Method of DNA Extraction from Pinus rigida Wood Pretreated with Sandpaper

Species identification of wood provides important information for archaeology, restoration of cultural assets, preventing illegal logging, and more. Wood species are usually identified based on their anatomical features with the use of a microscope. However, this method may not be able to distinguish between anatomically similar species or subspecies. To overcome this problem, wood species need to be identified at the molecular level using DNA sequencing. However, unlike living plant cells, wood is difficult to pulverize using a mortar, and DNA extraction from dried wood is challenging. To solve these problems, we propose a pretreatment method in which wood is pulverized using 60-grit sandpaper and hydrated with water for 2 days. Using this method, we were able to stably amplify the rpoB gene from the extracted DNA of Pinus rigida. In addition, sequence analysis of the rpoB gene revealed six single nucleotide polymorphisms (SNPs), which classified the rpoB sequences in the genus Pinus into five groups. Our data indicate that although these SNPs were not suitable for species identification, they can potentially be used to determine the origin of different wood subspecies or individual samples of wood.

본 연구는 국내 기능성 축산식품 관리제도 도입과 관련하여, 이를 반영하기 위한 기능성 물질(성분)의 과학적 분석기술 및 기능성 축산물별 검사방법 등 세부관리기준과 인증 방안을 마련하여 시행규칙을 제안하고자 한다. 기능성 축산식품을 관리하기 위한 새로운 법률 개정에 발맞추어 기능성 물질(성분)의 효용성(efficacy)과 안전성(safety)을 확인(검증)할 수 있는 분석.평가기술의 재정립을 통하여, 기능성 식품의 분석.평가 현황을 파악하고, 축산식품을 위한 기능성 물질의 범위를 정하여 이에 부합하는 표준검사법을 확립하고, 표준화를 제안한다. 본 연구를 통해 그 동안 우리나라에서 건강지향식품들의 지속적 노력에도 불 구하고, 질적인 발전을 이루지 못한 과거의 문제점을 해결하여, 합리적인 관리체계 안에서 안전하고 우수한 품질을 갖는 기능성 축산식품을 제조.판매하는데 이바지 하며, 소비자의 건강증진은 물론 잘못된 건강기능식품의 섭취로 인한 피해를 방지하고자 한다. This study aims to suggest a management system consisting of functional ingredients by establishing guidelines for theanalysis of functional animal products in Korea. Further, the guidelines can be used for validity tests of functional animalproducts. In addition, we can standardize the management system for details, and frame new laws for functional animalproducts. We researched the current studies on the analysis and assessment of functional foods, and established standardmethods of analysis. We devised new analysis and assessment techniques to determine the efficacy and safety of functionalingredients for the legislation and management of functional animal products. This study may contribute to not only thesafety and quality of functional animal products but also to enhancing the nation`s health and the national image as beinghealth-oriented, through improvements in the productivity and quality of functional animal products.

<P><B>Abstract</B></P> <P>The performance of the prototype modules of neutron detectors for the Large Acceptance Multi-Purpose Spectrometer (LAMPS) was investigated, using cosmic muons and neutron beams at 65 and 392 MeV, provided by the Research Center for Nuclear Physics (RCNP). The timing and position resolutions were estimated using cosmic muons as 309 ps and 4.8 cm, respectively. The energy resolution depended on the incident energy of neutrons: 1.3% at 65 MeV and 3.1% at 392 MeV. The neutron-detection efficiency also showed weak energy dependence as it decreased from (9.0 ± 1.6)% at 65 MeV to (6.3 ± 1.0)% at 392 MeV.</P>

γ-Aminobutyric acid (GABA) content in fermented plant products and their main plant materials (aerial part of Acanthopanax sessiliflorus, fruit of Crataegus pinnatifida, and whole plant of Morus alba) was determined by high-performance liquid chromatography. GABA was quantified using a reversephase column with a gradient elution program (water:acetonitrile =90:10 to 0:100 for 40 min). UV detection was conducted at 280 nm. GABA content was measured in fermented plant products (15.07 mg/g), aerial part of A. sessiliflorus (4.49 mg/g), fruit of C. pinnatifida (10.59 mg/g), and whole plant of M. alba (2.31 mg/g). The presence of GABA in fermented plant products, including A. sessiliflorus, C. pinnatifida, and M. alba is important in industrial application for health supplements.
Pure rate effect on the concrete compressive strength in the split Hopkinson pressure bar test

