      • Association of Increased Pulmonary Interleukin-6 with the Priming Effect of Intra-Amniotic Lipopolysaccharide on Hyperoxic Lung Injury in a Rat Model of Bronchopulmonary Dysplasia

        <P><I>Background:</I> The authors previously demonstrated the priming effect of intra-amniotic lipopolysaccharide (LPS) on hyperoxic lung injury in a rat model of bronchopulmonary dysplasia (BPD). <I>Objectives:</I> To investigate the mechanism underlying this priming effect by determining biochemical profiles in a rat model of BPD. <I>Methods:</I> The rat model involved intra-amniotic LPS administration and postnatal hyperoxia (85%). The mRNA expressions of <I>interleukin-6 (IL-6), vascular endothelial growth factor (VEGF), VEGF receptor-2 (VEGFR-2), basic fibroblast growth factor (bFGF),</I> and <I>transforming growth factor </I>β<SUB><I>1</I></SUB><I> (TGF-</I>β<SUB><I>1</I></SUB><I>),</I> as well as the protein levels of IL-6, VEGF, and protein carbonyl in lung tissue were compared between the LPS plus hyperoxia, the LPS only, the hyperoxia only, and the control groups. <I>Results:</I> Morphometric analysis of lung tissues demonstrated that alveolarization was significantly inhibited only in the LPS plus hyperoxia group. IL-6 protein levels and its mRNA expression in the lungs were significantly increased only in the LPS plus hyperoxia group. Neither LPS nor hyperoxia increased IL-6 in the lungs independently. <I>bFGF</I> mRNA expression was significantly decreased in the LPS-treated groups. VEGF protein levels were significantly reduced by hyperoxia, whereas protein carbonyl levels were increased by intra-amniotic LPS or hyperoxia. No additional significant change to VEGF or protein carbonyl levels was produced by intra-amniotic LPS or hyperoxia. There were no significant differences in the mRNA expressions of <I>VEGF, VEGFR-2,</I> and <I>TGF-</I>β<SUB><I>1</I></SUB>. <I>Conclusions:</I> The priming effect of intra-amniotic LPS on hyperoxic lung injury may be associated with IL-6 elevation in the lungs.</P><P>Copyright © 2009 S. The spirituality of Dail interests more in "Who am I" not in "What should I do in front of God" because the Dail focuses more on the being than doing. This is not appearance, but inner side and even reality, but a religious mystery. That means a life style which is faithful more to essence and value than to appearance and illusion. The spirituality of Dail is focused on Jesus who came to world as life. It deals with everywhere life for only life can save life. I love my life which is given to me, I love all the creature that has life. Thus everyday life becomes spiritual life.

        Immune-Enhancing Effect of Nanometric Lactobacillus plantarum nF1 (nLp-nF1) in a Mouse Model of Cyclophosphamide-Induced Immunosuppression

        Nanometric Lactobacillus plantarum nF1 (nLp-nF1) is a biogenics consisting of dead L. plantarum cells pretreated with heat and a nanodispersion process. In this study, we investigated the immune-enhancing effects of nLp-nF1 in vivo and in vitro. To evaluate the immunostimulatory effects of nLp-nF1, mice immunosuppressed by cyclophosphamide (CPP) treatment were administered with nLp-nF1. As expected, CPP restricted the immune response of mice, whereas oral administration of nLp-nF1 significantly increased the total IgG in the serum, and cytokine production (interleukin-12 (IL-12) and tumor necrosis factor alpha (TNF-α)) in bone marrow cells. Furthermore, nLp-nF1 enhanced the production of splenic cytokines such as IL-12, TNF-α, and interferon gamma (IFN-γ). In vitro, nLp-nF1 stimulated the immune response by enhancing the production of cytokines such as IL-12, TNF-α, and IFN-γ. Moreover, nLp-nF1 given a food additive enhanced the immune responses when combined with various food materials in vitro. These results suggest that nLp-nF1 could be used to strengthen the immune system and recover normal immunity in people with a weak immune system, such as children, the elderly, and patients.

        Background & Objective: Articular cartilage is a potential target for drugs designed to inhibit the activity of matrix metalloproteinases (MMPs) to stop or slow the destruction of the proteoglycan and collagen in the cartilage extracellular matrix. The purpose of this study was to investigate the effects of Aralia cordata Thunb. in inhibiting the release of glycosaminoglycan (GAG), the degradation of collagen, and MMP activity in rabbit articular cartilage explants. Methods : The cartilage-protective effects of Aralia cordata Thunb. were evaluated by using glycosaminoglycan degradation assay, collagen degradation assay, colorimetric analysis of MMP activity, measurement of lactate dehydrogenase activity and histological analysis in rabbit cartilage explants culture. Results : Interleukin-la (IL-1a) rapidly induced GAG, but collagen was much less readily released from cartilage explants. Aralia cordata Thunb. significantly inhibited GAG and collagen release in a concentration-dependent manner. Aralia cordata Thunb. dose-dependently inhibited MMP-3 and MMP-13 expression and activities from IL-1a-treated cartilage explants cultures when tested at concentrations ranging from 0.02 to 0.2 mg/ml. Aralia cordata Thunb. had no harmful effect on chondrocytes viability or cartilage morphology in cartilage explants. Histological analysis indicated that Aralia cordata Thunb. reduced the degradation of the cartilage matrix compared with that of IL -1a-treated cartilage explants.

