        정신분열병 환자에서 Haloperidol이 99m-Tc-ECD SPECT로 측정한 국소뇌혈류량에 미치는 영향

        연구목적 : 정신분열병 환자에서의 뇌혈류량은 약리학적 상태를 포함한 다양한 요소들에 의하여 영향을 받는다. 항정신병약물이 국소뇌혈류량에 미치는 영향에 관한 많은 연구가 행해져 왔으나 항정신병약물의 종류와 노출기간, 약물용량의 차이로 인해 다양하고도 상반된 결과를 보여왔다. 따라서 본 연구에서는 정신분열병 환자에서 항정신병약물의 종류와 노출기간을 통제한 상태에서 약물 투여시 국소뇌혈류량의 변화를 관찰하고자 하였다. 방 법 : 항정신병약물 비노출(antipsychotic drug-naive) 정신분열병 환자 18명과 haloperidol을 투여받은 환자 19명을 대상으로 안정 상태(resting state)에서 SPECT를 촬영하여 국소뇌혈류량을 측정한 후 두군 사이의 상대적 뇌혈류량을 비교하였다. Haloperidol은 D2 수용체에 비교적 선택적으로 작용하고 활성대사산물이 적기 때문에 실험약물로 선택되었고 약물노출기간은 1∼3주동안으로 제한하였다. 결 과 : 우측 저측두엽, 좌측 저전두엽, 좌·우측 기저신경절, 좌측 시상에서의 상대적 평균 혈류지표는 halo-peridol 투여군에서 비노출군에 비해 유의하게 증가되어 있었다. 또한 좌·우측 두정-후두엽과 우측 두정엽에서의 상대적 평균 혈류지표도 haloperidol 투여군에서 비노출군에 비해 유의하게 증가되어 있었다. 반면 좌측 저측두엽의 상대적 평균 혈류지표는 haloperidol 투여군에서 항정신병약물 비노출군에 비해 유의하게 감소되어 있었다. 그러나 우측 저전두엽, 우측 시상, 좌·우측 고측두엽, 좌·우측 고전두엽, 좌측 두정엽에서 두 군 사이의 상대적 평균 혈류지표는 유의한 차이를 보이지 않았다. 결 론 : 정신분열병 환자에서 항정신병약물이 국소뇌혈류량에 영향을 미치며, 항정신병약물 투여상태가 뇌혈류량 측정시 고려되어야할 중요한 변수임을 시사한다. Objectives : Regional cerebral blood flow(rCBF) in schizophrenics is confounded by various factors including medication status. Previously, there have been numerous studies regarding the effects of antipsychotics on rCBF. However, these works have shown contradictory and inconsistent findings due to the different of type, dose and exposed duration of antipsychotics. The aim of this study was to observe the effect of antipsychotic medication on rCBF and exposed duration of antipsychotics under control. Methods : Eighteen drug-naive schizophrenics and 19 schizophrenics medicated with halo-peridol were included in the study. Regional cerebral blood flow was assessed with single-photon emission computed tomography(SPECT) under a resting state. Relative rCBF was compared between two groups. Haloperidol was selected as the antipsychotic drug as it has relatively selective action at the D2 receptor and less active metabolites. Exposed duration was limited from one to three weeks. Results : Haloperidol-medicated schizophrenic patients had a significantly greater increase of relative cerebral perfusion in the right inferior temporal lobe, left inferior frontal lobe, both basal ganglia, left thalamus, both parieto-occipital lobes, and right parietal lobe than drug-naive schizophrenic patients. Haloperidol-medicated schizophrenic patients had a significant decrease of relative cerebral perfusion in left inferior temporal lobe. However, no significant differences in relative rCBF were found between drug-naive and haloperidol-medicated schizophrenic patients in right inferior frontal lobe, right thalamus, both superior temporal lobes, both superior frontal lobes, and left parietal lobe. Conclusion : These findings suggest that antipsychotics affect regional cerebral blood flow, and antipsychotic medication status must be considered in the relative rCBF studies of schizophrenic patients.

      • 감중에 존재하는 Tyrosinase의 정제 및 특성에 관한 연구 : Diospyros Kaki Thunb

        감으로부터 tyrosinase를 추출하여 ammonium sulfate에 의한 침전, Sephadex G-25와 DEAE-Sephacel column chromatography하여 isozyme P₁과 P₂그리고 P₃를 정제하였다. Isozyme P₁과 P₂그리고 P₃는 monohydroxypenol류의 기질에 대하여 거의 활성이 없었으나, o-dihydroxyphenol류와 trihydroxyphenol류 중 phrogallol에서 높은 활성을 나타내었다. m-dihydroxyphenol류는 catechin에서 높은 활성을 보였지만 그 외에서는 거의 활성을 보이지 않았고, p-dihydroxyphenol류에 대해서도 활성이 없었다. 또 L-DOPA에 대한 Km 값은 P₁이 9.1로 P₂는 11.2 그리고 P₃는 7.4mM이었고, Vmax 값은 P₁이 13.7로 P₂는 13.9 그리고 P₃는 10.0 nmoles/ml.min이었다. Tyrosinase isozyme 모두는 sodium diethyldithiocarbamate, sodium cyanide와 L-ascorbic acid가 10 mM에 대하여 100% 저해되었다. Sodium diethyldithiocarbamate는 비경쟁적 저해를 보였으며, P₁과 P₂그리고 P₃에 대한 Km값은 각각 25.0과 52.6 그리고 13.5mM이었다. P₁과 P₂그리고 P₃ isozyme의 반응 최적 pH와 반응 최적온도는 각각 pH7과 50℃였다. 금속이온중 Ca²+와 Hg²+와 Cu²+ 그리고 Mn²+는 모든 isozyme P₁과 P₂그리고 P₃의 활성을 증가 시키는 반면 Fe²+와 Zn²+와 Mg²+그리고Ba²+는 활성을 저해시켰다. Tyrosinase was purified from persimmon(Diospyros kaki Thunb) by ammonium sulfate precipitation, Sephadex G-25 and DEAE-Sephacel column chromatography. The enzyme has high substrate specificity on o-dihydroxyphenols and trihydroxyphenols. The Km values for L-DOPA were P₁;13.7, P₂;13.9 and P₃;10.0 nmoles/ml.min. Sodium diethyldithiocarbamate, sodium cyanide and L-ascorbic acid inhibited the activity of enzyme. Sodium diethyldithiocarbamate inhibited tyrosinase activity noncompetitively. The Km values for sodium diethyldithiocarbamate were P₁;25.0, P₂;52.6 and P₃;13.5mM. The pH of maximum enzyme activity was increased by addition of Ca2+, Hg2+, Cu2+ and Mn2+ ions, but it was decreased in the presence of Fe2+, Zn2+, Mg2+ and Ba2+ ions.

