      • HPV에 감염된 자궁경부 상피세포의 Keratin 아형 변화에 관한 연구

        The human papillomavirus (HPV) are wide spread in the world, causing proliferation of epidermal and mucosal surface. Certain papillomaviral types are oncogenic in vivo and in vitro. HPV DNA has been detected in most of cervical lesions such as condyloma acuminata, dysplasias, and invasive carcinomas. The materials for this study consisted of 5 dysplasias. 5 squamous cell carcinomas, and 5 exocervix for control, that resected for definite treatment of uterine disease at Chosen University Hospital from January, 1991 to December, 1993. All cases were performed of in situ hybridization for HPV DNA type 6/11, 16, and 18. And also immununohistochemical stain for panel of monoclonal cytokeratin (CK) antibodies were done. The result obtained is as fallows : 1. HPV DNA type 6/11 are detected in all dysplasias. HPV DNA type 16 and 18 are detected in squamous cell carcinomas. 2. Stratifed squamous epithelium of exocervix are positive staining for CK 1, CK 5/6, CK 8, CK 13, CK 14, and CK 19. HPV DNA type 6/11 infected cells are positive for CK 1, CK 13, CK 14, CK 17, CK 18, and CK 19. HPV DNA type 16/18 infected cells are positive for CK 1, CK 5/6, CK 10, CK 13, CK 14, CK 17, CK 18, and CK 19. In conclusion, HPV infection of uterine cervix is associated with change of normal cytokeratin expression pattern. Dysplastic cells are changed of CK 5/6, CK 8, CK 17 and CK18. Squamous cell carcinoma cells are changed of CK 8, CK 17, and CK 18.

      • 강원도지역에서 발생한 발진열과 쯔쯔가무시병

        In Korea, there has been an increasing concern on rickettsiosis as one of most prevailing acute febrile illness, since murine typhus and scrub typhus had not been reported for 2 decades until 1986 and thereafter a few studies were reported on these diseases. A microimmunofluorescence test was used to study antibodies to R. typhi and R. tsutsugamushi in urban and rural inhabitants students, and patiants suspected of leptospirosis or rickettsiosis from 1986 to 1987 in Kangwon-do Korea. 43% of inhabitatnts and 17% of students reacted to R. typhi. Only one inhabitant among inhabitants and students reacted to R. tsutsugamushi. Among 169 patients, 43(25%) reacted to R. typhi, 38(22%) to R. tsutsugamushi and 25(15%) to Leptospira interrogans. Sexual rate(male; female) in murine typhus was 1.7:1, that in scrub typhus was 1:2.2 and that in leptospirosis was 1.5;1. The age ditribution of patients with murine typhus was generally even over 20 years old. But 71% of patients with scrub typhus were over 40 years old. Scrub typhus was most prevailing in female over 40 years old. Murine typhus was relatively more prevailing in Choonchun city than rural area as compared with scrub typhus and leptospirosis. Most of scrub typhus occurred in rural area. Most of each disease occurred between September and November (murine typhus; 93%, scrub typhus; 95%, leptospirosis; 84%) with a peak in October. Scrub typhus considerably occurred not only October(58%) but also November(28%). All patients with scrub typhus were immediately diagnosed by antibody when they visited hospital. 53% of patients with murine typhus were diagnosed when they visited hospital within 7 days after onset of illness, and others were done by second sera obtained 8 days after. The patients in whom IgM was seronegative but antibodies of other class were seropsitive by 20 days after onset of illness, were 14% in murine typhus and 26% in scrub typhus. At least those patients were considered to be infected second. Murine typhus and scrub typhus were suggested to be endemic diseases in Kangwon-do as antibodies of inhabitants and characteristics of patients' sera were considered. By serologic finding of patients with scrub typhus in Kangwon-do, Gilliam was 59'% of etiologic strain of scrub typhus, Karp was 41q% and Kato was none. Most scrub typhus occurred in person to be related to agriculture, but murine typhus was not related to job and occurred more in small city than rural area. Rash was scarcely detected in murine typhus and scrub typhus, and eschar was in 50% of scrub typhus only. Serum transaminase elevated in many patients with scrub typhus and murine typhus. Leukocytosis and abnormal finding in chest X-ray were showed in some patients with scrub typhus but were scarcely in murine typhus.

        Usefulness of Skin Explants for Histologic Analysis after Fractional Photothermolysis

        Background: Fractional laser resurfacing treatment has been extensively investigated and is widely used. However, the mechanism underlying its effects is poorly understood because of the ethical and cosmetic problems of obtaining skin biopsies required to study the changes after laser treatment. Objective: To evaluate the usefulness of human skin explants for the investigation of fractional photothermolysis. Methods: Full-thickness discarded skin was treated in 4 ways: no treatment (control), fractional carbon dioxide laser, fractional Er:YAG laser, and fractional 1,550-nm erbium- doped fiber laser. Both treated and non-treated skin samples were cultured ex vivo at the air-medium interface for 7 days. Frozen tissue was sectioned and stained with hematoxylin & eosin for histologic examination and nitro blue tetrazolium chloride for viability testing. Results: Skin explants cultured for up to 3 days exhibited histologic changes similar to those observed in in vivo studies, including microscopic treatment zones surrounded by a thermal coagulation zone, re-epithelialization, and formation of microscopic epidermal necrotic debris. However, the explant structure lost its original form within 7 days of culture. The viability of skin explants was maintained for 3 days of culture but was also lost within 7 days. Conclusion: The skin explant model may be a useful tool for investigating the immediate or early changes following fractional photothermolysis, but further improvements are required to evaluate the long-term and dermal changes.       • 장기에 따른 편평상피암의 Cytokeratin 발현에 관한 연구

