        분얼형 옥수수의 식물학적 특성과 RAPD 분석에 의한 근연관계

        분얼형 옥수수 자석계통의 식물학적 특징, 유전적 거리를 조사하여 새로운 교잡종 육성을 위한 기초자료를 얻고자 실험 한 결과를 요약하면 다음과 같다. 1. 개화기에 있어서 조사된 20계통 중 K15가 가장 빨랐고 K07이 가장 늦은 개화소요일수를 보였다. 2. 간장과 착수고는 비슷한 경향으로 보며 K04가 가장 컸고, K15가 가장 작은 것으로 나타났다. 3. 상관 분석 결과 분열수는 개화기 및 출사기와 고도의 정의 상관을 보였으며, 100립중은 개화기, 이삭높이와 고도의 부의 상관을 가지고 있다. 4. RAPD 분석결과 17개의 BAND가 나타났고 이중 1개만이 monomolpic band를 보였으며, primer 당 평균 4개의 loci를 보였다. 5. 유전적 거리와 식물학적 특성간에는 개화기, 분얼수, 엽장과 100립중에서 관련이 있었다. This study was carried out to obtain a basic information for the development of a new corn hybrid with tillers. Materials used in this study were 20 lines having three to four tillers per plant including the PI213749 U S line with non-tillers. These 20 lines were compared for the botanical characteristics and genetic distances were measured using RAPD analysis. Flowering date of the K15 was very earlier. while the K07 was very late in flowering date. Stem height and ear height were similar except for KO4 and the K15. K06, K13 and K19 lines were appeared to be resistant to lodging due to decreased ear height. Number of tillers per plant of lines used were shown three to five on average. KO9 showed the highest kernel yield, while the K08 was low. Among characteristics measured tiller per plant and flowering dates, and silking dates showed a positive correlation, while 100 kernels weight, flowering date and ear height were shown a negative with tillers per plant. A total of 17 bands by RAPD analysis using four per primer were appeared and these lines were classified into three groups, especially the third group could be classified into of four sub-groups.

        Space Supervision을 이용한 치성 정중선 변위의 교정

        상하악 중절치 간의 정중선 변위는 골격성 변위,치성 변위 및 골격성과 치성의 복합적 변위 등으로 구분된다. 그 중 치성 정중선 변위는 치열의 발육시기에 따라 다양한 방법으로 치료 방법을 선택할 수 있다. 특히 혼합치열의 초기에는 치열 내에 존재하는 공간의 조건,인접 영구치의 맹출 시기 또는 맹출 방향에 따라 영구 절치들의 위치가 자발적으로 변화,개선될 수 있다. 따라서 이러한 치아의 생리적 이동을 이용하여 특별한 교정장치의 이용 없이 치열 내에서 치아의 배열을 개선시킬 수 있다. 본 증례보고에서는 특히 유견치의 발거 시기 및 좌우 유치의 발거 순서 등을 이용하여 치성 정중선 불일치를 개선시킨 증례틀을 보고하고자 한다. A midline deviation is divided into three types, such as a skeletal midline deviation, a dental midline deviation, and a combination. Among them, the midline deviation which appear in the early mixed dentition without any skeletal problem can be managed differently by the stage of dental development. The location of the permanent incisors can be improved without using artificial force in consideration of the condition of the space, the eruption timing or the eruption path of the adjacent permanent teeth in early mixed dentition, especially when the permanent incisors are erupting. Therefore. the space supervision technique, which can have the teeth align themselves utilizing physiologic events such as sequence or timing of the eruption of permanent teeth, can be an alternative to orthodontic movement. This case report describes midline deviation improvement by space supervision.

