        The Expression of IL-4 and Interferon-gamma Genes in Peripheral Blood Mononuclear Cells of Patients with Severe Atopic Dermatitis : Evaluation in Proportion to Serum IgE Levels

        Background : It is not yet clear whether the abnormal cytokine production in relation to serum IgE levels in atopic dermatitis (AD) is associated with the amount of mRNA of cytokine gene. Objective : Our purpose was to delineate the effect of reciprocal correlation in the level of mRNA between interleukin-4 (IL-4) and interferon-gamma (IFN-γ) in severe AD. Methods : We examined 1S cases including 5 AD patients with high serum IgE ($gt;2,000 kU /liter), 5 AD patients with low serum IgE ($lt;100 kU/liter), and 5 healthy controls. Using semi quantitative reverse transcription-polymerase chain reaction, IL-4 and IFN-γ gene expressions in peripheral mononuclear cells (PBMC) were examined. Results : 1) IL-4 gene expression in spontaneous PBMC was higher in AD patient groups than in control group, significantly higher only in AD patient group with high serum IgE level (p $lt; 0.05 ). 2) IFN-γ gene expression in spontaneous PBMC showed increased tendency in AD patient groups than in control group without statistical significance. 3) IL-4 and IFN-γ gene expressions in stimulated PBMC were not different among all three groups. Conclusion : In light of our results, high and low IgE subgroups in AD can exist and AD may not be characterized by the shift in the reciprocal relationship between IL-4 and IFN-γ when T cells are stimulated under antigen presenting cell-independent conditions.

      • A Study on the Resistance and Wake Characteristics of a Full Ship Series

        A series of towing tank tests were carried out for 18 full ship models of high block coefficients. The resistance coefficients and wake distribution at the propeller plane were measured and carefully examined. Regression analysis was employed to find out the relationships with the hull form parameters. Equations for wave resistance coefficient, form factor, and nominal wake are given. A harmonic analysis of measured wake was performed to look into the influence of the local stern shape on the magnitude of fluctuating wake components at three different radii. The amplitude of wake harmonics was also expressed by regression quations. It was found that the regression formulas were very useful in estimating resistance and circumferential wake characteristics of full ship models. It was also considered that the formulas presented in this paper could be utilized in the hull form improvement in a preliminary design.

        본 연구에서는 몬테칼로 시늉내기 방법을 사용하여 자기장에 의한 쩔쩔맴이 있는 경우 결합무질서가 있는 3차원 XY 모델의 상전이를 살펴본다. 그 결과 쩍쩔맴이 f = 1/2 인 경우 자기장 방향의 상관관계거리를 결정하는 임계지수로 z = 1.0이고, 자기장에 수직한 방향의 상관관계거리 임계지수로 v = 0.67을 얻었다. We have studied the phase transition in a bond-disordered 3D XY model using Monte Carlo methods in the presence of frustration due to a magnetic field. As a result, we obtain z = 1.0, which is the critical exponent characterizing the correlation length along the magnetic field, and v = 0.67, which is the correlation length exponent, when frustration is f = 1/2.

        IL-4 and IFN-γ gene expressions in peripheral mononuclear cells(PBMC) were compared by semi-quantitative reverse transcription-polymerase chain reaction (semi-quantitative RT-PCR) between atopic dermatitis(AD) patients showing high serum IgE level and those showing low serum IgE level. The subjects of this study were 15 cages; 5 AD patients with high serum IgE(>2,000kU/l), 5 AD patients with low serum IgE(<100kU/l), and 5 healthy controls. Cultures of PBMC were harvested at 0 hr(unstimulated), 3 hr, 6 hr, 12 hr, and 24 hr(stimulated). Phorbol 12-myristate 13-acetate in combination with calcium ionophore A231879(PMA / CA) was used as a stimulant. Semi-quantitative RT-PCR was performed. The results were as follws; 1) IL-4 gene expression in unstimulated PBMC was higher at AD patient groups than at control group but statistically higher only at AD patient group with high serum IgE level than at control group (P<0.05). 2) IFN-γ gene expression in unstimulnted PBMC was higher at AD patient groups than at control group without statistical significance. 3) IL-4 mRNA was first detected at 3 hr after stimulation in vitro and persisted until 6 hr, and slowly decreased in all three groups. IL-4 gene expression was higher at 3 hr than at 12 hr or 24 hr in AD patient group with high IgE level(P<0.05). 4) IFN-γ gene expression was first detected at 3 hr and Persisted until 24 hr. IFN-γ gene expression was lower at 24hr than at 3 hr, 6hr, and 12hr in AD patient group with low IgE level(P<0.05). 5) IL-4 and IFN-γ gene expressions in stimulated PBMC were not different among all three groups. In light of our results, it is suggested that T cells of AD patients are more activated in vivo because IL-4 and IFN-γ gene expressions are increased at AD patient groups than at control group. Since there are no differences of IL-4 and IFN-γ gene expressions in PBMC stimulated with PMA / CA, it may be suggested that AD is not characterized by the shift in the reciprocal relationship between IL-4 and IFN-γ production when T cells are stimulated under antigen presenting cell - independent conditions.

