산소조건 및 Catalase가 돼지난포란의 체외성숙과 배발달에 미치는 영향

본 연구는 돼지난포란의 체외배양액에 황화합물인 Catalase 첨가 및 저분압산소조건 하에서 첨가배양함으로서 돼지난포란의 체외성숙과 체외배발달에 미치는 영향을 구명하고자 실시하였다. 본 연구에서 얻어진 결과를 요약하면 다음과 같다. 1. 돼지난포란을 체외성숙배양액인 0.9mM cy-steine이 함유된 NCSU-23 배양액에 CAT를 각각 0, 100, 500 및 1000uni1s/m1 첨가하여 체외성숙 및 수정을 실시한 결과, 모든 평가요소에서 처리구가 대조구에 비하여 유의적으로 낮은 결과를 나타내어 체외수정에 좋지 않은 영향을 미치는 것으로 판단된다(P>0.05). 2. 체외수정을 실시한 후, 배발달배양액인 NCSU-23에 7일 동안 배양한 결과, 배반포형성률과 총세포수에 있어서 처리구가 대조구에 비하여 유의적으로 낮은 결과를 나타내어 체외성숙시 CAT 첨가배양은 배발달에 좋지 않은 것으로 판단된 다(P>0.05). 3. 배발달배양액인 NCSU-23에 CAT를 각각 0, 100, 500 및 1000units/m1을 첨가하고 5 및 20% 산소농도하에서 7일간 배양을 실시한 결과, 산소농도에 따른 차이는 인정되지 않았으며, CAT 첨가에 따른 배반포형성률과 총세포수에 있어서도 처리구가 대조구보다 유의적으로 낮은 결과를 나타냈다(P>0.05). 결론적으로 돼지난포란을 이용하여 체외수정란을 생산할 때, NCSU-23 배양액에 catalase의 첨가 및 산소조건은 영향을 미치지 않는 것으로 조사되었다. The present study was carried out to examine the effect of catalase (CAT) on in vitro maturation (IVM) of porcine follicular oocytes and oxygen concentration with CAT on in vitro development (IVD) of porcine IVM/IVF embryos. The results were summarized as follows - 1 . The rates of nuclear maturation, penetrated oocytes, pronucleus formation rates, polyspermic oocytes and mean numbers of the penetrated sperm were significantly lower in oocytes matured with 100, 500 and 1,000 units/ml CAT than those of central groups (P>0.05). 2. The rates of blastocyst formation and total cell numbers of blastocyst at day 7 after in vitro fertilization were significantly lower in CAT treatment groups than those of the central groups (P>0.05). 3. There were not significant difference in the blastocyst development and total cell numbers of blastocyst on in vitro culture of NCSU-23 media with 0, 100, 500 and 1000 units/ml CAT under the 5% and 20% O₂ concentrations. These results suggested that the addition of CAT was not helpful for porcine oocyte maturation and further development, also the rates of blastocyst formation and total cell numbers of blastocyst at day 7 of porcine IVM/IVF embryos were not significantly different in the NCSU-23 culture medium under the 5% and 20% O₂ concentrations.
β-Mercaptoethanol의 첨가배양이 돼지난포란의 체외성숙과 배발달에 미치는 영향

