        I studied Song, Gyu-ryeom`s life through his `Annual Report` and writing series, `Jewoldangjip` and the ideology of the society that he lived in through the country activity. Song, Gyu-ryeom was a famous Confucian scholar who lived in Hoedeok at the latter Choseon dynasty. He was called the one of the three prominent `Songs` with Uam, Song, Si-yeol, and Dongchundang, Song, Jun-gil. The reason why Song, Gyu-ryeom`s ancestors who lived in Hoedeok was that Song, Myung-eui(Song, Gyu-ryeom`s 10th ancestor) settled there as he became the son-in-law of Hwang, Su who lived in Hoedeok. Since then many famous Confucian scholars came out of Eunjin Songs` family members and they thrived through marrying the nobles. When Song, Gyu-ryeom was a boy, he experienced Byeonjahoran and the fall of the Ming Dynasty. He also spend his adolescence under the mood that people insisted to conquer the Cheong Dynasty. So it made him follow Song, Si-yeol and Song, Jun-gil`s political and ideologic way and he learned the basic of study from them. Song, Gyu-ryeom was respected as a Confucian scholar in Hoedeok. He emphasized `Hyangeumjurye`, `Hyangsarye`, `Reading methods` at the society of the country. `Hyangeumjurye` means that Confucian scholars in the country gathered at Hyanggyo or Seowon and had a feast with prominent Confucian scholars. The Confucians learned the manner to respect honest people, to take care of the old people, and to experience good courtesy there. Also he taught students devotionally at the Sunghyeonseowon. In 1672(Sukjong 2nd year), he built the Jewoldang inside his house and spent his whole life on studying Confucianism and teaching students. Teaching students at the Jewoldang and the Sunghyeonseowon was theoretical part and Hyangeumjurye was the reformation project which included the education of practical manner. Like this, Song, Gyu-ryeomtried to carry out `manner` through personality based on Neo-Confucianism And Song, Gyu-ryeom also had the social ideology of the autonomy of the country. That idea was shown in the preface of Hoedeok Hyangyack(1672) that he wrote. Hoedeok Hyangyack was made into one book with Hoedeok Hyangyan. Then Song, Si-yeol made the preface of Hoedeok Hyangyan and Song, Jun-gil wrote it. And Song, Gyu-ryeom wrote the preface of Hyangyack which was attached to the back of Hyangyan. He was the core member to make the Hoedeok Hyangyack with two Songs. The reason why he was called as the one of the three Songs of Hoedeok might be due to this work. Hyangyack was the regulation that Hyangwon made for education and cooperation of the country people. So the Hoedeok Hyangyack also had the content like that. The Jewoldang was the place where Song, Gyu-ryeom studied and taught students with all effort at his old age. Also it was the place where he built for just studying after he abandoned his will to have a government position. The education place like this was the sign that he wanted to live with the nature and worked hard for teaching students. Song, Gyu-ryeom`s country activity showed his social ideology to improve national customs from the country customs with the spread of Hyangyack.

        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.         Ethanol Extract of Ganoderma lucidum Augments Cellular Anti-oxidant Defense through Activation of Nrf2/HO-1

