In order to characterize the chemical change, of the constituents and pharmacological transformation of Glycyrrhizae Radix(GR) which may occur during its processing, processed GR has been investigated in comparison with unprocessed GR. It was found that glycyrrhizin(GL) and other several constituents contained in GR were remarkably changed by processing, and the decrease of contents of GL was identified by making use of thin layer chromatography-chromatoscanner. And also, it was found that glycyrrhetic acid (GA) was identified from processed GR. We recognized that commonly processed GR showed less hemolytic effect than unprocessed GR and processed GR at $190{\circ}c$. It was also found that commonly processed GR showed more effective analgesic effect and prolonged the duration of hypnosis induced by pentobarbital-Na in mice as compared with corresponding unprocessed GR and processed GR at $190{\circ}c$. And then, commonly processed GR significantly suppressed the increases of s-GOT, s-GPT and s-LDH activities in injured mice induced by $CCl_4$.

The water extract of Gamidaebo-Tang depressed the lethal toxicity of cis-platin$(45{\mu}M/kg,\;s.c.)$, and mitomycin C (6mg/kg, s.c.). Significant depression of renal toxicity(indicated by an increased blood urea nitrogen value and creatinine value) of cis-platin $(35{\mu}M/kg)\;s.c.)$, was observed in mice treated with Gamidaebo-Tang. In rats treated with mitomycin C and Gamidaebo-Tang, the increased LDH activity and reduced total protein and albumin contents by mitomycin C were significantly depressed. The number of WBC was significantly increased in rats treated with mitomycin C but in the rats treated with mitomycin C and Gamidaebo-Tang, the number of WBC was increased and the hematocrit value and the number of RBC and Hgb were significantly increased in the dose-dependent manner. Therefore, bone marrow toxicity of mitomycin C in the rats treated with Gamidaebo-Tang was depressed. In the end, alleviative effects of the side actions of cis-platin and mitomycin C were acknowledged by combined usage of Gamidaebo-Tang and these anti-neoplastic drugs.

Two kinds of gangliosides contained in deer horns were determined by integrating the peaks of TLC densitometry. Japanese deer horn originated from China showed the highest gangliosides among tested samples and the upper parts in deer horns showed higher gangliosides than the lower parts. In the case of graded samples, the best grade A showed the highest content and the worst grade E did the lowest content. Sixteen kinds of free amino acids were analyzed by auto amino acid analyzer. The lower region the deer horn was, the more the total content of free amino acids was and several kinds of amino acids were contained quite regularly.

The anti-diabetic effects of the methanol extracts of Cortex, Ramulus and Folium part of Eucommia ulmoides Oliver were investigated by the experimental diabetic model in rats. The increased blood glucose level in the diabetic rats induced by alloxan and streptozotocin was significantly lowered and a marked increase in body weight was shown with the treatments of the extracts of Cortex and Folium. The blood insulin level lowered by streptozotocin pretreatment was significantly recovered with the treatment of the extract of Folium. In glucose tolerance test, the extracts of Cortex and Folium displayed a significant inhibitory effect on the glucose level. The extracts of Cortex and Folium also caused a marked inhibition in increased blood pressure in the diabetic rats induced by alloxan.
생약복합제제의 약효연구(제35보) : 우황청심원이 순환기계에 미치는 영향

Woohwangchungsim-Won has been widely used for the treatment of appoplexy, hypertension, arteriosclerosis, insomnia and cerebrovascular accident, etc in oriental hospital and pharmacy. In order to investigate the efficacy of Woohwangchungsim-Won, the water extract of it were bioassayed for isolated ileum, blood vessels, blood pressure, heart and diuresis. The results of this studies were as follows; Spontaneous motilities of isolated ileum of mice were strongly suppressed, and contraction of isolated ileum of mice and guinea-pigs induced by acetylcholine chloride, barium chloride and histamine were inhibited. Vaso-dilating action due to vascular smooth muscle relaxation in frogs and rabbits, and hypotensive action in anesthetized rabbits were noted. Negative inotropic action on the isolated frog heart and diuretic effect in rabbits were shown.

(목적) 두충나무(Eucommia ulmoides ‘Oliv.)는 강장, 강정, 진통약으로 한방에서 널리 응용되는 생약으로 간장과 신장을 보한다라고 되어 있고, 민간에서는 당뇨병에 좋다고 하여 차(tea)로서 복용되기도 한다. 그러나 이 두충나무의 당뇨병에 대한 영향은 아직 보고된 바 없고 또 그 유효성분도 명확히 구명되어있지 못하므로, 실험적 당뇨병 병태모델에 미치는 두충나무의 영향 및 그 활성부위를 구명하고자 유효성분연구에 선행하여 본 실험에 착수하였다. (방법) 두충나무의 피부(Cortex), 소지부(Ramulus) 및 엽부(Folium)의 각 부위의 메탄올 엑기스가 Alloxan및 Streptogotocin에 의한 당뇨병 병태모델에 미치는 체중 및 혈당량 변화를 경시적으로 측정하였고, 또 Streptosotocin에 의하여 낮아진 insulin level에 미치는 영향과 당부하시험에 있어서의 영향도 관찰하였다. (결과) Alloxan 및 Streptogotocin에 의해 유발된 당뇨병 흰쥐에 있어서 두충나무 Cortex 및 Folium의 메탄올액기스가 혈중 g1ucose함량의 유의성있는 상승억제효과를 나타내었고 또 체중의 감소를 억제함을 알 수 있었다. 또 두충나무의 Folium은 낮아진 혈중 insulin 함량의 유의성있는 회복효과를 보였다. 당부하시험에서 두충나무의 Cortex와 Folium은 유의한 혈당상승억제효과를 나타내었다. 따라서 두충나무 Cortex와 Folium의 부위에서 항당뇨활성이 있음을 알 수 있었다.

These studies were conducted an attempt to investigate effects of Paery ungtang and Kamipaeryungtang water extracts on diuretic, antipyretic, antiinfla mmatory and analgesic actions. The results of these studies were summerized as follows; Increase in urinary volume, urinary Na+ excretions were significantly recognized in normal rat. Increase in urinary volume, urinary Na^+ and Cl^- excretions were significantly shown in rat with 2㎎/㎏ HgCl₂-induced acute renal failure. Antipyretic, anti-inflammatory and analgesic effects of Paeryungtang and Kamipaeryungtang were recognized in mice, rats and rabbits.
