        牧楡湯이 人體 肺癌細胞의 變化에 미치는 影響에 關한 分子生物學的 硏究

        This experimental study was performed to investigate the effects of Mokyou-Tang on cell cycle and gene expressions of human lung cancer cell. I examined the cell cycle progression and cell cycle-related gene expression in A549 using a flow cytometry and a quantitative PT-PCR analysis. The treatment groups, in accordance with concentration of Mokyou-Tang were classified as 3 group(MYT 100㎕/㎖, MYT 500㎕/㎖, MYT 1,000㎕/㎖), and according to treatment time, were classified as 2 group(24hours, 48hours). The results were summarized as follows; 1. In cell cycle stage, MYT 100㎕/㎖(48hours) and 1000㎕/㎖(48hours) groups were increased as compared with control group, especially 1000㎕/㎖(48hours) group showed eminent increase. 2. In p53 gene, MYT 500㎕/㎖(48hours) and 1000㎕/㎖(48hours) groups showed eminent suppressive effect to cancer cell. 4. CDKNI gene was decreased in all treatment groups, this mean that MYT has suppressive effect to proliferation of cancer cell. 4. In GADD45 gene was decreased in all treatment groups, this mean that MYT has suppressive effect to proliferation of cancer cell. The results suggest that MYT promotes G1 cell cycle arrest and suppresses the proliferation of cancer cell. Thus, it seems likely that MYT has suppressive effect to proliferation of lung cancer cell.

        Provisional implant의 임상적 적용례

        It has been reported that for implatns to become osseointegrated, they must heal in the absence of functional loads for 4 to 6 months. The provisional implants were designed to support provisional restorations and to allow for load-free osseointegration of definitive implants, while the was provided with immediate esthetic and function. In addition, the provisional implant protects graft site. Also the provisional implant provides anchorage for orthodontic treatment and reconstructs vertical dimension in edentulous condyle fracture to practice functional therapy. The provisional implant system is a sound and economical method to achieve patient's immediate masticatory and esthetic function. Three cases are presented to document and illustrate the clinical application.

        科學領域에서의 聯想辭典作成과 그 活用性

        The fundamental and important thing, for science education, is to clarify the concept that represented by language. Accordingly, we introduce "the associative method", which is largely treated on psychology and as we measure the associative frequency of the words containing scientific meaning, we make an associative dictionary. Finally it would suggest the utilizable method in the actual situation of instruction of a science course of secondary school. That is to say, as we measure the frequency of associative word on a given word, we catch the intuation of preconception concerned with that word. As we deal with it educationally, we will search the effective method which can be utilized in teaching of concept which is the most important task in the science education. It was identified that the frequency of experience in common things is the decisive factor of the frequency of the association for the given atimulating word. Therefore, we could conclude that the associative dictionary producted on this thesis, which is considered the considered the content structures of subject matter and students' cognitive structures, can be utilized as an useful material in a real situation of instruction.

      • 농산물중의 잔류농약 실태에 관한 연구(Ⅰ) : 고체상 추출에 의한 농산물중 잔류농약 분석에 관한 연구 Analysis of Residual Pesticides in Agricultural Products by Solid-Phase Extraction

        신속 · 정밀하고 효율적인 짠류농약 검출을 위하여 극싱칼럼 (Extrelut-3')을 사옹한 SPE법을 시도하였다. 시료는 채소류체 사용하는 농약중 잔류성이 있고 식품공전상 시험 전처리가 각각 다른 Chlorothalonil등 6종의 농약을 선정하고 그들 표준액에 대한 PPIR수준의 농도로추출용라의 종류 및 용매량 등의 추출조건을 달리하여 회수율 등을 측정한 결과 최적의 분석조건을 얻었다. 1) 추출 용매량은 증가할수록 추출 수율이 높았고,용매 종류에 대한 추출수율은 각 농약별로 다소 차이는 있었으나 극성 정도가 비교적 높은 Ethyl acetate애서 평균 추출수율이 가장 높았다 2) 최대 평균회수율은 Ethyl acetate 60mL를 사웅할 경우로서 93.6%였다. 3)표준액을 시료에 첨가한 시험에서도 역시 Ethyl ace늘to가 푼출수율이 가장 높았으며 불순물 분리제거 효과도 LLE법 보다 월등히 우수하였다. 4) 각 농약별 검출한계는 치소 0.001ppin으로 농산물 중 미량 잔류하는 농약을 검출할 수 있을 것으로 사료된파. 그러므로 SPE법은 LLE법보다 경제적인 방법으로 판명되었으며, 앞으로 농산물 중 잔류농약 분석에서 LEE법을 대치할 쑤 있을 것으로 기대된다. Solid-phase extraction by polar column(Extrelut-3') was attempted to develope the fast and efficient method of detecting pesticide of farm product.5·ix kinds of pesticide used in farming fieldfrequently and different in pretreatment, were chosen from Korea Food Cord. Optfmal analysis condi-tions were determiBed from t31e recovery rate of standard pesticides according to extraction solvent andextraction volurae. Extraction yietd was increased as solvent polarity and extraction volume. Maximumrecovery rate was acquired at 93.6% when 60mL of ethyl acetate was used as eluent. Tllis method wasmore effective than liquid-liquid partition extract·ion method to eliminated the impurity and had 0.001ppm of detection limit. Therefore. solid-phase eEl=raction was expected to be economical substitute fortraditional liquid-liquid extT·action method.

