        여지 전기영동법에 의한 난백 단백질의 분리

        A detailed separation of egg white protein into fractions was performed by paper-electrophoretic techniques. Four to six fractions in terms of general protein were obtained by virtue of this method employ ing barbiturate buffer (pH=8. 6 u=0. 1) with 150 V and 22 hours operation. The glycoproteins and lipoproteins of egg white were fractionated paper electrophoretically with the use of Schiff`s reagent and Sudan Black B, respectively. Quantitative estimation of each fraction was carried out with the egg whites laid by hens differing in age with each other. The egg whites of duck and uroloncha were subjected to the paper electrophoresis likewise and compared to that of hen.

        모노클론 항체에 대한 신생 , 어린 및 성숙된 흰쥐 간조직 마이크로좀 cytochrome P - 450 pattern 확인

        We observed the multiplicity as well as differential expression of cytochrome P-450 during the development of rats detected by RIA and Western blotting. Compared to non-specific MAbs (NBS 1-48-5 and HYEL-9), MAbs to cytochromes P-450 2c/RLM5(1-68-11) recognized the Corresponding cytochrome P-450 2c/RLM5 the most in young and adult male and the following cytochromes P-450 in their quantity detected by respective MAbs were ethanol-, isosafrole-and phenobarbital-inducible cytochrome P-450. When MAbs to cytochrome P-450 2c/RLM5 were purified and cytochrome P-450 was immunopurified from the liver microsomes of adult male rats with the purified MAbs, the concentration dependent MAb-bindings to both chemically and immunopurified cytochrome P-450 showed similar patterns. The highly expressed form of cytochrome P-450, P-450 2c/RLM5 was immunopurified from liver microsomes of adult male rats with purified MAbs and it was confimed that both chemically purified cytochrome P-450 were homologous or at least contained common epitope to cytochrome P-450 2c/RLM5. Similar results were obtained in Western blotting with MAbs to cytochrome P-450 2c/RLM5.

      • Identification of Microsomal Cytochrome P-450 Patterns in Neonatal, Young and Adult Rats by Monoclonal Antibodies

        흰쥐 성장과정 중에 cytochrome P-450의 달라지는 발현(expression)과 다양성을 RIA와 Western blotting에 의하여 관찰하였다. Non-specific MAbs (NBS 1-48-5와 HYEl-9), cytochrome P-450 2c/RLM5에 대한 MAbs (1-68-11)을 비교하였더니 대부분의 숫컷 어린쥐와 성숙된 쥐 cytochrome P-450 2c/RLM5와 일치 하는 것을 확인하였다. 그리고 양적인 순서는 ethanol-, isosafrole- 및 phenobarbital-로 유도된 cytochrome순이었다. 정제된 cytochrome P-450 2c/RLM5에 대한 MAbs와 숫컷 성숙된 쥐 간조직 마이크로좀에서 immunopurify한 cytochrome P-450은 같은 pattern을 나타내었다. 이런 결과는 정제된 cytochrome P-450이 homologus 하거나 화학적으로 정제된 cytochrome 2c/RLM5에 공통된 epitope를 갖는다는 것을 지적하는 것이다. 또한 cytochrome P-450 2c/RLM5에 대한 MAbs같이 Western blotting도 비슷한 결과를 얻었다.       • 여지 전기영동법에 의한 난백 단백질의 분리

        여지전기영동법에 의하여 단백질을 분리하는 조건을 모색하고 염색법을 달리하여 복합단백질인 당단백질, 지단백질, Flavoprotein 등의 탐색을 시도하였다. 계란난백의 총단백질은 Kjeldahl 방법으로 측정하고 그 각각의 성분은 여지전기영동법으로 정량하여 그 변화를 관찰하였다. 실험결과 난백단백질을 혈청단백질에 사용한 Barbiturate buffer pH=8.6 $\mu$=0.10와 150 V 22시간 전개에서 가장 잘 분리되었으며 각 fraction은 4~6으로 나누어졌고 동정 또는 추측되는 것은 ovalbumin ($A_1$, $A_2$), lysozyme ($G_2$, $G_3$), ovomucoid, conalbumin 등이 있다. 각 fraction의 당단백질과 지단백질은 Schiff's 시약과 Sudan Black B를 사용하여 염색하므로 분리가 가능하였다. 연령별 산란계의 계란 난백의 일반단백질, 당단백질, 지단백질의 정량을 실시하였던 바 ovalbumin의 비율이 연령에 따라 점증하고 그 외부분인 $G_3$, ${G_2}^+$ ovomucoid, conalbumin 등의 부위도 차츰 감소하였으며 지단백질에서도 이 부위에 위치하는 fraction이 같은 경향을 보였다. 즉 닭의 노화에 따라 총단백질과 ovalbumin 을 뺀 기타 각성분이 감소하는 경향을 보였다. 오리 십자매의 난백에 대하여 일반단백질, 당단백질, 지단백질의 분리를 시험하여 계란난백에 그것들과 비교하였다.         개구리에서 N - Acetyl 및 N - Carbamyl Glutamic Acid 의 암모니아 독에 대한 해독작용에 관한 연구

