        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.         밤나무 교잡종 과실의 질적 특성

        본 연구는 밤나무 교잡종 과실의 조합별 및 개체별 질적특성을 조사함으로써 신품종 육성의 기초 자료뿐만 아니라 품종검정 기준설정의 기초자료로 활용하는데 그 목적이 있다. 과실모양은 11 조합의 많은 개체에서 타원형(85.30%)을 나타냈으며 K×R 등 5조합에서 삼각형(3.78%), R×K 등 8조합에서 원형(5.46%), R×O 등 3조합에서 장타원형(5.46%)으로 각각 관찰되었다. 과피색은 모든 조합에서 암갈색과 적갈색이 각각 49.16%와 38.24%로 관찰되었으며 과피털은 K×R과 R×K만이 과피 전체에서 각각 1개체씩 관찰되었다. 과피털의 밀도는 대부분의 조합에서 92.86%가 중 이상이었으며 좌면의 크기는 E×O, E×R, K×O에서만 중 이상인 것으로 조사되었다. 과실의 광택은 K×O조합에서만 약인 개체가 출현하였으며 열과형태는 모든 조합에서 3가지 형태중 2가지 이상의 형태가 각각 혼재되어 있었다. 양적형질이 우수한 8개체 중 RO-22, RO-25 등 2개체만이 과실모양과 과피색이 각각 장타원형과 암갈색을 나타내었으며 OK-2 제외한 나머지 5개체는 각각 타원형과 적갈색으로 관찰되었다. 과피털의 위치는 OK-2 제외한 모든 개체는 과정부였으며 과피털의 밀도는 IO-15 제외한 모든 개체에서 밀하게 분포하였다. 좌면의 크기는 JO-7과 RO-25 개체가 가장 작았으며 광택은 K×O의 1개체를 제외한 모든 개체에서 중 이상으로 우수하였다. 열과형태는 JO-7과 JO-12, IO-15, OK-15 개체에서 각각 과정부 열과와 주두부 열과 등 각각 단일 형태를 나타낸 반면, OK-2 등 4개체는 과정부 열과와 측면부 열과가 혼재하고 있음을 알 수 있었다. 내피박피성을 조사한 결과, OK-2, OK- 4, OK-15 등 3개체는 내피박피율이 20% 내외로 불량한 반면 RO-25, JO-12 등 2개체의 내피박 피율이 각각 78.6%, 72.4%로 매우 우수한 것으로 나타났다. This study was aimed to applicate as the basic data for the presentation of International Union for the Protection of New Varieties of Plants (UPOV) guideline and for breeding new cultivars through qualitative characteristics of nut in chestnut hybrids. In nut shape, oval type, 85.30%, could be observed in all combinations, and triangle type, 3.78%, in K×R, K×O, E×R, I×R, J×O, and oblong type, 5.46%, in R×O, I×O, E×O, I×R, respectively. Both dark and reddish brown, 49.16% and 38.24%, in nut color were observed in all combinations, and nut pericarp hairs in only K×R, R×K could be observed in whole part. The density of its hair showed above middle rank (92.86%) in most of combinations, and the size of hilum area showed above middle rank in only E×O, K×O and E×R. Ten combinations except K×O showed a good condition in nut luster, and two or three types of three split type could be observed in all combinations, simultaneously. Superior individuals, RO-22 and RO-25, showed oblong and dark brown, but others except OK-2 showed oval and reddish brown in nut shape and nut color, respectively. In position of nut pericarp hair, only OK-2 showed stigma part, but all of superior individuals except IO-15 showed thickness in its density. Only JO-7 and RO-25 were smaller than others in size of hilum area, and JO-7, OK-4, OK-15, RO-22 and RO-25 had nuts with excellent luster. Most of individuals had nuts with two or three types in type of nut split, and JO-7, JO-12, IO-15 and OK-15 showed only single type. In peeling rate, OK-2, OK-4 and OK-15 showed around 20% while RO-25 and JO-12 showed 78.6% and 72.4% being considered as superior individuals for roasted chestnuts.

