L1210 세포증식에 대한 Glycyrrhizin의 억제작용 기전

The purpose of this research was to investigate the mechanism of inhibitory action of Glycyrrhizin(GZ) on the proliferation of mouse leukemia cell-line, L1210 cells. The cytotoxic activity was tested using a colorimetric tertrazolium assay(MTT assay), the apoptosis was tested using flow cytometry. Nitric oxide(NO) production form mouse peritoneal macrophage was tested using a Griess method and the phagocytosis of human polymorphonuclear cells was tested using a lucigenin chemiluminescence. GZ ingibited the proliferation of L1210, BALB/c 3T3 cells and mouse thymocytes at 50 ug/ml/ GZ did not affect nitric oxide production from mouse peritoneal macrophages in vitro, but ingibited nitric oxide production from lipopolysaccharide and y-interferon treated macrophages. Macrophages of GZ-administered mice accelerated NO production. The proliferation of L1210 cells apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. The apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. GZ increased the phagocytosis of human polymorphonuclear cells. These results suggest that GZ inhibit the proliferation of L1210 cells not only as a direct cytotoxic agent o tumor cells, but also by the enhancement of NO production and phagocytic activity.

간동맥 가성동맥류는 비교적 드문 질환으로, 과거에는 주로 외상에 의해 발생했으나, 최근에는 중재적 시술이나 복강경 수술의 합병증으로 인한 발생이 증가하고 있다. 하지만 자발성 가성동맥류는 매우 드물다. 복통, 종괴, 출혈 및 파열 등의 다양한 증상을 보이며, 파열 시 높은 사망률과 유병률을 보이기 때문에 조기 진단과 치료가 중요하다. 저자들은 지속적인 상복부 통증으로 내원한 환자에서 총간동맥에 발생한 자발성 가성동맥류를 진단하고, 경동맥 코일 색전술로 치료한 증례를 경험하여 보고하는 바이다. Hepatic pseudoaneurysms were once relatively uncommon and often associated with hepatic trauma. Currently, the main etiology of pseudoaneurysms is iatrogenic injuries with the increased use of percutaneous procedures and laparoscopic surgery. However, a spontaneous hepatic pseudoaneurysm is very rare. The clinical manifestations can include pain, swelling, thrombosis, bleeding, and rupture. Early detection is critical for the proper treatment, because ruptured pseudoaneurysms have high morbidity and mortality. We present a case of spontaneous pseudoaneurysm in the common hepatic artery that was treated successfully with transarterial coil embolization. (Korean J Med 77:95-98, 2009)
P222 : Changes of epidermal thickness in vitiligo

Background: Stratum corneum and pigmentation have protective role from ultraviolet radiation. Because vitiligo skin has no melanocyte and has no potential to make pigment, some studies suggested that the thickness of stratum corneum increase in vitiligo skin. Objectives: We investigated the thickness of stratum corneum and full thickness of epidermis at the vitiligo skin comparing with adjacent normal-appearing skin. Methods: Under the microscope, we took photos of routine hematoxylin and eosin stained slides in vitiligo patient. Then we measured the thickness of stratum corneum and full thickness of epidermis. Results: We examined 206 patients, composed with 96 men and 110 women. Their mean age was 32.01 years and mean duration of vitiligo was 18.14 months. The thickness of stratum corneum in lesional skin is 33.31 ± 30.38μm, and that in normal-appearing skin is 29.30 ± 21.08μm. There is no significant difference (p=0.12). However, in thickness of epidermis, there is significant difference between lesional (100.39 ± 48.46μm) and normal-appearing skin (91.28 ± 29.34μm, p=0.02). While there is no difference on nonsun-exposed area, the epidermal thickness of lesion is significant higher than that of normal-appearing skin on the sun-exposed area (p<0.05). Conclusion: We reveal the epidermal thickness is increased in vitiligo, and it could be photoprotective role to compensation for absent pigmentation.

