Background and Objectives There are several evidences of reduced cochlea blood flow after noise exposure in the cochlea. However, the pathophysiology of blood flow change is still obscure, and endothelins, proteins that constrict blood vessels and play a key role in vascular homeostasis using its receptors may have importance in this respect. In this study, we investigated the expression changes of endothelin-1 (ET-1), endothelin receptor A (ETAR) and B (ETBR) according to auditory threshold change after noise exposure. Materials and Method Mice were exposed to different noise to generate transient (group 2) and permanent threshold shift (group 3), respectively. Auditory threshold shifts were evaluated with auditory brainstem response and expression changes of ET-1, ETAR and ETBR after noise exposure were evaluated by immunohistochemistry and real time RT-PCR. Results After noise exposure, the increased ET-1, ETAR and ETBR immunoreactivities were observe in stria vascularis, spiral ligament and spiral ganglion neuron. ET-1 mRNA expressions increased after noise exposure in both group 2 and group 3 compared to those of the control group. At 2 weeks after noise exposure, however, the ET-1 mRNA expressions in group 3 increased compared to that of the control but decreased compared to that of group 2. On the other hand, ETAR mRNA expression increased at 2 weeks after noise exposure in both groups, just after noise exposure in group 2 and at 2 weeks after noise exposure in group 3. Conclusion These results suggest that expression changes of ET-1, ETAR and ETBR might be associated with hearing threshold shift and recovery after noise exposure in the cochlea.
백서 재생간에서의 NF-kB의 활성화에 따른 유도성 Nitric Oxide Synthase 유전자의 전사에 관한 연구

A concerted activation of transcription factors and inducible nitric oxide synthase (iNOS) gene expression by the transcription factors were observed after partial hepatectomy (PH) of rats. To define the molecular mechanism of transcriptional control of iNOS gene during the hepatic regeneration, expression of iNOS, interleukin-6 (IL-6) and tumor necrosis-alpha (TNF-α) mRNA and the binding pattern of nuclear transcriptional factor (NF-κB and STAT-3) in the iNOS gene promoter region have been investigated. The expression of iNOS mRNA was initiated at 1 h post-PH and reached a maximum at 3 h and 5 h post-PH. This induction was preceded by an early and transient activation of NF- KB complex composed of p50/p65 heterodimer, which was rapidly activated within 30 min post-PH and remained elevated for up to 1 h post-PH, and tend to decline after 3 h post-PH. Administration of N-acetylcysteine (NAC) significantly inhibited the expression of iNOS mRNA. Expression of IL-6 and TNF-α, activators of iNOS and STAT-3, were also increased after PH, but the activation of STAT-3 binding during hepatic regeneration was found at 3 h or 5 h post-PH, which shows a different time kinetic pattern of transcriptional factor activation of iNOS gene between NF-κB and STAT-3. These results suggest that there is an important role of the NF-κB complex (p50/p65 heterodimer) activation in the transcriptional control of iNOS gene during hepatic regeneration.
재생간에서의 Na, K-ATPase 활성변도에 관한 연구

To study the alterations of Na, K-AITPase in the regenerating liver cell membrane, after partial hepatectomy(70% ) of Sprague Dawley rat, the activity of Na, K-ATPase and ^3H-ouabain binding in the control or regenerated liver cell membrane were measured. Results are follows 1. In the cell membrane of regenrated liver for 3 days, the activities of Na, K-ATPase were decreased about 60% as compared to that of control liver. After 5 to 7 days from partial hepatectomy, the activities of Na. K-ATPase were restored. 2. Na, K-ATPase activities were increased by increase of Na concentration within reation medium in both control and regenerated liver cell membrance. EC_50, of the regenerated liver cell membrane was little different from control rat liver cell membrane. 3. Ouabain concentration for the inhibition of Na, K--ATPase activity(IC_50) in the regenerated liver was slightly higher than that in control liver. 4. In the ouabain-binding experiment, Kd in the regenerated liver was slightly lowered but maximum binding sites(Bmax) were decreased compared to that of control liver. 5. Na, K-ATPase activities per pump site were decreased about 40% in regenerated liver From these results, it is suggested that the decreased actvitv of Na-- pump in regenerating rat liver cell may due to the alterations of number and property of Na-pump.

