        Use of NaCl prevents aggregation of recombinant COMP–Angiopoietin-1 in Chinese hamster ovary cells

        <P><B>Abstract</B></P><P>To investigate the effect of hyperosmotic medium on production and aggregation of the variant of Angiopoietin-1 (Ang1), cartilage oligomeric matrix protein (COMP)–Ang1, in recombinant Chinese hamster ovary (CHO) cells, CHO cells were cultivated in shaking flasks. NaCl and/or sorbitol were used to raise medium osmolality in the range of 300–450mOsm/kg. The specific productivity of COMP–Ang1, <I>q</I><SUB>COMP–Ang1</SUB>, increased as medium osmolality increased. At NaCl-450mOsm/kg, the <I>q</I><SUB>COMP–Ang1</SUB> was 7.7-fold higher than that at NaCl-300mOsm/kg, while, at sorbitol-450mOsm/kg, it was 2.9-fold higher than that at sorbitol-300mOsm/kg. This can be attributed to the increased relative mRNA level of COMP–Ang1 at NaCl-450mOsm/kg which was approximately 2.4-fold higher than that at sorbitol-450mOsm/kg. Western blot analysis showed that COMP–Ang1 aggregates started to occur in the late-exponential phase of cell growth. When sorbitol was used to raise the medium osmolality, a severe aggregation of COMP–Ang1 was observed. On the other hand, when NaCl was used, the aggregation of COMP–Ang1 was drastically reduced at NaCl-400mOsm/kg. At NaCl-450mOsm/kg, the aggregation of COMP–Ang1 was hardly observed. This suggests that environmental conditions are critical for the aggregation of COMP–Ang1. Taken together, the use of NaCl-induced hyperosmotic medium to cell culture process turns out to be an efficient strategy for enhancing COMP–Ang1 production and reducing COMP–Ang1 aggregation.</P>

        Effects of modified LDPE on physico-mechanical properties of HDPE/CaCO3 composites

        HDPE/CaCO3/LDPE-g-MA composites of high density polyethylene (HDPE) and calcium carbonate (CaCO3) with maleic anhydride grafted low density polyethylene (LDPE-g-MA) as a compatibilizer were prepared by melt mixing. LDPE-g-MA was prepared using a solution process. The maximum grafting degree was obtained at 6 wt% maleic anhydride (MA) and 0.2 wt% dicumyl peroxide (DCP) at 120 oC and 240 min. Functional groups not found in pure LDPE were observed with the formation of LDPE-g-MA. The successful dispersion of CaCO3 particles in the HDPE matrix using LDPE-g-MA was revealed. The crystallite size of the composites was a little higher than that of CaCO3. The mechanical properties of the HDPE/CaCO3/LDPE-g-MA composites increased with decreasing CaCO3 content. Thermogravimetric analysis (TGA) revealed a higher thermal stability for the HDPE/CaCO3/LDPE-g-MA composites than for pure HDPE. Differential scanning calorimetry (DSC) revealed that the crystallization conditions were not substantially different. However, melting enthalpy and crystallinity increased with decreasing CaCO3 content. The thermal stability was greatly improved compared to that previously reported for nano CaCO3.

        예비 유아교사의 불안수준에 따른 심리적 독립이 정서지능에 미치는 영향

        본 연구는 예비 유아교사의 정서지능에 영향을 미치는 요인으로 심리적 독립을 선정하여 미치는 영향력을 규명하고자 하였다. 그리고 불안수준에 따라 예비 유아교사의 심리적 독립과 정서지능이 차이가 있는지를 알아보고자 하였다. 연구대상은 유아교육관련학과의 예비유아교사 703명 중 불안 수준에 따른 상위 10%, 하위 10%의 140명이다. 수집된 자료는 SPSS Statistics 18.0 통계프로그램을 사용하여 심리적 독립과 정서지능간의 관계를 알아보기 위해서 상관분석을 실시하였으며, 단계선택방식으로 위계적 회귀분석을 사용하였다. 연구결과, 불안수준에 상관없이 심리적 독립은 정서지능에 통계적으로 유의한 수준에서 영향을 미치는 요인으로 나타났다. 그리고, 불안수준이 높은 예비유아교사와 낮은 예비교사는 심리적 독립이 유의미하게 차이가 났다. 정서지능 중 사고촉진, 정서조절은 불안수준이 높은 예비유아교사의 경우 갈등적 독립만이 영향을 미쳤지만 불안수준이 낮은 예비교사의 경우 갈등적 독립과 기능적 독립이 영향을 미쳤다. 정서의 표현, 활용, 감정이입에서는 갈등적 독립이 불안수준이 높은 예비유아교사와 낮은 예비교사 모두 영향을 미쳤다. 마지막으로 본 연구의 의미와 시사점, 한계에 대해 논하였다. The purpose of tuis study is to examine psychological separation on emotional intelligence among early Childhood Teachers with different levels of state anxiety. The subjects of this study consisted of 703 male and female early childhood teachers sampled randomly in Daegu and Kyungbuk. The instruments used in this study were Psychological Separation Inventory(PSI), Symptom Checklist-90- Revised( SCL-90-R), Emotional intelligence inventory. This study estimate the effect of different levels of state anxiety. In analyzing data, Correlation Analysis and multiple regression analysis were done by using SPSS Statistics 18.0 program. Major results of this study are as follows. First, the significant correlations were found between psychological separation and emotional intelligence. Second, psychological separation effect emotional intelligence differed according to the anxiety level. Third, emotional intelligence. influenced psychological separation. Thus, psychological separation is the biggest influence emotional intelligence. Discussion of this study result, implication and limitations were described.