        Crateva unilocularis Buch‑Ham leaf extract improves glucose metabolism via regulation of insulin secretion and sensitivity in vitro and in vivo

        Crateva unilocularis Buch-Ham has traditionally been used in Nepal for the treatment and prevention of diabetes. However, scientific verification through studies on the preventive effect of C. unilocularis on diabetes has not been done properly. In this study we investigated the effect of C. unilocularis leaf extract (CULE) on glucose uptake and insulin resistance. The effect of CULE on glucose uptake in an in vivo system was measured using zebrafish. In the cell-free system, enzymes activities related to diabetes were measured. Moreover, in the cell-cultured system, RINm5F pancreatic beta cells, 3T3-L1 adipocytes, and L6 myotubes were used to measure the effect of CULE on insulin secretion and glucose metabolism. CULE effectively enhanced glucose uptake in zebrafish larvae, and inhibited the activity of dipeptidyl peptidase-IV (DPP-IV) and protein tyrosine phosphatase-1B (PTP-1B) enzymes related to insulin secretion and insulin signaling activation. Also, CULE significantly increased insulin secretion with suppression of NO production in RIN-m5F pancreatic beta cells. In L6 myotubes and TNF-α-induced insulin resistance model of 3T3-L1 adipocytes, CULE significantly increased glucose uptake and immunofluorescence staining of glucose transporter (GLUT)4 protein. Furthermore, the regulatory response of glucose metabolism by CULE was a close correlation with the activation of insulin signaling (IRβ, PI3K, AKT) and 5′-AMP-activated protein kinase (AMPK)α and the reduction of p38 mitogen-activated protein kinase (MAPK) in TNF-α-induced insulin resistance model of 3T3-L1 adipocytes. Thus, our results suggest that CULE may act as a potential agent for the prevention and treatment of metabolic syndrome associated with type 2 diabetes.

        옥덩굴 에탄올 추출물의 당 대사 및 인슐린 민감성 개선효과

        이 연구의 목적은 Caulerpa okamurae 에탄올 추출물(COE)이 제2형 당뇨병 치료의 약물 표적 중 하나인 당 대사 및 인슐린 민감성에 미치는 영향을 평가하는 것이다. COE는 in vitro 실험에서 단백질 티로신 포스타제 1B (PTP1B)와 디펩티딜 펩티데이즈-IV (DPP-IV) 효소 활성을 유의하게 억제시켰다. 또한, COE는 3T3-L1 지방세포와 제브라피쉬에서 당 흡수, 인슐린 수용체 기질(IRS-1) 및 당 수송체(GLUT4) 단백의 발현을 대조군에 비해 유의하게 향상시켰다. L6 근육세포의 덱사메타손(dexamethasone)으로 유도된 인슐린 저항성 모델에서도 COE는 인슐린 신호전달 및 당 흡수 단백의 발현을 효과적으로 증가시켰다. 더불어 인슐린 저항성 지표로 알려진 IRS-1 Ser307의 인산화 활성도 COE 첨가에 의해 유의하게 억제되었다. 그러나 COE는 췌장 베타세포의 인슐린 분비에는 아무런 영향을 미치지 않았다. 결론적으로 COE는 인슐린 표적세포와 제브라피쉬에서 인슐린 신호전달과 당 수송체 GLUT4 단백 발현의 조절을 통해 당 대사 및 인슐린 민감성을 개선시키는 것으로 밝혀졌다. The aim of this study is to examine the effect of Caulerpa okamurae ethanol extract (COE) on glucose metabolism and insulin sensitivity as one of the drug targets for treatment of type2 diabetes. COE significantly inhibited protein tyrosine phosphatase (PTP1B) and dipeptidyl peptidase-IV (DPP-IV) enzyme activities in vitro assay. Also, COE significantly enhanced the glucose uptake and the expression of insulin receptor substrate-1 (IRS-1) and glucose transporter4 (GLUT4) proteins in 3T3-L1 adipocytes or zebrafish larvae compared with control. In dexamethasone-induced resistance model of L6 myotubes, the protein expression of insulin signaling and glucose uptake was effectively increased by the treatment of COE. In contrast, the elevated phosphorylation of IRS-1 Ser307 was normally suppressed by treatment of COE. However, COE had no effect on insulin secretion in pancreatic beta cells. Thus, our results suggest that COE improves the glucose metabolism and insulin sensitivity through the regulation of insulin signaling and GLUT4 protein in insulin’s target cells and zebrafish larvae.

