상황버섯(Phellinus linteus)으로부터 Ergosterol의 분리

상황버섯을 80% MeOH 용액으로 추출하고, 추출물을 EtOAc, n-BuOH 및 물로 분배, 추출하였다. 이 중 EtOAc 분획을 silica gel 및 octadecylsilica gel(ODS) column chromatography로 정제하여 1종의 sterol 화합물을 분리하였다. 화합물의 화학구조는 NMR, MS 및 IR 등의 스펙트럼 데이터를 해석하여, ergosta-7,22E-diene-3β,5α,6β-triol(cerevisterol, 1)로 동정하였다. 이 화합물은 상황버섯에서 처음 분리 보고 되었다. Phellinus linteus was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and H_(2)O. The repeated silica gel and ODS column chromatographies of the EtOAc fraction led to the isolation of one sterol. From the result of spectral data including NMR, MS and IR, the chemical structure of the sterol was determined as ergosta-7,22E-diene-3β,5α,6β-triol (cerevisterol, 1). Compound 1 was isolated from the Phellinus linteus for the first time in this study.

To investigate the microfloral composition in activated sludge, more than six hundred bacterial and fungal strains isolated from influent sewage, return sludge and aeration tank of an activated sludge sewage treatment plant were identified to the genus level. During the holding period in the aeration tank development of diverse heterotrophic microbial populations occurred and bacterial and fungal numbers in the aeration tank were 10- to 100-fold higher than in the influent sewage and return sludge. A large number of Gram-negative rods, particularly Pseudomonas, Flavobacterium, Enterobacter, Klebsiella, and Alcaligenes were found to be the dominant genera in the aeration tank. The species of filamentous fungi Trichoderma, Aspergillus, Rhizopus and Lepiomitus, and various yeasts were frequently associated with activated sludge. The most commonly found yeast genera were Candida, Geotrichum, Saccharomyces, Torulopsis and Rhodotorula.
CRW형식 무인항공기 추진시스템의 밸브작동을 고려한 비행모드 전환에 따른 천이성능 특성 연구

본 연구는 CRW형식 무인항공기 추진시스템의 밸브작동을 고려한 비행모드에 따른 천이 성능특성을 파악하기 위해 SIMULINK를 이용하여 모델링 하였다. 주 엔진시스템의 천이 모사에는 ICV방법이 적용되었다. 그리고 밸브시스템은 로터리 덕트와 메인 덕트로 빠져나가는 유량을 제어하는 시스템으로서 밸브를 통해 로터리 덕트로 빠져나가는 유량과 메인 덕트를 빠져나가는 유량의 합은 터빈의 출구 유량과 같다는 가정하에 수행되었으며, 이 때 밸브각 변화에 따른 손실, 유량 및 유효면적 등이 고려되었다. 성능해석은 비행 천이 영역인 고도 1Km 비행마하수 0.1에서 엔진최대회전수시 회전익 모드에서 고정익 모드로 변환시외 고정익 모드에서 회전익 모드로 변환 경우들이 수행되었다. In order to investigate transient behaviour of the CRW(Canard Rotor Wing) type UAV(Unmanned Aerial Vehicles) propulsion system during flight mode transition considering flow control valve operation, the propulsion system was modelled using SIMULINK commercial program. For transient simulation of the main engine system, the ICV(Inter-Component Volume) method was applied. The valve system is to control the gas flow of the rotary duct system and the main duct system, and the analysis was performed with an assumption that the total gas mass flow of the main engine is the same as summation of the rotary duct flow and the main duct flow, and with consideration of valve loss, flow rate and effective area in valve angle variation. The performance analysis was carried out during flight mode transitions from the rotary flight mode to the fixed wing flight mode and vice versa mode at altitude of 1Km, flight Mach number 0.1 and maximum engine rpm.

