eco-educational spots that living thing inhabit to no-life spots.

Pathogenic Salmonella typhimurium can invade the intestinal epithelium and cause a wide range of diseases including gastroenteritis and bacteremia in human and animals. To identify the genes involved in the infection, the invasion determinant was obtained from S. typhimurium 82/6915 and was subcloned into pGEM-7Z. A subclone DHl (pSV6235) invaded HEp-2 and Chinese hamster ovary epithelial cells and contained a 4.4 kb fragment of S. typhimurium genomic region. Compared with the host strain E. coli DHl, the subclone DHl (pSV6235) invaded cultured HEp-2 and Chinese hamster ovary cells at least 75- and 68-fold higher, respectively. The invasion rate of E. coli DHl for the cells significantly increased by harbouring the genomic region derived from pathogenic S. typhimurium 82/6915.
Purification and biochemical characterization of a fibrinolytic enzyme from Bacillus subtilis BK-17

A fibrinolytic enzyme from Bacillus subtilis BK-17 has been purified to homogeneity by gel-filtration and ion exchange chromatography. Compared to the crude enzyme extract, the specific activity of the enzyme increased 929-fold with a recovery of 29%. The subunit molecular mass of the purified enzyme was estimated to be 31 kDa by SDS-PAGE. The N-terminal amino acid sequence of the purified fibrinolytic enzyme was; A-Q-S-V-P-Y-G-V-S-Q-I-K-A-P-A-A-H-N. The sequence was highly homologous to the fibrinolytic enzymes nattokinase, subtilisin J and subtilisin E from bacillus spp. However, there was a substitution of three amino acid residues in the N-terminal sequence. The amidolytic activity of the purified enzyme for several substrates was assessed. In comparison with nattokinase and CK (fibrinolytic enzyme from a Bacillus spp.), which showed strong fibrinolytic activity, the amidolytic activity of the enzyme for the synthetic substrate, kallikrein (H-D-Val-Leu-Arg-pNA, S-2266) increased 2.4- and 11.8-fold, respectively.

Effect of Cordycepin Purified from Cordyceps militaris on Th1 and Th2 Cytokines in Mouse Splenocytes

Cordycepin was purified from a mushroom, Cordyceps militaris, and its effect on Th1 and Th2 cytokines was examined. The level of cytokine induction in mouse splenocytes was estimated after co-inoculation of purified cordycepin and LPS. When 5 ug/ml of purified cordycepin was exposed to mouse splenocytes for 72 h, the level of a Th1 cytokine IL-12 increased by 2.9-fold. The addition of the purified cordycepin to splenocytes also increased the level of Th2 cytokines, IL-4 and IL-10, by 1.9- and 1.8-fold, respectively. Therefore, cordycepin increases the cytokine levels and may contribute to the up-regulation of cellular and humoral immunity.

Salmonella typhimurium is a causative agent of the common worldwide disease, salmonellosis. To identify putative invasion genes involved in Salmonella infections, a S. typhimurium cosmid library was constructed in non-invasive E. coli DH1. The invasion efficiencies of the cosmid library for cultured HEp-2 and HeLa cells were estimated by tissue-culture invasion assay. 2 out of 1,000 transductants, DH1 (pSI623) and DH1 (pSI511) were able to invade the cells. Compared to E. coli DH1, the invasion efficiencies of HEp-2 cells by DH1 (pSI511) increased 25- and 33 fold, respectively. The invasion efficiencies of HeLa cells by DH1 (pSI623) increased 31- and 35 fold, respectively. This illustrates that the cosmid clones, DH1 (pSI623) and DH1 (pSI623) and HD1 (pSI511) could harbor the invasion determinants derived from genomic DNA of S. typhimurium 82/6915, conferring the invasive characters for the cells.
Development of a biological delivery system for a live Salmonella vaccine

A cosmid library of S. typhimurium was constructed in E. coli by transduction. Cultured HeLa and Chinese hamster ovary cells were infected by the cosmid library. The genomic region of a clone, pSI623 which was able to penetrate the mammalian cells was cleaved with BglII completely and subcloned into a plasmid vector, pGEM-7Z, resulting a series of subclones conserving the genomic region representing the invasion genes. The subclone, DH1(pSV6235) carrying 4.65 kb of the genomic region was highly able to penetrate HeLa and Chinese hamster ovary epithelial cells. In addition, the invasion efficiency of pSV6235 was significantly higher than that of DH1(pSI623). However, the subclones carrying 7.4 kb, 6.3 kb, 5.5 kb and 1.4 kb of the genomic region from pSI623 were not able to penetrate the mammalian cells. pSV6235 was introduced into a live Salmonella vaccine, S. dublin aroA mutant. The invasiveness of HeLa and Chinese hamster ovary epithelial cells by the vaccine strain harboring pSV6235 was significantly increased, compared with the vaccine strain. These results indicated that the non-invasive E. coli was significantly able to invade HeLa and Chinese hamster ovary epithelial cells by harboring the invasion genes of pSV6235 which were originally derived from S. typhimurium and could be applied as a biological delivery vector to augment immune efficacy.