        Effects of Konjac, Isolated Soy Protein, and Egg Albumin on Quality Properties of Semi-dried Chicken Jerky

        The objective of this study was to examine the effect of adding various humectants (konjac, egg albumin, and isolated soy protein) on the properties of semi-dried chicken jerky. Jerky samples were prepared as follows: control with no humectants and treatments with 0.05, 0.1, and 0.2% of added humectants. Adding the humectants influenced the increase in pH, processing yields, moisture contents, water activity, mechanical tenderness, and sensorial properties (tenderness, juiciness, and overall acceptability) of chicken jerky. Additionally, the konjac treatment most improved the yields, tenderness, and sensorial traits, among the humectant treatments tested. Furthermore, adding 0.1% konjac during jerky manufacture resulted in similar quality properties as adding 0.2% konjac.

        Design and Fabrication of Si Subwavelength Structures for Broadband Antireflection in Mid-Infrared Ranges

        <P>Broadband antireflection in infrared ranges is essential for various applications such as photovoltaics, light-emitting diodes, and optical lenses for thermal imaging. We performed numerical simulations to find the optimal design of Si subwavelength structures for broadband antireflection in the mid-infrared wavelength ranges from 3 mu m to 20 mu m. By using the simulation results as a design guide, we fabricated Si subwavelength structures in the form of ellipsoids and paraboloids with self-assembled silica bead monolayers as dry etch masks. The silica bead monolayers on the Si wafers in large area were prepared by colloidal lithography based on spin coating of silica beads dispersed in organic binary solvents. A two-step dry etch process with combination of isotropic and anisotropic etching enables fabrication of the Si subwavelength structures of a high aspect ratio, and we demonstrated broadband antireflection in the mid-infrared wavelength ranges.</P>

      • Enhanced blue responses in nanostructured Si solar cells by shallow doping

        <P>Optimally designed Si nanostructures are very effective for light trapping in crystalline silicon (c-Si) solar cells. However, when the lateral feature size of Si nanostructures is comparable to the junction depth of the emitter, dopant diffusion in the lateral direction leads to excessive doping in the nanostructured emitter whereby poor blue responses arise in the external quantum efficiency (EQE). The primary goal of this study is to find the correlation of emitter junction depth and carrier collection efficiency in nanostructured c-Si solar cells in order to enhance the blue responses. We prepared Si nanostructures of nanocone shape by colloidal lithography, with silica beads of 520 nm in diameter, followed by a reactive ion etching process. c-Si solar cells with a standard cell architecture of an Al back surface field were fabricated varying the emitter junction depth. We varied the emitter junction depth by adjusting the doping level from heavy doping to moderate doping to light doping and achieved greatly enhanced blue responses in EQE from 47%–92% at a wavelength of 400 nm. The junction depth analysis by secondary ion mass-spectroscopy profiling and the scanning electron microscopy measurements provided us with the design guide of the doping level depending on the nanostructure feature size for high efficiency nanostructured c-Si solar cells. Optical simulations showed us that Si nanostructures can serve as an optical resonator to amplify the incident light field, which needs to be considered in the design of nanostructured c-Si solar cells.</P>

        High-precision quality assurance of robotic couches with six degrees of freedom

        <P>The mean error and standard deviation of linear and rotational motions, as well as couch sagging were analyzed from continuously acquired images in the moving couch. The accuracy of VTS was 0.024 mm deviation for the sinusoidal motion, and the accuracy of the transformation matrix was 0.02 mm. In a cross-comparison, the difference between Laser Tracker (FARO) measurements was 0.14 +/- 0.12 mm for translation and 0.032 +/- 0.026 degrees on average for yaw rotation. The new system provides QA of yaw, pitch and roll motion as well as sagging of the couch and sub-millimeter/degree accuracy together with precision.</P>

        Effects of Rice Bran Fiber on Quality of Low-fat Tteokgalbi

        This study evaluated the effects of dietary fiber extracted from rice bran (rice bran fiber) on the quality of low-fat tteokgalbi. The controls were formulated with 20% added fat. Test samples of tteokgalbi were produced with 5 different formulations containing 1, 2, 3, 4, and 5% rice bran fiber as a fat replacer, in addition to 10% fat. The control had the highest fat content, energy value, cooking loss, reduction in diameter, reduction in thickness, CIE L<sup>*</sup>-value (lighness), and color of sensory properties. The tteokgalbi containing rice bran fiber had higher moisture, protein, ash, and carbohydrate contents than the controls. Tteokgalbi with 3% rice bran fiber had the lowest cooking loss, reduction in diameter, and reduction in thickness. Meat products containing 2 and 3% rice bran fiber had a higher overall acceptability similar to the high fat control.
