        백서의 실험적 치아이동시 connexin 43의 발현에 관한 연구

        치아이동시 골세포간 활성 조화에서 세포간 교통의 중요성이 차츰 인식되고 있으며 세포간 교통을 위한 교통반의 존재가 확인되고 있다. 동물실험 모델에서 실험적 치아이동을 통해 조성된 압박 및 견인측 치주인대에서 교통반 단백의 일종인 connexin 43의 발현을 관찰함으로써 인접 연조직 및 골 조직 내에서 세포 신호 전달 양상의 한 부분을 파악하고자 하였다. Sprague-Dawley계 백서 27마리를 대조군(3마리)과 실험군(24마리)으로 나누었으며, 실험군은 견인력(75g)을 가한 후 12시간, 1일, 4일, 7일, 14일, 28일이 경과한 후 각각 4마리씩 희생시켜, connexin 43의 발현을 면역조직화학적으로 관찰한 바 다음과 같은 결과를 얻었다. 1.대조군에서의 connexin 43 발현은 치은, 상아질, 치주인대, 치수, 골세포에서 경미하였다. 2.교정적 치아이동에 반응하는 치수, 치주인대, 골모세포, 파골세포, 골세포부위에서 connexin 43의 발현이 많았으나 치은, 상아질, 상아모세포에서의 발현은 견인력 적용기간에 관계없이 대조군과 큰 차이가 없었다. 3.치수에서 connexin 43의 발현은 교정력을 가한 4일에서부터 증가하여 7일째까지 크게 증가한 후 14일째부터 감소되었으며 28일째에는 대조군과 차이가 없었다. 4.치주인대에서 connexin 43의 발현은 주로 혈관을 중심으로 치조골 면을 따라 압박측에서 주로 나타났는데 교정력을 가한 12시간에서부터 증가하여 4일에서 가장 많은 발현을 보인 후 7일부터 감소하기 시작하였다. 5.골모세포 및 골세포에서 발현은 실험 1일째부터 출현해 4일째에 가장 많은 발현을 보였고 7일 이후에는 크게 감소하였으며 파골세포에서의 발현은 골모세포와 골세포보다 실험 4일째에 더 많이 관찰할 수 있었다. Bone remodeling in response to force requires coordinated actions of osteoblasts, osteoclasts, osteocytes, and periodontal ligament cells. Coordination among these cells may be mediated, in part, by cell-to-cell communication via gap junctions. This study was designed to evaluate the expression of gap junction, connection 43 in periodontal tissue during the experimental movement of rat's incisors, by LSAB(labelled streptavidine biotin) immunohistochemical staining fort connexin 43. Twenty seven Sprague-Dawley rats were divided into a control group(3 rats), and 6 experimental groups(24 rats) where 75g of force was applied from helical springs across the maxillary incisors. Rats of experimental groups were sacrificed at 12 hours, 1, 4, 7, 14 and 28 days after force application, respectively. And the tissues of a control group and experimental groups were studied immunohistochemically. The results were as follows : 1.In control group, the expression of connexin 43 was rare in gingiva, dentin, cementum, periodontal ligament and bone cells. 2.In experimental group, the expression of connexin 43 was increased in pulp, periodontal ligament, osteoblasts, and osteoclasts, comparing to that in control. And it was rare in gingiva, dentin, and odontoblasts regardless of the duration of force application, which was not different from that of control group. 3.The expression of connexin 43 in pulp of experimental group began to increase in 4-day after force application and got to the highest degree at 7-day. And it decreased after 14-day to be similar to that of control group at 28-day. 4.The expression of connexin 43 in periodontal ligament was noted in small capillaries adjacent to alveolar bone, showing higher intensity of immunolabelling after 4-day. And it was stronger in the pressure side than in tension side of periodontal ligament. After 7-day, decrease in connexin 43 expression was observed. 5.The expression of connexin 43 in alveolar bone began to increase 1-day, reached to the highest degree at 4-day, and decreased at 7-day. And the expression in osteoclasts was more than that in osteoblasts or osteocyte at 7-day.

      • 결핵군 55 kDa 항원의 면역학적 분석

        Identification and characterization of mycobacterial antigens are critical for evaluation of their role in diagnosis, vaccination, and pathogenesis of mycobacterial diseases. An attempt has been made to immunologic characterize the 55 kDa protein antigen of Mycobacterium tuberculosis H37Rv which attracted our interest because it is present in high concentration in 50-80% ammonium sulfate fraction of the culture filtrate. Two monoclonal antibodies (MAbs) directed to 55 kDa antigen were produced. MAbs MT55-1 and MT55-2 reacted with a single 55 kDa protein band. On examination of degree for cross-reactivity with other mycobacterial species by enzymelinked immunosorbent assay (ELISA) and immunoblotting, these antibodies reacted strongly with M. tuberculosis and M. bovis BCG, and reacted weakly with M. marinum and M. smegmatis. To investigated the subcellular distribution of MAbs defined epitopes in the 55 kDa antigen within the mycobacterium, we isolated three major subcellular factions of M. tuberculosis, namely, cell wall, cytoplasmic membrane, and cytosol, by a simple fractionation procedure. MAb MT55-1 reactive cpitope was found in the cytosol when tested by immunoblotting. A sandwich ELISA was initially developed for detecting 55 kDa antigen using MT55-1 MAb in mycobacterial culture filtrate before detecting it in clincal specimens. The minimal detectable concentration was 1.0 ㎍/m1 for M. tuberculosis culture filtrate and 100 ㎍/ml for sonic extracts of M. bovis BCG and M. marinum, respectively. But the 55 kDa antigen was not detected in sonic extracts of other mycobacterial species examined. Although further evaluations are required, this study suggests that the 55 kDa antigen may be of interest as potential diagnostic reagent.