        SJO¨GREN'S SYNDROME 환자에게 즉시 임프란트 술식을 행한 치험례

        The old edentulous patients experience xerostomia commonly. They have considerable difficulty in wearing tissue-supported prosthesis and high risk rate of caries. One of them is Sj□gren's syndrome patient. Sj□gren's syndrome is a chronic inflammatory autoimmune disorder that results in a lymphocyte-mediated destruction of exocrine glands. We planned to place implants immediately after extraction in this case of autoimmune disease. The advantages of immediate implant placement include considerable decrease in time from tooth extraction to placement of the final prosthesis, fewer surgical procedures, and better acceptance of the overall treatment plan. We present a case of Sj□gren's syndrome patient treated with immediate implant placement & immediate loading in anterior region and immediate implant placement in posterior region.

        The seeds of 163 taxa at front the diverse wild populations in south Korea were collected by our staffs of the Arboretum for six years (1995-2000). More then 138 taxa (ca, 85%) were collected mainly from September to October. Successful long-terml survey is recommended to determine the befit dates for seed collections of woody plants in Korea.

        A role for Leu247 residue within transmembrane domain 2 in ginsenoside-mediated α7 nicotinic acetylcholine receptor regulation

        <P>Nicotinic acetylcholine receptors (nAChRs) play important roles in nervous system functions and are involved in a variety of diseases. We previously demonstrated that ginsenosides, the active ingredients of Panax ginseng, inhibit subsets of nAChR channel currents, but not alpha7, expressed in Xenopus laevis oocytes. Mutation of the highly conserved Leu247 to Thr247 in the transmembrane domain 2 (TM2) channel pore region of alpha7 nAChR induces alterations in channel gating properties and converts alpha7 nAChR antagonists into agonists. In the present study, we assessed how point mutations in the Leu247 residue leading to various amino acids affect 20(S)-ginsenoside Rg(3) (Rg(3)) activity against the alpha7 nAChR. Mutation of L247 to L247A, L247D, L247E, L247I, L247S, and L247T, but not L247K, rendered mutant receptors sensitive to Rg(3). We further characterized Rg(3) regulation of L247T receptors. We found that Rg(3) inhibition of mutant alpha7 nAChR channel currents was reversible and concentration-dependent. Rg(3) inhibition was strongly voltage-dependent and noncompetitive manner. These results indicate that the interaction between Rg(3) and mutant receptors might differ from its interaction with the wild-type receptor. To identify differences in Rg(3) interactions between wild-type and L247T receptors, we utilized docked modeling. This modeling revealed that Rg(3) forms hydrogen bonds with amino acids, such as Ser240 of subunit I and Thr244 of subunit II and V at the channel pore, whereas Rg(3) localizes at the interface of the two wild-type receptor subunits. These results indicate that mutation of Leu247 to Thr247 induces conformational changes in the wild-type receptor and provides a binding pocket for Rg(3) at the channel pore.</P>

        Effect of Sesbania Incorporation as Nitrogen Source on Growth and Yield of Whole Crop Barley and Reduction of N Fertilizer in Saemangeum Reclaimed Tidal Land

        It is known that the poor soil fertility of newly reclaimed saline soils is due to the lack of organic matter and available mineral nutrients for crop production. The effect of green manuring with Sesbania aculeata in combination with five rates of urea-N treatments (N0. N25, N50, N75, N100) on the productivity of a subsequent whole-crop barley and the fertility of the reclaimed saline soil in Saemangeum was evaluated in the field during 2013-2014 growing season. Sesbania was grown during summer season (June to October). The amount of Sesbania incorporated was 16.2 Mg ha<SUP>-1</SUP>. Sesbania contributed to 393 kg N ha<SUP>-1</SUP> to the soils when ploughed down and incorporated before whole-crop barley cultivated. The performances of whole-crop barley following sesbania incorporation were significantly affected by a combination of Sesbania manuring and different N rates. The N fertilizer equivalence without N fertilizer following Sesbania was 42.6% (63.9 kgN ha<SUP>-1</SUP>), compared with N100(150 kg N ha<SUP>-1</SUP>) in fallow soils. The whole-crop barley yield responded to N fertilizer rates in both sesbaniaamended and fallow soil. The yield response to nitrogen rates in fallow soil was linear (Y=0.0586X+3.3011, R<SUP>2</SUP> =0.9534), whereas that in sesbania-amended soils was quadratic (Y= -0.001X<SUP>2</SUP>+0.1322X+5.7143, R<SUP>2</SUP>=0.9576). The yield of whole-crop barley in sesbania-amended with increasing N rates was increased up to SN75 (115 kgN) 10.3 Mg ha<SUP>-1</SUP>. Apparent N recovery (ANR) of whole-crop barely showed decreased with sesbania plus increasing rates of N fertilizer. Despite higher yield with sesbania manuring plus increasing N rates, the contributions of N from Sesbania with increasing N rates to whole-crop barley were decreased, whereas those from fertilizer increment due to excessively mineralized Nitrogen. Considering yield, ANR, N contribution from Sesbania and nitrogen fertilizer, the optimum N rate was N50 rate following sesbania incorporation.