        삼색싸리 메탄올 추출물의 3T3-L1지방세포와 db/db 마우스에서의 PPARγ 작용제와 인슐린 유사효과를 통한 혈당조절 개선효과

        이 연구의 목적은 당 대사에 대한 삼색싸리(Lespedeza maximowiczii var. tricolor Nakai; LMTN)의 효과를 조사하는 것이다. LMTN 추출물은 대조군과 비교하여 3T3-L1 지방 세포에서 당 섭취능 및 지질축적을 유의하게 향상시켰다. 또한, 3T3-L1 지방 세포에서 LMTN 추출물은 퍼옥시좀 증식제 활성화 수용체(PPAR)γ, 인슐린수용체기질-1 (IRS-1) 및 포도당수송체(GLUT)4의 단백질 발현을 유의하게 증가시켰다. LMTM 추출물의 당 섭취능 또는 인슐린 신호 전달계의 조절 효과는 양성 대조물질인 트로글리타존 또는 피니톨보다 낮았지만 PPARγ 단백 활성화는 증가하였다. 또한, LMTM 추출물은 인슐린 유사효과를 나타냈다. db/db 마우스에서, LMTN 추출물(250 mg/kg BW)은 물과 식이 섭취량, 혈당, 중성지방과 총 콜레스테롤 함량을 유의적으로 감소시켰다. 더불어 지방과 근육조직에서의 PPARγ 및 GLUT4 mRNA의 발현도 LMTN 추출물 투여군에서 유의적으로 증가되었다. 따라서, 본 연구의 결과는 LMTN 추출물이 3T3-L1 지방세포 및 db/db 마우스에서 PPARγ 및 인슐린 유사효과를 통해당 대사를 조절하는 것으로 밝혀졌다. The aim of this study is to investigate the effect of Lespedeza maximowiczii var. tricolor Nakai (LMTN) on glucose metabolism. LMTN extract significantly enhanced the glucose uptake and lipid accumulation in 3T3-L1 adipocytes compared with control. Also, LMTN extract in 3T3-L1 adipocytes significantly increased the protein expression of peroxisome proliferatoractivated receptor (PPAR)γ, insulin receptor substrate-1, and glucose transporter (GLUT)4. The regulatory effect on glucose uptake or insulin signal transduction of LMTM extract was lower than troglitazone or pinitol such as the positive control, but increased PPARγ activation. Additionally, LMTM extract has an insulin-mimetic effect. In db/db mice, LMTN extract (250 mg/kg BW) significantly reduced water and food intake, blood glucose, and level of plasma triglyceride and total cholesterol. Furthermore, the expression of PPARa and GLUT4 mRNA in adipose or muscle tissue effectively was increased by oral treatment of LMTN extract. Thus, our results suggest that LMTN extract improves the glucose metabolism through PPARγ and insulinmimetic effect in 3T3-L1 adipocytes and db/db mice.

        수종의 상용 세치제들의 향균 및 향염효과 비교연구

        It is known that some natural extracts from plants have a various range of antimicrobial and antiinflammatory activity. There are lots of clinical trials to develop toothpastes containing natural extracts for prevention of dental caries and gingival inflammation. The purpose of this study was to evaluate antimicrobial and antiinflammatory activity of magnolol containing toothpastes and other commercial toothpastes. Eleven kinds oftoothpastes were used. They include magnolol, sanguinarine, Myrrha, Mori radicis cortex,Cimicifugae rhizoma, sodium fluoride, aminocaprolactic acid etc. Six strains of bacteria were used for this test, ego Porphylomonas gingivalis, Prevotellain-termedia, Actinobacillus actinomy cetemcomitans, Streptococcus mutans, Stretococcus sanguis, and Actinomyces species. Antimicrobial activity was determined by an agar dillution method and a broth microdillution method. Antiinflammatory activity was assessed by the inhibition of $PGE_2$ production from gingival fibroblast with the addition of rHIL-1 and centrifuged solution of toothpastes. Control group was only rHIL-1 additive sample. $PGE_2$ enzyme immunoassay systemfAmersham, In. Buckinghamshire, U.K). $PGE_2$ level was measured by ELISA reader with 450 nm, The results from the study revealed that toothpastes containing natural extracts generally had high antimicrobial and antiinflammatory activity. Especially magnolol containing toothpaste showed higher antimicrobial activity than other toothpastes, and sanguinarine containing toothpaste showed particularly high antimicrobial activity in A. actinomicetemcomitans and A. viscosus. In some degree all toothpastes inhibited $PGE_2$ production, but magnolol containing toothpaste was potent inhibitor of $PGE_2$. Sodium chloride containing toothpaste had also effective result. The results suggested that toothpastes containing natural extracts were promising in plaque control and prevention of dental caries and gingivitis.