본 연구는 돼지 난포란의 체외배양액에 황화합물인 β-mercaptoethanol(β-ME)을 첨가배양함으로서 돼지난포란의 체외성숙과 체외배발달에 미치는 향을 구명하고자 실시하였다. 본 연구에서 얻어진 결과를 요약하면 다음과 같다. 1. 돼지난포란을 체외성숙배양액인 NCSU-23 배양액에 β-ME를 각각 0, 25, 50 및 100μM 첨가배양하여 성숙을 유기한 다음, 체외수정을 실시한 결과, 모든 처리구에서 핵성숙률, 정자침투율, 웅성전핵형성률, 다정자침입률 및 평균침입정 수에서 유의적인 차이가 인정되지 않았다 P>0.05). 2. 체외성숙 및 수정을 실시한 후, 배발달 배양액인 NCSU-23에 7일간 배양한 결과, 배반포 형성률은 25μM의 β-ME 처리구가 유의적(P<0.05)으로 높은 결과를 나타냈고, 총세포수에 있어서는 대조구와 처리구간 유의적인 차이가 인정되지 않았다. 3. 배발달배양액인 NCSU-23에 β-ME를 각각 0, 12.5, 25 및 5μM 첨가하고 5 및 20%산소조건 하에서 7일간 배양한 결과, 처리구별 산소농도에 따른 배발달률 및 총세포수에 있어서는 유의 적 인 차이가 없었으나, 25μM β-ME 처리구에서 유의적으로 높은 결과를 나타냈다(P<0.05). 그러나 총세포수에서는 처리구 및 대조구간 유의성이 인정되지 않았다. 결론적으로 돼지난포란의 체외성숙 및 배발달시 황화합물인 β-ME의 첨가는 25μM이 가장 적합하며, 산소농도에 따라서는 영향을 미치지 않는 것으로 조사되었다. However, no differences ware found in total cell numbers of blastocyst among the treatments. 3. The rates of blastocyst formation at day 7 after in vitro fertilization were higher in the NCSU-23 Culture medium With 25 μM β-ME (23.6±2.8%) than in those Cultured With 0 (15.4±4.4%), 12.5 (17.5±2.3%) and 50 μM β-ME (18.6±2.1%) Under the 5% and 20% O₂ Concentrations (P<0.05). However, no differences was found in total cell numbers of blastocyst among the treatments. These results suggested that the addition of 25 μM β-ME in the IVM/IVD media were effective on the porcine embryo production. However, the rates of blastocyst formation and total cell numbers of blastocyst at day 7 of porcine IVM/IVF embryos were not significantly different in the NCSU-23 culture medium under 5% and 20% 02 concentrations.
MEMS 기술을 이용한 미소 리블렛 필름 제작 및 항력 감소에의 응용

This paper presents the fabrication method of a micro-riblet film (MRF) using MEMS technology and the experimental results of the drag reduction of an airfoil with MRFs. Riblets having grooved surface in the streamwise direction has been proven as an effective passive control technique of the drag reduction. A V-grooved pattern on (100) silicon wafer is etched with anisotropic bulk micromachining. The MRF is completed by replicating the V-grooved pattern with polydimethylsiloxane (PDMS). Experiments were performed by measuring a velocity field behind the trailing edge of a NACA 0012 airfoil with and without MRFs in a closed-type subsonic wind tunnel using particle image velocimetry (PlV) technique. The MRF provides about 3.8 % drag reduction compared to the drag on a smooth airfoil when the freestream velocity of wind tunnel is 3.3 m/s.
산소농도 및 Superoxide Dismutase가 돼지 난포란의 체외성숙과 배발달에 미치는 영향

본 연구는 돼지 난포란의 체외성숙배양액에 superoxide dismutase(SOD)를 첨가배양하여 체외성숙과 수성이후 배발달에 미치는 영향과 산소농도 및 SOD가 체외생산된 돼지초기수정란치 배발달에 미치는 영향을 규명하고자 실시하였다. 본 연구에서 얻어진 결과를 요약하면 다음과 같다. 1. 돼지 난포란을 체외성숙배양액인 NCSU-23에 SOD를 각각 0, 100,500 및 1,000uni1s/m1 첨가하여 성숙시킨 다음, 체외수정을 실시한 결과, 핵 The present study was carried out to examine the effect of superoxide dismutase (SOD) on in vitro maturation (IVM) of porcine follicular oocytes and oxygen concentration with SOD on in vitro development (IVD) of porcine IVM/IVF embryos. The rates of blastocyst formation at day 7 after in vitro fertilization were higher in the NCSU-23 culture medium with 100 units/ml SOD than in those cultured with 0, 500 and 1,000 units/ml SOD under the 5% and 20% concentrations. However, no differences was found in the total cell numbers of blastocyst among the treatments. In conclusion, these results suggested that the addition of SOD was not adequate for porcine oocyte maturation and further development, also the rates of blastocyst formation and total cell numbers of blastocyst at day 7 of porcine IVM/IVF embryos were not different in the NCSU-23 culture medium under the 5% and 20% concentrations.