        Objectives: The mushroom Ganoderma lucidum has been widely used as a traditional herbal medicine for many years. Although several studies have focused on the anti-oxidative activity of this mushroom, the molecular mechanisms underlying its activity have not yet been clearly established. The present study investigated the cytoprotective effect of ethanol extract of Ganoderma lucidum (EGL) against oxidative stress (hydrogen peroxide, $H_2O_2$) and elucidated the underlying mechanisms in a C2C12 myoblast cell line. Methods: Oxidative stress markers were determined by using the comet assay to measure reactive oxygen species (ROS) generation and deoxyribonucleic acid (DNA) damage. Cell viability and Western blotting analyses were employed to evaluate the cellular response to EGL and $H_2O_2$ in C2C12 cells. Transfection with nuclear factor erythroid 2-related factor 2 (Nrf2)-specific small interfering ribonucleic acid (siRNA) was conducted to understand the relationship between Nrf2 expression and $H_2O_2$-induced growth inhibition. Results: The results showed that EGL effectively inhibited $H_2O_2$-induced growth and the generation of ROS. EGL markedly suppressed $H_2O_2$-induced comet-like DNA formation and phosphorylation of histone H2AX at serine 139 ($p-{\gamma}H2AX$), a widely used marker of DNA damage, suggesting that EGL prevented $H_2O_2$-induced DNA damage. Furthermore, the EGL treatment effectively induced the expression of Nrf2, as well as heme oxygenase-1 (HO-1), with parallel phosphorylation and nuclear translocation of Nrf2 in the C2C12 myoblasts. However, zinc protoporphyrin IX, a HO-1 inhibitor, significantly abolished the protective effects of EGL against $H_2O_2$-induced accumulation of ROS and reduced cell growth. Notably, transient transfection with Nrf2-specific siRNA attenuated the cytoprotective effects and HO-1 induction by EGL, indicating that EGL induced the expression of HO-1 in an Nrf2-dependent manner. Conclusion: Collectively, these results demonstrate that EGL augments the cellular anti-oxidant defense capacity through activation of Nrf2/HO-1, thereby protecting C2C12 myoblasts from $H_2O_2$-induced oxidative cytotoxicity.

        Protective Effect of Heme Oxygenase-1 on High Glucose-Induced Pancreatic β-Cell Injury

        <P><B>Background</B></P><P>Glucose toxicity that is caused by chronic exposure to a high glucose concentration leads to islet dysfunction and induces apoptosis in pancreatic β-cells. Heme oxygenase-1 (HO-1) has been identified as an anti-apoptotic and cytoprotective gene. The purpose of this study is to investigate whether HO-1 up-regulation when using metalloprotophyrin (cobalt protoporphyrin, CoPP) could protect pancreatic β-cells from high glucose-induced apoptosis.</P><P><B>Methods</B></P><P>Reverse transcription-polymerase chain reaction was performed to analyze the CoPP-induced mRNA expression of HO-1. Cell viability of INS-1 cells cultured in the presence of CoPP was examined by acridine orange/propidium iodide staining. The generation of intracellular reactive oxygen species (ROS) was measured using flow cytometry. Glucose stimulated insulin secretion (GSIS) was determined following incubation with CoPP in different glucose concentrations.</P><P><B>Results</B></P><P>CoPP increased HO-1 mRNA expression in both a dose- and time-dependent manner. Overexpression of HO-1 inhibited caspase-3, and the number of dead cells in the presence of CoPP was significantly decreased when exposed to high glucose conditions (HG). CoPP also decreased the generation of intracellular ROS by 50% during 72 hours of culture with HG. However, decreased GSIS was not recovered even in the presence of CoPP.</P><P><B>Conclusion</B></P><P>Our data suggest that CoPP-induced HO-1 up-regulation results in protection from high glucose-induced apoptosis in INS-1 cells; however, glucose stimulated insulin secretion is not restored.</P>

      • Ln₃Ba?Co₄O? (Ln = Sm, Pr, Er, Gd, Ho) 상의 합성과 결정구조에 관한 연구

        The Ln₃Ba?Co₄O? phases were synthesized with Pr?O? Sm₂O₃, Er₂O₃, Gd₂O₃, Ho₂O₃, BaCO₃, and Co₃O₄by solid state reaction at 1200℃ with intermittent grinding. The phases with Pr and Sm formed hexagonal structure isostructural with Nd₃Ba?Co ₄O? phase Whereas the phases with Er, Ho, and Gd formed unknown cubic phases. The crystal structure of Sm₃Ba?Co₄O? phase were refined by X-ray diffraction powder data by means of Rietveld method. The starting model was based on the Nd₃Ba?Co₄O? structure. The Crystal system was hexagonal, space group P6₃mc, a=11.634(6)A, c=6.861(4)A for pr₃Ba?Co₄O? ; a=11.556(6)A, c=6.788(3)A for Sm₃Ba?Co₄O?. Final R values are : Rwp=0.121, Rp=0.084 for pr₃Ba?Co₄O?, Rwp=0.101, Rp=0.077 for Sm₃Ba?Co₄O? .