        골절단을 이용한 급속 치아이동 후 치수 및 치주조직 변화

        본 연구는 골절단을 이용한 치아-치조골 분절의 급속 치아이동 후 치수, 치주인대 및 치조골의 변화를 유성견에서 평가하고자 하였다. 하악 제4소구치의 근심, 원심, 치근단 부위에서 피질골을 절단하여 치아-골 분절을 형성하고 하악 제3소구치를 발거하면서 협측, 설측 부위의 피질골을 삭제하였다. 1주 휴지기 부여에 따라 휴지기군과 비휴지기군으로 나누어 치아의존형 견인장치로 6일 동안 견인하고 강화기를 거친 후 0주, 1주, 2주, 4주, 6주, 8주에 희생시켜 치수, 치주인대 및 치조골의 조직 변화를 임상적, 방사선적, 조직학적 및 면역화학적으로 평가하여 다음과 같은 결과를 얻었다. 1. 치아이동 양과 강화기 동안의 조직 치유 양상에서 휴지기 유무에 따른 차이는 없었다. 2. 견인측에서 골형성은 강화기 8주까지 계속되었는데 강화기 1-2주에 가장 활발하였으며 6-8주간의 변화는 적었다. 3. 비휴지기군의 강화기 1주에서 치조골 흡수 및 파골세포 출현, 염증세포 침윤이 가장 많았으며, 특징적으로 파상아세포가 압박측의 치주인대와 치수 내에 나타났다. 4. TGF-β는 치조골의 골기질 및 골모세포, 파골세포, 치수 내 파상아세포에서 강양성 발현을 보인 반면 치수, 백악모세포, 무세포성 백악질에서 경미한 양성 발현을 나타내는 부위 특이성이 있었다. 5. TGF-β는 견인측 치주인대의 치조골에 인접한 혈관 및 치주인대세포, 골모세포에서 강화기 초기 1-2주에 주로 발현되었으며 6주 이후에는 발현이 크게 감소하였다. The movement of tooth-bone segments by osteotomy can simultaneously shift tooth and surrounding alveolar bone in a relatively short period. The purpose of this study was to evaluate the tissue changes in pulp, periodontal ligament, and alveolar bone in rapid tooth bone movement with osteotomy. The mandibular 3rd premolar of a dog was extracted and cortical bones of the buccal and lingual area were eliminated, and then cortical bones around the mesial and distal area of root, and below the root apex of the mandibular 4th premolar were osteotomized. After a one week latency period, a tooth-borne distraction device was activated for 6 days. And pulp, periodontal ligament and alveolar bone were evaluated clinically, radiologically, histologically and immunohistochemically at 0, 1, 2, 4, 6, 8 weeks of the consolidation period and conclusions were reached as follows. 1. Latency period didn't affect total amount of tooth movement and healing process of tissue during consolidation period. 2. Bone formation continued through 8 weeks of consolidation in distracted side, with a high peak at 1-2 weeks, and the lowest at 6-8 weeks of consolidation. 3. At 1 week of consolidation, alveolar bone resorption, osteoclast appearance and inflammatory cell infiltration were the most active, and dentinoclasts characteristically appeared on the pulp and pressure side of the periodontal ligament. 4. The expression of TGF-β was area-specific, as it was strong-positive at bone matrix, osteoblast, osteoclast of alveolar bone, and dentinoclast inside pulp, but weak in pulp, cementoblast and acellular cementum. 5. The expression of TGF-β was generally observed at the initial 1-2 weeks of consolidation at vessels, periodontal ligament cells, and osteoblast near alveolar bone on the distraction side of the periodontal ligament, and was significantly decreased after 6 weeks of consolidation.

        Acinar cell carcinoma is a rare tumor that represents 1~2% of al1 pancreatic cancers. Clinical and radiologic findings are inconclusive in this disease Acinar cell carcinoma is characterized by rapid progression and early metastasis, which lead to its poor prognosis. A 41-year-o1d man was admitted to our hospital for abdominal pain. Abdominal computed tomography (CT) and positron emission tomography-computed tomography (PET-CT) showed a splenic mass, which was being invaded by a pancreatic tail mass and which had increased ^(18)F-fluorodeoxyglucose (FDG) uptake Primary radical distal pancreatectomy and splenectomy were performed. Pathologic findings revealed an acinar cell carcinoma of the Pancreas The patient underwent a total gastrectomy three months later because of gastric recurrence Four months later, multiple hepatic metastases were discovered, and the patient underwent a left hepatectomy During treatment with capecitabine, there was no evidence of tumor progression for 14 months. We report a case of metastatic pancreatic acinar cell carcinoma, which did not progress for an extended period while the patient was being treated with capecitabme.