        Keratin is a major class of epithelial intermediate filaments and comprises a group of at least 20 different multigene derived proteins. It is expressed in different epithelia with specific combinations. The expression of cytokeratins (CK) 1.5/6, 7, 8, 10, 13, 17, 18, 19, and 20 in 35 cases of squamous cell carcinomas from uterine cervix (5 cases), lung (5 cases), esophagus (5 cases), skin (5 cases), oral cavity (5 cases), and metastatic Iymph nodes (2 cases of each organs) were examined. Some cases were examined by electron microscopy. The results obtained is as follows. 1. The CK 1 positive tumors is from keratinizing skin tumor and is show weak positivity. The CK 5/6 positlve tumor is from uterine cervix, lung and oral cavity. The CK 8 positive tumor is from uterine cervix, lung, esophagus, skin and oral cavity. The CK 13 positive tumors is from uterine cervix and lung. The CK 17 show positive staining in uterine cervix and lung. The CK 18 positive tumors is from uterine cervix, lung, esophagus, skin, and oral cavity. The CK 19 positive tumors is from uterine cervix, lung, esophagus, skin, and oral cavity. The CK 20 shows no reactivity in all cases. 2. Ultrastructurally, the tumor cells showed bundles of tonofibriIs, electron-dense tonofilaments, and desmosomes. In conclusion, CK 1,8 18 and 19 did not show orgn specificity. So it used simple epithelial markers in the majority of squamous cell carcinomas. The absence of CK 20 immunoreactivity may helpful in differential diagnosis of squamous cell tumors.

        Thanatophoric dysplasia is a severe skeketal disorder characterized by extremely short limbs and folds of extra skin on the arms and legs, narrow chest, small ribs and underdeveloped lungs. Affected infants are usually stillborn or die shortly after birth from respiratory failure. We experienced a case of thanatophoric dysplasia died shortly after birth from respiratory failure.

      • 노랑초파리 집단내 세대변화에 다른 Cytotype 및 P factor activity의 변화에 관한 연구

        Isofemale lines were estabilished from YJ island in August and September, 1990. After 15 generation, they tested for the G.D sterility by P-M system. And then, the results were compared G_1 and G_15 to cytotype conversion and P factor activity change. The results were as follows. It was analyzed to consist of M(0.3%), M' (56%), Q(42.6%) and no P strain in isofemale line(G_15). It was mainly consist of M' and Q strain like any other Korea region. And the cytotype are prevalent, though the P activity are very low in this population. During the period of 15 generation labortary culture, cytotype was transformed M cytotype (28.2% → 25.5%). And P factor activity decreased from 1.0% to 0.3%.

      • Drosophila immigrans의 韓國集團에 있어서의 同位酸素 多型現象

        In order to clarify the genetic structure and variability in Korean populations of D. immigrans, for isozymes are assayed by means of starch gel electrophoresis. The results are follow : 1. Polymorphism appeared in four isozyme loci. 1. In Korean natural population, average heterozygosities are estimated, 0.9% (AcPH), 50.1%(APH), 47.7%(Est), 0.8%(GOT) respectively. 3. The frequencies of three isozymes except GOT locus had significant differences among five Korean natural populations. 4. Through the Roger's similarity index analysis, the value between Gangreung and Daejeon seemed to be low, but the value among other populations and estimated between 0.611 and 0.925. 5. The maintenance mechanism of isozyme variability in Korean populations of D. immigrans may be explained by genetic drift or founder effects.

      • 한국전통 식품의 원료인 메주와 누룩에서 분리된 접합균에 대한 연구

        The fungal isolates of Mucorales, directly collected from Korean traditional raw materials of Nuruk (raw material for Korean rice wine) and Meju (raw material for Korean soysauces), were compared with those of Rhizopus oryzae purchased. The fungal isolates of Rhizopus, Mucor, and Absidia mostly identified as based on the morphological observations, were evaluated with the PCR-polymorphic bands. The PCR-polymorphic bands of the genomic DNA reacted with the primers of OPD series tenmer were various, but showed averaged 4 to 6 in the agarose-electrophoresis. The dissimilarity coefficient (DC) between two isolates were compared by the cluster analysis, dendrogams and polar ordinations. The isolates of R. oryzae known showed several groupings within the lower value of DC and were divided to two groups of amylo-process and other fungi with other purposes. The isolates unidentified were identified by the DC made of this results. Taxonomy of these isolates made by the morphological observations were consistent with those resulted above in most case but not in all aspects. More works were needed with the isolates known for detail informations of Mucorales.

        β-CuZn 합금의 항복강도 이상거동에 미치는 변형률 속도와 온도의 영향

        A series of tensile and load relaxation behaviors in B2 type β-CuZn alloy have been examined to clarify the strain rate & the temperature dependence of yield stress anomaly. It has been found in tensile tests that the region showing anomalous peak of yield stress moves to a higher stress and higher temperature region with increasing strain rate. The anomalous yielding region below peak temperature(T_p) revealed slightly positive strain rate dependence, while the region above T_p showed considerably positive strain rate dependence. The slip lines formed below peak temperatures (T_P) were clearly visible with naked eye(coarse slip lines), and became finer near T_p(fine slip lines), while they disappeared completely above T_p. Based on the different tendency of temperature and strain rate dependence of anomalous yielding as the above-mentioned results, two types of deformation modes were suggested and incorporated into the flow curve analysis, which was in accord well with the experimental results. The slightly positive strain rate dependence of yield stress anomaly indicated that the yield stress anomaly in β-CuZn alloy mainly seemed to be attributed to the vacancy hardening.