        ERK1/2 activation in quercetin-treated BEAS-2B cell plays a role in Nrf2-driven HO-1 expression

        Quercetin, a member of the flavonoids,has been heralded as chemopreventive agent, generally because of its free radical-scavenging and antiproliferative activities. In this process, NF-E2-related factor-2 (Nrf2) plays a role in protecting cells from oxidative damage; however, the exact mechanisms in which bronchial epithelial cells respond to quercetin by activating Nrf2 are not fully understood. Here, we report the role of extracellular signal-regulated kinase (ERK) activation in response to quercetin. Quercetin treatment decreased the viability of human bronchial epithelial BEAS-2B cells in a dose- and time-dependent manner. ERK1/2 was activated within 10 min of quercetin addition and then remained increased above control level throughout 48 h, whereas AKT phosphorylation was suppressed from the first 1 h of quercetin treatment. Inhibition of Erk1/2 phosphorylation attenuated quercetin-induced loss of cell viability. Nrf2levels in both nuclear and whole cell lysates increased after quercetin treatment and was accompanied by the increase of heme oxygenase-1 (HO-1) protein level,indicating that increased HO-1 expression is Nrf2-mediated. Activation of the Nrf2/HO-1 system after quercetin treatment was suppressed by the pretreatment with MEK inhibitor, PD98059. Also, silencing Nrf2 with siRNA decreased cell viability and augmented cytotoxic effect of anticancer drug LBH589. Overall, our results indicate that quercetin induces the up-regulation of Nrf2-mediated HO-1 expression, at least in part, via MEK/ERK1/2 signaling, as an adaptive mechanism which may provide advantages for cell proliferation and enhance survival despite endogenous and/or exogenous oxidative insults.

        Morin exerts cytoprotective effects against oxidative stress in C2C12 myoblasts via the upregulation of Nrf2-dependent HO-1 expression and the activation of the ERK pathway

        <P>In the present study, we investigated the cytoprotective efficacy of morin, a natural flavonoid, against oxidative stress and elucidated the underlying mechanisms in C2C12 myoblasts. Our results indicated that morin treatment prior to hydrogen peroxide (H2O2) exposure significantly increased cell viability and prevented the generation of reactive oxygen species. H2O2-induced comet-like DNA formation and gamma H2AX phosphorylation were also markedly suppressed by morin with a parallel inhibition of apoptosis in C2C12 myoblasts, suggesting that morin prevented H2O2-induced cellular DNA damage. Furthermore, morin markedly enhanced the expression of heme oxygenase-1 (HO-1) associated with the induction and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the inhibition of Kelch-like ECH-associated protein 1 (Keapl) expression. Notably, these events were eliminated by transient transfection with Nrf2-specific small interfering RNA. Additional experiments demonstrated that the activation of the Nrf2/HO-1 pathway by morin was mediated by the extracellular signal-regulated kinase (ERK) signaling cascade. This phenomenon was confirmed with suppressed Nrf2 phosphorylation and consequently diminished HO-1 expression in cells treated with a pharmacological inhibitor of ERK. Collectively, these results demonstrated that morin augments the cellular antioxidant defense capacity through the activation of Nrf2/HO-1 signaling, which involves the activation of the ERK pathway, thereby protecting C2C12 myoblasts from H2O2,-induced oxidative cytotoxicity.</P>

        Stain improvement in the white button mushroom ‘Seolgang’ and its varietal characteristics in Agaricus bisporus