It is becoming increasing1y difficult and expensive for construction contractors not only to dispose of building waste and demolition rubble but also to obtain natural aggregates of good quality for concrete product and building construction. As one of solutions for the above-mentioned problems, this study was planned to suggest a fundamental data and method for producing recycled aggregate concrete using crushed cement bricks as fine aggregates. In this study, one normal concrete with natural river sand as fine aggregate and crushed stone as coarse aggregate and three recycled aggregate concretes according to the mix proportion of recycled fine aggregate substitution ratio of 35%, 65%, 100% were made using tile water-cement ratio of 45% and 65%.
Squalene Adjuvant를 이용한 단백질 항원의 세포성 면역반응 유도

To investigate the role of adjuvant for cytotoxic T lymphocytes (CTL) induction, squalene-based adjuvant was studied its capability eliciting to the induction of cellular immunity as well as humoral immunity to exogeneous proteins. Ovalbumin (OVA) was used as model proteins. It was demonstrated that antigen formulation consisted of metabolizable oil squalene mixed with Tween 80 and pluronic L121 (S/TJ121) could induced the antigen specific CTL responses and antibodies irrespective of immunization routes in mice. We also demonstrated that this antigen formulation was a inducer of CD8+, major histocompatibility complex(MHC) class I-restricted, and antigen-specific C;1'Ls. These data suggest that the squalene-based emulsion system is a potent adjuvant inducing cellular immunity as well as humoral immunity.

조사 구역 상류 지점의 평균 수질은 pH가 7.1, DO가 8.3mg/ℓ, BOD가 1.7mg/ℓ, COD가 1.7mg/ℓ, SS가 15.4mg/ℓ, 분변성 대장균이 1.1x10³ MPN/100ml고 상수원 수 1급 내지 2급의 양호한 상태이고 하류지점의 수질은 pH가 6.8, DO가 7.7mg/ℓ, BOD가 2.2mg/ℓ, COD가 2.4mg/ℓ, 분변성 대장균이 1.7x10³MPN/100ml로 상수원 수 2급 내지 3급의 상태를 나타내 하류로 내려가면서 오염도가 증가하는 것으로 나타났다. 청주시의 영향을 크게 받는 미호천 하류의 평균수질은 pH가 7.2, DO가 6.3mg/ℓ, BOD가 16.2mg/ℓ, COD가 15.1mg/ℓ, 분변성 대장균이 1.6x10??MPN/100ml로 심각한 오염 상태를 보여 청주시와 대전시가 금강 본류의 주 수질 오염원으로 나타났다. 영양 염류 (NH₃-N, NO₂-N, NO₃-N, PO₄-P)와 무기성 유해 성분 (Cl-, CN-, F-)의 함량은 지천의 일부 지점을 제외하고는 극히 낮은 수준으로 검출되었으며, 유기인, 페놀류, PCB 성분은 조사기간 중 전혀 검출되지 않았다. 또한 Fe, Zn, Cu, Mn, Cr, Pb, Cd, Hg등의 중금속은 검출되지 않거나 하천 수질 기준 또는 식용수 수질 기준의 허용치를 초과하지 않는 양호한 수준으로 나타났다.

This study investigated effects of five consumption values of family restaurants, that is to say, functional value, economic value, social value, creative value and emotional value, upon consumers’satisfaction and brand reliability, and effects of satisfactions upon brand reliability, and effects of satisfaction and brand reliability upon consumers’ behavioral intention to help develop family restaurant industry. Frequency analysis was done. The interviewees had demographic characteristics of gender: 156 men (54.4%) and 131 women (45.6%). The interviewees live in the places: 136 persons (47.4%) in Gyeonggi and Metropolitan area, 57 persons (27.0%) in Gangbuk, Seoul, 38 persons (13.2%) in other areas in Seoul, 34 persons (11.8%) in Gangnam, Seoul, 18 persons (6.3%) in Incheon and 4 persons (1.4%) in other regions. Values of the use of family restaurants, for instance, functional value, economic value, social value, emotional value and rarity value had influence upon satisfaction and brand reliability, had influence upon satisfaction as well as brand reliability. This study investigated consumption values of which consumers thought much, effects of consumption values upon satisfaction and brand reliability, effects of consumer’s satisfaction upon brand reliability, and consumer’s satisfaction and brand reliability upon behavioral intention, and to verify effects having influence upon local and foreign family restaurant.
Development of marker-based diagnosis for estimating the inflow time of beer-contaminating insects