        One of the contributory causes to poor pregnancy rates in assisted reproduction has been the decreased viability of transferred embryos and the transfer of four-cell embryos into and environment that naturally would be receptive only to 5-day-old blastocysts. In this paper, we have reviewed on the role of helper cells(cocultures) to mimic the in vivo, environment to bring about improved embryo quality and an increased number of blastocysts for replacement in IVF patients. The behavior of helper cell is first presented, followed by their use as cocultures for fertilization and cleavage of embryos. The mode of action and specificity of helper cells are also discussed. The currently available results of pregnancies after cocultures are presented together with future aspects of research that are necessary to refine the coculture system, The ultimate aim is to mimic in vivo conditions in vitro, so that at least the pregnancy rates of assisted conception can be parallel to normal fecundity in the human. Therefore, a very attractive future includes the freezing of blastocysts generated from coculture, thawing, and replacing them in natural cycles.

        Lung Microbiome Analysis in Steroid-Naive Asthma Patients by Using Whole Sputum

        Background: Although recent metagenomic approaches have characterized the distinguished microbial compositions in airways of asthmatics, these results did not reach a consensus due to the small sample size, non-standardization of specimens and medication status. We conducted a metagenomics approach by using terminal restriction fragment length polymorphism (T-RFLP) analysis of the induced whole sputum representing both the cellular and fluid phases in a relative large number of steroid naive asthmatics. Methods: Induced whole sputum samples obtained from 36 healthy subjects and 89 steroid-naive asthma patients were analyzed through T-RFLP analysis. Results: In contrast to previous reports about microbiota in the asthmatic airways, the diversity of microbial composition was not significantly different between the controls and asthma patients (p=0.937). In an analysis of similarities, the global R-value showed a statistically significant difference but a very low separation (0.148, p=0.002). The dissimilarity in the bacterial communities between groups was 28.74%, and operational taxonomic units (OTUs) contributing to this difference were as follows: OTU 789 (Lachnospiraceae ), 517 (Comamonadaceae, Acetobacteraceae , and Chloroplast ), 633 (Prevotella ), 645 (Actinobacteria and Propionibacterium acnes ), 607 (Lactobacillus buchneri, Lactobacillus otakiensis, Lactobacillus sunkii , and Rhodobacteraceae ), and 661 (Acinetobacter, Pseudomonas , and Leptotrichiaceae ), and they were significantly more prevalent in the sputum of asthma patients than in the sputum of the controls. Conclusion: Before starting anti-asthmatic treatment, the microbiota in the whole sputum of patients with asthma showed a marginal difference from the microbiota in the whole sputum of the controls.

      • Thematic Poster : TP-20 ; Lung Function Impairment According to Prior Pulmonary Tuberculosis

        Background: Previous pulmonary tuberculosis (TB) can affect lung function, but a few data exist on long-term follow-up. This study was conducted to evaluate whether prior pulmonary TB was related with lung function impairment in Korean adult population and risk factors of decreased lung function in subjects with prior pulmonary TB. Methods: We used data from the fourth and fifth annual South Korean National Health and Nutrition Examination Survey (Korean NHANES IV) conducted in 2008, 2009, 2010, 2011 and 2012. From a total of 45,811 subjects, 14,967 adult subjects who were at least 40 years of age were included in the study population. The correlation between prior pulmonary TB and chest x-ray abnormality and impaired lung function using questionnaires about diagnosis of pulmonary tuberculosis, chest x-ray and pulmonary lung function were analyzed. The results of pulmonary functions were all adjusted by age, sex and smoking pack-years. Results: Mean age was 57.1years and males were 43.3%. Subjects with preciously diagnosed and treated pulmonary TB were 5.5%. Chest PA with sequale of pulmonary tuberculosis was 7.9%. FEV1%, FEV1/FVC in subjects with prior pulmonary TB were significantly low compared with those without TB (84.89±17.19 vs. 92.58±13.51, P<0.001, and 77.91±7.52 vs. 77.91±7.52, P<0.001, respectively). Subjects with prior pulmonary TB had higher prevalence of COPD compared with those without previous TB (19.5% vs. 6.5%, OR=2.862, 95% CI: 2.344-3.496), also in subjects with normal chest x-ray (9.1% vs. 5.0%, OR=1.702, 95% CI: 1.112-2.607). In subject with prior pulmonary TB, TB sequale on chest x-ray was risk factor of COPD development (OR=2.244, 95% CI: 1.578-3.191). Conclusion: In the current study, previous pulmonary TB morbidity was significantly related with decreased lung function in Korean adults. The evaluation and follow up of pulmonary function in subjects with previous pulmonary TB should be needed.