An activation of transcription factors involved in the transactivation of inducible nitric oxide synthase (iNOS) gene occurred after partial hepatectomy (PH), resulting in the transient expression of iNOS. The effect of serum after PH on iNOS gene expression has been investigated with RT-PCR, elctrophoretic mobility shift assay (EMSA) and chloramphenicol acetyltransferase (CAT) assay. Stimulation of iNOS mRNA expression reached a maximum with serum from 3 h and 5 h of PH. This stimulation was preceded by an early and transient activation of NF-κB complex composed of p50/p65 heterodimer, which was rapidly activated with serum from 1 h and 3 h of PH, and tend to decline with serum after 5 h of PH. The stimulatory effects of serum after PH were does-dependently inhibited by treatment of PDTC. Transfection of BNL-CL2 cells with iNOS promoter linked to a CAT reporter gene was followed by stimulation with serum after PH. Site-specific mutation of three conserved nucleotide within the upstream NF-κB site showed no difference in CAT activity by serum after PH compared with that of cells transfected with wild type iNOS promoter. In contrast, site-specific mutation of the downstream NF-κB caused remarkable reduction in the reporter gene expression by serum after PH. These results suggest that serum after PH regulates the iNOS gene transcription via NF-κB complex (p50/p65 heterodimer) activation, and downstream NF-κB site is the essential component in transcription of iNOS gene during hepatic regeneration.
Overproduction of GABA in Apple Pomace with Auricularia auricula-judae by Lactic Acid Fermentation

Lactic acid fermentation of apple pomace was performed using Lactobacillus plantarum EJ2014 to produce gamma-aminobutyric acid (GABA), in the presence of 3% MSG and 0.5% yeast extract. After fermenting for 5 days, apple pomace showed pH and acidity of 6.26 and 0.48%, respectively. Fermentation of apple pomace in the presence of wood ear mushroom (Auricularia auricula-judae) resulted in low acidity (0.16%) and a pH of 6.26. Initially, the number of viable cells was ~1.91×107 CFU/mL, which increased to >3.15×109 CFU/mL after one day. Addition of wood ear mushroom resulted in the highest bacterial stability during lactic acid fermentation for 5 days, resulting in higher number of viable cells (3.86×108 CFU/mL). In particular, GABA production using apple pomace was considerably low; however, it increased following the addition of 2% wood ear mushroom. After 5 days, glutamic acid concentration decreased from 19,552 μg/mL to 1,532 μg/mL, resulting in the production of 13,442 μg/mL GABA. Thus, lactic acid fermentation of apple pomace containing wood ear mushroom resulted in the formation of multifunctional ingredients, including GABA, dietary fibers, and probiotics.

In the area of dynamic driving control and sensor monitoring, the parameters, which characterize the vehicle current state, play an important role. In practice, the vehicle mass, characteristicspeed, road grade and therefore the parameters change occasionally. This fact influences the quality of both the control and the sensor monitoring. To ensure the efficiency of such control and sensor monitoring systems, there exist several possibilities. One possibility is to estimate the parameters on-line and insert them into the related models continuously. The estimated parameters can be regarded as available vehicle parameters for the total electronic control system. The contribution of this paper is to develop a concept for estimating these parameters.

An analysis was conducted to predict the behavior of gasket by applying an optimal-strain energy-density function selected through a uniaxial tensile test and an analysis of the gasket used in an actual hydrogen fuel cell. Among the models compared to predict the materials" properties, the Mooney-Rivlin secondary model showed the behavior most similar to the test results. The maximum stress of the gasket was not significantly different, depending on the location. The maximum surface pressure of the gasket was higher at positions “T” and “Y” than at other positions, owing to the branch-shape effect. In the future, a jig that can measure the surface pressure will be manufactured and a comparative verification study will be conducted between the test results and the analysis results.
