SecM에서 유래한 접착펩타이드에 의한 라이보솜 정지를 우회하는 SSU rRNA 돌연변이체 발굴을 위한 유전학적 시스템 개발

최근 단백질 합성 과정 중 라이보솜의 일시적인 정지에 의한 라이보솜의 A자리에서 전사체가 분해되는 현상이 여러 생명체에서 보고되었다. 이러한 현상이 라이보솜의 작은 소단위체를 이루고 있는 SSU rRNA의 기능과 관련 있는지를 알아보기 위해, SecM에서 유래한 접착펩타이드에 의한 라이보솜 정지를 우회하는 SSU rRNA 돌연변이체 발굴을 위한 유전학적 시스템을 개발하였다. 이 시스템에서는 SecM에서 유래한 접착펩타이를 포함하는 CAT 단백질을 코딩하는 CAT-SecM 전사체가 플라스미드에서 유래한 SSU rRNA를 포함한 재조합 라이보솜에 의해서만 해독된다. 이러한 재조합 라이보솜은 접착펩타이드를 합성한 후 CAT-SecM mRNA 상에서 일시 정체하며, 재조합 라이보솜의 발현은 이 전사체의 양을 감소시키는 것을 확인하였다. 이러한 결과는 개발된 시스템을 이용해 라이보솜 검지를 우회하는 SSU rRNA 돌연변이체의 선별이 가능하다는 것을 보여주며, 이러한 변이체에 대한 연구는 단백질 합성 단계에서 일어나는 라이보솜 정지와 전사체 절단 현상에 있어서, SSU rRNA의 역할을 규명하는데 기여할 것이다. Ribosome stalling by nascent sticky peptide has been reported in several organisms across the kingdom. To test whether small subunit (SSU) rRNA is involved in this phenomenon, we developed a genetic system that utilized the specialized ribosome system to isolate SSU rRNA mutants that enable ribosomes to bypass the SecM-derived sticky peptide in protein synthesis. In this system, CAT-SecM mRNA, which encodes CAT protein containing the sticky peptide derived from SecM, is only translated by specialized ribosomes. These ribosomes were shown to transiently stall on CAT-SecM mRNA followed by the synthesis of the sticky peptide. Expression of specialized ribosomes resulted in the decreased steady-state level of CAT-SecM mRNA, which is consistent with a notion that ribosome stalling induces mRNA degradation. Isolation and characterization of SSU rRNA mutations using this genetic system that are sufficient to circumvent ribosome stalling induced by the SecM-derived sticky peptide will provide evidence of SSU rRNA function in mRNA cleavage.

Most of the people have some prejudices against morality. The first prejudice is that morality means excluding self-interest and caring for others. Secondly, morality means obligation which must be always done. Thirdly, morality is related to not an actual human life but an abstract notion. Accordingly critical comments on moral education are connected with these three biases. Especially dichotomy separating morality and self-interest is pointed out to be a major cause for obstructing moral practice. Because, so far in moral education, we stressed on that most important character of morality is the duty and care for other people. But self-interest is one of the most important things that causes one's behavior, so it can not be ignored completely. To be the practical moral education, such problems must be solved. Recently, moral philosophy of Adam Smith, who is widely known as economist, is being paid attention by most of the today's economists. For they believe his moral philosophy -「The Theory of Moral Sentiments」- can help overcome current crisis. In this paper I tried to find out current moral educational messages implied on Adam Smith's moral philosophy. They are as follows. Firstly, not only altruism but also self-interest can be morality. Secondly, individual passion can be universalized to social sympathy. Thirdly, obligation and supererogation should be distinguished. Finally, morality should be based on human being nature. 경제학자로 널리 알려진 아담 스미스의 도덕이론이 최근에 새롭게 주목받고 있다. 스미스는 자기 이전 시대의 제 도덕학설들을 비판적으로 검토하고 그것들을 종합적으로 취사선택하여 도덕의 통합화를 시도한 인물이다. 특히 자기이익을 동감의 원리를 통하여 보편적인 도덕과 긍정적으로 접목시킴으로써, 이타심뿐만 아니라 이기심도 덕이 될 수 있음을 강조하였다. 또한 도덕을 자신을 위한 덕과 타인을 위한 덕으로 구분하여 설명하면서, 자연스러운 인간의 본성에 기반한 자신을 위한 덕의 중요성을 역설하고 있다. 타인에 관한 덕에 있어서도 의무의 도덕과 의무 이상의 도덕으로 구분하여 설명함으로써, 도덕에 관한 오래된 오해 중의 하나인 ‘자신의 희생이나 타인들을 위한 의무’라는 편견을 불식시킬 수 있는 단서를 제공하고 있다. 이러한 스미스의 도덕에 관한 입장은 오늘날 도덕교육에 가해지는 주요 비판 중의 하나인 비현실성과 추상성의 한계를 극복할 수 있는 가능성을 제공한다는 점에서 의의가 있다.

남강 본류로 유입되는 경상남도 진주시 관내에 소재한 13개 지류에서 습지의 보전 현황을 평가하고 우수한 습지생태계를 발굴하며 인위적인 훼손과 오염으로부터 습지생태계를 보호하기 위한 보전 대책을 제시하기 위하여 습지생태계의 생물 서식지로서의 물리적 특성과 식생의 자연도를 조사하였다. 지류 주변의 토지 이용, 제방 상단의 생태적 기능, 제방 사면의 구조, 둔치의 인공화 정도 및 이용 강도, 호안의 인공화 정도, 하상 재료의 다양성, 하천의 횡단 구조물 등으로부터 산정한 물리적 특성의 자연도 평가지수는 오미천이 가장 높고 반성천, 향양천, 그리고 나불천, 영천강 및 수곡천, 중촌천, 대곡천 등의 순이었으며, 문산천이 가장 낮았다. 식생의 다양성, 외래종의 우점면적, 1년생 초본의 우점면적, 현존식생의 자연도 및 특이성, 종다양성 등으로부터 산정한 식생 자연도의 평가지수는 오미천이 가장 높고, 수곡천, 반성천, 영천강, 중촌천, 지내천, 나불천, 지수천 등의 순이었으며 용아천이 가장 낮았다. 물리적 특성과 식생 자연도의 평가지수로부터 산정한 하천 자연도는 오미천이 Ⅰ등급으로 가장 높고, Ⅱ등급인 반성천, 수곡천, 영천강, 나불천 및 중촌천, 향양천, 지내천, 지수천, 대곡천, 문산천 및 독산천 등의 순이었으며, 용아천이 Ⅲ등급으로 가장 낮았다. 지류의 습지생태계를 복원하여 하천의 자연도를 향상시키기 위한 보전대책이 제시되었다.

Current technology's potential to prolong and sustain life raises profound questions of quality versus sanctity of life. One of the most highly debated legal-ethical issues in the administration of healthcare concerns the right of patients or their surrogates to refuse life-sustaining treatment. Professional ethics for nurses requires members to practice with compassion and to respect human dignity and the patient's autonomy. Because autonomy is a basic human right based on society's principle of self-determination. The decision to withdraw life support incorporates the two ethical standards of substituted judgement-where an attempt is made to determine the patient's preferences based on previous oral or written statements- and making a decision based on protection of patient welfare, taking into account such factors as suffering and quality/length of life.