        <P>NK cells are a key component of innate immune systems, and their activity is regulated by cytokines and hormones. Adiponectin, which is secreted from white adipose tissues, plays important roles in various diseases, including hypertension, cardiovascular diseases, inflammatory disorders, and cancer. In this study the effect of adiponectin on NK cell activity was investigated. Adiponectin was found to suppress the IL-2-enhanced cytotoxic activity of NK cells without affecting basal NK cell cytotoxicity and to inhibit IL-2-induced NF-kappaB activation via activation of the AMP-activated protein kinase, indicating that it suppresses IL-2-enhanced NK cell cytotoxicity through the AMP-activated protein kinase-mediated inhibition of NF-kappaB activation. IFN-gamma enhances NK cell cytotoxicity by causing an increase in the levels of expression of TRAIL and Fas ligand. The production of IFN-gamma, one of the NF-kappaB target genes in NK cells, was also found to be suppressed by adiponectin, accompanied by the subsequent down-regulation of IFN-gamma-inducible TRAIL and Fas ligand expression. These results clearly demonstrate that adiponectin is a potent negative regulator of IL-2-induced NK cell activation and thus may act as an in vivo regulator of anti-inflammatory functions.</P>

      • 비정렬격자 격자점 중심 압력기반해법

        본 논문에서는 연속류 영역과 미끄럼류 영역의 유동을 수치계산하기 위한 혼합 비정렬격자 압력기반해법을 제안한다. 압축성 효과를 반영하는 수정된 SIMPLE법을 비정렬격자 격자점 중심법에 적용하였다. 대류항은 2차의 중앙차분법을 지연수정법으로 이산화 하였으며, 확산항은 2차 정화도의 유사 정렬격자 기법을 사용하였다. 또한, 혼합 비정렬격자에서 기억장소의 절약과 계산 효율을 저하여 모서리 자료구조와 격자점 중심법을 사용하였다. 연속류 영역 뿐 만 아니라, 미끄럼 유동의 벽면에서의 미끄럼 현상을 해석할 수 있도록 미끄럼 경계조건을 부과하였으며, 여기에는 널리 쓰이는 Maxwell 경계조건과 명에 의해 새로이 제안된 Langmuir 경계조건을 사용하였다. 제안된 수치기법을 이용하여 연속류 영역의 유동과 미끄럼류 영역의 유동에 대해 수치 계산을 수행하였으며, 본 기법의 정확도와 효율성을 입증하였다. This paper proposes a pressure based method to compute both continuum flow and slip flow on hybrid unstructured grids. The compressible version of SIMPLE algorithm is extended to unstructured grids. Convection terms are discretized using second order central differencing with deferred correction approach and diffusion terms by second-order structured grids analogy. This method also uses cell vertex scheme with edge based data structure in order for memory savings and computing efficiency for arbitrary grid topologies. As well as continuum flow, slip flows are solved by adopting slip boundary conditions, which are well known Maxwell condition and Langmuir condition recently proposed by Myong.(30) The flows in both continuum regime and slip flow regime are calculated using the above methodology, which shows that the proposed method is well-devised with respect to accuracy and efficiency.

      • 도로교통소음의 수평·수직적 전달 및 감쇠 특성

        This study has been conducted to achieve the following objectives: First, in order to understand the horizontal propagation and attenuation characteristics for the road traffic noise, we selected actively developed areas within 100 meters away from the road to measure the level of traffic noise near a road. To measure the noise near a railroad, we selected areas within 100 meters away from the road. In this way, we analyzed the horizontal propagation and attenuation characteristics for the traffic noise occurring in diversified areas. Second, in order to understand the vertical propagation and attenuation characteristics for the road traffic noise, we measured and analyzed the distributional characteristics of vertical sound pressure levels on each floor of multi-storied apartment buildings according to changes of traffic load and types, and the existence or nonexistence of soundproof walls. For the case of the road traffic noise, we also selected we measured and analyzed the different noise levels on each floor of multi-storied apartment buildings from the soundproof wall. The results of Horizontal propagation and attenuation characteristics for the road traffic noise are as follows: In cases of the soundproof wall + raising ground, raising ground, cutting land, and flat land as distance increases, the sound pressure level steadily decreases. The soundproof wall + raising ground has an especially high distance decrease compared to the other areas, and the sound pressure level for the raising ground is higher than that of the cutting land with a measurement of 10.7~12.1dB(A). Vertical propagation and attenuation characteristics for the road traffic noise are as follows: The amount of sound pressure level decrease is 12.6- 14.5dB(A) for first floor when measuring the vertical sound pressure levels at soundproof wall, which indicates a fairly large decrease. At fifth and sixth floor, the amount of decrease is 2.3dB(A), which indicates a small decrease.
