UHRF2 is a member of the ubiquitin plant homeo domain RING finger family, which has been proven to be frequently up-regulated in colorectal cancer cells and play a role as an oncogene in breast cancer cells. However, the role of UHRF2 in glioma cells remains unclear. In this study, we performed real-time quantitative PCR on 32 pathologically confirmed glioma samples (grade I, 4 cases; grade II, 11 cases; grade III, 10 cases; and grade IV, 7 cases; according to the 2007 WHO classification system) and four glioma cell lines (A172, U251, U373, and U87). The expression of UHRF2 mRNA was significantly lower in the grade III and grade IV groups compared with the noncancerous brain tissue group, whereas its expression was high in A172, U251, and U373 glioma cell lines. An in vitro assay was performed to investigate the functions of UHRF2. Using a lentivirus-based RNA interference (RNAi) approach, we down-regulated UHRF2 expression in the U251 glioma cell line. This down-regulation led to the inhibition of cell proliferation, an increase in cell apoptosis, and a change of cell cycle distribution, in which S stage cells decreased and G2/M stage cells increased. Our results suggest that UHRF2 may be closely related to tumorigenesis and the development of gliomas.

Background: Panax ginseng Meyer (P. ginseng), a herb distributed in Korea, China and Japan, exerts benefits on diverse inflammatory conditions. However, the underlying mechanism and active ingredients remains largely unclear. Herein, we aimed to explore the active ingredients of P. ginseng against inflammation and elucidate underlying mechanisms. Methods: Inflammation model was constructed by lipopolysaccharide (LPS) in C57BL/6 mice and RAW264.7 macrophages. Molecular docking, molecular dynamics, surface plasmon resonance imaging (SPRi) and immunofluorescence were utilized to predict active component. Results: P. ginseng significantly inhibited LPS-induced lung injury and the expression of proinflammatory factors, including TNF-a, IL-6 and IL-1b. Additionally, P. ginseng blocked fluorescence-labeled LPS (LPS488) binding to the membranes of RAW264.7 macrophages, the phosphorylation of nuclear factor-kB (NF-kB) and mitogen-activated protein kinases (MAPKs). Furthermore, molecular docking demonstrated that ginsenoside Ro (GRo) docked into the LPS binding site of toll like receptor 4 (TLR4)/myeloid differentiation factor 2 (MD2) complex. Molecular dynamic simulations showed that the MD2-GRo binding conformation was stable. SPRi demonstrated an excellent interaction between TLR4/MD2 complex and GRo (KD value of 1.16 × 10<SUP>-9</SUP> M). GRo significantly inhibited LPS488 binding to cell membranes. Further studies showed that GRo markedly suppressed LPS-triggered lung injury, the transcription and secretion levels of TNF-α, IL-6 and IL-1β. Moreover, the phosphorylation of NF-kB and MAPKs as well as the p65 subunit nuclear translocation were inhibited by GRo dose-dependently. Conclusion: Our results suggest that GRo exerts anti-inflammation actions by direct inhibition of TLR4 signaling pathway.

The breeding of domestic rabbits (Oryctolagus cuniculus) for human consumption has a long tradition in China. Infections that can affect the production of meat or even be transmitted from animals to humans are important to monitor, especially for public health reasons as well as for their impact on animal health. Thus, a total of 1,132 domestic rabbit sera from 4 regions in China were collected for serological screening for Encephalitozoon cuniculi and for Toxoplasma gondii by ELISA and modified agglutination test (MAT), respectively. Antibodies to E. cuniculi were detected in 248/1,132 (21.9%) sera tested while antibodies against T. gondii revealed a seroprevalence of 51/1,132 (4.5%). We believe that the present results are of epidemiological implications and public health importance due to the acknowledged susceptibility of humans to E. cuniculi and T. gondii infections. Therefore, routine screening tests of domestic rabbits are proposed considering the zoonotic potential of these parasites.

This study combines the feedback linearization method and our recent researches of Chen electrical unifying approach to propose a global tracking controller design for nonlinear uncertain diff erential-diff erence systems with time delay. One example, which cannot be solved by the fi rst paper on the almost disturbance decoupling problem, is presented in this study to show the essential points that the tracking performance is easily solved by the proposed approach. The study applies Chen electrical unifying approach to take the place of using the disorganized virtual ground technique and the Kirchhoff ’s law for those traditional approaches. In order to show its signifi cant practicability, the study has fi rstly designed an easy-to-implement adder and a time-delay circuit, and an unstable time-delay operational amplifi er control system based on the Chen electrical unifying approach, and then has proposed a stable controller.

To explore the ability of MaxPol-T/S, a novelbiodegradable poly-lactic-co-glycolic acid(PLGA) scaffold, in providing a cell-matrix interaction interface for cell growth, we generated GFP positive adipose derived stem cell(ADSCGFP+) and fibroblasts(FCGFP+) for investigating the cellular growth on this biomaterial. The MaxPol-T/S was produced through salt-leaching/particulate-leaching technology for tissue engineering, which provides a modified surface for the best cellular attachment sites and nutrient supply and waste exchange conditions. The morphological features of MaxPol-T/S were studied with a scanning electron microscope and the ADSCGFP+ and FCGFP+ were confirmed by auto-fluorescence microscopy and flow cytometry assay. Being seeded onto the MaxPol-T/S in vitro, the growth and morphology of ADSCGFP+ and FCGFP+ were further verified by auto-fluorescence microscopy and MMT test. The ADSCs and fibroblasts, expressed strong GFP signals in the cytoplasm and nucleus, adhered and proliferated on the surface of scaffold MaxPol-T/S. Both cell lines survived on the scaffold more than 21 days in vitro and formed three-dimensional colonies on the surface of the MaxPol-T. In conclusion, MaxPol-T/S is a novel PLGA scaffold for ADSCGFP+ and FCGFP+ and implies a promising technique for tissue engineering.