        Membrane proteomic analysis of human mesenchymal stromal cells during adipogenesis

        <P>Mesenchymal stromal cells (MSCs) have proven useful for cell and immune therapy, but the molecular constituents responsible for their functionalities, in particular, those on the plasma membrane, remain largely unknown. Here we employed both gel and nongel based MS to analyze human MSCs' membrane proteome before and after adipogenesis. 2-DE of cells that were pretreated with membrane impermeable fluorescent dyes revealed that both the whole cell proteome and the cell surface subproteome were independent of donors. LC coupled with tandem MS analysis of the plasma membrane-containing fraction allowed us to identify 707 proteins, approximately half of which could be annotated as membrane-related proteins. Of particular interest was a subset of ectodomain-containing membrane-bound proteins that encompass most known surface markers for MSCs, but also contain a multitude of solute carriers and ATPases. Upon adipogenic differentiation, this proteomic profile was amended to include several proteins involved in lipid metabolism and trafficking, at the expense of, most noticeably, ectoenzymes. Our results here provide not only a basis for future studies of MSC-specific molecular mechanisms, but also a molecular inventory for the development of antibody-based cell isolation and identification procedures.</P>

        The author present a modified multi-loop algorithm including feedforward for controlling a 55kW step down chopper in the power supply of Maglev. The control law for the duty cycle consists of three term. The first is the feedforward term which compensates for variations in the input voltage. The second term consists of the difference between the slowly moving inductor current and output current. The third term consists of proportional and integral terms involving the perturbation in the output voltage. This perturvation is derived by subtracting the desired output voltage from the actual output voltage. The proportional and integral action stabilizes the system and minimizes output voltage error. In order to verify the validity of the proposed multi-loop controller, simulation study was tried using Matlab simulink.

        Genome-Wide Differential Gene Expression Profiling of Human Bone Marrow Stromal Cells

        <P>Bone marrow stromal cells (BMSCs) reside in bone marrow and provide a lifelong source of new cells for various connective tissues. Although human BMSCs are regarded as highly suitable for the development of cell therapeutics and regenerative medicine, the molecular factors and the networks of signaling pathways responsible for their biological properties are as yet unclear. To gain a comprehensive understanding of human BMSCs at the transcriptional level, we have performed DNA microarray-based, genome-wide differential gene expression analysis with the use of peripheral blood-derived mononuclear cells (MNCs) as a baseline. The resulting molecular profile of BMSCs was revealed to share no meaningful overlap with those of other human stem cell types, suggesting that the cells might express a unique set of genes for their stemness. By contrast, the distinct molecular signature, consisting of 92 different genes whose expression strengths are at least 50-fold higher in BMSCs compared with MNCs, was shown to encompass largely a gene subset of umbilical cord blood-derived adherent cells, suggesting that adherent cells derived from bone marrow and umbilical cord blood may be defined by a common set of genes, regardless of their origin. Intriguingly, a large number of these genes, particularly ones for extracellular matrix products, coincide with normal or tumor endothelium-specific markers. Taken together, our results here provide a BMSC-specific genetic catalog that may facilitate future studies on molecular mechanisms governing core properties of these cells.</P>

      • 45,XI 46,XXr 핵형의 터너 증후군 1례

        A 16-years-old-girl was presented with chief complaints of dizziness and exertional dyspnea. She was tall 146 cm (less than the fifth percentile) and sexually developed normally. But she experienced intermittent menorrhagia. She had a shield chest, a low-set nuchal hairline and a short left metatarsal bone. There were no other dysmorphic facial features, no webbed neck, no cubitus valgus, & no mental retardation. The blood hemoglobin was 6.2 g/dl and peripheral blood smear showed hypochromic microcytic red blood cells. Bone marrow showed moderate erythroid hyperplasia and normoblast with scant cytoplasm, and it was not stained by Prussian Blue stain. She had a mosaicism of 45.X/46.XXr in the cells, cultured from the peripheral blood. Fifty five cells were examined. Forty-two cells had 45 chromosome, with one X. The remaining thirteen cells had 46 chromosomes per cell, with a normal X and a ring X chromosome. The mother was 155 cm tall and had normal karyotype. We herein report a Turner syndrome patient with a ring X chromosome with a brief review of literature.