        The Macrophage-Specific Transcription Factor Can Be Modified Posttranslationally by Ubiquitination in the Lipopolysaccharide-Treated Macrophages

        Background: Macrophages are one of the most important inflammatory cells in innate immunity. PU.1 is a macrophage-specific transcription factor. Ubiquitins are the ultimate regulator of eukaryotic transcription. The ubiquitination process for PU.1 is unknown. This study investigated the lipopolysaccharide (LPS)-induced activation of PU.1 and its relation to ubiquitins in the macrophages. Methods: Raw264.7 cells, the primary cultured alveolar, pulmonary, and bone marrow derived macrophages were used. The Raw264.7 cells were treated with MG-132, NH4Cl, lactacytin and LPS. Nitric oxide and prostaglandin D2 and E2 were measured. Immunoprecipitation and Western blots were used to check ubiquitination of PU.1. Results: The PU.1 ubiquitination increased after LPS (1ug/mL) treatment for 4 hours on Raw264.7 cells. The ubiquitination of PU.1 by LPS was increased by MG-132 or NH4Cl pretreatment. Two hours of LPS treatment on macrophages, PU.1 activation was not induced nor increased with the inhibition of proteasomes and/or lysosomes. The ubiquitination of PU.1 was increased in LPS-treated Raw264.7 cells at 12- and at 24 hours. LPS-treated cells increased nitric oxide production, which was diminished by MG-132 or NH4Cl. LPS increased the production of PGE2 in the alveolar and peritoneal macrophages of wild type mice; however, PGE2 was blocked or diminished in Rac2 null mice. Pretreatment of lactacystin increased PGE2, however it decreased the PGD2 level in the macrophages derived from the bone marrow of B57/BL6 mice. Conclusion: LPS treatment in the macrophages ubiquitinates PU.1. Ubiquitination of PU.1 may be involved in synthesis of nitric oxide and prostaglandins.

        Background: Many studies investigated that smoking is the major cause of various physical illnesses, but the relationship between smoking status and mental health including suicidal ideation and suicidal attempts was not studied. This study was conducted to evaluate whether cigarette smoking status was related with mental health including suicidal ideation and suicidal attempts in Korean adult population. Methods: We used data from the fourth and fifth annual South Korean National Health and Nutrition Examination Survey (Korean NHANES IV) conducted in 2008,2009, 2010, 2011 and 2012. From a total of 45,811 subjects, 32,184 adult subjects who were at least 19 years of age were included in the study population. The correlation between smoking status and mental health using questionnaires about mental health, cigarette smoking and diagnosis of major depressive disorder were analyzed. They were adjusted by age, income, education and marital state. Results: In male, current smoker was 42.4%, former smoker 38.9% and non smoker18.7% and in female, 5.9%, 5.6% and 88.4%. Age, quartile of income, education state and marital state were different according to smoking status (P<0.001). In male and female, perceived stress was higher and EQ-VAS in current smoker was lower than those in non smoker (P<0.001). The incidence of major depressive disorder in current smoker female was higher than that in non smoker female (8.4% vs. 4.5%, OR=1.945, 95% CI: 1.536-2.463). In female, current smoker had higher prevalence of suicidal ideation and suicidal attempts in last year compared with non smoker (OR=1.912, 95% CI: 1.659-2.203, OR=3.930, 95% CI: 2.701-5.719, respectively).Conclusion: In the current study, current smoking was signifi cantly related with major depressive disorder, depression symptom and suicidal ideation and attempts in Korean females.
