      • 집토끼의 시상하부내에서 Vasopressi과 Oxytocin 분비세포의 미세구조에 대한 면역세포화학적 연구

        An immunocytochemical study was performed using the specific monoclonal antibodies against arginine vasopressin (VP) and oxytocin(OT) to demonstrate vasopressinergic and oxytocinergic neurons in the hypothalamus of the domestic rabbit. 1. VP and OT neurons were distributed mainly in the supraoptic and paraventricular nuclei. 2. The VP and OT neurons were round, oval, spindle and polygonal in shape, and the diameters of the neurons were 15-25um in domestic rabbit. 3. VP neurons were dominant in the supraoptic nucleus and OT neurons were in the paraventricular nucleus. 4. From the electron microscopic observations, the rough endoplasmic reticulum, mitochondna, Golgi apparatus and microtubules were found in the VP and OT neurons.

        치수제거후 흰쥐 삼차신경절에서 VIP 면역반응세포의 변화: 공초점레이저주사현미경적 연구

        말초신경 손상에 의한 VIP의 변화를 연구하기 위해 흰쥐 하악대구치 치수제거 후 삼차신경 절에서 VIP의 분포 및 반응강도를 공초점레이저주사현미경을 이용하여 관찰하였다. 체중 200g내외의 Sprague-Dawley계 흰쥐를 대조군과 하악대구치 치수제거 후 14일군으로 분리하여 희생시켰다. 1차 항체로 rabbit anti-VIP, 2차 항체로 fluorescein isothiocyanate(FITC) conjugated anti-rabbit IgG를 사용하여 면역형광염색을 시행한 후 공초점레이저주사현미경으로 관찰하여 다음과 같은 결론을 얻었다. 1.삼차신경절 하악부위에서 VIP 양성반응세포의 비율은 대조군에서 7.40%를, 실험군에서는 28.42%를 보였다. 대조군에 비해 실험군에서 양성반응세포의 증가를 보였다. 2.삼차신경절 하악부위에서 VIP 면역반응세포체에 대한 상대성 형광강도는 대조군에서 87.78을, 실험군에서는 138.65를 보였다. 대조군과 비교하였을 때 실험군에서 상대성 형광강도의 증가를 보였다. 3.실험군의 광연속절편(1㎛) 관찰에서 VIP 면역반응세포는 9개의 절편 대부분에서 강하게 나타났다. 축삭의 면역반응을 살펴보면, 대조군의 축삭에서는 약한 반응을 보였으며, 실험군의 축삭에서는 강한 면역반응을 보였다. 또한 양성반응 세포체의 크기는 20∼25㎛의 중간 크기의 세포체에서 강한 면역반응을 보였다. 위의 결과로 보아 치수제거 후에 삼차신경절 하악부위에서 VIP면역반응세포의 증가와 함께 상대성 형광강도가 높아졌음을 알 수 있었다. The purpose of this study was to investigate the distribution and fluorescence intensity of vasoactive intestinal polypeptide immunoreactive(VIP-IR) cells in rat trigeminal ganglion following pulp extirpation of rat mandibular molar. The animals were divided into control group(n=6) and experimental group(n=6). The experimental animals were sacrificed at 14 days after pulp extirpation. The trigeminal ganglion was removed and immersed in the 4% paraformaldehyde in 0.1M phosphate buffer. Serial frozen sections about 20㎛ in thickness were cut with a cryostat The immunofluorescence staining was performed. The rabbit anti-VIP(1:8,000) was used as primary antibody and fluorescene isothiocynate(FITC) cojugated anti-rabbit IgG(1:80) as secondary antibody. The slides were observed under confocal laser scanning microscope(CLSM). Unprocessed optical sections were obtained and stored on a optical disk. Color pictures were printed by a video copy processor. The results were as follows ; 1. The positive ratio of VIP-IR cells in mandibular part of trigeminal ganglion were 7.40% in control group and 28.42% in experimental group(14 days affter pulp extirpation). 2. The relative fluorescence intensity of VIP-IR cells in mandibular part of trigeminal ganglion were 87.78 in control group and 138.65 in experimental group. The relative fluorescence intensity of experimental group was 58% higher than that of control group. 3. In optical serial section analysis of VIP-IR cells of experimental group, most of the 9 section showed high fluorescence intensity. At high magnification, axons of the experimental group displayed greater VIP-IR than in the control group, and the positive cells were mainly of medium size. The result indicate that number and fluorescence intensity of VTP-IR cells were increased in the mandibular part of trigeminal ganglion following pulp extirpation of mandibular molar, and it suggests that VIP could play a role in processing of nociception.