        15d-PGJ2 inhibits NF-kB and AP-1-mediated MMP-9 expression and invasion of breast cancer cell by means of a heme oxygenase-1-dependent mechanism

        Activation of peroxisome proliferator-activated receptor  (PPAR) serves as a key factor in the proliferation and invasion of breast cancer cells and is a potential therapeutic target for breast cancer. However, the mechanisms underlying this effect remain largely unknown. Heme oxygenase-1 (HO-1) is induced and overexpressed in various cancers and is associated with features of tumor aggressiveness. Recent studies have shown that HO-1 is a major downstream target of PPAR. In this study, we investigated the effects of induction of HO-1 by PPAR on TPAinduced MMP-9 expression and cell invasion using MCF-7 breast cancer cells. TPA treatment increased NF-B /AP-1 DNA binding as well as MMP-9 expression. These effects were significantly blocked by 15d-PGJ2, a natural PPAR ligand. 15d-PGJ2 induced HO-1 expression in a dose-dependent manner. Interestingly, HO-1 siRNA significantly attenuated the inhibition of TPA-induced MMP-9 protein expression and cell invasion by 15d-PGJ2. These results suggest that 15d-PGJ2 inhibits TPA-induced MMP- 9 expression and invasion of MCF-7 cells by means of a heme oxygenase-1-dependent mechanism. Therefore, PPAR/HO-1 signaling- pathway inhibition may be beneficial for prevention and treatment of breast cancer.

        In vitro and in vivo anti-inflammatory effects of pegmatite

        Pegmatite is a coarse-grained intrusive igneous rock rich in rare elements such as uranium, tungsten, and tantalum with Ca, K, Mg, Fe, Se, Ge, and Ho. We tested in vitro and in vivo assays for the anti-inflammatory activity of pegmatites. We firstly evaluated the suppressive effects of pegmatite on macrophage cell line RAW 264.7 cells stimulated with proinflammatory stimuli lipopolysaccharide (LPS) to determine nitric oxide (NO) production and TNF-$\alpha$ and IL-6 release. The $IC_{50}$ values of pegmatite exceeded $5,000\;{\mu}g/mL$. Treatment of RAW 264.7 cells with pegmatite significantly inhibited LPS-stimulated NO production and proinflammatory cytokines such as TNF-$\alpha$ and IL-6 secretion in a dose-dependent manner (P<0.05). In vivo studies were tested with two animal models of arachidonic acid-induced mouse ear edema and an acetic acid-induced increase in capillary permeability. The pegmatite significantly attenuated ear edema induced by arachidonic acid and reduced the acetic acid-induced increase in capillary permeability in mice (P<0.05) when the pegmatite was administered topically (10 mg per ear) for 24 h. Therefore, pegmatite potentially shows an anti-inflammatory activity in the in vitro and in vivo mice and in the development of newer anti-inflammatory agents as mineral materials.

      • 감마선과 Methyl Bromide 처리가 사과의 생리화학적 품질에 미치는 영향

        감마선(0, 0.5, 1, 2, 3 kGy) 조사와 methyl bromide(MeBr, 26 g/㎏, 4hr) 훈증 및 이들의 처리시기가 사과의 생리화학적 품질특성에 영향을 비교하여, 검역처리를 위한 ◁그림 삽입▷ (원문을 참조하세요) 감마선 조사의 응용성을 검토하였다. 감마선과 MeBr 처리에 따른 사과의 생리화학적 품질평가에서 2kGy 이상의 감마선 조사와 MeBr 훈증은 사과의 일부 생리화학적 품질특성의 변화를 촉진시키는 것으로 나타났다. 감마선과 MeBr의 처리시기별로는 수확 직후보다는 저온(0℃)에서 40일 정도 보관 후 처리한 대체적으로 품질손상이 적은 것으로 나타났다. 따라서 1kGy 이하의 감마선 조사와 지연조사는 사과 과실의 품질을 비교적 양호한 상태로 유지할 수 있어 검역처리 기술로 활용성이 있는 것으로 생각된다. 'Fuji' apples were treated by gamma irradiation (0∼3 kGy) or methyl bromide fumigation at commercial conditions (MeBr, 25 g/㎏, 4 hr) for quarantine purpose at two different treatment timing after harvest and stored under air a 0℃ for 4 months. Associated with the treatment timing of irradiation or fumigation, after 40 days of storage at 0℃ following harvest was more adequate than immediately after harvest in keeping qualities of stored apples. However, more than 2 kGy irradiation and MeBr fumigation were detrimental to physiological and chemical qualities of the fruits. These results show that less than 1 kGy irradiation at the delayed timing has a possibility to be applied as a quarantine procedure without significant changes in the quality of apples.

