        Extracellular acidity enhances tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-mediated apoptosis via DR5 in gastric cancer cells

        The tumor microenvironment greatly influences cancer cell characteristics, and acidic extracellular pH has been implicated as an essential factor in tumor malignancy and the induction of drug resistance. Here, we examined the characteristics of gastric carcinoma (GC) cells under conditions of extracellular acidity and attempted to identify a means of enhancing treatment efficacy. Acidic conditions caused several changes in GC cells adversely affecting chemotherapeutic treatment. Extracellular acidity did inhibit GC cell growth by inducing cell cycle arrest, but did not induce cell death at pH values down to 6.2, which was consistent with downregulated cyclin D1 and up-regulated p21 mRNA expression. Additionally, an acidic environment altered the expression of atg5, HSPA1B, collagen XIII , collagen XXAI , slug, snail , and zeb1 genes which are related to regulation of cell resistance to cytotoxicity and malignancy, and as expected, resulted in increased resistance of cells to multiple chemotherapeutic drugs including etoposide, doxorubicin, daunorubicin, cisplatin, oxaliplatin and 5-FU. Interestingly, however, acidic environment dramatically sensitized GC cells to apoptosis induced by tumor necrosis factor-related apoptosisinducing ligand (TRAIL). Consistently, the acidity at pH 6.5 increased mRNA levels of DR4 and DR5 genes, and also elevated protein expression of both death receptors as detected by immunoblotting. Gene silencing analysis showed that of these two receptors, the major role in this effect was played by DR5. Therefore, these results suggest that extracellular acidity can sensitize TRAIL-mediated apoptosis at least partially via DR5 in GCs while it confers resistance to various type of chemotherapeutic drugs.

        Squamous Cell Carcinoma of an Ileal Neobladder - A Case Report -

        Bladder reconstruction using bowel segments, especially the ileum, has become a realistic option for urinary diversion. There is only one prior case of squamous cell carcinoma of the ileal neobladder that has been reported in the clinical literature. Here we report a patient with a spectrum of squamous cell lesions, including squamous cell carcinoma, sarcomatoid carcinoma, squamous papilloma and squamous metaplasia that developed in the ileal neobladder. A 46-year-old woman underwent a hysterectomy, cystectomy and ileocystoplasty for tuberculosis 25 years previously complained of urinary frequency and gross hematuria for one week. A pelvic CT revealed a 6.3 cm mass in the neobladder. The histopathological examination showed an 11×8 ㎝ polypoid fragile mass with a microscopically well-differentiated squamous cell carcinoma, squamous papilloma and non-tumor squamous metaplasia.

        수종 고정액의 상피세포에 대한 고정효과에 관한 연구

        The author thought that the results of cytologic tests could be changed according to the fixative methods. So the author has studied several methods of fixing exfolaiative cells collected from normal adult cheek mucosa with a solution of equal parts of ether and 95% ethylalcohol, 95 ethylalcohol and Isoprophyl alcohol, and compared the results with each other. The results were as follows : The effects of fixation, and staining efficiency of nucleus and protoplasm are similar in each method, but the staining efficiency of cytoplasm and the conversation of cytoplasmic membrane were best in 95% ethylalcohol.

        샘암종과 동반된 위 난황막종양- 1예보고와 문헌고찰 및 면역조직학적 연구 -

        Gastric yolk sac tumors are extremely rare with only a few cases reported in the literature. Here, we present the case of a 50-year-old man with an adenocarcinoma and a yolk sac tumor of the stomach, without metastasis to the lymph nodes. The tumor was an early gastric carcinoma confined to the submucosa. Twelve months post-operation the patient was alive with no complications. The yolk sac tumor showed positive labeling for -fetoprotein (AFP), 1-antitrypsin ( 1-AT), cytokeratin (CK) and carcinoembryonic antigen (CEA), but was negative for human chorionic gonadotrophin (hCG), placental alkaline phosphatase (PLAP), epithelial membrane antigen (EMA) and p53. The adenocarcinoma was positive for 1-AT, CK, EMA, and CEA, but was negative for AFP, hCG, PLAP, and p53. These findings suggest that the yolk sac tumor and the adenocarcinoma components are closely related and may represent distinct phenotypes that arise from a common mucosal epithelial cell.