      • Microprocessor를 利用한 直流電動機의 速度制御

        This paper is concerned with the seperately excited dc motor speed control using a microprocessor. The voltage, necessary for dc motor drive, is adjusted by a chopper and its control circuits are fired with digital circuits. Therefore, no D/A converter is needed between power circuits and a microprocessor. A voltage that relates to the motor speed, is measured through a tachogenerator. The analog voltage, measured through a tachogenerator, is converted into the digital signal by the A/D converter and the converted digital signal goes into the microprocessor. For the purpose of maintaining tho motor speed constant, the digital signal is compared with a reference setting, and the error is used to adjust the on-off time of the chopper. Throughout the experiments, we recognized that the dc motor reaches at the desired speed rapidly and maintains the reference speed in spite of a variable load.

        상아모세포 관련 유전자, OD314의 발현과 기능 연구 : OD314

        Odontoblasts are responsible for the formation and maintenance of dentin. They are known to synthesize unique gene products including dentin sialophosphoprotein (DSPP). Another unique genes of the cells remain unclear. OD314 was isolated from the odontoblasts/pulp cells of rats and partially characterized as an odonto-blast-enriched gene (Dey et al., 2001). This study aimed to elucidate the biological function of OD314, relating to odontoblast differentiation and dentinogenesis. After determining the open reading frame (ORF) of OD314 by transient transfection analysis using green fluorescent protein (GFP) expression vector, mRNA in-situ hybridization, immunohistochemistry, reverse transcription-polymerase chain reaction (RT-PCR) and western analysis were performed. The results were as follows: 1. In in-situ hybridization, OD314 mRNAs were expressed in odontoblasts of developing coronal and root pulp. 2, OD314 was a novel protein encoding 154 amino acids, and the protein was mainly expressed in cyto-plasm by transient transfection analysis. 3. Mineralized nodules were associated with multilayer cell nodules in the culture of human dental pulp cells and first detected from day 21 using alizarin-red S staining. 4. In RT-PCR analysis, OD314, osteocalcin (OC) and DSPP strongly expressed throughout 28 days of culture. Whereas, osteonectin (ON) mRNA expression stayed low up to day 14, and then gradually decreased from day 21. 5. Western blots showed an approximately 17 kDa band. OD314 protein was expressed from the start of culture and then increased greatly from day 21. In conclusion, OD314 is considered as an odontoblast-enriched gene and may play important roles in odontoblast differentiation and dentin mineralization.

        The objective of this study was to help decision-makers to be able to make a decision on a social tourism investment, based on the reasonable estimate standard. This was possible by introducing the estimate method for the effectiveness of investment on social tourism project and by using this method on the analysis of the alternative investment projects. For the analysis and estimate of welfare tourism investment, the advanced cost-benefit model was developed, based on the previous traditional cost-benefit analysis. This new cost-benefit model supplemented the weakness fo traditional cost-benefit model by utilizing the concept of total-social-benefit and total-social-cost.

      • 단지양 근위축증(Monomelic Amyorophy) 3례 : 증례 보고 Case Report

        Monomelic amyotrophy is a rare form of motor neuron disease which is insidious onset in the second and fourth decades, male preponderance, sporadic occurrence, wasting and weakness confined to one limb, and absence of involvement of the cranial nerves, cerebrum, brain, stem, and sensory system. Symptoms and signs often progress for several years before spontaneously arresting. Neurologic findings and electrophysiologic studies are only useful for the diagnosis of monomelic amyotrophy and other laboratory testing are frequently normal or nonspecific. We reported three cases of typical monomelic amyotrophy with review of the articles.