Insects or insect remains found in beer are one of major issues in consumer claim. Accurate estimation of inflow time isa critical factor for the settlement of such claims related with beer-contaminating insects but no reliable methods have been developed. In an attempt to establish a molecular marker-based diagnostic method, the degradation rates of 18S rRNA genes in the insectssoaked in 500 ml beer were investigated by quantitative real-time PCR (qPCR) over one month period at room temperature. Among the six insect species tested, the house fly (Musca domestica) and honey bee (Apis mellifera) revealed high correlations (r2=0.974-0.990) between the degradation of 18S rRNA gene and inflow time. In these insects, statistically significant distinction was possible between the samples stored in beer less than 14 days and more than 14 days. Other insects, including the fruit fly, common house mosquito, German cockroach and Indian meal moth, displayed poor correlations, which appeared attributed to the inefficient genomic DNA extraction likely due to small sample size or disintegration of body parts during storage in beer. With proper improvement in DNA extraction, this 18S rRNA-based diagnostic method would be applicable for estimating the inflow time of beer-contaminating insects.

<P><B>Abstract</B></P> <P><B>Objective</B></P> <P>Renal cell carcinoma (RCC) is the most typical type of kidney cancer in adults. Hypercalcemia is a well known paraneoplastic syndrome associated with RCC and recent studies have reported that hypercalcemia is closely related to the poor prognosis of RCC patients. Clear cell RCC (ccRCC) is the most common and aggressive subtype of RCC. Although the histological classification of RCC is important for determination of appropriate treatment strategies, effective biomarkers for predicting prognosis of ccRCC have not yet been identified. Since calcium levels affect the prognosis of RCC patients, we evaluated whether the calcium-sensing genes on the plasma membrane, including those encoding calcium channels, CaSR, GPRC6a, and <I>DYSF</I>, could be used as biomarkers to predict the prognosis of ccRCC patients.</P> <P><B>Methods</B></P> <P>Information from 537 patients from The Cancer Genome Atlas (TCGA; <I>n</I> = 446) and International Cancer Genome Consortium (ICGC; <I>n</I> = 91) was used in this study. Among these genes, <I>DYSF</I> was the only gene whose expression correlated with overall survival of both TGCA and ICGC patients.</P> <P><B>Results</B></P> <P>Although <I>DYSF</I> gene expression was higher in ccRCC tissue than in normal kidney tissue, Kaplan-Meier curves showed that the survival rate of ccRCC patients with high <I>DYSF</I> expression was significantly higher than that of patients with low <I>DYSF</I> expression (TCGA, <I>P</I> < 0.0001; ICGC, <I>P</I> = 0.0011). We also validated the potential of <I>DYSF</I> as a prognostic biomarker for ccRCC by conducting a time-dependent area under the curve (AUC) analysis and 5-years receiver operating characteristic curve analysis. Finally, multivariate regression analysis revealed that the expression of <I>DYSF</I> is independent of other prognostic parameters (TCGA, <I>P</I> = 0.017; ICGC, <I>P</I> = 0.006).</P> <P><B>Conclusions</B></P> <P>These results suggested that <I>DYSF</I> may play a suppressive role in the progression of ccRCC and could act as a promising prognostic biomarker for predicting the survival of ccRCC patients.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Higher expression of <I>DYSF</I> is associated with better prognosis of ccRCC patients. </LI> <LI> <I>DYSF</I> expression in tumor tissue is higher than that in normal tissue. </LI> <LI> <I>DYSF</I> gene might be a potential prognostic biomarker for ccRCC patients. </LI> </UL> </P>