Concern about detecting gastric carcinoma in its early stages has increased the incidence of detecting multiple synchronous gastric carcinomas. Although gastric carcinomas may present with various gross features, those showing the features of submucosal tumor (SMT) are rare. We report on a case of synchronous gastric carcinomas comprising one lesion with typical features of superficial early gastric carcinoma and the other with atypical features that mimicked SMT. Even though synchronous gastric carcinoma is rare, it may be worthwhile to make a pathological diagnosis of coexisting SMT using endoscopic-ultrasound-guided fine-needle aspiration or endoscopic mucosal resection. The study surveyed 350 dental hygienists in Seoul, Gyeonggi-do, and other regions to assess the correlation between their emotional labor and depression level and their impact on job satisfaction and commitment to the organization. In order to find out the factors affecting job satisfaction, monthly income, place of work, and leisure activities were treated as variable numbers, emotional labor, depression, and organizational committment were independent variables, and multiple regression analysis was conducted with job satisfaction as dependant variables, and organizational committment and emotional labor were shown as factors affecting job satisfaction. To identify the factors affecting organizational committment, monthly income, place of work, and leisure activities were treated as variable numbers, emotional labor, depression, and multi-recovery analysis using organizational committment as independent variables, and the results showed that job satisfaction, emotional labor, and depression were the factors affecting organizational committment. In this study, dental hygienists have shown tendencies for commitment to the organization in the order of job satisfaction, emotional labor, and depression. The results have confirmed that emotional labor and depression experienced in the health care environment are closely linked to job satisfaction and commitment to the organization, which will help with other research to deal with and solve emotional labor and depression of dental hygienists.

Backgrounds: Drug-induced liver injury (DILI) is a major causal factor for failure in clinical trials and withdrawal of drugs from pharmaceutical markets. Therefore, a human cell-based in vitro system has been established to overcome the limitations of preclinical trials. However, previous studies focused on the drug metabolism in each cell line or described to be superior in one single cell line for assessing DILI. In this study, we used Huh7, HepaRG, and Hepatosight-S cells to evaluate the liver toxicity and dysfunction driven by DILI-inducing drugs. Methods: Cytotoxicity of 17 DILI-inducing drugs was assessed by LDH release assay and the drug-induced liver dysfunction was confirmed by albumin secretion. Furthermore, we analyzed the expression levels of phase I and phase II enzymes and nuclear receptors in each cell lines. Results: DILI-inducing drugs were differently detected in each cell line because each cell line differs on the expression of drug metabolic enzymes which associated with reactivity on DILI inducing drugs. Conclusion: To predict the responses of DILI-inducing drugs in human liver cells, using diverse cell lines having different drug metabolic capabilities is more suitable for predicting DILI.

Background/Aims: Although various solutions are being tested for submucosal injection during endoscopic resection, ideal solution has not been established yet. We performed an animal study to evaluate the possibility of sodium alginate as an ideal submucosal injection solution for endoscopic mucosal resection (EMR). Methods: To compare the lesion-lifting properties of different solutions, injection was done to the submucosal layer of porcine stomach. Then the height of mucosal elevation was measured. In addition, EMR was performed after submucosal injection of sodium alginate solution in six dogs. Two were euthanized after 30 minutes of endoscopic observation while the others were euthanized after 1-4 weeks. Retrieved stomachs were examined microscopically. Results: Sodium alginate and sodium hyaluronate solutions maintained longer elevation of the submucosal layer than other solutions. There was no significant difference in the height between two solutions. A clear separation of the mucosal layer from the proper muscle layer was achieved by injecting sodium alginate solution. Histological examination of EMR-induced artificial ulcers revealed no apparent tissue damage and showed normal healing process. Conclusions: Sodium alginate solution creates a sufficient submucosal fluid cushion without apparent tissue damage. It can be considered as an effective submucosal injection material.