      • X-관련성 만성육아종성질환의 진단을 위한 PMA-activated NBT 검사

        목적 : Activated NBT 검사는 만성육아종성질환을 선별하는 검사이다. 활성제로 PMA를 사용하면 X-linked CGD의 경우 보인자를 감별할 수 있다. 방법 : CGD의 가족력을 부인하는 정상인 30명을 대상으로 PMA activated NBT 슬라이드 검사를 실시하여, 정상대조에서의 formazan 형성율을 보았으며, 동일 검사를 CGD가 강력히 의심되는 환자와 그 가족에 대하여 실시하였다. 결과 : 정상대조 30명 전원에서 98% 이상의 formazan 형성세포가 관찰되었다. 환자에게 실시한 결과 세환자 모두에서 0%의 결과를 보였다. 환자의 어머니에서는 각각 57, 70, 67%의 결과를 보였다. 결론 : 본 연구를 통하여 세 환자는 X-관련성 만성육아종성질환이라는 진단을 내릴 수 있었다. PMA activated NBT 슬라이드 검사는 시행하기가 쉽고 재현성이 좋으므로, 어러 검사실에서 소아기 면역결핍증을 선별하는 검사의 하나로 시행되는게 바람직하다고 사료된다. Background : Chronic granulomatous disease(CGD) is uncommon inherited disorder in which phagocytic cells fail to produce antimicrobial superoxide(O₂) due to NADPH oxidase deficiency. This disease can be easily screened by activated nitroblue tetrazolium(NBT) test. In case of X-linked CGD, carriers also can be detected. Methods : PMA(phorbol myristate acetate) activated NBT test was performed as previously described. Test performance was evaluated in 30 normal persons who denied family history mimicking CGD. Test was also applied to the three strongly suspected cases of CGD. Two of the cases were 12 year old boy and 7 year old boy, who suffered from many infectious diseases since 6 month old and 4 month old, respectively. The other remaining case was 22 year old man who suffered from many infectious diseases since 6 year old, but now relatively in health compared to the other two cases. Results : More than 98% formazan formation was observed in all normal controls. No formazan positive cell was shown in all of the three patients. However, applied to the mothers, the test revealed 57%, 70%, 67% positive cells, respectively. Conclusion:X-linked transmission of CGD could be determined with the results of PMA activated NBT slide test. This test can be easily performed. It would be better to do this test routinely as one of the tests for the screening of the immmunodeficiency syndrome in childhood.

      • 전환전략: Marriott 기업의 리스트락쳐링에 관한 사례연구

        현재의 복잡하고 급변하는 기업환경하에서 경영상 심각한 어려움에 처해있는 많은 기업들을 볼 수 있다. 특히 호텔산업에서도 기업 도산이라는 최악의 상태로 까지 가는 경우를 흔히 접하게 된다. 그러나 어려움에 처한 기업이라고 반드시 도산한다고는 볼 수 없다. 쇠퇴의 이유에 대한 정확한 인식과 적절한 전환전략의 사용으로 기업은 회생할 수 있게 된다. 기업의 쇠퇴기에서 회복국면으로의 전환에 관한 학문적인 접근의 필요성에도 불구하고, 호텔산업분야에 있어서 전환전략들에 관한 체계적인 연구는 아직 미흡하다고 볼 수 있다. 본 연구의 목적은 기존 연구 문헌 조사와 현재 리스트락쳐링(기업구조개편) 중에 있는 미국의 MARRIOTT 기업에 대한 사례연구를 통해 전환전략개념을 이해하는 데 있다. On the current complex and volatile environment, many hospitality companies are confronting severely declining performance and are in serious trouble. However, troubled companies are not usually destined to fail: a successful turnaround is possible by understanding the cause of decline and using proper turnaround strategies. In spite of the increasing need of in-depth study on corporate turnarounds, yet there is a lack of systematic analysis about turnaround strategies in hospitality industry. The purpose of this paper is to synthrsize strategic concepts concering turnaround strategies through literature review and case study of Marriott corporation which is under restructuring.