        The button mushroom (Agaricus bisporus) is one of the most widely cultivated important edible mushroom species. In the breeding of new button mushroom, ‘Seolgang’ was developed by crossing two monokaryons ‘CM020913-27’ and ‘SSU423-31’. Because of the secondarily homothallism, only a small percentage of the basidia produce 3 or 4 spores, which are mostly haploid (n) and do not fruit. Single spore cultures derived from these types of spores produce a vegetative mycelium that also contain a variable number of genetically identical nuclei per cell called monokaryon. The lack of clamp connections between monokaryon and dikaryon required a series of mycelial culture and fruiting test. After crossing, hybrids were cultivated on a small scale and on a commercial scale at a farm. For this, the spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of ‘Seolgang’ on CDA was better at 20℃ and 25℃ when it was compared with that of ‘505 Ho’. The mature cap shape of new strain ‘Seolgang’ is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 41.2 mm on average. In comparison with white strain ‘505 Ho’, the strain had a yield that was 9% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 19% firmer with a good shelf life. Days of fruiting body were 3-4 days later than those of ‘505 Ho’. The physical characteristics such as elasticity, chewiness, adhesiveness were better than that of ‘505 Ho’. Genetic analysis of the new strain ‘Seolgang’ showed different profiles compared to ‘505 Ho’, CM02913-27, SSU413-31, when RAPD primers A02 and O04 were used.

      • Plant Regeneration and Bulblet Formation of Allium wakegi Araki

        Allium wakegi was cultured shoot tip in the condition of light culture. The Allium wakegi added plant growth regulator was observed of plant regeneration and bulblet formation. Callus Induction and growing rate was the best of 78% when added alone 2,4-D 0.5mg/L. In the formation of shoot, its regeneration rate was 96% when added BA 0.5mg/L in the light culture condition. When BA 0.5mg/L and NAA 0.5mg/L mixed and BA 0.5 mg/L and NAA 1.0mg/L mixed, the rates were 99% and 97% respectively, and these conditions were suitable for forming shoot. In the formation of roots, when added NAA 2.0mg/L in the light culture condition, the regeneration rate was 90.6 % and the roots were abnormal. When added NAA 1.0mg/L, the rate was 82 % and the highest. In the formation of bulbs, when BA 05mg/L and NAA 1.0mg/L mixed, the root generantion and its size in the bulbs was the best compare to other treatment experiments.

      • Genetic Diversity of Korean Rice Breeding Parents as Measured by DNA Fingerprinting with Simple Sequence Repeat (SSR) Markers

        Molecular markers are useful tools for evaluating genetic diversity and determining cultivar identity. Present study was conducted to evaluate the genetic diversity within a diverse collection of rice accessions used for Korean breeding programs. Two hundred eighty-seven rice cultivars, composed of temperate japonica, tropical japonica, indica, and Tongil-type of Korean crossing parents were evaluated by means of 15 simple sequence repeat (SSR) markers. A total of 99 alleles were detected, and the number of alleles per marker ranged from 4 to 11, with an average of 6.6 per locus. Polymorphism information content (PIC) for each of the SSR markers ranged from 0.2924 to 0.8102 with an average of 0.5785. These results, with the result that use of only 15 SSR markers made all rice cultivars examined could be uniquely distinguished, imply the efficiency of SSR markers for analysis of genetic diversity in rice. Cluster analysis was performed on similar coefficient matrics calculated from SSR markers to generate a dendogram in which two major groups corresponding to japonica (Group I) and indica and Tongil type rice (group II) with additional subclasses within both major groups. The narrowness of the Korean breeding germplasm was revealed by the fact that most of the Korean-bred and Japan-bred temperate japonica cultivars were concentrated into only 2 of the sub-group I-1 (143 cultivars) and I-2 (58 cultivars) among six sub-groups in major group of japonica. This is because of the japonica accessions used in this study was a very closely related ones because of frequent sharing of the crossing parents with similar genetic background with synergy effect of the inherited genetic difference between indica and japonica. A rice breeding strategy with the use of molecular markers was discussed for overcoming of genetic vulnerability owing to this genetic narrowness.