        The Differential Expressions of the Epithelial-Mesenchymal Transition Regulator, Slug and the Cell Adhesion Molecule, E-cadherin in Colorectal Adenocarcinoma

        Background : Slug is a member of the Snail family of transcription factors, and it plays a crucial role in the regulation of the epithelial-mesenchymal transition by suppression of several epithelial proteins and adhesion molecules, including E-cadherin. Methods : The aim of the present study was to examine the significance between the expression of Slug in colorectal adenocarcinoma (CRA) specimens and the clinicopathological parameters of CRA, as determined by immunohistochemical analysis, and to determine the correlation between the Slug and E-cadherin expressions in non-neoplastic colorectal mucosa (n=45), primary CRA (n= 109) and metastatic CRA (n=17). A semiquantitative scoring system was applied based on the intensity and extent of the positive immunohistochemical staining. Results : The expressions of Slug and E-cadherin were associated with the depth of tumor invasion (pT) (p=0.019, p=0.001, respectively), and these expressions showed a significant inverse correlation (p< 0.001) each other. Conclusions : Our results demonstrated a positive role for Slug in the development of CRA, and Slug is a mediator of tumor invasion in CRA. In addition, an up-regulated Slug expression is significantly correlated with the loss of an E-cadherin expression, which suggests that Slug may play some role in the epithelial-mesenchymal transition (EMT) by down-regulating the E-cadherin expression.

        Survey of ERETIC2 NMR for quantification

        The ERETIC (Electronic REference To access In vivo Concentrations)2 method is a new qNMR experimental technique to measure analytes based on the signal of the reference compound without additional hardware equipment. In this study, ERETIC2 method was validated, and we sought to identify whether it would be possible to apply this method to a specific compound analysis of metabolites in plant. The 90° pulse value (P1) and spin-lattice relaxation time (T1) of each compound were measured for ERETIC2. The 91H of 3-(trimethylsilyl) propionic-2,2,3,3-d4 acid (TSP) was used as a reference peak for ERETIC 2, and then, a suitable solvent and pulse sequence for each compound were selected. Under the NOESY-presat sequence, the relative accuracy error for quantitative analyses of primary metabolites was within the range of 5%, with the exception of glucose, which showed ≥ 55% error due to saturation. It showed excellent results for the quantification of glucose by using a 30° pulse sequence, which did not suppress the water peak. In addition, the quantitative accuracy for secondary metabolites was extremely accurate, with an error ≤5% when considering the purity of the standard sample. The ERETIC2 method showed outstanding linearity, precision, and accuracy.

        A new approach to quantify paraquat intoxication from postmortem blood sample by using 1H qNMR method

        For a case study of suspected paraquatintoxication, we developed a simple and rapidmethod of 1H qNMR to determine the mili-molaramount of paraquat in postmortem blood samples. There were no interfering signals from endogenouscompounds in the chemical shift of paraquat anddiquat (internal standard). The amount of sampleused ranged from 0.25 mM to 10.0 mM. Diquat,which has similar physicochemical properties withparaquat, was chosen as an internal standard. TheNMR experimental conditions, relaxation delay timeand CPMG spin-echo pulse sequence were optimized. The developed method was validated in terms ofspecificity, accuracy, precision, matrix effect,recovery, limit of detection (LOD), and low limit ofquantification (LLOQ). The proposed qNMR methodprovided a simple and rapid assay for theidentification and quantification of the quaternaryammonium herbicide, “paraquat” in postmortemblood samples. This method was tested by using theblood from the heart of a man who was intoxicatedwith paraquat. In this particular case, the level ofparaquat was 1.07 mM in the blood. For thedetermination of quaternary ammonium herbicides,qNMR could also be used to provide a betterunderstanding of the currently available techniques.