        Using N-acetyl-L-glutamate, N-carbamyl-L-glutamate, lysine and other amino acids, the antidotal effect of these substances against ammonia was investigated in frogs. And the following conclusion was obtained: lysine and glutamic acid showed similar antidotal effect against ammonia. N-carbamy-L-glutamate showed stronger antidotal effect than N-acetyl-L-glutamate and it was more effective when administered with arginine than when administered alone.

        개구리에서 암모니아의 $LD_{99.9}$은 체중 20gm 당(當) 0.65mM 이었다(32.5mM/kg). 백서에서의 $LD_{99.9}$는 이에 비하면 1/3 밖에 아니된다. 그리고 개구리에서 체중 20gm 당 2.0mM의 arginine은 암모니아 $LD_{99.9}$량의 독성을 완전히 해독하였다. 그외 glutamic acid, aspartic acid 그리고 glycine을 주사했을 때는 암모니아 독에 대한 해독작용은 없었고 아마노산 자체의 독성을 나타내었다. The $LD_{99.9}$ of ammonium acetate in frogs was 0.65 mM/20 gm of the body weight. The $LD_{99.9}$ of ammonium acetate in rats when compared with frogs is about one-third of that in frogs, and 2 mM of arginine/20 gm of the body weight completely prevented the toxicity of $LD_{99.9}$ of ammonium acetate in frogs.

        개구리에서 암모니아 독에 대한 Arginine 의 해독작용에 관한 연구

        Th.e LD_(99.9) of ammonium acetate in frogs was 0. 65 mM/20 gm of the body weight. The LD_(99.9) of ammonium acetate in rats when compared with frogs is about one-third of that in frogs, and 2 mM of arginine/20 gm of the body weight completely prevented the toxicity of LD_(99.9) of ammonium acetate in frogs.

        N-acetyl-L-glutamate, N-carbamyl-L-glutamate 및 lysine과 그밖에 다른 아미노산들을 사용하여 암모니아에 대한 이들 물질에 대한 해독효과를 개구리에서 연구하였다. 그리고 다음 결론을 얻었다 : lysine과 glutamic acid는 암모니아에 대해 비슷한 해독효과를 보여 주었다. N-carbamyl-L-glutamate는 N-acetyl-L-glutamate 보다 강한 해독효과를 보여주었으며, 이것은 단독으로 처리되었을 해보다 arginine 과 같이 처리되었을 때 더 효과적이었다.         Vitamin A Isomer 에 의한 적혈구 용혈에 관한 연구

        The suspensions of erythrocytes from the rat, ox, pig, human, and rabbit were treated with vitamin A isomers (9-cis, 13-cis ar all-tracts retinal and 13-cis or all-trans retinol) and hemolysis and released potassium were determined. The results were as follows : 1. Lysis of erythrocytes and release of potassium from the erythrocytes were caused by all the vitamin A isomers tested. 2. When the concentration of vitamin A isomer increased, hemolysis and released potassium from the erythrocytes increased. 3. The hemolysis and potassium release from ery throcy tes of animals by vitamin A isomers were promoted by increasing incubation time and temperature. 4. The potency of hemolysis was shown in the order of retinol and retinal of all-traps forms; 9-cis retinal; and the retinol and retinal of 13-cis forms. 5. When the sera of respective animals were added to the erythrocytes of respective animals, they inhibited strongly the hemolysis and release of potassium by vitamin A isomers. 6. When cholesterol and hydrocortisone were added to the erythrocytes of the human and rabbit, no particular effect was shown on the hemolysis and potassium release by vitamin A isomers but hydrocortisone increased hemolysis, especially in the erythrocytes of the rabbit treated with all-trans retinal.