        Antioxidant and hepatoprotective effects of Korean ginseng extract GS-KG9 in a D-galactosamine-induced liver damage animal model

        BACKGROUND/OBJECTIVES: This study was designed to investigate the improvement effect of white ginseng extract (GS-KG9) on D-galactosamine (Ga1N)-induced oxidative stress and liver injury. SUBJECTS/METHODS: Sixty Sprague-Dawley rats were divided into 6 groups. Rats were orally administrated with GS-KG9 (300, 500, or 700 mg/kg) or silymarin (25 mg/kg) for 2 weeks. The rats of the GS-KG9- and silymarin-treated groups and a control group were then intraperitoneally injected Ga1N at a concentration of 650 mg/kg for 4 days. To investigate the protective effect of GS-KG9 against GalN-induced liver injury, blood liver function indicators, anti-oxidative stress indicators, and histopathological features were analyzed. RESULTS: Serum biochemical analysis indicated that GS-KG9 ameliorated the elevation of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) in GalN-treated rats. The hepatoprotective effects of GS-KG9 involved enhancing components of the hepatic antioxidant defense system, including glutathione, glutathione peroxidase (GPx), superoxide dismutase (SOD), and catalase (CAT). In addition, GS-KG9 treatment inhibited reactive oxygen species (ROS) production induced by GalN treatment in hepatocytes and significantly increased the expression levels of nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) proteins, which are antioxidant proteins. In particular, by histological analyses bases on hematoxylin and eosin, Masson"s trichrome, α-smooth muscle actin, and transforming growth factor-β1 staining, we determined that the administration of 500 mg/kg GS-KG9 inhibited hepatic inflammation and fibrosis due to the excessive accumulation of collagen. CONCLUSIONS: These findings demonstrate that GS-KG9 improves GalN-induced liver inflammation, necrosis, and fibrosis by attenuating oxidative stress. Therefore, GS-KG9 may be considered a useful candidate in the development of a natural preventive agent against liver injury.

        최근 고품질의 음성합성과 코딩을 위한 연구의 일환으로 성문폐쇄시점(GCI:Glottal Closure Instant) 검출을 위한 다양한 기법들의 개발이 진행 중이다. 본 연구에서는 GCI 검출에 가장 효과적인 기법으로 알려진 힐버트 변환법을 이용한 EFLPR(Epoch Filtering of Linear Prediction Residual)법을 제안하였다. 본 연구에서 제안된 SGCID법은 합성음과 남성, 여성 화자의 자연음의 단모음과 숫자음 그리고 연속음 등 다양한 음성에 적용한 결과 안정된 유성음 구간에서 기존 방법들에 비해 정확한 GCI를 검출할 수 있었다.

        The pulling apparatus for the growth of single crystalline solids was made by sing the Czochralskis' method. The sing1e crystaline solids with 4Agl-NaI,4Ag I-KI,4AgI-RbI and 4Ag I-NH_4I system have been grown without seeds of crystalline solids cooling at various temperatures of growth and considering the pulling and rotating rates of shaft from the appanatus mentioned above. The single crystalline solids grown were indentified by using the analytical data measured in the visible range of the X-ray diffractometer, polarimeter and polarizing microscope. All of the crystalline solids were grown at the range of 6.5~10.3cm lengthand 0.8~1.5cm diameter, respectivity and these crystalline solids have the structures of d and ℓorientation.