      • KS H ISO 1242 표준의 평가 : 라반딘 정유의 산값 측정

        The acid value (AV) as well as the ester value of essential oil is an important index to evaluate quality of essential oil. The official standard analytical method of KS H ISO 1242 dealing with “Essential oils - Determination of acid value” was assessed experimentally using two lavandin essential oils obtained in France as an example. One French lavandin oil sample showed the results of AV= 0.9 by both indicator method and potentiometry. Another sample showed AV = 1.9 by indicator method and AV = 1.6 by potentiometry. Latter lavandin oil sample exhibited significantly higher acid value than maximum value of 1 described in the standard. This result suggests that latter lavandin oil was contaminated, because this oil was contained in an orange juice bottle. Results of this study proved that KS H ISO 1242 could be used as a suitable scientific standard for the determination of acid value of essential oils.

        The purpose of this investigation was to develop a new carrier using chitin and glucan as the model for Aspergillus oryzae cell wall. In case of the ratio of chitin and glucan(3/1), the activity of the immobilized protease had higher than that of model(chitin/glucan=1/3) and carriers were formed gel. In case of soluble chitin and glucan treated with calcium carbonate, there were a 1.8-fold increase in activity of immobilized protease compared with untreated chitin. The scanning electron microscope image showed that a pore size of this carrier had a homogenized pore size and a smooth surface.

      • 흰쥐 선조체에서 6-OHDA-유도 도파민 고갈 및 SH-SY5Y 세포주에서 6-OHDA-유도 산화적 스트레스에 대한 I-Deprenyl의 신경 보호효과

        A neurotoxin, 6-hydroxydopamine (6-OHDA) has long been used to form a Parkinson's disease (PD) model by inducing the lesion in catecholaminergic pathways, particularly the nigrostriatal dopamine (DA) pathway. Whereas I-deprenyl, a selective inhibitor of monoamine exidase (MAO) type B, is now widely used in the treatment of PD, the precise action mechanism of the drug remains elusive. In this studym, we investigated whether I-deprenyl shows protective effect against the DA depletion induced by 6-OHDA in rat brain, and against 6-OHDA-induced neurotoxicity and oxidative stress in catecholaminergic neuroblastoma SH-SY5Y cells that are known to lack MAO-B activity. Pretreatment fo I-deprenyl significantly enhanced the striatal DA, 3,4-dihydroxyphenylacetic acid, homovanilic acid, and 3-methoxytyramine levels compared to the untreated 6-OHDA-lesioned rat, indicating that I-deprenyl pretreatment prevents 6-OHDA-induced depletion of not only striatal dopamine but also its metabolites. Treatment of 6-OHDA for 24 hrs decreased the cell viability and increase the generation of ROS in dose-dependent manners.We further investigated whether caspase activity is involved in the action of I-deprenyl. Treatment of I-deprenyl (0.1~100μM) did not produce any changes in 6-OHDA-induced cleavage of poly (ACP-ridose) polymerase in SH-SY5Y cells. Our result suggest that the neuroprotective effect of I-deprenyl against 6-OHDA is due to its incresed scavenger activity, but independent of inhibition of MAO-B or caspase-3activation.

        NaCl이 보리(Hordeum vulgare L.) 잎의 엽록소 형광에 미치는 영향

        This study was conducted to investigate the changes of chlorophyll contents and chlorophyll fluorescence in barley(Hordeum vulgare L.) 7 day old seedling treated with 0.2M, 0.4M, 0.6M, 0.8M, and 1.0M NaCl concentration containing Hepes buffer(pH 7.5). Barley was affected by NaCl treatment. The chlorophyll a, b and carotenoid of barley decreased with an increase in NaCl concentration. However, chlorophyll a, b and carotenoid of barley were not greatly influenced by o.8M and 1.0M NaCl. Fv, Fv/Fm and qP were gradually decreased by higher concentration of NaCl. qP, qNP, qR and qE were gradually decreased by 6hr. During barley chloroplast was development NaCl affected chlorophyll synthesis than photosynthetic activity. Whereas barley seedling leaves were more influenced photosynthetic activity than chlorophyll contents by NaCl.

      • 백금(Ⅳ)착체의 약리학적 연구

        The platinum complex, PtCl₄(2-CEen), was previously synthesized and shown to have anti-tumor activity. In the present study, toxicologic profile of PtCl₄(2-CEen) and its acute anti-inflammatory activity on carrageenin edema was observed with oral administration. The ef-4feet of this agent on acetic acid-induced writhing in mice were also examined. In male and female mice given a single intragastrical dose of PtCl₄(2-CEen), we determined that LD_(50) values were over lOOOmg/kg B.W. In the method of carrageenin edema and writhing test, the PtCl₄(2-CEen), orally administered in a single dose, tended to inhibit the development of hind paw edema in rats, and the number of writhing induced by acetic acid in mice. These facts suggest that PtCl₄(2-CEen) may have weak anti-inflammatory effect.
