Occurrence and Analysis of Apple Blotch-like Symptoms on Apple Leaves

Apple blotch-like symptoms (ABLS) were observed on ‘Fuji’ apple leaves in Cheongsong, Gunwi and Yeongcheon apple orchards located in Gyeongbuk Province during 2010-2014. Characteristics of ABLS were yellowing, brown spots on leaves, and defoliation, similar to apple blotch diseased (ABD) leaves, which are infected with Marssonina coronaria. It is difficult to differentiate by eye between ABLS and ABD, which has led to misdiagnosis and overuse of fungicides. The present study was conducted to investigate the cause of ABLS using stereomicroscopy, culture isolation, cross-sectional analysis of leaves, and PCR. No acervuli were found on the surface of ABLS leaves and no growth was observed on potato dextrose agar (PDA) plates in culture. Furthermore, cross-sectional analysis revealed similar results, and mycelia were absent in ABLS leaves. By contrast, all these characteristics were present in ABD leaves. Furthermore, no fungi or viruses were detected in ABLS leaves by PCR, suggesting that the disease is not caused by these agents. These findings suggest that ABLS might be a physiological disorder in plants that is distinct from ABD.

In this study, we developed a species-specific PCR assay for rapid and accurate detection of three Xanthomonas species, X. axonopodis pv. poinsettiicola (XAP), X. hyacinthi (XH) and X. campestris pv. zantedeschiae (XCZ), based on their draft genome sequences. XAP, XH and XCZ genomes consist of single chromosomes that contain 5,221, 4,395 and 7,986 protein coding genes, respectively. Species-specific primers were designed from variable regions of the draft genome sequence data and assessed by a PCR-based detection method. These primers were also tested for specificity against 17 allied Xanthomonas species as well as against the host DNA and the microbial community of the host surface. Three primer sets were found to be very specific and no amplification product was obtained with the host DNA and the microbial community of the host surface. In addition, a detection limit of $1pg/{\mu}l$ per PCR reaction was detected when these primer sets were used to amplify corresponding bacterial DNAs. Therefore, these primer sets and the developed species-specific PCR assay represent a valuable, sensitive, and rapid diagnostic tool that can be used to detect three specific pathogens at early stages of infection and may help control diseases.

From June to October 2012, severe outbreaks of rust disease on Solidago virgaurea var. gigantea were observed in 5different regions of Ulleung-do, Korea. Typical rust symptoms appeared as yellow spots, uredinia on leaves, and massiveure diniospores covering the entire plant. Severely infected plants became discolored and died. On the basis of its morphological and molecular characteristics, the causal agent was identified as Coleosporium asterum. This pathogen has been reported as rust disease on S. virgaurea in the Canada and United States. In Korea, rust disease on S. virgaurea var. gigantea was reported as Uromyces rudbeckia in 1992. To the best of our knowledge, C. asterum is a novel rust pathogen on S. virgaurea var. gigantea in Korea.

Four Cladobotryum isolates were collected from four different commercially grown mushroom types infected with cobweb disease in Cheongdo-gun and Chilgok-gun of Gyeongbuk Province, Korea in 2010. The isolates were identified as C. mycophilum from Agaricus bisporus and Pleurotus eryngii, C. varium from Flammulina velutipes and Hypsizygus marmoreus. The cultural characteristics of the four isolates were investigated using potato dextrose agar (PDA) media under nine different temperatures ranging from 5~32oC. Rapid growth of the isolates to colony diameters of 47~82 mm was observed at conditions of 18~22oC. No growth was observed at 32oC. C. mycophilum produced a yellowish red pigment while C. varium produced a cream colored pigment after cultivation for 25 days on PDA. Phylogenetic analysis of the internal transcribed spacer region and partial 28S rDNA from the four isolates confirmed they were C. mycophilum and C. varium. Cross pathogenicity tests revealed that the two isolates of C. mycophilum were highly pathogenic toward three mushroom types, but not toward H. marmoreus. The two isolates of C. varium were less pathogenic than those of C. mycophilum, but were pathogenic toward all mushroom types evaluated.

우리나라에 발생하는 잿빛곰팡이병균 Botrytis cinerea의 분자계통학적 유연관계

잿빛곰팡이병의 전형적인 병징을 나타내는 병든 사과, 고추, 딸기, 오이, 토마토에서 곰팡이를 분리하고, 그들의 배양학적 특성과, 형태적 특성 및 PCR-RFLP을 통해 이 병원성 곰팡이를 모두 Botrytis cinerea로 동정하였다. 또한, 배양학적 특징에 따라 사과, 고추, 오이에서 분리한 잿빛곰팡이병균의 표현형은 균핵형이며, 딸기와 토마토에서 분리한잿빛곰팡이병균은 균사형이었다. 각각의 잿빛곰팡이병균의 ITS 영역 염기서열을 포함한 4종의 유전자(RPB2, HSP60, G3PDH)의 염기서열을 결정하고 분자계통학적 유연관계를 분석하였다. RPB2 유전자 염기서열을 제외한 ITS 영역, HSP60유전자 및 G3PDH 유전자의 염기서열은 Botrytis cinerea 종 내 뿐만 아니라 Botrytis 속 종간에도 매우 높은 상동성을 나타내어 계통학적 유연관계 분석이 어려웠다. 하지만, 3종의 유전자(RPB2, HSP60, G3PDH)를 결합한 유전자 염기서열을 이용한 분자 계통수 작성 결과, 본 연구에서 분리한 잿빛곰팡이병균은 Botrytis 속의 다른 종들과 구별되며, 사과, 고추, 오이, 토마토의 분리주는 아주 높은 근연관계에 있고, 딸기잿빛곰팡이병균은 다른 분리주와 달리 종내 다른 lineage를 형성하였다. Several isolates were collected from apple, pepper, strawberry, cucumber and tomato having typical gray mold symptoms. All the isolates were identified as Botrytis cinerea by using morphological characteristics and PCR-RFLP method. It was difficult to analyze the phylogenetic relationship of these isolates by using ITS region, HSP60 and G3PDH because these genes were highly homologous in their nucleotide in inter-species of B. cinerea and intra-species of genus Botrytis. However, phylogenetic analysis using combined sequences (RPB2, HSP60 and G3PDH genes) clearly showed that all isolate of B. cinerea were different from Botrytis spp. Furthermore, it was also confirmed that strawberry isolate was distantly related to apple, pepper, cucumber and tomato isolates that were closely related to each other in nucleotide level.