<P><B>Abstract</B></P> <P>The dynamic increase factor (DIF) has been widely used to consider the rate effect in the analysis and design of concrete structures that are subject to impact loads. A variety of DIFs have been proposed by many researchers based on the results of dynamic material tests such as the split Hopkinson pressure bar (SHPB) test. These DIFs have been adopted in authoritative design guidelines and model codes such as the ACI 349–13, ACI 370R-14, fib MC2010, and UFC 3-340-02. However, previous studies did not properly consider the strain acceleration and the geometrical characteristics of the test specimens that cause the axial and radial inertia forces which influence the test results. For this reason, predictions can become non conservative when these DIFs are used in the analysis and design of concrete structures that are subject to impact or impulsive loads. In this study, to overcome the limitations of existing DIFs, a new concrete DIF that excludes inertia effects by considering the strain acceleration and geometry of the specimens has been proposed based on SHPB test results. The proposed DIF was numerically validated using finite element analyses. Compared with other existing DIFs, the results show improved predictions of the enhancement of the concrete compressive strength due to rate effect.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Strain acceleration and geometry of a specimen lead to inertia effects in SHPB test. </LI> <LI> Concrete SHPB tests were performed. </LI> <LI> DIF considering the pure rate effect was proposed. </LI> <LI> Proposed DIF provided an accurate prediction for the apparent dynamic strength. </LI> </UL> </P>
Silicon nanowire-based tunneling field-effect transistors on flexible plastic substrates

<P>A technique to implement silicon nanowire (SiNW)-based tunneling field-effect transistors (TFETs) on flexible plastic substrates is developed for the first time. The p-i-n configured Si NWs are obtained from an Si wafer using a conventional top-down CMOS-compatible technology, and they are then transferred onto the plastic substrate. Based on gate-controlled band-to-band tunneling (BTBT) as their working principle, the SiNW-based TFETs show normal p-channel switching behavior with a threshold voltage of −1.86 V and a subthreshold swing of 827 mV/dec. In addition, ambipolar conduction is observed due to the presence of the BTBT between the heavily doped p<SUP>+</SUP> drain and n<SUP>+</SUP> channel regions, indicating that our TFETs can operate in the n-channel mode as well. Furthermore, the BTBT generation rates for both the p-channel and n-channel operating modes are nearly independent of the bending state (strain = 0.8%) of the plastic substrate. </P>

<P><I>Background:</I> Although there have been reports on threadlike structures inside the heart, they have received little attention. We aimed to develop a method for observing such structures and to reveal their ultrastructures. <I>Methods:</I>An in situ staining method, which uses a series of procedures of 0.2-0.4% trypan blue spraying and washing, was applied to observe threadlike structures on the surfaces of endocardia. The threadlike structures were isolated and observed by using confocal laser scanning microscopy (CLSM) and transmission electron microscopy (TEM). <I>Results:</I> Networks of endocardial vessels (20 μm in thickness) with expansions (40-100 μm in diameter) were visualized; they were movable on the endocardium of the bovine atrium and ventricle. CLSM showed that (1) rod-shaped nuclei were aligned along the longitudinal direction of the endocardial vessel and (2) there were many cells inside the expansion. TEM on the endocardial vessel revealed that (1) there existed multiple lumens (1-7 μm in diameter) and (2) the extracellular matrices mostly consisted of collagen fibers, which were aligned along the longitudinal direction of the endocardial vessel or were locally organized in reticular structures. <I>Conclusion:</I> We investigated the endocardial circulatory system in bovine cardiac chambers and its ultrastructures, such as nucleic distributions, microlumens, and collagenous extracellular matrices.</P><P>Copyright © 2011 S.
